1Age and sex specific timing, frequency, and spatial distribution of horseshoe crab spawning in Delaware Bay:Insights from a large-scale radio telemetry array显示文摘To study horseshoe crab Limulus polyphemus spawning behavior and migration over a large-spatial extent (>100 km),we arrayed fixed station radio receivers throughout Delaware Bay and deployed radio transmitters and archival tags on adulthorseshoe crabs prior to their spawning season.We tagged and released 160 females and 60 males in 2004 and 217 females in2005.The array covered approximately 140 km of shoreline.Recapture rates were >70% with multi-year recaptures.We categorizedadult age by carapace wear.Older females tended to spawn earlier in the season and more frequently than young females,but those tendencies were more apparent in 2004 when spawning overall occurred earlier than in 2005 when spawning was delayedpossibly due to decreased water temperatures.Timing of initial spawning within a year was correlated with water temperature.After adjusting for day of first spring tide,the day of first spawning was 4 days earlier for every 1 degree (℃) rise in meandaily water temperature in May.Seventy nine % of spawning occurred during nighttime high tides.Fifty five % of spawning occurredwithin 3 d of a spring tide,which was slightly higher than the 47% expected if spawning was uniformly distributed regardlessof tidal cycle.Within the same spawning season,males and females were observed spawning or intertidally resting atmore than one beach separated by >5 km.Between years,most (77%) did not return to spawn at the same beach.Probability ofstranding was strongly age dependent for males and females with older adults experiencing higher stranding rates.Horseshoecrabs staging in the shallow waters east of the channel spawned exclusively along the eastern (NJ) shoreline,but those stagingwest of the channel spawned throughout the bay.Overall,several insights emerged from the use of radio telemetry,which advancesour understanding of horseshoe crab ecology and will be useful in conserving the Delaware Bay horseshoe crab populationandDavid R.SMITH Lorne J.BROUSSEAU Mary T. 11Fabrication of biomimetic placental barrier structures within a microfluidic device utilizing two-photon polymerization显示文摘The placenta is a transient organ,essential for development and survival of the unborn fetus.It interfaces the body of the pregnant woman with the unborn child and secures transport of endogenous and exogenous substances.Maternal and fetal blood are thereby separated at any time,by the so-called placental barrier.Current in vitro approaches fail to model this multifaceted structure,therefore research in the field of placental biology is particularly challenging.The present study aimed at establishing a novel model,simulating placental transport and its implications on development,in a versatile but reproducible way.The basal membrane was replicated using a gelatin-based material,closely mimicking the composition and properties of the natural extracellular matrix.The microstructure was produced by using a high-resolution 3D printing method– the two-photon polymerization(2PP).In order to structure gelatin by 2PP,its primary amines and carboxylic acids are modified with methacrylamides and methacrylates(GelMOD-AEMA),respectively.High-resolution structures in the range of a few micrometers were produced within the intersection of a customized microfluidic device,separating the x-shaped chamber into two isolated cell culture compartments.Human umbilical-vein endothelial cells(HUVEC)seeded on one side of this membrane simulate the fetal compartment while human choriocarcinoma cells,isolated from placental tissue(BeWo B30)mimic the maternal syncytium.This barrier model in combination with native flow profiles can be used to mimic the microenvironment of the placenta,investigating different pharmaceutical,clinical and biological scenarios.As proof-of-principle,this bioengineered placental barrier was used for the investigation of transcellular transport processes.While high molecular weight substances did not permeate,smaller molecules in the size of glucose were able to diffuse through the barrier in a time-depended manner.We envision to apply this bioengineered placental barrier for pathophysiological research,where altered nutrient transport is associated with health risks for the fetus.Denise Mandt Peter Gruber Marica Markovic Maximillian Tromayer Mario Rothbauer Sebastian Rudi Adam Kratz Syed Faheem Ali Jasper Van Hoorick Wolfgang Holnthoner Severin Mühleder Peter Dubruel Sandra Van Vlierberghe Peter Ertl Robert Liska Aleksandr Ovsianikov 2018International Journal of Bioprinting2018,4,2:1
