| 1 | Helicobacter pylori : Friend or foe?显示文摘Helicobacter pylori(H. pylori) is a Gram-negative spiral bacterium that is present in nearly half the world's population. It is the major cause of peptic ulcer disease and a recognized cause of gastric carcinoma. In addition, it is linked to non-ulcer dyspepsia, vitamin B12 deficiency, iron-deficient anemia and immune thrombocytopenic purpura. These conditions are indications for testing and treatment according to current guidelines. An additional indication according to the guidelines is 'anyone with a fear of gastric cancer' which results in nearly every infected person being eligible for eradication treatment. There may be beneficial effects of H. pylori in humans, including protection from gastroesophageal reflux disease and esophageal adenocarcinoma. In addition, universal treatment will be extremely expensive(more than $32 billion in the United States), may expose the patients to adverse effects such as anaphylaxis and Clostridium difficile infection, as well as contributing to antibiotic resistance. There may also be an as yet uncertain effect on the fecal microbiome. | 2 | 高通量透析膜蛋白质通透性的研究显示文摘目的评估高通量透析膜的蛋白质通透性。方法所有的透析模式均设为单纯超滤,在应用不同的透析膜透析时于透析器的出液口留取滤出液标本,同时进行蛋白质浓度测定和十二烷基硫酸钠-聚丙烯酰氨凝胶电泳(SDS-PAGE)作蛋白质电泳分析。SDS-PAGE采用银染色的方法并根据Melzer的方法作了改良,结果应用激光吸光度测定仪进行条带分析。结果透析膜滤出液中蛋白质条带与经肾小球膜滤出的蛋白质相似,含IgG、转铁蛋白、视黄醇结合蛋白和β2-微球蛋白。由于不同的透析膜的分子截留量、电荷和吸附能力不同,其蛋白质条带的分布亦不同。随着透析的进行,所有膜的通透性都逐渐下降。结论高通量透析膜的蛋白质通透性与肾小球滤过膜相似,但受孔径、表面所带电荷、吸附能力和透析时间的影响而发生改变。与人体的肾小球滤过膜不同,随着透析的进行,透析膜的蛋白质通透性下降。 | 徐筱琪 Siegfried Stiller Helmut Mann 钱家麒 Heinrich Melzer | 2005 | 中华肾脏病杂志2005，21，9： | 6 |
| 3 | 慢性髓性白血病发病机制及药物研究进展显示文摘目的对慢性髓性白血病发病机制、治疗药物、研究中药物进行了综述。方法应用检索Pudmed、CNKI及万方等数据库检索系统,以“慢性髓性白血病、慢性粒细胞白血病和BCR-ABL”等为关键词,检索2000-01—2020-10的相关文献,根据纳入标准和剔除标准进行筛选,符合纳入标准的中文文献75篇,英文文献121篇,最后纳入分析196篇文献。结果TKI作为一线治疗慢性髓性白血病药物,联合阿糖胞苷、或干扰素α、或高三尖杉酯碱等有助于提高细胞遗传学缓解率。长期治疗产生耐药性,以及骨髓抑制、基础疾病患者引起的心肺毒性和肾脏毒性等严重不良反应仍是亟待关注的问题。结论精准诊断、合理用药、开发更具靶向性的小分子药物和筛选基于信号通路进行候选药物是今后努力的方向。 | 郑一敏 刘晶晶 胥秀英 郭牡丹 丁锐 ANDREASE Melzer | 2021 | 重庆理工大学学报(自然科学)2021，35，9： | 4 |
| 4 | PDX1.1-dependent biosynthesis of vitamin B6 protects roots from ammonium-induced oxidative stress显示文摘Despite serving as a major inorganic nitrogen source for plants,ammonium causes toxicity at elevated con-centrations,inhibiting root elongation early on.While previous studies have shown that ammonium-inhibited root development relates to ammonium uptake and formation of reactive oxygen species(ROS)in roots,it remains unclear about the mechanisms underlying the repression of root growth and how plants cope with this inhibitory effect of ammonium.In this study,we demonstrate that ammonium-induced apo-plastic acidification co-localizes with Fe precipitation and hydrogen peroxide(H_(2)O_(2))accumulation along the stele of the elongation and differentiation zone in root tips,indicating Fe-dependent ROS formation.By screening ammonium sensitivity in T-DNA insertion lines of ammonium-responsive genes,we identified PDX1.1,which is upregulated by ammonium in the root stele and whose product catalyzes de novo biosyn-thesis of vitamin B6.Root growth of pdx1.1 mutants is hypersensitive to ammonium,while chemical complementation or overexpression of PDX1.1 restores root elongation.This salvage strategy requires non-phosphorylated forms of vitamin B6 that are able to quench ROS and rescue root growth from ammo-nium inhibition.Collectively,these results suggest that PDX1.1-mediated synthesis of non-phosphorylated B6 vitamers acts as a primary strategy to protect roots from ammonium-dependent ROS formation. | Ying Liu Rodolfo A.Maniero Ricardo F.H.Giehl Michael Melzer Priscille Steensma Gabriel Krouk Teresa B.Fitzpatrick Nicolaus von Wirén | 2022 | Molecular Plant2022，15，5： | 3 |
| 5 | Reduced striatal volumes in Parkinson’s disease:a magnetic resonance imaging study显示文摘Background:The presence and extent of structural changes in the brain as a consequence of Parkinson’s disease(PD)is still poorly understood.Methods:High-resolution 3-tesla T1-weighted structural magnetic resonance images in sixty-five PD and 27 age-matched healthy control participants were examined.Putamen,caudate,and intracranial volumes were manually traced in the axial plane of 3D reconstructed images.Striatal nuclei volumes were normalized to intracranial volume for statistical comparison.Disease status was assessed using the Unified Parkinson’s Disease Rating Scale and Hoehn and Yahr scale.Cognitive status was assessed using global status tests and detailed neuropsychological testing.Results:Both caudate and putamen volumes were smaller in PD brains compared to controls after adjusting for age and gender.Caudate volumes were reduced by 11%(p=0.001)and putamen volumes by 8.1%(p=0.025).PD striatal volumes were not found to be significantly correlated with cognitive or motor decline.Conclusion:Small,but significant reductions in the volume of both the caudate and putamen occur in PD brains.These reductions are independent of the effects of age and gender,however the relation of these reductions to the functional loss of dopamine,which is characteristic of PD,remains unclear. | Toni L Pitcher Tracy R Melzer Michael R MacAskill Charlotte F Graham Leslie Livingston Ross J Keenan Richard Watts John C Dalrymple-Alford Tim J Anderson | 2012 | Translational Neurodegeneration2012，1，1： | 2 |
