| 1 | Differential control of growth,apoptotic activity and gene expression in human colon cancer cells by extracts derived from medicinal herbs,Rhazya stricta and Zingiber officinale and their combination显示文摘AIM:To investigate the effects of extracts from Rhazya stricta(R.stricta) and Zingiber officinale(Z.officinale) on human colorectal cancer cells.METHODS:Human colorectal cancer cells(HCT116) were subjected to increasing doses of crude alkaloid extracts from R.stricta(CAERS) and crude flavonoid extracts from Z.officinale(CFEZO).Cells were then harvested after 24,48 or 72 h and cell viability was examined by trypan blue exclusion dye test; clonogenicity and soft agar colony-forming assays were also carried out.Nuclear stain(Hoechst 33342),acridine orange/ethidium bromide double staining,agarose gel electrophoresis and comet assays were performed to assess pro-apoptotic potentiality of the extracts.Quantitative reverse transcription-polymerase chain reaction(qRT-PCR),using gene-specific primers and Western blot analyses were performed to assess the impact of CAERS and CFEZO on the expression levels of key regulatory proteins in HCT116 cells.RESULTS:Treatment with a combination of CAERS and CFEZO synergistically suppressed the proliferation,colony formation and anchorage-independent growth of HCT116 cells.Calculated IC50,after 24,48 and 72 h,were 70,90 and 130 μg/mL for CAERS,65,85 and 120 μg/mL for CFEZO and 20,25 and 45 μg/mL for both agents,respectively.CAERS- and CFEZO-treated cells exhibited morphologic and biochemical features of apoptotic cell death.The induction of apoptosis was associated with the release of mitochondrial cytochrome c,an increase in the Bax/Bcl-2 ratio,activation of caspases 3 and 9 and cleavage of poly ADP-ribose polymerase.CAERS and CFEZO treatments downregulated expression levels of anti-apoptotic proteins including Bcl-2,Bcl-X,Mcl-1,survivin and XIAP,and upregulated expression levels of proapoptotic proteins such as Bad and Noxa.CAERS and CFEZO treatments elevated expression levels of the oncosuppressor proteins,p53,p21 and p27,and reduced levels of the oncoproteins,cyclin D1,cyclin/cyclin-dependent kinase-4 and c-Myc.CONCLUSION:These data suggest that a combination of CAERS and CFEZO is a promising treatment for the prevention of colon cancer. | Ayman I Elkady Rania Abd El Hamid Hussein Osama A Abu-Zinadah | 2014 | World Journal of Gastroenterology2014，20，41： | 12 |

| 3 | Endophytic bacteria associated with endangered plant Ferula sinkiangensis K. M. Shen in an arid land: diversity and plant growth-promoting traits显示文摘Ferula spp. are traditional medicinal plants found in arid land. Large-scale excavation for extracting bioactive compounds from the plants in arid regions of Xinjiang over the last few years has, however, significantly decreased their distributions. Due to the urgent need for preservation of these plant resources, along with the need of searching for alternative source of the useful metabolites, it is important to screen the endophytic microbial resources associated with the plant Ferula sinkiangensis K. M. Shen. In the study, a total of 125 endophytic bacteria belonging to 3 phyla, 13 orders, 23 families, and 29 genera were isolated based on 16 SrRNA gene sequencing data. Among the different isolates, three strains isolated from roots were potential novel species of the genera Por phyrobacter, Paracoccus and Amycolatopsis. In this study, 79.4% and 57.1% of the total isolates were capable of producing indole-3-acetic acid(IAA) and siderophore, respectively. And, 40.6% of the strains inhibit the growth of fungal pathogen Alternaria alternata, 17.2% and 20.2% strains were positive for antagonism against Verticillium dahlia 991 and V. dahlia 7, respectively. | 4 | Rituximab for troublesome cases of childhood nephrotic syndrome显示文摘Nephrotic syndrome(NS) is the most common glomerular disease of childhood. Steroid-dependent and steroid-resistant nephrotic syndrome present challenges in their pharmaceutical management; patients may need several immunosuppressive medication for optimum control, each of which medication has its own safety profile. Rituximab(RTX) is a monoclonal antibody that targets B cells and has been used successfully for management of lymphoma and rheumatoid arthritis. Recent clinical studies showed that rituximab may be an efficacious and safe alternative for the treatment of complicated nephrotic syndrome. In this review article, we aim to review the efficacy and safety of RTX therapy in nephrotic syndrome. We reviewed the literature pertaining to this topic by searching for relevant studies on Pub Med and Medline using specific keywords. The initial search yielded 452 articles. These articles were then examined to ensure their relevance to the topic of research. We focused on multicenter randomized controlled trials with relatively large numbers