| 1 | 肺炎链球菌呼吸道感染分离株的耐药性显示文摘目的 :研究肺炎链球菌呼吸道感染分离株的耐药性及其传播情况。方法 :肺炎链球菌分离株进行药物敏感试验和血清学分型 ,并结合BOXPCR、脉冲场电泳 (pulse-fieldgelelectrophoresis,PFGE)和青霉素结合蛋白基因 (penicillinbindingpro teingene,pbp)指纹等分子生物学方法分析菌株间亲缘关系。 结果 :呼吸道感染分离株 1 2 8株中有青霉素低度耐药株 (peni cillinintermediatedStreptococcuspneumoniae,PISP) 1 0株 ,未发现青霉素高度耐药株 (penicillinresistantStreptococcuspneumoni ae,PRSP)。 1 1 1株存活菌株分属 2 5种血清型 ,主要为 2 3F、3、1 9F、1 4、9V、6A、6B等。 9株存活PISP中有 6种PFGE谱型 ,2株PFGE谱型相同菌株的耐药谱和血清型相仿 ;2株不同PFGE谱型菌株pbp指纹相似 ,对青霉素和头孢曲松敏感性相似 ;上述呼吸道感染分离PISP中未发现与世界主要流行耐药克隆代表株DNA指纹或 pbp基因相似菌株。 结论 :上述肺炎链球菌呼吸道感染分离株青霉素耐药率尚低 ,存在青霉素耐药克隆和基因的传播 ,其中尚未发现世界主要流行耐药克隆。 | 杨帆 张婴元 Lesley McGee Avril Wasas Keith Klugman 吴卫红 周乐 朱德妹 汪复 | 2001 | 中国抗感染化疗杂志2001,1,1: | 26 |
| 2 | AMPK调节骨骼肌细胞GLUT4基因表达的机制研究显示文摘腺苷酸活化蛋白激酶(AMPK)能调节运动/肌肉收缩所引起的骨骼肌细胞葡萄糖转运蛋白4(GLUT4)基因的表达,但至今它的调节机制不清。研究显示在非运动刺激引起的细胞信号事件中由组蛋白去乙酰化酶(HDACs)以及组蛋白乙酰化酶(HATs)控制的组蛋白乙酰化状态是调节基因表达的重要机制,所以我们假设AMPK信号途径是通过征用HDACs中的HDAC5(在骨骼肌细胞内高表达)来实现对运动/肌肉收缩引起的GLUT4基因表达控制。细胞分为正常浓度葡萄糖对照组(NGLU组)、正常浓度AICAR组(NGLU+AICAR组)、高浓度对照组(HGLU组)、高浓度AICAR组(HGLU+AICAR组)。用5 mmol/L和20 mmol/L葡萄糖浓度培养骨骼肌细胞后,NGLU+AICAR组和HGLU+AICAR组与肌肉收缩模拟信号刺激5-氨基-4-甲酰胺咪唑核糖核苷酸(AICAR)孵育。AICAR能激活NGLU组骨骼肌细胞AMPKα2、减少骨骼肌细胞核HDAC5蛋白、促使HDAC5与骨骼肌细胞加强因子(MEF2)蛋白分离和上调GLUT4基因的表达;相反,高浓度葡萄糖延迟由AICAR引起的AMPKα2磷酸化、AMPKα2向细胞核转入、HDAC5向细胞核转出和GLUT4基因的表达。实验结果说明在不同葡萄糖浓度下的骨骼肌细胞GLUT4基因表达变化都对应着上游AMPK蛋白和下游HDAC5蛋白的变化,AMPK可能是征用转录抑制子HDAC5来调节MEF2的活性而达到控制肌肉收缩所引起的GLUT4基因表达。 | 李良刚 陈槐卿 Sean L. | 7 | Helicobacter pylori arginase mutant colonizes arginase Ⅱ knockout mice显示文摘AIM: To investigate the role of host and bacterial arginases in the colonization of mice by Helicobacter pylori (H.pylori).METHODS: H.pylori produces a very powerful urease that hydrolyzes urea to carbon dioxide and ammonium,which neutralizes acid.Urease is absolutely essential to H.pylori pathogenesis;therefore,the urea substrate must be in ample supply for urease to work efficiently.The urea substrate is most likely provided by arginase activity,which hydrolyzes L-arginine to L-ornithine and urea.Previous work has demonstrated that H.pylori arginase is surprisingly not required for colonization of wild-type mice.Hence,another in vivo source of the critical urea substrate must exist.We hypothesized that the urea source was provided by host arginase Ⅱ,since this enzyme is expressed in the stomach,and H.pylori has previously been shown to induce the expression of murine gastric arginase Ⅱ.To test this hypothesis,wild-type and arginase (rocF) mutant H.pylori strain SS1 were inoculated into arginase Ⅱ knockout mice.RESULTS: Surprisingly,both the wild-type and rocF mutant bacteria still colonized arginase Ⅱ knockout mice.Moreover,feeding arginase Ⅱ knockout mice the host arginase inhibitor S-(2-boronoethyl)L-cysteine (BEC),while inhibiting > 50% of the host arginase Ⅰ?activity in several tissues,did not block the ability of the rocF mutant H.pylori to colonize.In contrast,BEC poorly inhibited H.pylori arginase activity.CONCLUSION: The in vivo source for the essential urea utilized by H.pylori urease is neither bacterial arginase nor host arginase Ⅱ;instead,either residual host arginase Ⅰ?or agmatinase is probably responsible. | Songhee H Kim Melanie L Langford Jean-Luc Boucher Traci L Testerman David J McGee | 2011 | World Journal of Gastroenterology2011,17,28: | 3 |

