| 3 | Changes in circulating Foxp3^+ regulatory T cells and interleukin-17-producing T helper cells during HBV-related acute-on-chronic liver failure显示文摘AIM:To longitudinally investigate cytokine gene expression and protein levels in Th17 and Treg cells,to observe T-cell phenotypes during hepatitis B virus(HBV)-related acute-on-chronic liver failure(ACHBLF)and to analyze changes in Th17 and Treg phenotypes during disease progression.METHODS:We measured the expression of seven Th17/Treg differentiation-related genes and serum concentrations of the corresponding cytokines in 18ACHBLF,18 chronic hepatitis B(CHB)disease controls and 10 healthy controls(HCs)by real-time quantitative pCR and enzyme linked immunosorbent assay.peripheral Th17 and Treg cell frequencies were analyzed by flow cytometry.RESULTS:From the onset of ACHBLF,patients presented with a conductive Th17 differentiation cytokine environment accompanied by high Th17 frequency and high serum IL-17 levels,which were sustained throughout the disease course.The Treg-related cytokine IL-2and Foxp3 were also up-regulated from disease onset,and Foxp3 gene expression showed a gradually increasing trend during ACHBLF.The circular phenotype of Treg and Th17 cells showed changes from the onset of ACHGLF.At disease onset,Th17 frequency increased significantly compared with both CHB and HCs,but Treg cell frequency decreased significantly compared with CHB.During the ACHBLF event,Th17 frequency remained higher compared with HCs,but decreased sharply from the peak point to the recovery point;Treg cell frequency increased gradually during the ACHBLF event.Treg and Th17 cell counts correlated with ACHBLF development;in all patients,serum IL-17 levels significantly correlated with patient serum ALT levels.In survivors,Th17 frequency at the onset point and the Treg to Th17 ratio at the peak point correlated with the patient’s model for end stage liver disease(MELD)plus sodium(MELD-Na)score.The Treg to Th17 ratio and the Th17 frequency at onset were significant predictors of patient survival.Low Treg/Th17 cell ratios at the onset predicted poor survival.Survivors exhibited an initial decrease in the circulating Treg/Th17 ratio from the onset to the peak time,and subsequently displayed a continuous increase.CONCLUSION:Treg and Th17 cells showed changes in genes,protein levels and T cell phenotypes during ACHBLF events.An increased Treg/Th17 ratio was associated with the survival of ACHBLF patients. | Xue-Song Liang Cheng-Zhong Li Yin Zhou Wei Yin Ya Yun Liu Wen-Han Fan | 2014 | World Journal of Gastroenterology2014,20,26: | 44 |
| 4 | Role of Epstein-Barr Virus Encoded Latent Membrane Protein 1 in the Carcinogenesis of Nasopharyngeal Carcinoma显示文摘Epstein-Barr virus (EBV)-encoded latent membrane protein 1 (LMP1) has been known to have oncogenic properties during latent infection in nasopharyngeal carcinoma (NPC). Our studies focused on the role of LMP1 in NPC, and showed that LMP1 triggers the NF-κB, AP-1 and STAT signaling pathways. Strikingly, LMP1 was found to mediate the formation of a new heterodimer between c-Jun and JunB. Also, we have identified JAK/STAT and PI-PLC-PKC activation triggered by LMP1 through upregulating the expression of JAK3 and enhancing the phosphorylation of STAT. The constitutive activation of these signaling cascades explains LMP1’s ability to induce such a diverse array of morphological and phenotypic effects in cells and provides insight into how LMP1 may induce cell transformation, in which multihit targeted genes in the downstream play an essential role. All signaling cascades triggered by LMP1 ultimately lead to the disruption of the cell cycle: the acceleration of G1/S phase and the arrest of G2/M phase. We also found that LMP1 induced the expression of hTERT and promoted cell immortalization. Importantly, by intervening physical intracellular signal transduction pathways and disturbing the progression of the cell cycle, LMP1, an important oncoprotein encoded by EBV, is thought to be a key modulator in the pathogenesis of NPC. | 5 | Symptom-based tendencies of Helicobacter pylori eradication in patients with functional dyspepsia显示文摘AIM: To investigate whether there were symptom-based tendencies in the Helicobacter pylori (H. pylori) eradication in functional dyspepsia (FD) patients. METHODS: A randomized, single-blind, placebo-controlled study of H. pylori eradication for FD was conduct- ed. A total of 195 FD patients with H. pylori infection were divided into two