1Bombyx mori U-shaped regulates the melanization cascade and immune response via binding with the Lozenge protein显示文摘Zinc finger protein,an important transcription factor,regulates gene expression associated with various physiological and pathological processes.U-shaped,belong to the Friend of GATA(FOG)transcription factor,plays a crucial role in hematopoiesis by interacting with the GATA transcription factor as a co-factor.However,little is known about its functions in insects.In the present study,a U-shaped cDNA was identified and characterized from the silkworm Bombyx mori and its potential roles in innate immunity investigated.The predicted silkworm U-shaped amino acid sequence contained a classical nuclear localization signal(NLS)motif“GESSPKRRRR”at position 45CU459,and arginine residues at position 456 and 478 are the critical sites of the NLS.U-shaped mRNA was detected in all tested tissues of the B.mori;however,the highest levels were found in the hemocytes.U-shaped mRNA expression levels were upregulated in the hemocyte after the Escherichia coli and Staphylococcus aureus challenge.Furthermore,U-shaped knockdown significantly reduced the melanization process and suppressed the expression of melanization-associated genes,including PPO1,PPO2,PPAE and BAEE.In addition,U-shaped interacts with Lozenge protein to regulate the innate immune response of the insect.Our results revealed that U-shaped binds directly to Lozenge protein to modulate the melanization process and innate immune responses in silkworm.Kui Zhang Juan Tan Xiangwei Hao Houyi Tang Muhammad Nadeem Abbas Jingjing Su Yongyue Su Hongjuan Cui 2022Insect Science2022,29,3:1
2Methylation across the central dogma in health and diseases: new therapeutic strategies显示文摘The proper transfer of genetic information from DNA to RNA to protein is essential for cell-fate control,development,and health.Methylation of DNA,RNAs,histones,and non-histone proteins is a reversible post-synthesis modification that finetunes gene expression and function in diverse physiological processes.Aberrant methylation caused by genetic mutations or environmental stimuli promotes various diseases and accelerates aging,necessitating the development of therapies to correct the disease-driver methylation imbalance.In this Review,we summarize the operating system of methylation across the central dogma,which includes writers,erasers,readers,and reader-independent outputs.We then discuss how dysregulation of the system contributes to neurological disorders,cancer,and aging.Current small-molecule compounds that target the modifiers show modest success in certain cancers.The methylome-wide action and lack of specificity lead to undesirable biological effects and cytotoxicity,limiting their therapeutic application,especially for diseases with a monogenic cause or different directions of methylation changes.Emerging tools capable of site-specific methylation manipulation hold great promise to solve this dilemma.With the refinement of delivery vehicles,these new tools are well positioned to advance the basic research and clinical translation of the methylation field.Ruochen Liu Erhu Zhao Huijuan Yu Chaoyu Yuan Muhammad Nadeem Abbas Hongjuan Cui 2023Signal Transduction and Targeted Therapy2023,8,9:0
3Unsteady Flow and Heat Transfer of a Casson Micropolar Nanofluid over a Curved Stretching/Shrinking Surface显示文摘We present the results of an investigation into the behavior of the unsteady flow of a Casson Micropolar nanofluid over a shrinking/stretching curved surface,together with a heat transfer analysis of the same problem.The body force acting perpendicular to the surface wall is in charge of regulating the fluid flow rate.Curvilinear coordinates are used to account for the considered curved geometry and a set of balance equations for mass,momentum,energy and concentration is obtained accordingly.These are turned into ordinary differential equations using a similarity transformation.We show that these equations have dual solutions for a number of different combinations of various parameters.The stability of such solutions is investigated by applying perturbations on the steady states.It is found that high values of the Micropolar and Casson parameters cause the flow to move more slowly.However,when compared to a shrunken surface,a stretched surface produces a greater Micro-rotation flux.Muhammad A.Sadiq Nadeem Abbas Haitham M.S.Bahaidarah Mohammad Amjad 2023Fluid Dynamics & Materials Processing2023,19,2:0
4Enabling Smart Cities with Cognition Based Intelligent Route Decision in Vehicles Empowered with Deep Extreme Learning Machine显示文摘The fast-paced growth of artificial intelligence provides unparalleled opportunities to improve the efficiency of various industries,including the transportation sector.The worldwide transport departments face many obstacles following the implementation and integration of different vehicle features.One of these tasks is to ensure that vehicles are autonomous,intelligent and able to grow their repository of information.Machine learning has recently been implemented in wireless networks,as a major artificial intelligence branch,to solve historically challenging problems through a data-driven approach.In this article,we discuss recent progress of applying machine learning into vehicle networks for intelligent route decision and try to focus on this emerging field.Deep Extreme Learning Machine(DELM)framework is introduced in this article to be incorporated in vehicles so they can take human-like assessments.The present GPS compatibility issues make it difficult for vehicles to take real-time decisions under certain conditions.It leads to the concept of vehicle controller making self-decisions.The proposed DELM