2Formation control and collision avoidance for multi-UAV systems based on Voronoi partition显示文摘This paper deals with the formation control problem of multiple unmanned aerial vehicles(UAVs) with collision avoidance. A distributed formation control and collision avoidance method is proposed based on Voronoi partition and conventional artificial potential field. The collision avoidance is achieved by partitioning the whole space into non-overlapping regions based on Voronoi partition theory, which is taken as the task region to confine the movement of each UAV. The general motion control law is designed based on the conventional artificial potential field. As this often leads to local optimum when two UAVs are going to collide with each other and they may stay still where the repulsive force is adversely equivalent to the attractive force. To address this problem,the destination switch scheme is further proposed to let UAVs switch destinations when they reach the local equilibrium. 3Effects of recombinant human canstatin protein in the treatment of pancreatic cancer显示文摘AIM: To examine the effect of canstatin, a newly discovered endogenous inhibitor of angiogenesis, in the treatment of pancreatic cancer in vivo. METHODS: The canstatin cDNA fragment was synthesized and amplified from the total RNA extracted from human placenta tissues by RT-PCR. The resulting product was firstly cloned into pUCm-T vector, then into plasmid pET-22b (+) and transformed into E. coli BL21. Isopropyl-1-thio-b-Dgalactopyran-oside (IPTG) was used to induce the expression of canstatin protein and affinity chromatography was used to purify the protein. To determine the activity of purified recombinant human canstatin (rhCanstatin), orthotopic xenograft human pancreatic cancer models were established. Human pancreatic cancer cells (SW1990) were injected into the pancreas of BALB/c nude mice. Twenty-four nude mice with orthotopic xenograft tumor were randomly divided into 3 groups 10 d after the inoculation, and were treated with PBS 0.3 mL, or canstatin 5 mg/kg, or 10 mg/kg per day for 3 wk intraperitoneally. When the experiment was over, all tumors were resected and the effects of rhCanstatin on tumor growth, microvessel density (MVD) were analyzed. RESULTS: After IPTG induction, SDS-PAGE showed a new monomeric 24 kDa protein band. This protein was purified through affinity chromatography and refolded through dialysis with a final concentration of 60 mg/L. In orthotopic pancreatic cancer models, the final tumor volume in groups treated with PBS, canstatin 5 mg/ kg, 10 mg/kg were 355.21 ± 39.54 mm3, 112.73 ±10.47 mm3, and 61.75 ± 6.99 mm3 respectively. The immunohistochemical examination showed that the MVD in tumors treated with canstatin was significantly less than that in other group. 4Effects of elevated atmospheric CO_2 concentration and temperature on the soil profile methane distribution and diffusion in rice–wheat rotation system显示文摘The aim of this experiment was to determine the impacts of climate change on soil profile concentrations and diffusion effluxes of methane in a rice–wheat annual rotation ecosystem in Southeastern China. We initiated a field experiment with four treatments:ambient conditions(CKs), CO_2 concentration elevated to ~ 500 μmol/mol(FACE),temperature elevated by ca. 2°C(T) and combined elevation of CO_2 concentration and temperature(FACE + T). A multilevel sampling probe was designed to collect the soil gas at four different depths, namely, 7 cm, 15 cm, 30 cm and 50 cm. Methane concentrations were higher during the rice season and decreased with depth, while lower during the wheat season and increased with depth. Compared to CK, mean methane concentration was increased by 42%, 57% and 71% under the FACE, FACE + T and T treatments, respectively, at the 7 cm depth during the rice season(p < 0.05). Mean methane diffusion effluxes to the 7 cm depth were positive in the rice season and negative in the wheat season, resulting in the paddy field being a source and weak sink, respectively. Moreover, mean methane diffusion effluxes in the rice season were 0.94, 1.19 and 1.42 mg C/(m^2·hr) in the FACE,FACE + T and T treatments, respectively, being clearly higher than that in the CK. 5Hypoxia-mimicking 3D bioglass-nanoclay scaffolds promote endogenous bone