groups: 98 patients in the treatment group were treated with rabeprazole 10 mg twice daily for 2 wk, amoxicillin 1.0 g and clarithromycin 0.5 g twice daily for 1 wk; 97 patients in the placebo group were given placebos as control. Symptoms of FD, such as postprandial fullness, early satiety, nausea, belching,epigastric pain and epigastric burning, were assessed 3 mo after H. pylori eradication. RESULTS: By per-protocol analysis in patients with successful H. pylori eradication, higher effective rates of 77.2% and 82% were achieved in the patients with epigastric pain and epigastric burning than those in the placebo group (P < 0.05). The effective rates for postprandial fullness, early satiety, nausea and belching were 46%, 36%, 52.5% and 33.3%, respectively, and there was no significant difference from the placebo group (39.3%, 27.1%, 39.1% and 31.4%) (P > 0.05). In 84 patients who received H. pylori eradication therapy, the effective rates for epigastric pain (73.8%) and epigastric burning (80.7%) were higher than those in the placebo group (P < 0.05). The effective rates for postprandial fullness, early satiety, nausea and belch- ing were 41.4%, 33.3%, 50% and 31.4%, respective- ly, and did not differ from those in the placebo group (P > 0.05). By intention-to-treat analysis, patients with epigastric pain and epigastric burning in the treatment group achieved higher effective rates of 60.8% and 65.7% than the placebo group (33.3% and 31.8%) (P < 0.05). The effective rates for postprandial fullness, early satiety, nausea and belching were 34.8%, 27.9%, 41.1% and 26.7% respectively in the treatment group, with no significant difference from those in the placebo group (34.8%, 23.9%, 35.3% and 27.1%) (P > 0.05). CONCLUSION: The efficacy of H. pylori eradication has symptom-based tendencies in FD patients. | 9 | Low-temperature SCR activity and SO_2 deactivation mechanism of Ce-modified V_2O_5–WO_3/TiO_2 catalyst显示文摘The promotion effect of ceria modi fi cation on the low-temperature activity of V2O5-WO3/TiO2 catalyst was evaluated for the selective catalytic reduction of NO with NH3(NH3-SCR). The catalytic activity of 1 wt% V2O5-WO3/TiO2 was signi fi cantly enhanced by the addition of 8 wt%ceria, which exhibited a NO x conversion above 80% in a broad temperature range 190–450 1C. This performance was comparable with 3 wt%V2O5-WO3/TiO2, indicating that the addition of ceria contributed to reducing the usage of toxic vanadia in developing low-temperature SCR catalysts. Moreover, V1 Ce WTi exhibited approximately 10% decrease in NOx conversion in the presence of 60 ppm SO2. The characterization results indicated that active components of V, W and Ce were well dispersed on TiO2 support. The synergetic interaction between Ce and V species by forming V–O–Ce bridges enhanced the reducibility of VCe WTi catalyst and thus improved the low-temperature activity. The sulfur poisoning mechanism was also presented on a basis of the designed TPDC(temperature-programmed decomposition) and TPSR(temperatureprogrammed surface reaction) experiments. The deposition of(NH4)2SO4on V1 Ce WTi catalyst was much smaller compared with that on V1 Ti.On the other hand, the oxidation of SO2 to SO3was signi fi cantly promoted on the CeO2-modi fi ed catalyst, accompanied by the formation of cerium sulfates. | 11 | Analysis of DNA methylation in different maize tissues显示文摘DNA methylation plays an important role in gene expression regulation during biological development and tissue differentiation in plants. This study adopted methylation-sensitive Amplified fragment length polymorphism (AFLP) to compare the levels of DNA cyto- sine methylation at CCGG sites in tassel, bracteal leaf, and ear leaf from maize inbred lines, 18 White and 18 Red, respectively, and also examined specific methylation patterns of the three tissues. Significant differences in cytosine methylation level among the three tissues and the same changing tendency in two inbred lines were detected. Both MSAP (methylation sensitive amplification polymorphism) ratio and full methylation level were the highest in bracteal leaf, and the lowest in tassel. Meanwhile, different methylation levels were ob- served in the same tissue from the inbred lines, 18 White and 18 Red. Full methylation of internal cytosine was the dominant type in the maize genome. The differential methylation patterns in the three tissues were observed. In addition, sequencing of nine differentially me- thylated fragments and the subsequent blast