based system for self-intelligent vehicle decision makes use of the cognitive memory to store route observations.This overcomes inadequacy of the current in-vehicle route-finding technology and its support.All the relevant route-related information for the ride will be provided to the user based on its availability.Using the DELM method,a high degree of precision in smart decision taking with a minimal error rate is obtained.During investigation,it has been observed that proposed framework has the highest accuracy rate with 70%of training(1435 samples)and 30%of validation(612 samples).Simulation results validate the intelligent prediction of the proposed method with 98.88%,98.2%accuracy during training and validation respectively.Dildar Hussain Muhammad Adnan Khan Sagheer Abbas Rizwan Ali Naqvi Muhammad Faheem Mushtaq Abdur Rehman Afrozah Nadeem 2021Computers, Materials & Continua2021,,1:0
5Angiotensin converting enzyme inhibitor associated spontaneous herniation of liver mimicking a pleural mass:A case report显示文摘BACKGROUND Spontaneous diaphragmatic herniation of the liver is a rare entity.It may mimic pulmonary mass especially in the absence of trauma.Cough is a common side effect of angiotensin converting enzyme(ACE)inhibitors that may cause diaphragmatic rupture due to a sudden increase in trans-diaphragmatic pressure.We present a case of ACE-inhibitor associated spontaneous herniation of the liver mimicking pleural mass.CASE SUMMARY An 80-year-old woman presented with dry cough for 1 mo and sudden onset of cramping abdominal pain for 1 d.She denied history of trauma,prior surgeries,smoking,alcohol or illicit drug use.She has a history of diabetes and was started on an ACE inhibitor 6 mo ago for the management of hypertension.Examination was remarkable for right upper quadrant tenderness.Lab work-up was unremarkable.Chest X-ray showed a right lower lung opacity suspecting right pleural mass.Chest computed tomography scan ruled out pleural mass,however,revealed herniated right lobe of the liver(3.9 cm×3.6 cm×3.4 cm)into the thoracic cavity through the posterolateral diaphragmatic defect.Laparoscopic repair of the diaphragmatic defect was performed and the ACE inhibitor was stopped.Patients’symptoms had completely resolved on follow-up.CONCLUSION ACE inhibitor-associated cough may cause diaphragmatic liver herniation mimicking pleural mass.Early diagnosis,surgical repair and addressing the triggering factors improve patients’outcomes.Sameer Saleem Tebha Zain Ali Zaidi Sehrish Sethar Muhammad Asif Abbas Virk Muhammad Nadeem Yousaf 2022World Journal of Hepatology2022,14,4:0
6Hedgehog promotes cell proliferation in the midgut of silkworm,Bombyx mori显示文摘The Hedgehog(Hh)signaling pathway is one o f the major regulators of embryonic development and tissue homeostasis in multicellular organisms.However,the role of this pathway in the silkworm,especially in the silkworm midgut,remains poorly understood.Here,we report that Bombyx mori Hedgehog(BmHh)is expressed in most tissues of silkworm larvae and that its functions are well-conserved throughout evolution.We further demonstrate that the messenger RNA o f four Hh signaling components,BmHh ligand,BmPtch receptor,signal transducer BmSmo and transcription factor BmCi,are all upregulated following Escherichia coli or Bacillus thuringiensis infection,indicating the activation o f the Hh pathway.Simultaneously,midgut cell proliferation is strongly promoted.Conversely,the repression of Hh signal transduction with double-stranded RNA or cyclopamine inhibits the expression o f BmHh and BmCi and reduces cell proliferation.Overall,these findings provide new insights into the Hh signaling pathway in the silkworm,B.mori.Shun-Qin Zhu Ya-Jun Zhang Muhammad Nadeem Abbas Xiang-Wei Hao Yu-Zu Zhao Hang-Hua Liang Hong-Juan Cui Li-Qun Yang 2020Insect Science2020,27,4:0

8Integrin β2 and β3:Two plasmatocyte markers deepen our understanding of the development of plasmatocytes in the silkworm Bombyx mori显示文摘Insect hemocytes play important biological roles at developmental stages,metamorphosis,and innate immunity.As one of the most abundant cell types,plasmatocytes can participate in various innate immune responses,especially in encapsulation and node formation.Here,2 molecular markers of plasmatocytes,consisting of integrinβ2 andβ3,were identified and used to understand the development of plasmatocytes.Plasmatocytes are widely distributed in the hematopoietic system,including circulating hemolymph and hematopoietic organs(HPOs).HPOs constantly release plasmatocytes with high proliferative activity in vitro;removal of HPOs leads to a dramatic reduction in the circulating plasmatocytes,and the remaining plasmatocytes gradually lose their ability to proliferate in vivo.Our results demonstrated that the release of plasmatocytes from HPOs is regulated by insulin-mediated signals and their downstream pathways,including PI3K/Akt and MAPK/Erk signals.The insulin/PI3K/Akt signaling pathway can significantly irritate the hematopoiesis,and its inhibitor LY294002 could inhibit the hemocytes discharged from HPOs.While the insulin/MAPK/Erk signaling pathway plays a negative regulatory role,inhibiting its activity with U0126 can markedly promote the discharge of plasmatocytes from HPOs.Our results indicate that the circulating plasmatocytes are mainly generated and discharged by HPOs.This process is co-regulated by the PI3K/Akt and MAPK/Erk signals in an antagonistic manner to adjust the dynamic balance of the hemocytes.These findings can enhance our understanding of insect hematopoiesis.Kui Zhang Jingjing Su Xin Hu Xiaomin Yan Siyuan Chen Chongyang Li Guangzhao Pan Hongbo Chang Wenli Tian Muhammad Nadeem Abbas Hongjuan Cui 2022Insect Science2022,29,6:0