regeneration显示文摘Large bone defect repair requires biomaterials that promote angiogenesis and osteogenesis.In present work,a nanoclay(Laponite,XLS)-functionalized 3D bioglass(BG)scaffold with hypoxia mimicking property was prepared by foam replication coupled with UV photopolymerization methods.Our data revealed that the incorporation of XLS can significantly promote the mechanical property of the scaffold and the osteogenic differentiation of human adipose mesenchymal stem cells(ADSCs)compared to the properties of the neat BG scaffold.Desferoxamine,a hypoxia mimicking agent,encourages bone regeneration via activating hypoxia-inducible factor-1 alpha(HIF-1α)-mediated angiogenesis.GelMA-DFO immobilization onto BG-XLS scaffold achieved sustained DFO release and inhibited DFO degradation.Furthermore,in vitro data demonstrated increased HIF-1αand vascular endothelial growth factor(VEGF)expressions on human adipose mesenchymal stem cells(ADSCs).Moreover,BG-XLS/GelMA-DFO scaffolds also significantly promoted the osteogenic differentiation of ADSCs.Most importantly,our in vivo data indicated BG-XLS/GelMA-DFO scaffolds strongly increased bone healing in a critical-sized mouse cranial bone defect model.Therefore,we developed a novel BG-XLS/GelMA-DFO scaffold which can not only induce the expression of VEGF,but also promote osteogenic differentiation of ADSCs to promote endogenous bone regeneration.Xiao Zheng Xiaorong Zhang Yingting Wang Yangxi Liu Yining Pan Yijia Li Man Ji Xueqin Zhao Shengbin Huang Qingqing Yao 2021Bioactive Materials2021,6,10:4
6Risk factors for de novo hepatitis B infection in pediatric living donor liver transplantation显示文摘AIM: To investigate the incidence of de novo hepatitis B virus(HBV) infection after pediatric living donor liver transplantation(LDLT) and to analyze the risk factors associated with this de novo HBV infection. METHODS: The clinical and laboratory data of children who underwent LDLT from June 2010 to September 2012 in First Center Hospital in Tianjin, China, were retrospectively included in the study. Intrahepatic HBV DNA in donors and recipients was quantified by realtime polymerase chain reaction using DNA extracted from formalin-fixed, paraffin-embedded tissues. RESULTS: Between June 2010 to September 2012, 32 consecutive pediatric patients underwent LDLT in our institute. Thirty LDLT patients(13 girls and 17 boys) were followed up for a median of 15 mo, of whom 53.3%(16/30) were hepatitis B core antibody(HBcAb) positive and 36.7%(11/30) were hepatitis B surface antibody(HBsAb)/HBcAb positive before transplantation. Sixteen of the children received HBcAb-positive allografts, and 43.7%(7/16) of the grafts were found to be intrahepatic HBV DNA positive. De novo HBV infection developed in 16.1%(5/30) of the children within a median of 11 mo after transplantation. All five of the HBV-infected children had received HBcAb-positive allografts, four of which were intrahepatic HBV DNA positive. Two of the children developed de novo HBV infection despite the preoperative presence of both HBsAb and HBcAb CONCLUSION: In pediatric recipients, positive intrahepatic HBV DNA in allografts could be a risk factor for de novo HBV infection from HBcAb-positive allografts. 8Application of phosphate-containing materials affects bioavailability of rare earth elements and bacterial community in soils显示文摘The exploitation and smelting of rare earths can cause serious pollution to the farmland around the mining area. The rare earth elements(REEs) are absorbed by crops and enter the human body through the food chain, which threatens people’s health. The effects of four phosphorus-containing materials-calcium superphosphate(SSP), phosphate rock(PR), calcium magnesium phosphate(CMP) and bone charcoal(BC) on rice growth and bacterial community structure in REE mining area of Xinfeng County were studied by pot experiment. The soil solution was collected during rice transplanting and harvest periods respectively, the rice and soil samples were collected and sequenced. The concentrations of water-soluble REEs were measured by inductively coupled plasma mass spectrometry(ICP-MS), and bacteria in soil was deeply sequenced by the Illumina Miseq sequencing platform. PR, CMP and BC promoted the growth of rice, improved the biomass of rice roots, shoots and grains, and