search revealed that the cytosine methylated 5′-CCGG-3′sequences were distributed in re- peating sequences, in the coding and noncoding regions. Southern hybridization was used to verify the methylation polymorphism. These results clearly demonstrated the power of the MSAP technique for large-scale DNA methylation detection in the maize genome, and the complexity of DNA methylation change during plant growth and development. | 12 | Pollution characteristics of organic and elemental carbon in PM_(2.5) in Xiamen,China显示文摘Xiamen,located on the southeastern coastal line of China,is undergoing rapid urbanization and industrialization,so its air quality has a trend of degradation.However,studies on level,temporal and spatial changes of fine particles (PM 2.5) and their carbonaceous fractions are scarce.In this article,abundance,sources,seasonal and spatial variations,distribution of organic carbon (OC) and elemental carbon (EC) in PM 2.5,were studied at suburban,urban and industrial sites in Xiamen during four season-representative months in 2009-2010.PM 2.5 samples were collected with middle volume sampler and were analyzed for OC and EC with thermal optical transmittance (TOT) method.Results showed that the annual average PM 2.5 concentrations were 63.88-74.80 μg/m 3 at three sites.While OC and EC concentrations were in the range of 15.81-19.73 μg/m 3 and 2.74-3.49 μg/m 3,respectively,and clearly presented the summerminima and winter maxima in this study.The carbonaceous aerosol accounted for 42.8%-47.3% of the mass of PM 2.5.The annual average of secondary organic carbon (SOC) concentrations in Xiamen were 9.23-11.36 μg/m 3,accounting for approximately 56% of OC.Strong correlations between OC and EC was found in spring (R 2=0.50) and autumn (R 2=0.73),suggesting that there were similar emission and transport processes for carbonaceous aerosols in these two seasons,while weak correlations were found in summer (R 2=0.33) and winter (R 2=0.41).The OC/EC ratios in PM 2.5 varied from 2.1 to 8.7 with an annual average of 5.7,indicating that vehicle exhaust,coal smoke and biomass burning were main source apportionments of carbonaceous fractions in Xiamen. | Fuwang Zhang Jinping Zhao Jinsheng Chen Ya Xu Lingling Xu | 2011 | Journal of Environmental Sciences2011,23,8: | 27 |

| 18 | Crustal P-wave velocity structure of the Longmenshan region and its tectonic implications for the 2008 Wenchuan earthquake显示文摘The P-wave velocity structure of the crust in the Longmenshan region has been imaged by seismic travel time tomography us ing local and regional first P-wave arrivals recorded from 2000 to 2008. The tomographic model provides a way to analyze the deep tectonics of the Longmenshan fault belt and the tectonic implications for the 2008 Ms8.0 Wenchuan earthquake. The P-wave velocity images indicate that the initial rupture site and focal depth of the Wenchuan earthquake, together with the di rection of rupture propagation, closely relate to the crustal structure of the Longmenshan region. The Pengguan massif to the west of the Longmenshan fault belt is characterized by high velocity anomalies, suggesting that the crust has a strong strain strength that can accumulate large stresses over a long period. The Ms8.0 Wenchuan earthquake is located at the southwestern end of the Pengguan massif and the western edge of the Sichuan Basin. The collision between the Pengguan massif and the Sichuan Basin becomes the primary reason for the occurrence of the Ms8.0 Wenchuan earthquake. To the north of Wenchuan, the occurrence and propagation of rupture benefit from low velocity anomalies along the Longmenshan fault belt; whereas to the south of Wenchuan, the brittle rupture can occur with more difficulty in relatively weak crust with low velocities. This may be one of the reasons for the absence of aftershocks to the south of Wenchuan, and the rupture induced by the Ms8.0 Wenchuan earthquake propagating from the north to the south along the Longmenshan fault belt. The deep geodynamics of the Ms8.0 Wenchuan earthquake may occur due to the discrepancy of crustal structures on the two sides of the Longmenshan fault belt. Ductile deformation and crustal flow can easily occur in the weak middle-lower crust beneath the Songpan-Garze orogenic belt. The eastward movement of the Tibetan Plateau is obstructed by the rigid lithosphere of the Sichuan Basin, and then the thick ening of the middle-lower crust and vertical deformation occur in the crust of the Longmenshan fault belt.
