| 2 | Two RING-Finger Ubiquitin E3 Ligases Regulate the Degradation of SPX4, An Internal Phosphate Sensor, for Phosphate Homeostasis and Signaling in Rice显示文摘SPX-domain-containing proteins (SPXs) play an important role in inorganic phosphate (Pi) sensing,signaling,and transport in eukaryotes.In plants,SPXs are known to integrate cellular Pi status and negatively regulate the activity of Pi central regulators,the PHOSPATE STARVATION RESPONSE proteins (PHRs).The stability of SPXs,such as SPX4,is reduced under Pi-deficient conditions.However,the mechanisms by which SPXs are degraded remain unclear.In this study,using a yeast-twhybrid screen we iden.tified two RING-finger ubiquitin E3 ligases regulating SPX4 degradation,designated SDEL1 and SDEL2,which were post-transcriptionally induced by Pi starvation.We found that both SDELs were located in the nucleus and cytoplasm,had ubiquitin E3 ligase activity,and directly ubiquitinated the K^213 and K^299 lysine residues in SPX4 to regulate its stability.Furthermore,we found that PHR2,a Pi central regulator in rice,could compete with SDELs by interacting with SPX4 under Pi-sufficient conditions,which protected SPX4 from ubiquitination and degradation.Consistent with the biochemical function of SDEL1 and SDEL2,overexpression of SDEL1 or SDEL2 resulted in Pi overaccumulation and induced Pi-starvation signaling even under Pi-sufficient conditions.Conversely,their loss-of-function mutants displayed decreased Pi accumulation and reduced Pi-starvation signaling.Collectively,our study revealed that SDEL1 and SDEL2 facilitate the degradation of SPX4 to modulate PHR2 activity and regulate Pi homeostasis and Pi signaling in response to external Pi availability in rice. | Wenyuan Ruan Meina Guo Xueqing Wang Zhenhui Guo Zhuang Xu Lei Xu Hongyu Zhao Haiji Sun Chengqi Yan Keke Yi | 2019 | Molecular Plant2019,12,8: | 14 |

| 4 | Traffic Prediction in 3G Mobile Networks Based on Multifractal Exploration显示文摘Traffic prediction plays an integral role in telecommunication network planning and network optimization. In this paper, we investigate the traffic forecasting for data services in 3G mobile networks. Although the Box-Jenkins model has been proven to be appropriate for voice traffic (since the arrival of calls follows a Poisson distribution), it has been demonstrated that the Internet traffic exhibits statistical self-similarity and has to be modeled using the Fractional AutoRegressive Integrated Moving Average (FARIMA) process. However, a few studies have concluded that the FARIMA process may fail in modeling the Internet traffic. To this end, we conducted experiments on the modeling of benchmark Internet traffic and found that the FARIMA process fails because of the significant multifractal characteristic inherent in the traffic series. Thereafter, we investigate the traffic series of data services in a 3G mobile network from a province in China. Rich multifractal spectra are found in this series. Based on this observation, an integrated method combining the AutoRegressive Moving Average (ARMA) and FARIMA processes is applied. | 5 | A reciprocal inhibitory module for Pi and iron signaling显示文摘Phosphorous(P)and iron(Fe),two essential nutrients for plant growth and development,are highly abundant elements in the earth's crust but often display low availability to plants.Due to the ability to form insoluble complexes,the antagonistic interaction between P and Fe nutrition in plants has been noticed for decades.However,the underlying molecular mechanism modulating the signaling and homeostasis between them re-mains obscure.Here,we show that the possible iron sensors HRZs,the iron deficiency-induced E3 ligases,could interact with the central regulator of phosphate(Pi)signaling,PHR2,and prompt its ubiquitination at lysine residues K319 and K328,leading to its degradation in rice.Consistent with this,the hrzs mutants dis-played a high Pi accumulation phenotype.Furthermore,we found that iron deficiency could attenuate Pi star-vation signaling by inducing the expression of HRZs,which in turn trigger PHR2 protein degradation.Inter-estingly,on the other hand,rice PHRs could negatively regulate the expression of HRZs to modulate iron deficiency responses.Therefore,PHR2 and HRZs form a reciprocal inhibitory module to coordinate Pi and iron signaling and homeostasis in rice.Taken together,our results uncover a molecular link between Pi and iron master regulators,which fine-tunes plant adaptation to Pi and iron availability in rice. | Meina Guo Wenyuan Ruan Yibo Zhang Yuxin Zhang Xueqing Wang Zhenhui Guo Long Wang Tian Zhou Javier Paz-Ares Keke Yi | 2022 | Molecular Plant2022,15,1: | 5 |

| 8 | Survival difference between EGFR Del19 and L858R mutant advanced non-small cell lung cancer patients receiving gefitinib:a propensity score matching analysis显示文摘Objective: Although superior clinical benefits of epidermal growth factor receptor(EGFR) tyrosine kinase inhibitors(TKIs) in the treatment of advanced non-small-cell lung cancer(NSCLC) had been reported,the survival difference between exon 19 deletion(Del19) and exon 21 Leu858 Arg substitution(L858 R) remains controversial.The purpose of this study is to investigate the differences in progression-free survival(PFS) and overall survival(OS) between different EGFR mutant subtypes among advanced NSCLC patients receiving gefitinib.Methods: There were 204 advanced NSCLC patients with EGFR mutations treated with gefitinib were enrolled in this retrospective cohort study.Patients were divided into the EGFR Del19 group and the L858 R mutated group according to their mutant subtype.Propensity score matching(PSM) was conducted by using a nearest-neighbor algorithm(1:1) to adjust for demographical and clinical covariates.Survival curves were constructed with the Kaplan-Meier method and compared by using the log-rank test.Results: The PFS in Del19 group was similar to that in the L858 R group [before PSM 8.6 vs.7.2 months,P=0.072; after PSM 7.3 vs.7.2 months,P=0.155].No differences were detected in OS between the L858 R and the Del19 group(before PSM 17.8 vs.13.1 months,P=0.253; after PSM 16.9 vs.13.1 months,P=0.339).The Del19 group was significantly younger compared with the L858 R mutation group in age(P=0.015).Conclusions: No significant difference was found in the PFS or OS between the Del19 and L858 R mutant NSCLC patients receiving gefitinib.The age gap might contribute to the survival differences between Del19 and L858 R groups.PSM is of important value to the elimination of potential bias. | Minglei Zhuo Qiwen Zheng Jun Zhao Meina Wu Tongtong An Yuyan Wang Jianjie Li Shuhang Wang Jia Zhong Xue Yang Hanxiao Chen Bo Jia Zhi Dong Emei Gao JingjingWang Ziping Wang | 2017 | Chinese Journal of Cancer Research2017,29,6: | 4 |

