| 3 | Ectopic osteogenesis and angiogenesis regulated by porous architecture of hydroxyapatite scaffolds with similar interconnecting structure in vivo显示文摘The macro-pore sizes of porous scaffold play a key role for regulating ectopic osteogenesis and angiogenesis but many researches ignored the influence of interconnection between macro-pores with different sizes.In order to accurately reveal the relationship between ectopic osteogenesis and macro-pore sizes in dorsal muscle and abdominal cavities of dogs,hydroxyapatite(HA)scaffolds with three different macro-pore sizes of 500–650,750–900 and 1100–1250 mm were prepared via sugar spheres-leaching process,which also had similar interconnecting structure determined by keeping the d/s ratio of interconnecting window diameter to macro-pore size constant.The permeability test showed that the seepage flow of fluid through the porous scaffolds increased with the increase of macro-pore sizes.The cell growth in three scaffolds was not affected by the macro-pore sizes.The in vivo ectopic implantation results indicated that the macro-pore sizes of HA scaffolds with the similar interconnecting structure have impact not only the speed of osteogenesis and angiogenesis but also the space distribution of newly formed bone.The scaffold with macro-pore sizes of 750–900 mm exhibited much faster angiogenesis and osteogenesis,and much more uniformly distribution of new bone than those with othermacro-pore sizes.This work illustrates the importance of a suitable macro-pore sizes in HA scaffolds with the similar interconnecting structure which provides the environment for ectopic osteogenesis and angiogenesis. | Jinyu Li Wei Zhi Taotao Xu Feng Shi Ke Duan JianxinWang Yandong Mu Jie Weng | 2016 | Regenerative Biomaterials2016,3,5: | 16 |
| 4 | UDP-glucose pyrophosphorylase2 (OsUgp2), a pollen-preferential gene in rice, plays a critical role in starch accumulation during pollen maturation显示文摘UDP-glucose pyrophosphorylase (UGPase) is predominantly present and plays significant role in carbohydrate metabolism in plants. Two homologous UGPase genes, OsUgp1 and OsUgp2, exist in rice genome. OsUgp1 has recently been reported to be essential for callose deposition during pollen mother cell and meiosis stages as well as for seed carbohydrate metabolism. In this study, a full-length cDNA of OsUgp2 was isolated from rice anther. Northern blot and RNA in situ hybridization indicated that the expression of OsUgp2 was preferentially in pollen and developmentally regulated. No transcripts were found in leaf, stem, lemma/palea, ripening grain and florets before the uninucleate microspore developmental stage, but a large quantity of OsUgp2 mRNA was found in pollen at the binucleate and mature stages. The immunolocalization of OsUgp2 showed a similar expression pattern to that by RNA in situ hybridization. The function of OsUgp2 was investigated by dsRNA-mediated transcriptional gene silencing. The pollen fertility of 16 independent transgenic rice plants was found between 25% and 90%, which was correlated with the amount of OsUgp2 mRNA. The results of morphological changes and starch variation during pollen development in transgenic rice showed that the abnormal feature of pollen development appeared after the uninucleate microspore stage. Starch failed to accumulate in pollen and thus led to sterile pollens. These results demonstrated that OsUgp2 is a pollen-preferential 'late gene' and plays a key role during pollen maturation, especially for starch accumulation. | 6 | Overexpression of potato miR482e enhanced plant sensitivity to Verticillium dahliae infection显示文摘Verticillium wilt of potato is caused by the fungus pathogen Verticillium dahliae. Present s RNA sequencing data revealed that miR482 was in response to V. dahliae infection but the function in potato is elusive. Here, we characterized potato miR482 family and its putative role resistance to Verticillium wilt. Members of the potato mi R482 superfamily are variable in sequence, but all variants target a class of disease-resistance proteins with nucleotide binding site(NBS)and leucine-rich repeat(LRR) motifs. When potato plantlets were infected with V. dahliae, the expression level of miR482e was downregulated, and that of several NBS-LRR targets of miR482e were upregulated. Transgenic