| 1 | Epalrestat improves motor symptoms by reducing oxidative stress and inflammation in the reserpine induced mouse model of Parkinson’s disease显示文摘Background:Parkinson's disease(PD)is a progressive neurodegenerative disorder affecting a large number of elderly people worldwide.The current therapies for PD are symptom-based;they do not provide a cure but improve the quality of life.Muscular dysfunction is the hallmark clinical feature of PD and oxidative stress and inflammation play a critical role in its pathogenesis.Epalrestat is used for the treatment of diabetic neuropathy and is known to improve antioxidative defense mechanisms in the CNS.Therefore,in this study,we investigated the role of Epalrestat in the reserpine induced mouse model of PD.Method:We used Swiss Albino mice for the PD model and tested for akinesia/bradykinesia,muscular rigidity,palpebral ptosis,and tremor,as well as conducting swim and open field tests.Brain samples were used to determine oxidative stress parameters and infiltration of immune cells.Results:Epalrestat treatment significantly improved akinesia and bradykinesia,muscular dysfunctions,tremor level,and gait functions compared to the reserpine group.It also improved the latency in the swim test.Eplarestat significantly reduced lipid peroxidation and NO concentration in different brain tissues and increased the activity of antioxidative enzymes,glutathione,catalase,and superoxide dismutase.Furthermore,Epalrestat reduced neuroinflammation by reducing the number of infiltrating immune cells.Conclusion:Eplarestat improves muscular dysfunction in PD by reducing oxidative stress and inflammation. | Md.Mahbubur Rahman Rupali Rani Chakraborti Md.Abdullah Potol Ariful Haque Abir Ozayra Sharmin Mahabub Alam Md.Fazlur Rahman Khan Rownock Afrin Humayra Jannat Rasiqh Wadud Zaki Farhad Habib | 2020 | Animal Models and Experimental Medicine2020,3,1: | 3 |
| 2 | Hepatoprotective effects of Nigella sativa seed extract against acetaminophen-induced oxidative stress显示文摘Objective:To investigate the protective effects of Nigella sativa seed extract(NSSE) against acetaminophen(APAP)-induced hepaloloxicity in TIB-73 cells and rats.Methods:Toxicity in TIB-73 cells was induced with 10 μmol/L APAP and the protective effects of NSSE were evaluated at 25.50.75,100 μg/mL.For in rim examination,a total of 30 rals were equally divided into five experimental groups:normal control(vehicle),APAP(800 mg/kg body weight single IP injection) as a hepatotoxic control,and three APAP and NS pretreated(2 weeks) groups(APAP+NSSE 100 mg:APAP+NSSE 300 mg and APAP+NSSK 900 mg/kg).Results:TIB-73 cell viability was drastically decreased by(49.0±l.9)%after the 10 μmol/L APAP treatment,which also increased reactive oxygen species production.Co-treatment with NSSE at 25.50.75,and 100 μg/mL significantly improved cell viability and suppressed reactive oxygen species generation.In viro the APAP induced alterations in blood lactate levels,pH,anionic gap,and ion levels(HCO_3^-,Mg^(2+) and K^+),which tended to normalize with the NSSE pretreatment.The NSSE also significantly decreased elevated serum levels of alanine aminotransferase,aspartate aminotransferase,lactate dehydrogenase,and alkaline phosphatase induced by APAP,which correlated with decreased levels of hepatic lipid peroxidation(nialondialdehyde),increased superoxide dismutase levels,and reduced glutathione concentrations.Improved hepatic histology was also found in the treatment groups other than APAP group.Conclusions:The in vitro and in vim findings of this study demonstrated that the NSSE has protective effects against APAP-induced hepalotoxicity and metabolic disturbances by improving antioxidant activities and suppressing both lipid peroxidation and ROS generation. | Gareeballa Osman Adam Md.Mahbubur Rahman Sei-Jin Lee Gi-Beum Kim Hyung-Sub Kang Jin-Shang Kim Shang-Jin Kim | 2016 | Asian Pacific Journal of Tropical Medicine2016,9,3: | 2 |
| 3 | Aeromonas veronii isolated from climbing perch(Anabas testudineus)suffering from epizootic ulcerative syndrome(EUS)显示文摘Motile Aeromonads are responsible for various diseases in fishes that cause significant economic losses in aquaculture.Aeromonas spp.are also frequently isolated from fish suffering from Epizootic Ulcerative Syndrome(EUS).The present study was conducted to isolate and identify A.veronii from Vietnam climbing perch(Anabas testudineus)suffering from EUS,evaluate the pathogenicity of the isolates,and find out antibiogram profile and susceptibility of the isolates to medicinal plant extracts.Vietnam climbing perch(A.testudineus)suffering from EUS was collected from fish farms located at Savar,Dhaka and bacteria were isolated from the external ulcerative lesion,and kidney and liver of the infected fish on nutrient agar plates.Preliminary phenotypic identification of the bacterial isolates was performed following morphological,physiological and biochemical characterization tests.The 16S rRNA gene sequencing was accomplished for further molecular identification of five selected isolates.To evaluate the pathogenicity of the isolates in vivo artificial infection challenge test was conducted through immersion method.Another in vivo pathogenicity test was done at three different temperatures(18◦C,28◦C,and 37◦C)to examine the effects of temperature on the pathogenicity of the isolates.Ten commercial antibiotics were used to find out the antibiogram profile of the isolates.Finally,susceptibility of the isolates was screened against 19 medicinal plants extracts.A total of 38 out of 46 isolates isolated from Vietnam climbing perch(A.testudineus)suffering from EUS were phenotypically identified as Aeromonas sp.Five randomly selected isolates were further confirmed as A.veronii by analyzing the 16S rRNA gene sequence homology.The pathogenicity of the isolates was found moderate to high(41%-80%mortality)in the artificially challenged fish.The highest and lowest mortality was obtained in the fish challenged with the isolates BFKA33 and BFKA12,respectively.The isolates were found resistant to several antibiotics,including amoxicillin,ampicillin,penicillin-G,and vancomycin.Nevertheless,extracts of three medicinal plant(Allium sativum,Syzygium aromaticum,and Tamarindus indica)demonstrated antibacterial activity to the isolates(BFKA12,BFKA15,BFKA18,BFKA29,and BFKA33). | Rakib Ehsan Ashikur Rahman Sulav Indra Paul Md.Ali Arman Ador Md.Shameul Haque Tasmina Akter Md.Mahbubur Rahman | 2023 | Aquaculture and Fisheries2023,8,3: | 0 |