| 10 | AVP(4-8) Improves Cognitive Behaviors and Hippocampal Synaptic Plasticity in the APP/PS1 Mouse Model of Alzheimer’s Disease显示文摘Memory deficits with aging are related to the neurodegeneration in the brain, including a reduction in arginine vasopressin(AVP) in the brain of patients with Alzheimer’s disease(AD). AVP(4-8), different from its precursor AVP, plays memory enhancement roles in the CNS without peripheral side-effects. However, it is not clear whether AVP(4-8) can improve cognitive behaviors and synaptic plasticity in the APP/PS1 mouse model of AD. Here, we investigated for the first time the neuroprotective effects of AVP(4-8) on memory behaviors and in vivo long-term potentiation(LTP) in APP/PS1-AD mice.The results showed that:(1) APP/PS1-AD mice had lower spontaneous alternation in the Y-maze than wild-type(WT) mice, and this was significantly reversed by AVP(4-8);(2) the prolonged escape latency of APP/PS1-AD mice in the Morris water maze was significantly decreased by AVP(4-8), and the decreased swimming time in target quadrant recovered significantly after AVP(4-8) treatment;(3) in vivo hippocampal LTP induced by high-frequency stimulation had a significant deficit in the AD mice, and this was partly rescued by AVP(4-8);(4) AVP(4-8)significantly up-regulated the expression levels of postsynaptic density 95(PSD95) and nerve growth factor(NGF)in the hippocampus of AD mice. | 13 | A phase I study of nimotuzumab plus docetaxel in chemotherapy- refractory/resistant patients with advanced non-small-cell lung cancer显示文摘Background: To determine the safety and therapeutic efficacy of nimotuzumab(h-R3) combined with docetaxel in advanced non-small-cell lung cancer(NSCLC) patients who have failed to respond to prior first-line chemotherapy.Methods: In this single-center, open-label, dose-escalating phase I trial, patients with epidermal growth factor receptor(EGFR)-expressing stage IV NSCLC were treated with nimotuzumab plus docetaxel according to a dose escalation schedule. The safety and efficacy of the combination treatment were observed and analyzed.Results: There were 12 patients with EGFR-expressing stage IV NSCLC enrolled. The dose of nimotuzumab was escalated from 200 to 600 mg/week. The longest administration of study drug was 40 weeks at the 600 mg/week dose level. Grade III–IV toxicities included neutropenia and fatigue, and other toxicities included rash. Dose-limiting toxicity occurred with Grade 3 fatigue at the 200 mg dose level of nimotuzumab and Grade 4 neutropenia with pneumonia at the 600 mg dose level of nimotuzumab. No objective responses were observed, and stable disease was observed in eight patients(66.7%). The median progression-free survival(PFS) was 4.4 months in all patients, 1.3 months in patients with the EGFR mutation, and 4.4 months in those with wild type EGFR(EGFR WT). The median survival time(MST) was 21.1 months in all patients, 21.1 months in patients with EGFR mutation, and 26.4 months in patients with EGFR WT.Conclusions: Nimotuzumab and docetaxel combination therapy was found to be well tolerated and efficacious. | 18 | High Expression of ERCC1 Is a Poor Prognostic Factor in Chinese Patients with Non-small Cell Lung Cancer Receiving Cisplatin-based Therapy显示文摘Objective: to determine the role of excision repair cross-complementing group 1 gene (ERCC1) and ribonucleotide reductase subunit M1 (RRM1) expression in predicting response and survival in Chinese patients with advanced stage non-small cell lung cancer (NSCLC) treated with platinum-based chemotherapy. Methods: Formalin-fixed, paraffin-embedded biopsy tissues were retrospectively obtained from 160 advanced NSCLC patients. mRNA expression levels of ERCC1 and RRM1 were determined by real-time PCR. Results: Associations between mRNA expression level of ERCC1 and RRM1 and clinic-pathologic parameters were analyzed. One handred and forty two (88.75%) specimens were successfully amplified. mRNA expression level of ERCC1 and RRM1 was negatively associated with tumor response. ERCC1 expression levels ranged from 0.01 to 78.79 [median 0.63, mean 4.25 and standard deviation (SD) 9.23] and RRM1 from 0.00 to 30.91 (median 0.63, mean 0.87 and SD 3.36). By adopting cut-off values according to median expression levels, we found that MST in patients with low ERCC1 mRNA levels was significantly longer in overall population than in patients with higher levels (18.63 versus 13.69 months, log-rank 5.73, P=0.017), but we did not found the survival benefit of the patients with low expression level of RRM1. (19.07 versus 14.88, log-rank 1.66, P=0.197). Further, in the multivariable Cox regression model analysis we found that low level of ERCC1 expression, the presence of weight loss and target therapy, were significant prognostic factors for survival.
