| 1 | Arsenic Disulfide Induced Apoptosis and Concurrently Promoted Erythroid Differentiation in Cytokine-Dependent Myelodysplastic Syndrome-progressed Leukemia Cell Line F-36p with Complex Karyotype Including Monosomy 7显示文摘Objective:Acute myeloid leukemia progressed from myelodysplastic syndrome(MDS/AML)is generally incurable with poor prognosis for complex karyotype including monosomy 7(-7).Qinghuang Powder(青黄散,QHP),which includes Qing Dai(Indigo naturalis)and Xiong Huang(realgar)in the formula,is effective in treating MDS or MDS/AML even with the unfavorable karyotype,and its therapeutic efficacy could be enhanced by increasing the Xiong huang content in the formula,while Xiong huang contains>90%arsenic disulfide(As_2S_2).F-36p cell line was established from a MDS/AML patient with complex karyotype including-7,and was in cytokine-dependent.The present study was to investigate the effects of AS_2S_2 on F-36p cells.Methods:Cell proliferation was measured by an3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay.Cell apoptosis was identified by Annexin V-staining.Cell viability was determined by a propidium iodide(PI)exclusion.Erythroid differentiation was evaluated by the expression of cell surface antigen CD235a(GpA).Results:After treatment with AS_2S_2,at concentrations of 0.5to 16μmol/L for 72 h,As_2S_2 inhibited the proliferation of F-36p cells.The 50%inhibitory concentrations(IC_(50))of As_2S_2against the proliferation of F-36p cells was 6μmol/L.The apoptotic cells significantly increased in a dose-dependent mannar(P<0.05).The cell viabilities were significantly inhibited by As_2S_2 dose-dependent in a dose-dependent manner(P<0.05).Significant increases of CD235a-positive cells were concurrently observed(P<0.05)also in a dosedependent manner.Conclusions:As_2S_2 could inhibit proliferation and viability,induce apoptosis,and concurrently promote erythroid differentiation dose-dependently in F-36p cells.AS_2S_2 can inhibit proliferation and viability,induce apoptosis,and concurrently promote erythroid differentiation in cytokine-dependent MDS-progressed human leukemia cell line F-36p with complex karyotype including-7.The data suggest that QHP and/or As_2S_2 could be a potential candidate in the treatment of MDS or MDS/AML even with unfavorable cytogenetics. | 胡晓梅 Sachiko Tanaka Kenji Onda 袁博 Hiroo Toyoda 麻柔 刘峰 Toshihiko Hirano | 2014 | Chinese Journal of Integrative Medicine2014，20，5： | 20 |
| 2 | Effects of slope gradient on runoff from bare-fallow purple soil in China under natural rainfall conditions显示文摘Purple soil is highly susceptible for overland flow and surface erosion, therefore understanding surface runoff and soil erosion processes in the purple soil region are important to mitigate flooding and erosion hazards. Slope angle is an important parameter that affects the magnitude of runoff and thus surface erosion in hilly landscapes or bare land area. However, the effect of slope on runoff generation remains unclear in many different soils including Chinese purple soil. The aim of this study was to investigate the relationship between different slope gradients and surface runoff for bare-fallow purple soil, using 5 m × 1.5 m experimental plots under natural rainfall conditions. Four experimental plots(10°, 16°, 20° and 26°) were established in theYanting Agro-ecological Experimental Station of Chinese Academy of Science in central Sichuan Basin. The plot was equipped with water storage tank to monitor water level change. Field monitoring from July 1 to October 31, 2012 observed 42 rainfall events which produced surface runoff from the experimental plots. These water level changes were converted to runoff. The representative eight rainfall events were selected for further analysis, the relationship between slope and runoff coefficient were determined using ANOVA, F-test, and z-score analysis. The results indicated a strong correlation between rainfall and runoff in cumulative amount basis. The mean value of the measured runoff coefficient for four experimental plots was around 0.1. However, no statistically significant relationship was found between slope and runoff coefficient. We reviewed the relationship between slope and runoff in many previous studiesand calculated z-score to compare with our experimental results. | 3 | Imaging findings of biliary hamartomas显示文摘AIM: To evaluate the imaging findings of biliary hamartomas (von Meyenburg complexes, VMCs) and discuss the differential diagnosis with other related diseases.METHODS: Imaging findings of biliary hamartomas on ultrasonography (US), computed tomography (CT), magnetic resonance imaging (MRI), MR cholangiopancreatography (MRCP)and hepatobiliary scintigraphy were retrospectively analyzed in six patients.RESULTS: On ultrasound images, five of the six cases showed multiple small hyper- and hypo-echoic lesions with comet-tail echoes, especially when magnified by US with the usage of zoom function. In all the six cases,multiple tiny hypodense lesions less than 10 mm in diameter were revealed as scattered throughout the liver with no enhancement on CT. These tiny lesions were demonstrated to be hyper- and hypo-intensity on T2- and TI-weighed images, respectively, in three patients who underwent MRI examinations. MRCP was performed in two patients, and clearly showed multiple tiny irregular- and round-shaped hyper-intensity lesions.MRCP and hepatobiliary scintigraphy showed normal appearances of intra- and extra-hepatic bile ducts in two and one patients, respectively.CONCLUSION: Imaging modalities are useful in the diagnosis and differential diagnosis of VMCs.
