2Antioxidative and anti-endotoxin effects of propofol on endothelial cells显示文摘Objective To investigate the antioxidant and anti-endotoxin effects of propofol on endothelial cells and the possible mechanisms.Methods Cultured endothelial cells were treated with hydrogen peroxide (H 2O 2), propofol + H 2O 2, lipopolysaccharide (LPS) and propofol + LPS, respectively. Endothelial cell damage was monitored for possible lactic dehydrogenase (LDH) release. The transcription and the protein expression levels of endothelial nitric oxide synthase (eNOS) were measured.Results LDH release was higher in groups treated with H 2O 2 or LPS than in the control group. After pretreatment with propofol, the effects induced by H 2O 2 were attenuated, but propofol did not decrease the LDH release induced by LPS. Both H 2O 2 and LPS significantly increased the eNOS transcript levels and the increases were significantly attenuated after pretreatment with propofol. Both H 2O 2 and LPS significantly increased the eNOS protein expression and the increase was attenuated after pretreatment with propofol. 5Antimicrobial Agents from Selected Medicinal Plants in Libya显示文摘Objective:To test the in vitro antimicrobial efficacy of water and methanol extracts of 23 plan species that are commonly used in Libyan folk medicine.Methods:The antimicrobial activity was determinec using the well-diffusion method.Four test microorganisms were used namely,Escherichia coli,Salmonella species,Staphylococcus aureus and Bacillus subtilis.The minimum inhibitory concentration(MIC) was determined for the high biologically active crude plant extracts.Results:Among 23 medicinal plants used in the study,only 5 methanolic extracts[Rosmarinus offcinalis L,Carduus marianium L.,Lantana camara L.,Rhus tripartite(ueria) Grande,and Thymus capitatus(L.) Hoffm(link)]showed the highest antimicrobial activity againsl Staphylococcus aureus,Bacillus subtilis and Salmonella species,while 22 methanolic and aqueous extracts showed moderate to weak antimicrobial activity on all tested organisms.However 19 of the extracts showed no activity at all against Gram-ve and Gram +ve microorganisms.MIC was found to be 1.25 mg/mL(Thymus capitatus),3 mg/mL(Rhus tripartite),4 mg/mL(Carduus marianium),5 mg/mL(Rosamarinus officinalis) and5 mg/mL(Lantana camara),respectively.Conclnsions:The present results revealed that,crude methanolic extracts of the investigated Libyan folk medicinal plants exhibited mild to high in vitro antibacterial activities against Gram-positive and Gram-negative microorganisms.Hasan MH Muhaisen Miftah Mailoud Ab-Mous Fadel A Ddeeb Aboclaid All Rtemi Omer M Taba Mehtab Parveen 2016Chinese Journal of Integrative Medicine2016,22,3:2

10Pit-and trench-forming osteoclasts:a distinction that matters显示文摘Osteoclasts(OCs)seeded on bone slices either drill round pits or dig long trenches.Whereas pits correspond to intermittent resorption,trenches correspond to continuous and faster resorption and require a distinct assembly of the resorption apparatus.It is unknown whether the distinction between pits and trenches has any biological relevance.Using OCs prepared from different blood donors,we found that female OCs achieved increased resorption mainly through pit formation,whereas male OCs did so through trench formation.Trench formation went along with high collagenolytic activity and high cathepsin K(CatK)expression,thereby allowing deeper demineralization.A specific CatK inhibitor abrogated the generation of trenches,while still allowing the generation of pits.OCs obtained from bone marrow were more prone to generate trenches than those obtained from blood.Scanning electron microscopy of bone surfaces eroded in vivo showed trenches and pits of similar size as those made by OCs in culture.We conclude that the distinction between trench-and pit-forming OCs is relevant to the differences among OCs from different skeletal sites,different individuals,including gender,and results from differences in colIagenolytic power.This indicates a biological relevance and highlights the importance of discriminating between pits and trenches when assessing resorption.Ditte MH Merrild Dinisha C Pirapaharan Christina M Andreasen Per Kjxrsgaard-Andersen Anas MJ Mùller Ming Ding Jean-Marie Delaissé Kent Sùe 2015Bone Research2015,3,4:2
11Prognostic Value of Promoter Hypermethylation of Retinoic Acid Receptor Beta (RARB) and CDKN2 (p16/MTS1) in Prostate Cancer显示文摘Objective: The molecular mechanism of prostate cancer is poorly understood. The aim of the study was to investigate the prevalence and prognostic value of promoter hypermethylation of retinoic acid receptor beta (RARB) and p16 among benign prostatic hyperplasia (BPH) and prostate cancer patients. Methods: In this case-control study, 63 patients were included in three groups; 21 with BPH as the control group, 21 with prostate cancer and good prognostic factors (based on prostate-specific antigen, Gleason score and stage) as good prognosis group, and 21 with prostate cancer and poor prognostic features as poor prognosis group. The prostate biopsy specimen of each individual was examined for hypermethylation of RARB and p16 promoters by methylation specific PCR (MSPCR). Results: Seven (33.3%) patients with good prognosis and 15 (71.4%) patients with poor prognosis were positive for RARB methylation, which were significantly higher than controls (P <0.0001). p16 promoter methylation was shown in 19.0% and 47.6% patients with good and poor prognosis, respectively. The RARB and p16 promoter methylation in the poor prognosis group was significantly higher than that in the good prognosis group (P =0.02 for RARB and P<0.0001 for p16).