significantly reduced absorption and accumulation of REEs in rice roots, shoots and grains. SSP treatment reduced the pH value of soil, significantly improved the concentration of REE solution in soil and improved biomass of rice roots, shoots and grains,and significantly improved the concentration of REEs in grain. The effects of phosphorus-containing materials on the absorption and accumulation of 15 REEs in rice roots, shoots and grains were very different, and significantly influenced the soil bacterial community. SSP reduced richness and diversity of bacteria. CMP improved the diversity of soil bacteria, but reduced their richness. PR and BC treatment improved the richness and diversity of soil bacteria, and significantly increased the abundance of Bacillus. 9Clinical characteristics and clinicopathological correlations of bilateral breast cancer in China:A multicenter study from Chinese Society of Breast Surgery(CSBrS-006)显示文摘Objective:To investigate the clinical characteristics and clinicopathological correlations of bilateral breast cancer(BBC)in China.Methods:Data of 440 patients diagnosed with BBC in 2018 were collected from 33 centers of the Chinese Society of Breast Surgery.Demographic characteristics,bilateral tumor characteristics,and comprehensive treatment data were obtained.Correlations between the clinicopathological characteristics of bilateral tumors were analyzed.Results:The proportion of BBC was 0.22%-3.08%.A total of 33(7.5%)patients had a family history of malignant tumors,304(69.1%)patients had synchronous BBC.Only 1(0.2%)patient was male.More than half of all patients received concurrent or asynchronous endocrine/chemotherapy,32.5%of all human epidermal growth factor receptor 2(HER2)-positive patients received HER2-targeted therapy,and approximately 21.6%of all patients received radiotherapy.The most common pathological cancer type was invasive ductal cancer(>60%).Approximately 70%of all patients had bilateral hormone receptor(HR)-positive tumors and presented with a single breast mass.Significant correlations were found with pathological type,histological grade,locations of tumor,molecular subtype,Ki-67 index,tumor site and size of bilateral tumors.Results of the subgroup analysis showed more clinicopathological characteristics when synchronous BBC was compared with metachronous BBC.Conclusions:In China,the clinicopathological characteristics of bilateral tumors showed significant correlations,and more significant clinicopathological correlations were observed when synchronous BBC was compared with metachronous BBC.Xuelu Li Yitong Wang Bo Pan Man Li Jiyue Gao Yi Zhao Zuowei Zhao Chinese Society of Breast Surgery 2021Chinese Journal of Cancer Research2021,33,1:2
10Prediction of tumor biological characteristics in different colorectal cancer liver metastasis animal models using^(18)F-FDG and^(18)F-FLT显示文摘Background: Positron emission tomography(PET) is a noninvasive method to characterize different metabolic activities of tumors, providing information for staging, prognosis, and therapeutic response of patients with cancer. The aim of this study was to evaluate the feasibility of^(18)F-fludeoxyglucose(^(18)F-FDG) and 3'-deoxy-3'-^(18)F-fluorothymidine(^(18)F-FLT) PET in predicting tumor biological characteristics of colorectal cancer liver metastasis.Methods: The uptake rate of^(18)F-FDG and^(18)F-FLT in SW480 and SW620 cells was measured via an in vitro cell uptake assay. The region of interest was drawn over the tumor and liver to calculate the maximum standardized uptake value ratio(tumor/liver) from PET images in liver metastasis model. The correlation between tracer uptake in liver metastases and VEGF, Ki67 and CD44 expression was evaluated by linear regression.Results: Compared to SW620 tumor-bearing mice, SW480 tumor-bearing mice presented a higher rate of liver metastases. The uptake rate of^(18)F-FDG in SW480 and SW620 cells was 6.07% ± 1.19% and2.82% ± 0.15%, respectively(t = 4.69, P = 0.04); that of^(18)F-FLT was 24.81% ± 0.45% and 15.57% ± 0.66%, respectively(t = 19.99, P < 0.001). Micro-PET scan showed that all parameters of FLT were significantly higher in SW480 tumors than those in SW620 tumors. A moderate relationship was detected between metastases in the liver and^(18)F-FLT uptake in primary tumors(r = 0.73, P = 0.0019).^(18)F-FLT uptake was also positively correlated with the expression of CD44 in liver metastases(r = 0.81, P = 0.0049).Conclusions: The uptake of^(18)F-FLT in metastatic tumor reflects different biological behaviors of colon cancer cells.^(18)F-FLT can be used to evaluate the metastatic potential of colorectal cancer in nude mice.Hai-Long Xu Man Li Rong-Jun Zhang Hui-Jie Jiang Ming-Yu Zhang Xin Li Yi-Qiao Wang Wen-Bin Pan 2018Hepatobiliary & Pancreatic Diseases International2018,17,2:2
