Rolling Bearing Condition Monitoring Technique Based on Cage Rotation Analysis and Acoustic Emission显示文摘In this paper,we present an alternative technique for detecting changes in the operating conditions of rolling element bearings(REBs)that can lead to premature failure. The developed technique is based on measuring the kinematics of the bearing cage. The rotational motion of the cage is driven by traction forces generated in the contacts of the rolling elements with the races. It is known that the cage angular frequency relative to shaft angular frequency depends on the bearing load, the bearing speed, and the lubrication condition since these factors determine the lubricant film thickness and the associated traction forces.Since a large percentage of REB failures are due to misalignment or lubrication problems, any evidence of these conditions should be interpreted as an incipient fault. In this paper,a novel method for the measurement of the instantaneous angular speed(IAS)of the cage is developed. The method is evaluated in a deep groove ball bearing test rig equipped with a cage IAS sensor,as well as a custom acoustic emission(AE)transducer and a piezoelectric accelerometer. The IAS of the cage is analyzed under different bearing loads and shaft speeds, showing the dependence of the cage angular speed with the calculated lubricant film thickness.Typical bearing faulty operating conditions(mixed lubrication regime, lubricant depletion, and misalignment)are recreated. It is shown that the cage IAS is dependent on the lubrication regime and is sensitive to misalignment. The AE signal is also used to evaluate the lubrication regime.Experimental results suggest that the proposed technique can be used as a condition monitoring tool in industrial environments to detect abnormal REB conditions that may lead to premature failure. | Matías Marticorena Oscar García Peyrano | 2022 | Journal of Dynamics, Monitoring and Diagnostics2022,1,2: | 0

Online Unbalance Detection and Diagnosis on Large Flexible Rotors by SVR and ANN trained by Dynamic Multibody Simulations显示文摘Multiple-stage steam turbine generators, like those found in nuclear power plants, pose special challenges with regards to mechanical unbalance diagnosis.Several factors contribute to a complex vibrational response, which can lead to incorrect assessments if traditional condition monitoring strategies are used without considering the mechanical system as a whole. This,in turn, can lead to prolonged machinery downtime.Several machine learning techniques can be used to integrally correlate mechanical unbalance along the shaft with transducer signals from rotor bearings.Unfortunately, this type of machinery has scarce data regarding faulty behavior. However,a variety of fault conditions can be simulated in order to generate these data using computational models to simulate the dynamic response of individual machines. In the present work,a multibody model of a 640MWsteam turbine flexible rotor is employed to simulate mechanical unbalance in several positions along the shaft.Synchronous components of the resulting vibration signals at each bearing are obtained and utilized as training data for two regression models designed for mechanical unbalance diagnosis. The first approach uses an artificial neural network and the second one utilizes a support vector regression algorithm. In order to test their performance, the stiffness of each bearing in the multibody simulation was altered between 50%and 150%of the training model values, random noise was added to the signal and several dynamic unbalance conditions were simulated. Results show that both approaches can reliably diagnose dynamic rotor unbalance even when there is a typical degree of uncertainty in bearing stiffness values. | Oscar García Peyrano Juan Vignolo Rodrigo Mayer Matías Marticorena | 2022 | Journal of Dynamics, Monitoring and Diagnostics2022,1,3: | 0

Drug repurposing for COVID-19 using machine learning and mechanistic models of signal transduction circuits related to SARS-CoV-2 infection显示文摘Dear Editor,Drug repurposing is a convenient alternative when the need for new drugs in an unexpected medical scenario is urgent,as is the case of emerging pathogens. In recent years, approaches based on network biology have demonstrated to be superior to genecentric ones.1 Here,we use an innovative methodology that combines mechanistic modeling of the signal transduction circuits related to SARS-CoV-2 infection(the COVID-19 disease map)with a machine-learning algorithm that learns potential causal interactions between proteins, already targets of drugs, and specific signaling circuits in the COVID-19 disease map,to suggest potentially repurposable drugs. | Carlos Loucera Marina Esteban-Medina Kinza Rian Matías M.Falco Joaquín Dopazo María Peña-Chilet | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 1