of patients. A total of 29 articles were finally identified and will be summarized in this review. The majority of clinical studies of RTX in complicated pediatric NS showed that rituximab is effective in approximately 80% of patients with steroid-dependent NS, as it decreases the number of relapses and steroid dosage. However, RTX is less effective at achieving remission in steroid-resistant NS. RTX use was generally safe, and most side effects were transient and infusion-related. | 5 | In vitro and in vivo effects of ferulic acid on gastrointestinal motility:Inhibition of cisplatin-induced delay in gastric emptying in rats显示文摘AIM: To study the effects of ferulic acid on gastrointe-stinal motility both in vitro and in vivo.METHODS: Ferulic acid induced concentration-dependent stimulation of the basal tone of isolated guinea pig ileum (2-20 μmol/L) and isolated rat fundus (0.05-0.4 mmol/L). RESULTS: Ferulic acid significantly accelerated the gastrointestinal transit and gastric emptying in rats in a dose-dependent manner (50-200 mg/kg, po). Cisplatin (2.5-20 mg/kg, ip) induced a dose-dependent delay in gastric emptying in rats. Pretreatment with ferulic acid dose-dependently, significantly reversed the cisplatin-induced delay in gastric emptying. CONCLUSION: The endogenous prostaglandins (PGs) are involved in mediating the stimulant effects of ferulic acid. | 6 | Inter-ethnic marriages and severity of coronary artery disease:A multicenter study of Arabian Gulf States显示文摘AIM To assess the association of inter-ethnic vs intra-ethnic marriage with severity of coronary artery disease(CAD) in men undergoing angiography.METHODS We conducted a prospective multicenter,multi-ethnic,cross sectional observational study at five hospitals in Saudi Arabia and the United Arab Emirates,in which we used logistic regression analysis with and without adjustment for baseline differences. RESULTS Data were collected for 1068 enrolled patients undergoing coronary angiography for clinical indications during the period of April 1^(st),2013 to March 30^(th),2014. Ethnicities of spouses were available only for male patients. Of those enrolled,687 were married men and constituted the cohort for the present analysis. Intra-ethnic marriages were reported in 70% and inter-ethnic marriages in 30%. After adjusting for baseline differences,interethnic marriage was associated with lower odds of having significant CAD [adjusted odds ratio 0.52(95%CI:0.33,0.81)] or multi-vessel disease(MVD) [adjusted odds ratio 0.57(95%CI:0.37,0.86)]. The adjusted association with left main disease showed a similar trend,but was not statistically significant [adjusted odds ratio 0.74(95%CI:0.41,1.32)]. | 13 | Diversity of cultivable endophytic bacteria associated with halophytes in Xinjiang of China and their plant beneficial traits显示文摘Endophytic bacteria from halophytes have a wide range of application prospects in various fields,such as plant growth-promoting,biocontrol activity and stress resistance.The current study aimed to identify cultivable endophytic bacteria associated with halophytes grown in the salt-affected soil in Xinjiang Uygur Autonomous Region,China and to evaluate their plant beneficial traits and enzyme-producing activity.Endophytic bacteria were isolated from Reaumuria soongorica(PalL Maxim.),Artemisia carvifolia(Buch.-Ham.ex Roxb.Hort.Beng.),Peganum harmala L.and Suaeda dendroides(C.A.Mey.Moq.)by using the cultural-dependent method.Then we classified these bacteria based on the difference between their sequences of 16S rRNA(16S ribosomal RNA)gene.Results showed that the isolated bacteria from R.soongorica belonged to the genera Brucella,Bacillus and Variovorax.The bacteria from A.carvifolia belonged to the genera Micromonospora and Brucella.The bacteria from P.harmala belonged to the genera Paramesorhizobium,Bacillus and Peribacillus.The bacteria from S.dendroides belonged to the genus Bacillus.Notably,the genus Bacillus was detected in the three above plants,indicating that Bacillus is a common taxon of endophytic bacteria in halophytes.And,our results found that about 37.50%of the tested strains showed strong protease-producing activity,6.25%of the tested strains showed strong cellulase-producing activity and 12.50%of the tested strains showed moderate lipase-producing activity.Besides,all isolated strains were positive for IAA(3-Indoleacetic acid)production,31.25%of isolated strains exhibited a moderate phosphate solubilization activity and 50.00%of isolated strains exhibited a weak siderophore production activity.Our findings suggest that halophytes are valuable resources for identifying microbes with the ability to increase host plant growth and health in salt-affected soils. | LI Li GAO Lei LIU Yonghong FANG Baozhu HUANG Yin Osama A A MOHAMAD Dilfuza EGAMBERDIEVA LI Wenjun MA Jinbiao | 2021 | Journal of Arid Land2021，13，8： | 1 |