11Rnf20 deficiency in adipocyte impairs adipose tissue development and thermogenesis显示文摘RNF20,an E3 ligase critical for monoubiquitination of histone H2B at lysine 120(H2Bub),has been implicated in the regulation of various cellar processes;however,its physiological roles in adipocytes remain poorly characterized.Here,we report that the adipocyte-specific knockout of Rnf20(ASKO)in mice led to progressive fat toss,organomegaly and hyperinsulinemia.Despite signs of hyperinsulinemia,normal insulin sensitivity and improved glucose tolerance were observed in the young and aged CD-fed ASKO mice.In addition,high-fat diet-fed ASKO mice developed severe liver steatosis.Moreover,we observed that the ASKO mice were extremely sensitive to a cold environment due to decreased expression levels of brown adipose tissue(BAT)selective genes,including uncoupling protein 1(l/cpf),and impaired mitochondrial functions.Significantly decreased levels of peroxisome proliterator-activated receptor gamma(Ppary)were observed in the gonadal white adipose tissues(gWAT)from the ASKO mice,suggesting that Rnf20 regulates adipogenesis,at least in part,through Ppary.Rosiglitazone-treated ASKO mice exhibited increased fat mass compared to that of the non-treated ASKO mice.Collectively,our results illustrate the critical role of RNF20 in control of white and brown adipose tissue development and physiological function.Xiaojuan Liang Cong Tao Jianfei Pan Lilan Zhang Lulu Liu Ying Zhao Yiping Fan Chunwei Cao Jiali Liu Jin Zhang Sin Man Lam Guanghou Shui Wanzhu Jin Wei Li Jianguo Zhao Kui Li Yanfang Wang 2021Protein & Cell2021,12,6:1

20Genome assembly and resequencing analyses provide new insights into the evolution, domestication and ornamental traits of crape myrtle显示文摘Crape myrtle(Lagerstroemia indica)is a globally used ornamental woody plant and is the representative species of Lagerstroemia.However,studies on the evolution and genomic breeding of L.indica have been hindered by the lack of a reference genome.Here we assembled the first high-quality genome of L.indica using PacBio combined with Hi-C scaffolding to anchor the 329.14-Mb genome assembly into 24 pseudochromosomes.We detected a previously undescribed independent whole-genome triplication event occurring 35.5 million years ago in L.indica following its divergence from Punica granatum.After resequencing 73 accessions of Lagerstroemia,the main parents of modern crape myrtle cultivars were found to be L.indica and L.fauriei.During the process of domestication,genetic diversity tended to decrease in many plants,but this was not observed in L.indica.We constructed a high-density genetic linkage map with an average map distance of 0.33 cM.Furthermore,we integrated the results of quantitative trait locus(QTL)using genetic mapping and bulk segregant analysis(BSA),revealing that the major-effect interval controlling internode length(IL)is located on chr1,which contains CDL15,CRG98,and GID1b1 associated with the phytohormone pathways.Analysis of gene expression of the red,purple,and white flower-colour flavonoid pathways revealed that differential expression of multiple genes determined the flower colour of L.indica,with white flowers having the lowest gene expression.In addition,BSA of purple-and green-leaved individuals of populations of L.indica was performed,and the leaf colour loci were mapped to chr12 and chr17.Within these intervals,we identified MYB35,NCED,and KAS1.Our genome assembly provided a foundation for investigating the evolution,population structure,and differentiation of Myrtaceae species and accelerating the molecular breeding of L.indica.Yang Zhou Tangchun Zheng Ming Cai Lu Feng Xiufeng Chi Ping Shen Xin Wang Zhiting Wan Cunquan Yuan Man Zhang Yu Han Jia Wang Huitang Pan Tangren Cheng Qixiang Zhang 2023Horticulture Research2023,10,9:1