Involvement of parathyroid hormone-related peptide in the aggressive phenotype of colorectal cancer cells显示文摘Colorectal cancer(CRC)remains one of the leading causes of mortality from malignant diseases worldwide. In general terms,CRC presents high heterogeneity due to the influence of different genetic and environmental factors;also, the neoplastic cells are strongly influenced by the extracellular matrix and several surrounding cells, known together as the tumor microenvironment(TME).Bidirectional communication takes place between the tumor and the TME through the release of autocrine and paracrine factors.Parathyroid hormone-related peptide(PTHrP)is a cytokine secreted by a wide variety of tissues and is able to regulate several cellular functions both in physiological as well as in pathological processes. It exerts its effects as a paracrine/autocrine factor, although its mode of action is mainly paracrine. It has been shown that this peptide is expressed by several tumors and that the tumor secretion of PTHrP is responsible for the malignant humoral hypercalcemia.Eight years ago, when our research group started studying PTHrP effects in the experimental models derived from intestinal tumors, the literature available at the time addressing the effects of PTHrP on colorectal tumors was limited, and no articles had been published regarding to the paracrine action of PTHrP in CRC cells.Based on this and on our previous findings regarding the role of PTH in CRC cells, our purpose in recent years has been to explore the role of PTHrP in CRC.We analyzed the behavior of CRC cells treated with exogenous PTHrP,focalizing in the study of the following events:Survival, cell cycle progression and proliferation,migration,chemoresistance,tumor-associated angiogenesis, epithelial to mesenchymal transition program and other events also associated with invasion, such us the induction of cancer stem cells features. This work summarizes the major findings obtained by our investigation group using in vitro and in vivo CRC models that evidence the participation of PTHrP in the acquisition of an aggressive phenotype of CRC cells and the molecular mechanisms involved in these processes.Recently,we found that this cytokine induces this malignant behavior not only by its direct action on these intestinal cells but also through its influence on cells derived from TME,promoting a communication between CRC cells and surrounding cells that contributes to the molecular and morphological changes observed in CRC cells. These investigations establish the basis for our next studies in order to address the clinical applicability of our findings.Recognizing the factors and mechanisms that promote invasion in CRC cells, evasion to the cytotoxic effects of current CRC therapies and thus metastasis is decisive for the identification of new markers with the potential to improve early diagnosis and/or to predict prognosis,to predetermine drug resistance and to provide treatment guidelines that include targeted therapies for this disease. | María Belén Novoa Díaz Pedro Matías Carriere María Julia Martín Natalia Calvo Claudia Gentili | 2021 | World Journal of Gastroenterology2021,27,41: | 0
Mitochondrial protection by low doses of insulin-like growth factor-Ⅰin experimental cirrhosis显示文摘AIM: To characterize the mitochondrial dysfunction in experimental cirrhosis and to study whether insulin-like growth factor-Ⅰ(IGF-Ⅰ) therapy (4 wk) is able to induce beneficial effects on damaged mitochondria leading to cellular protection. METHODS: Wistar rats were divided into three groups: Control group, untreated cirrhotic rats and cirrhotic rats treated with IGF-Ⅰtreatment (2 μg/100 g bw/d). Mitochondrial function was analyzed by flow cytometry in isolated hepatic mitochondria, caspase 3 activation was assessed by Western blot and apoptosis by TUNEL in the three experimental groups. RESULTS: Untreated cirrhotic rats showed a mitochon- drial dysfunction characterized by a significant reduction of mitochondrial membrane potential (in status 4 and 3); an increase of intramitochondrial reactive oxigen species (ROS) generation and a significant reduction of ATPase activity. IGF-Ⅰtherapy normalized mitochondrial func-tion by increasing the membrane potential and ATPase activity and reducing the intramitochondrial free radical production. Activity of the electron transport complexes Ⅰand Ⅲ was increased in both cirrhotic groups. In addition, untreated cirrhotic rats showed an increase of caspase 3 activation and apoptosis. IGF-Ⅰtherapy reduced the expression of the active peptide of caspase 3 and resulted in reduced apoptosis. | Raquel Pérez María García-Fernández Matías Díaz-Sánchez Juan E Puche Gloria Delgado Marian Conchillo Jordi Muntané Inma Castilla-Cortázar | 2008 | World Journal of Gastroenterology2008,14,17: | 11