potato plantlets overexpressing miR482e showed hypersensitivity to V. dahliae infection. Using s RNA and degradome datasets, we validated that miR482e targets m RNAs of NBS-LRR disease-resistance proteins and triggers the production of trans-acting(ta)si RNAs, most of which target m RNAs of defense-related proteins. Thus, the hypersensitivity of transgenic potato could be explained by enhanced miR482e and miR482e-derived tasi RNA-mediated silencing on NBS-LRR-disease-resistance proteins. | 9 | Sonographic appearance of anal cushions of hemorrhoids显示文摘AIM To evaluate the diagnostic value of different sonographic methods in hemorrhoids.METHODSForty-two healthy volunteers and sixty-two patients with gradesⅠ-Ⅳhemorrhoids received two different sonographic examinations from January 2013 to January2016 at the First and Second Hospitals of Xinjiang Medical University in a prospective way.We analyzed the ultrasonographic findings of these participants and evaluated the outcomes.Resected gradesⅢandⅣhemorrhoid tissues were pathologically examined.The concordance of ultrasonographic results with pathologyresults was assessed with the Cohen’s kappa coefficient.RESULTS All healthy volunteers and all patients had no particular complications related to sonography.There were no statistically significant differences between the participants regarding age(P=0.5919),gender(P=0.4183),and persistent symptoms(P>0.8692).All healthy control participants had nospecial findings.However,30 patients with hemorrhoids showed blood signals around the dentate line on ultrasonography.When gradesⅠandⅡhemorrhoids were analyzed,there were no significant differences between transrectal ultrasound(TRUS),transperianal ultrasound(TPUS),and transvaginal ultrasound(TVUS)(P>0.05).GradesⅢandⅣhemorrhoids revealed blood flow with different directions which could be observed as a'mosaic pattern'.In patients with gradesⅢandⅣhemorrhoids,the number of patients with'mosaic pattern'as revealed by TRUS,TPUS and TVUS was 22,12,and 4,respectively.Patients with gradesⅢandⅣdisease presented with a pathologically abnormal cushion which usually appeared as a'mosaic pattern'in TPUS and an arteriovenous fistula in pathology.Subepithelial vessels of resected gradesⅢandⅣhemorrhoid tissues were manifested by obvious structural impairment and retrograde and ruptured changes of internal elastic lamina.Some parts of the Trietz’s muscle showed hypertrophy and distortion.Arteriovenous fistulas and venous dilatation were obvious in the anal cushion of hemorhoidal tissues.After pathological results with arteriovenous fistulas were taken as the standard reference,we evaluated the compatibility between the two methods according to the Cohen’s kappa coefficiency calculation.The compatibility(Cohein kappa co-efficiency value)between'mosaic pattern'in the TPUS and arteriovenous fistula in pathology was very good(?=0.8939).When compared between different groups,TRUS presented the advantage that the mosaic pattern could be confirmed in more patients,especially for group A.There was a statistical difference when comparing group A with group B or C(P<0.05 for both).There were obvious statistical differences between group A and group B with regard to the vessel diameter and blood flow velocity measured by TRUS(P<0.05).CONCLUSION Patients with gradesⅢandⅣhemorrhoids present with a pathologically abnormal cushion which usually appears as a'mosaic pattern'in sonography,which is in accord with an arteriovenous fistula in pathology.There are clearly different hemorrhoid structures shown by sonography.'Mosaic pattern'may be a parameter for surgical indication of gradesⅢandⅣhemorrhoids. | Adilijiang Aimaiti Ma Mu Ti Jiang A Ba Bai Ke Re Irshat Ibrahim Hui Chen Maimaitituerxun Tuerdi Mayinuer | 2017 | World Journal of Gastroenterology2017,23,20: | 11 |