| 9 | Residual feed intake phenotype and gender affect the expression of key genes of the lipogenesis pathway in subcutaneous adipose tissue of beef cattle显示文摘Background: Feed accounts for up to 75% of costs in beef production systems,thus any improvement in feed efficiency(FE) will benefit the profitability of this enterprise.Residual feed intake(RFI) is a measure of FE that is independent of level of production.Adipose tissue(AT) is a major endocrine organ and the primary metabolic energy reservoir.It modulates a variety of processes related to FE such as lipid metabolism and glucose homeostasis and thus measures of inter-animal variation in adiposity are frequently included in the calculation of the RFI index.The aim of this study was to determine the effect of phenotypic RFI status and gender on the expression of key candidate genes related to processes involved in energy metabolism within AT.Dry matter intake(DMI) and average daily gain(ADG) were measured over a period of 70 d for 52 purebred Simmental heifers(n = 24) and bulls(n = 28) with an initial BW±SD of 372±39.6 kg and 387±50.6 kg,respectively.Residual feed intake was calculated and animals were ranked within gender by RFI into high(inefficient; n = 9 heifers and n = 8 bulls)and low(efficient; n = 9 heifers and n = 8 bulls) groups.Results: Average daily gain ±SD and daily DMI ±SD for heifers and bul s were 1.2±0.4 kg and 9.1±0.5 kg,and 1.8±0.3 kg and 9.5±1 kg respectively.High RFI heifers and bulls consumed 10% and 15% more(P < 0.05) than their low RFI counterparts,respectively.Heifers had a higher expression of all genes measured than bulls(P < 0.05).A gender × RFI interaction was detected for HMGCS2(P < 0.05) in which high RFI bulls tended to have lower expression of HMGCS2 than low RFI bulls(P < 0.1),whereas high RFI heifers had higher expression than low RFI heifers(P < 0.05) and high RFI bulls(P < 0.05).SLC2 A4 expression was consistently higher in subcutaneous AT of low RFI animals across gender.Conclusion: The findings of this study indicate that low RFI cattle exhibit upregulation of the molecular mechanisms governing glucose metabolism in adipose tissue,in particular,glucose clearance.The decreased expression of SLC2 A4 in the inefficient cattle may result in less efficient glucose metabolism in these animals.We conclude that SLC2 A4 may be a potential biomarker for RFI in cattle. | Clare McKenna Richard K.Porter Kate A.Keogh Sinead M.Waters Mark McGee David A.Kenny | 2019 | Journal of Animal Science and Biotechnology2019,10,1: | 2 |
