1Coordinated AR and microRNA regulation in prostate cancer显示文摘The androgen receptor(AR)remains a key driver of prostate cancer(PCa)progression,even in the advanced castrate-resistant stage,where testicular androgens are absent.It is therefore of critical importance to understand the molecular mechanisms governing its activity and regulation during prostate tumourigenesis.MicroRNAs(miRs)are small w22 nt noncoding RNAs that regulate target gene,often through association with 30 untranslated regions(30UTRs)of transcripts.They display dysregulation during cancer progression,can function as oncogenes or tumour suppressors,and are increasingly recognised as targets or regulators of hormonal action.Thus,understanding factors which modulate miRs synthesis is essential.There is increasing evidence for complex and dynamic bi-directional cross-talk between the multi-step miR biogenesis cascade and the AR signalling axis in PCa.This review summarises the wealth of mechanisms by which miRs are regulated by AR,and conversely,how miRs impact AR’s transcriptional activity,including that of AR splice variants.In addition,we assess the implications of the convergence of these pathways on the clinical employment of miRs as PCa biomarkers and therapeutic targets.Ieva Eringyte Joanna N.Zamarbide Losada Sue M.Powell Charlotte L.Bevan Claire E.Fletcher 2020Asian Journal of Urology2020,7,3:2

3Mechanical suppression of breast cancer cell invasion and paracrine signaling to osteoclasts requires nucleo-cytoskeletal connectivity显示文摘Exercise benefits the musculoskeletal system and reduces the effects of cancer.The effects of exercise are multifactorial,where metabolic changes and tissue adaptation influence outcomes.Mechanical signals,a principal component of exercise,are anabolic to the musculoskeletal system and restrict cancer progression.We examined the mechanisms through which cancer cells sense and respond to low-magnitude mechanical signals introduced in the form of vibration.Low-magnitude,high-frequency vibration was applied to human breast cancer cells in the form of low-intensity vibration(LIV).LIV decreased matrix invasion and impaired secretion of osteolytic factors PTHLH,IL-11,and RANKL.Furthermore,paracrine signals from mechanically stimulated cancer cells,reduced osteoclast differentiation and resorptive capacity.Disconnecting the nucleus by knockdown of SUN1 and SUN2 impaired LIV-mediated suppression of invasion and osteolytic factor secretion.LIV increased cell stiffness;an effect dependent on the LINC complex.These data show that mechanical vibration reduces the metastatic potential of human breast cancer cells,where the nucleus serves as a mechanosensory apparatus to alter cell structure and intercellular signaling.Xin Yi Laura E.Wright Gabriel M.Pagnotti Gunes Uzer Katherine M.Powell Joseph M.Wallace Uma Sankar Clinton T.Rubin Khalid Mohammad Theresa A.Guise William R.Thompson 2020Bone Research2020,8,4:1

5Arabidopsis ADC1 functions as an Nδ-acetylornithine decarboxylase显示文摘Polyamines are small aliphatic amines found in almost all organisms,ranging from bacteria to plants and animals.In most plants,putrescine,the metabolic precursor for longer polyamines,such as spermidine and spermine,is produced from arginine,with either agmatine or ornithine as intermediates.Here we show that Arabidopsis thaliana(Arabidopsis)arginine de-carboxylase 1(ADC1),one of the two known arginine decarboxylases in Arabidopsis,not only synthesizes agmatine from arginine,but also converts N-acety.lornithine to N-acetylputrescine.Phylogenetic analyses indicate that duplication and neofunctionalization of ADC1 and NATA1,the enzymes that synthesize Nδ-acetylormithine in Arabidopsis,co-occur in a small number of related species in the Brassicaceae.Unlike ADC2,which is localized in the chloroplasts,ADC1 is in the endoplasmic reticulum together with NATA1,an indication that these two enzymes have access to the same substrate pool.Together,these results are con-sistent with a model whereby NATA1 and ADC1 to-gether provide a pathway for the synthesis of N-acetylputrescine in Arabidopsis.Yann-Ru Lou Sheaza Ahmed Jian Yan Adewale M.Adio Hannah M.Powell Paul F.Morris Georg Jander 2020Journal of Integrative Plant Biology2020,62,5:0

7Dynamic energy system modeling using hybrid physics-based and machinelearning encoder–decoder models显示文摘Three model configurations are presented for multi-step time series predictions of the heat absorbed by thewater and steam in a thermal power plant. The models predict over horizons of 2, 4, and 6 steps into thefuture, where each step is a 5-minute increment. The evaluated models are a pure machine learning model, anovel hybrid machine learning and physics-based model, and the hybrid model with an incomplete dataset. Thehybrid model deconstructs the machine learning into individual boiler heat absorption units: economizer, waterwall, superheater, and reheater. Each configuration uses a gated recurrent unit (GRU) or a GRU-based encoder–decoder as the deep learning architecture. Mean squared error is used to evaluate the models compared totarget values. The encoder–decoder architecture is over 11% more accurate than the GRU only models. Thehybrid model with the incomplete dataset highlights the importance of the manipulated variables to the system.The hybrid model, compared to the pure machine learning model, is over 10% more accurate on averageover 20 iterations of each model. Automatic differentiation is applied to the hybrid model to perform a localsensitivity analysis to identify the most impactful of the 72 manipulated variables on the heat absorbed in theboiler.