| 10 | Insight into atmospheric corrosion evolution of mild steel in a simulated coastal atmosphere显示文摘The corrosion behavior of mild steel in a simulated coastal atmosphere environment has been investigated by the indoor accelerated wet/dry cyclic corrosion acceleration test(CCT),scanning electron microscopy(SEM),Raman spectroscopy and electrochemical measurements.During the CCT test of 60 cycles,the evolution of logarithmic(corrosion rate)vs.logarithmic(CCT cycles)presents a turning point at the 5th cycle,presenting a tendency to increase first and then decrease to gradually stabilize as the CCT cycle prolonged.Before the 5th cycle,γ-FeOOH andβ-FeOOH and Fe_(3)O_(4)were detected,respectively.And then,α-FeOOH as a new chemical composition was detected in the subsequent corrosion cycles.It is found that,after long term corrosion,the rust separated into a relatively dense inner layer rich withα-FeOOH and a loose outer layer rich withγ-FeOOH,both of which have poor electrical conductivity.The rapid increase of corrosion rate in the early stage since reducible corrosion products are involved in the reduction process of the cathode which promotes the dissolution of the anodic metal substrate.Afterward,as the rust layer thickens,the resistance of the rust increases,and the aggressive ions diffusion is blocked,gradually suppressing the electrochemical corrosion process.At last,when the composition and distribution of the rust layer remain stable,the corrosion presents a fluctuating speed around a certain value during the cracking and self-repairing process of the rust layer. | Yue Wang Xin Mu Junhua Dong Aniefiok Joseph Umoh Wei Ke | 2021 | Journal of Materials Science & Technology2021,,17: | 7 |
| 11 | Effect of tin addition on corrosion behavior of a low-alloy steel in simulated costal-industrial atmosphere显示文摘The effect of tin addition on the atmospheric corrosion behavior of a low-alloy steel in sim ulated coastalindustrial atm osphere has been investigated by indoor wet/dry cyclic corrosion test (CCT). The results indicate that tin addition can obviously make the steel substrate more resistant to atmospheric corrosion by suppressing the cathodic H^+ reduction reaction, and but tin addition is not of obvious beneficial effect when the steel is covered with a thicker rust layer during long-term corrosion process. The reason lies in the fact that the presence of un-reduced H^+ can lower the electrolyte pH value and lead to a loose and porous rust layer on tin-containing steel sample than that on tin-free steel sample. In addition, the 120 CCT cycles corrosion process of the two steels can be divided into three stages. Both the tin-free and tin-containing steels show an increasing corrosion rate during the initial corrosion stage and then exhibit a decreasing corrosion rate during the second and third corrosion stages. | 13 | High levels of circulating GM-CSF^(+) CD4^(+) T cells are predictive of poor outcomes in sepsis patients: a prospective cohort study显示文摘Granulocyte colony-stimulating factor(GM-CSF),produced by CD4^(+)T cells,has recently been implicated in the pathogenesis of inflammatory diseases,such as multiple sclerosis and juvenile arthritis.However,the role of GM-CSF-producing CD4^(+)T cells in sepsis remains unknown.This study reports peripheral changes in GM-CSF-producing CD4^(+)T cells in septic patients and the possible underlying mechanism by which GM-CSF influences the outcome of sepsis.Forty-three septic patients,20 SIRS patients,and 20 healthy controls were enrolled in this study and followed for 28 days to assess mortality.We measured the peripheral frequency of GM-CSF^(+)CD4^(+)T cells and recorded their associated relationship with disease progression.Our data demonstrated that peripheral GM-CSF-producing CD4^(+)T cells were significantly higher in septic patients than in both SIRS patients and healthy controls.These cells exhibit a memory phenotype and impaired IFN-γ-secreting capacity in sepsis patients.Using a receiver operating curve analysis with 8.01%as a cut-off point,the percentage of GM-CSF^(+)CD4^(+)T cells could predict the outcome of septic patients.Combined with the increase in GM-CSF-producing CD4^(+)T cells,inflammatory cytokines IL-1βand IL-6 were also upregulated.Using an in vitro neutrophil model,we found that GM-CSF inhibited C3aR expression,while inducing IL-8 production.Furthermore,this effect was transferrable in plasma from sepsis patients and was attenuated by inhibition of GM-CSF using an anti-GM-CSF antibody.These results indicate that GM-CSF-producing CD4^(+)T cells may serve as a marker of sepsis severity.Thus,targeting GM-CSF overproduction may benefit sepsis patients. | Huihuang Huang Siyu Wang Tianjun Jiang Rong Fan Zheng Zhang Jinsong Mu Ke Li Yonggang Wang Lei Jin Fang Lin Jie Xia Lijian Sun Biao Xu Chengcheng Ji Jing Chen Juan Chang Bo Tu Bing Song Chao Zhang Fu-Sheng Wang Ruonan Xu | 2019 | Cellular & Molecular Immunology2019,16,6: | 6 |

