1郾城郝家台遗址的发掘显示文摘郝家台遗址位于河南省漯河市偃城县东3公里石槽赵村东北的台地上。南去1公里为漯河市,沙河在遗址南1公里处由西向东流去,京广铁路从遗址的西南部穿过(图一)。遗址平面呈椭圆形,东西长260、南北宽250米,遗址总面积65000平方米。遗址高出附近地面2—3米,海拔62.1米。郝家台俗称“磕脚台”,其上原有“裴晋公庙”,HPAI and Yancheng County Memorial Hall of Xu Shen, 1992华夏考古1992,,3:57

3Clinicopathological analysis of 14 patients with combined hepatocellular carcinoma and cholangiocarcinoma显示文摘BACKGROUND:Combined hepatocellular carcinoma and cholangiocarcinoma (cHCC-CC) is an uncommon subtype of primary hepatic carcinoma,and its prognosis is poor.This study was undertaken to investigate the prognosis and the clinicopathological characteristics of cHCC-CC,including their possible cellular origin.METHODS:Among 852 patients with a primary hepatic carcinoma who underwent hepatectomy from January 1998 to April 2008 at our hospital,cHCC-CC was identified in 14 patients The clinicopathological characteristics of the 14 patients were analyzed retrospectively.The expression of the liver stem cell markers (c-kit,CD90,CD133 and CK19) in the tumor tissue was detected by immunohistochemistry,and the Kaplan-Meier method was used to evaluate survival.RESULTS:Among the 14 patients,9 presented with abdominal pain,3 with anorexia and debilitation,and the remaining two patients were asymptomatic.The mean age was 53.6±3.0 (range 38-74) years.Among the included patients,11 had an elevated serum alpha-fetoprotein level,13 were infected with hepatitis B virus,9 had vascular invasion and 1 had lymph node metastasis The average diameter of the tumors was 9.9±1.1 (range 5.0-16.0) cm.The median overall survival time was 7.9±1.0 months In addition,the presence of the liver stem cell markers,c-kit CD90,CD133 and CK19 was 71.4%,85.7%,92.9% and 78.6% respectively.All four markers were simultaneously expressed in eight cases.CONCLUSIONS:cHCC-CC has aggressive characteristics and the prognosis is extremely dismal.The high expression of liver stem cell markers in the tumor tissue suggests that these tumors may derive from liver stem cells.Xian-Huan Yu,Lei-Bo Xu,Hong Zeng,Rui Zhang,Jie Wang and Chao LiuAuthor Affiliations:Department of Hepato-Pancreato-Biliary Surgery and Department of Pathology,Sun Yat-Sen Memorial Hospital,Sun Yat-Sen University,Guangzhou 510120,China 2011Hepatobiliary & Pancreatic Diseases International2011,10,6:7
4Effect of Ionizing Radiation on the Expression of p16, CyclinDI and CDK4 in Mouse Thymocytes and Splenocytes显示文摘Objective:To investigate the effect of ionizing radiation on the expression of p16,CyclinD1,and CDK4 in mouse thymocytes and splenocytes.Methods:Fluorescent staining and flow cytometry analysis were employed for the measurement of protein expression.Results:In time course experiments,it was found that the expression of p16 protein was significantly increased at 8,24,and 48h for thymocytes(P<0.05,P<0.01,and P<0.05,respectively)and at 24h for splenocytes (P<0.05) after whole body irradiation(WBI) with 2.0Gy X-rays.However,the expression of CDK4 protein was significantly decreased from 8h to 24h for thymocytes(P<0.05-P<0.01) and from 8h to 72h for splenocytes(P<0.05-P<0.01).In dose effect experiments,it was found that the expression of P16 protein in thymocytes and splenocytes was significantly increased at 24h after WBI with 1.0,2.0 ,and 4.0GY(P<0.05-P<0.01),whereas the expression of CDK4 protein was significantly decreased with 2.0Gy for thymocytes(P<0.05)and 0.5-6.0 Gy for splenocytes(P<0.05-P<0.01),Results also showed that the expression of CyclinD1 protein decreased markedly in both thymocytes and splenocytes after exposure.Conclusion:the results indicate that the expression of p16 protein in thymocytes and splenocytes can be induced by ionizing radiation,and the p16-CyclinD1/CDK4 pathway may play an important role for G1 arrest of thymocytes induced by X-rays.GUI-ZHI JU, XIAO-MEI WANG, SHI-Bo FU, AND SHU-ZHENG LIUDepartment of Radiation Biology, Medical Center in Memorial of Norman Bethune, Jilin University, Changchun 130021, China 2003Biomedical and Environmental Sciences2003,16,1:7
5Functional distinction of rat liver natural killer cells from spleen natural killer cells under normal and acidic conditions in vitro显示文摘BACKGROUND: The microenvironment within solid tumors has often been shown to exhibit an acidic extracellular pH. Although the morphologic and functional differences in natural killer (NK) cells of the liver and spleen have been reported previously under physiological conditions, the difference under acidic conditions is still unclear. This study was to investigate the differences in the morphological and functional characteristics between rat liver and spleen NK cells under normal and acidic conditions in vitro. METHODS: Liver and spleen NK cells were isolated and purified from Sprague-Dawley rats by density gradient centrifugation and the Dynabeads FlowComp TM Flexi system, and stimulated for 4 days with or without IL-2 or treated with low pH or control for different times. Morphology was examined by scanning electron microscopy (SEM) and transmission electron microscopy (TEM), cell