1真鲷脂蛋白脂肪酶基因顺式元件PPRE及在肝脏活体调控作用显示文摘从血液提取总DNA并构建基因组文库,克隆海水鱼真鲷(Pagrosomusmajor)脂蛋白脂肪酶(LPL)基因5′侧翼区序列,再通过竞争性聚合酶链式反应(PCR)方法,定量研究饲料中添加不同饱和度脂肪酸对其肝脏LPL基因mRNA表达水平的影响。结果表明,真鲷LPL基因5′侧翼区序列中存在顺式元件过氧化物酶体增殖物反应元件,其序列为TGAAGA TGACAT,与TATA(CATAA)盒序列相隔211bp;与喂饲料对照组比较,添加10%油酸可增加真鲷肝脏LPL基因表达水平,但差异并不显著(p>0.05)。用亚油酸或n 3高不饱和脂肪酸混合物取代添加油酸的一半(占饲料添加量的5%)后,导致真鲷肝脏LPL基因表达水平升高并出现显著性变化(p<0.05)。对真鲷肝脏LPL基因诱导表达随饲料中的脂肪酸不饱和度增加而增加,表明在活体条件下脂肪酸对真鲷肝脏LPL基因表达的诱导作用可能与脂肪酸的氧化代谢有关。n 3高不饱和脂肪酸通过过氧化物酶体增殖物激活受体诱导真鲷肝脏LPL基因表达,使真鲷食物脂质供应水平与其肝脏脂肪酸氧化代谢强度调控偶联起来,可能是真鲷对高脂食物(特别是富含n 3高不饱和脂肪酸食物)的一种适应。梁旭方 李月琴 李贵生 白俊杰 劳海华 Hiromi Oku Hiroshi Y.Ogata 2004热带海洋学报2004,23,4:16
2海水鱼真鲷脂蛋白脂肪酶基因cDNA序列与组织表达显示文摘为研究脊椎动物真鲷脂蛋白脂肪酶 (LPL)结构与功能关系以及探讨动脉粥样硬化形成机理 ,通过构建cDNA文库 ,克隆对动脉粥样硬化表现抗性的海水鱼真鲷LPL基因cDNA全序列 .再通过PCR方法扩增基因组DNA ,获取内含子 9及其两侧序列以确定外显子 10的大小 ,最后通过RT PCR ,以 β肌动蛋白为外参照 ,比较真鲷在食用两种脂肪含量不同饲料和摄食状态不同的处理条件下 ,肝脏和腹腔肠系膜脂肪组织LPLmRNA的相对水平 .从腹腔肠系膜脂肪组织cDNA文库中克隆出LPLcDNA序列 ,其完整的开放阅读框架由 15 36bp组成 ,编码 5 11个氨基酸残基 .与哺乳类不同 ,真鲷LPL基因外显子 10的开始部分是翻译的 .LPL的催化位点、二硫键位点、N 糖基化位点、肝素结合区、脂质结合位点、介导脂蛋白与低密度脂蛋白受体结合位点、二聚体形成位点等主要功能域在真骨鱼类真鲷与其它脊椎动物间基本保守 ,但肝素结合区的碱性氨基酸残基含量较人类减少 ,并在结合脂质底物的疏水环套中出现插入片段 .与哺乳类不同 ,真鲷LPL基因在成体肝脏存在诱导性表达 ,而在其腹腔肠系膜脂肪组织则存在与哺乳类相似的组成性表达 .当真鲷喂食高脂饲料时 ,其饱食状态下肝脏LPLmRNA水平升高 ,但对其腹腔肠系膜脂肪组织LPL表达没有影响 .当真鲷喂食标准商业饲料时 ,?梁旭方 Hiromi OKU Hiroshi Y.OGATA 李月琴 白俊杰 罗建仁 2002中国生物化学与分子生物学报2002,18,6:15

4日新制钢汽车排气系统用耐热铁素体不锈钢的开发显示文摘为了减轻空气污染和缓解全球变暖,各个国家均出台了尾气排放标准,且限制标准越来越严格。在此背景下,多种技术已被应用以满足相关要求,如提高燃油燃烧率、催化效率,降低排气管热容量等。而这一系列措施均会导致汽车尾气的最高温度上升。近些年,汽车排气系统零件主要用铁素体不锈钢制作,因为铁素体不锈钢的热膨胀系数小,耐循环氧化和耐高温疲劳性能均优于奥氏体不锈钢。介绍了日新制钢新研发的汽车排气系统用耐热不锈钢,以及该钢种的研发现状和发展趋势。Nb、Mo、Cu在700℃时的固溶强化和析出强化对提高铁素体不锈钢的高温强度非常有效,而添加Nb、Mo、W主要是在900℃时起到固溶强化作用。Cr,Si和Mn可降低铁素体不锈钢(含14%Cr)在大气中950℃保温200h的氧化程度。特别是添加0.8%及以上的Mn可有效提高氧化皮的附着力。事实证明,日新制钢开发的NSSHR-1(14Cr-1Mn-0.9Si-Nb)和NSSHR-2(10Cr-0.9Si-Nb-Ti)铁素体不锈钢与441和439不锈钢相比,具有更高的耐热性和成形性。Manabu Oku 李鑫(译) 毕洪运(校) 2011世界钢铁2011,11,5:4