| 17 | Numerical Approach for Atmospheric Corrosion Monitoring Based on EIS of a Weathering Steel显示文摘Electrochemical impedance spectroscopy(EIS) and film thickness measurement have been employed to study the atmospheric corrosion of a weathering steel covered with a thin electrolyte layer in a simulated coastal–industrial atmosphere. The results indicate that the corrosion rate is a function of the covered electrolyte thickness and the wet/dry cycle. Within each wet/dry cycle, the increased corrosion rate is related to the increased Cl-and SO2-4concentration and an enhancement of oxygen diffusion rate with the evaporation of the electrolyte. In addition, the corrosion rate increases during the initial corrosion stage and then decreases as the wet/dry cycle proceeds. | 18 | Acetate-assistant efficient cation-exchange of halide perovskite nanocrystals to boost the photocatalytic CO_(2) reduction显示文摘The judicious implantation of active metal cations into the surface of semiconductor nanocrystal(NC)through cation-exchange is one of the facile and viable strategies to enhance the activity of catalysts for photocatalytic CO_(2)reduction,by shortening the transfer pathway of photogenerated carriers and increasing the active sites simultaneously.However,cation-exchange is hard to achieve for halide perovskite NCs owing to the stable octahedron of[PbX6]4−with strong interaction between halogen and lead.Herein,we report a facile method to overcome this obstacle by replacing partial Br−with acetate(Ac−)to generate CsPbBr_(3) NC(coded as CsPbBr_(3−x)Ac_(x)).A small amount of Ac−instead of Br−does not change the crystal structure of halide perovskite.Owing to the weaker interaction between acetate and lead in comparison with bromide,the corresponding octahedron structure containing acetate in CsPbBr_(3−x)Ac_(x) can be easily opened to realize efficient cation-exchange with Ni^(2+) ions.The resulting high loading amount of Ni^(2+) as active site endows CsPbBr_(3−x)Ac_(x) with an improved performance for photocatalytic CO_(2)reduction under visible light irradiation,exhibiting a significantly increased CO yield of 44.09μmol·g^(−1)·h^(−1),which is over 8 and 3 times higher than those of traditional pristine CsPbBr_(3) and nickel doped CsPbBr_(3) NC,respectively.This work provides a critical solution for the efficient metal doping of low-cost halide perovskite NCs to enhance their photocatalytic activity,promoting their practical applications in the field of photocatalysis. | Jialin Cheng Yanfei Mu Liyuan Wu Zhaolei Liu Ke Su Guangxing Dong Min Zhang Tongbu Lu | 2022 | Nano Research2022,15,3: | 3 |

The results show that zinc coating has a greater corrosion rate during the initial period and a lower corrosion rate during the subsequent period, and the patina composition and structure can greatly affect the corrosion kinetics evolution of zinc coating. | 20 | mTOR regulates TLR-induced c-fos and Th1 responses to HBV and HCV vaccines显示文摘Although IL-12 plays a critical role in priming Th1 and cytotoxic T lymphocyte(CTL) responses, Toll-like receptor(TLR) signaling only induces low amounts of IL-12 in dendritic cells and macrophages, implying the existence of stringent regulatory mechanisms. In this study, we sought to uncover the mechanisms underlying TLR-induced IL-12 expression and the Th1 response. By systemic screening, we identified a number of protein kinases involved in the regulation of TLRinduced IL-12 expression. In particular, PI3 K, ERK, and m TOR play critical roles in the TLR-induced Th1 response by regulating IL-12 and IL-10 production in innate immune cells. Moreover, we identified c-fos as a key molecule that mediates m TOR-regulated IL-12 and IL-10 expression in TLR signaling. Mechanistically, m TOR plays a crucial role in c-fos expression, thereby modulating NFκB binding to promoters of IL-12 and IL-10. By controlling the expression of a special innate gene program, m TOR can specifically regulate the TLR-induced T cell response in vivo. Furthermore, blockade of m TOR by rapamycin efficiently boosted TLR-induced antigen-specific T and B cell responses to HBV and HCV vaccines.