death and proliferation assays were performed by flow cytometry, IFN-γ production was tested by ELISA, and cytotoxic activity was evaluated by lactate dehydrogenase (LDH) release assay. RESULTS: Liver NK cells had significantly higher levels of cytotoxic activity than spleen NK cells under normal and acidic conditions, and the maximum difference was observed at pH 5.6. Further analysis revealed that the cytotoxic activity of NK cells was correlated with morphology, cell death, proliferative activity and IFN-γ production. By TEM, liver NK cells contained a greater number of electron-dense granules per cell at pH 5.6.Moreover, a modest elevation of cell death and reduction of proliferation of liver NK cells occurred within a range of 5.6-7.2. Interestingly, an acidic extracellular pH only marginally, and not significantly, suppressed IFN-γ production by liver NK cells. 6Inducible resistance to Fas-mediated apoptosis in B cells显示文摘Apoptosis produced in B cells through Fas (APO-1, CD95) triggering is regulated by signals derived from other surface receptors: CD40 engagement produces upregulation of Fas expres. sion and marked susceptibility to Fas-induced cell death, whereas antigen receptor engagement, or IL-4R engagement, inhibits Fas killing and in so doing induces a state of Fas-resistance, even in otherwise sensitive, CD40-stimulated targets. Surfaceim. munoglobulin and IL-4R utilize at least partially distinct pathways to produce Fas-resistance that differentially depend on PKC and STAT6, respectively. Further, surface immunoglob- 'lin signaling for inducible Fas-resistance bypasses Bib, requires NF-GB, and entails new macromolecular synthesis. Terminal effectors of B cell Fas-resistance include the known anti-apoptotic gene products, Bcl-xL and FLIP, and a novel anti-apoptotic gene that encodes FAIM (Fas Apoptosis Inhibitory Molecule). faim was identified by differential display and exists in two alternatively spliced forms; maim-S is broadly expressed, but faim-L expression is tissue-specific. The FAIM sequence is highly evolu tionarily conserved, suggesting an important role for this molecule throughout phylogeny. Inducible resistance to Fas killing is hypothesized to protect foreign antigen-specific B cells during potentially hazardous interactions with FasL-bearing T cells, whereas autoreactive B cells fail to become Fas-resistant and are deleted via Fas-dependent cytotoxicity. 7Experimental study of gene therapy with angiostatin gene in pancreatic cancer显示文摘Objective: To study the effects of angiostatin (AS)gene mediated by liposome on human pancreaticcancer cell line SW1990.Methods: Angiostatin gene was cloned into the eu-karyotic expression vector pRC/CMV. The recombi-nant of pRC/CMV-AS was introduced into the pan-creatic cancer cell line, SW1990. The mechanism ofanti-tumor was studied and tested.Results: The eukaryotic expression vector pRC/CMV-AS was identified by the restriction digest.pRC/CMV-AS was stably integrated into the targetcells and expressed by Western blot and drug-sensi-tivity tests, and inhibited the vascular endothelialcells proliferation in vitro. 8Donor liver natural killer cells alleviate liver allograft acute rejection in rats显示文摘BACKGROUND:Liver enriched natural killer (NK) cells are of high immune activity.However,the function of donor liver NK cells in allogeneic liver transplantation (LTx) remains unclear.METHODS:Ten Gy of whole body gamma-irradiation (WBI) from a 60 Co source at 0.6 Gy/min was used for depleting donorderived leukocytes,and transfusion of purified liver NK cells isolated from the same type rat as donor (donor type liver NK cells,dtl NKs) through portal vein was performed immediately after grafting the irradiated liver.Post-transplant survival observation on recipients and histopathological detection of liver grafts were adoptive to evaluate the biological impact of donor liver NK cells on recipients' survival in rat LTx.RESULTS:Transfusion of dtl NKs did not shorten the survival time among the recipients of spontaneous tolerance model (BN to LEW rat) after rat LTx,but prolonged the liver graft survival among the recipients depleted of donor-derived leukocytes in the acute rejection model (LEW to BN rat).Compared to the recipients in the groups which received the graft depleted of donor-derived leukocytes,better survival and less damage in the allografts were also found among the recipients in the two different strain combinations of liver allograft due to transfusion of dtl NKs.CONCLUSIONS:Donor liver NK cells alone do not exacerbate liver allograft acute rejection.Conversely,they can alleviate it,and improve the recipients' survival.Jian-Dong Yu,Tian-Zhu Long,Guo-Lin Li,Li-Hong Lv,Hao-Ming Lin,Yong-Heng Huang,Ya-Jin Chen and Yun-Le Wan Department of Hepatobiliary Surgery,Sun Yat-Sen Memorial Hospital,Sun Yat-Sen University,Guangzhou 510120,China Department of Breast Surgery,Guangzhou Women and Children’s Medical Center,Guangzhou 510120,China 2011Hepatobiliary & Pancreatic Diseases International2011,10,4:2
