1Altered Endothelin-1 Signaling in Production of Thromboxane A_(2) in Kupffer Cells from Bile Duct Ligated Rats显示文摘Kupffer cells(KCs),the liver resident macrophages accounting for 80-90%of the total population of fixed tissue macrophages in the body,not only play a key role in host defense via removing particulate materials from the portal circulation,but may also contribute to the pathogenesis of various liver diseases.We have previously demonstrated that KCs play an important role in controlling portal hypertension and hepatocellular injury via releasing thromboxane A_(2)(TXA_(2))in early fibrosis induced by one-week bile duct ligation(BDL).Production of TXA_(2) is controlled by cytosolic phospholipase A_(2)(cPLA_(2))that is activated by the interaction of entothelin-1(ET-1)with its G-protein coupled ET receptor B(ETBR).However,the signaling pathways that contribute to the ET-1-induced activation of cPLA_(2) and production of TXA_(2) in KCs in the normal healthy or injured livers are not yet clear,which are investigated in the present study using isolated KCs from one-week BDL or sham rats.The pharmacological inhibition of cPLA_(2) or chelation of intracellular calcium abrogated the ET-1 induction of TXA_(2) from KCs.Compared to those from sham rats,KCs from BDL animals displayed a significantly enhanced responsiveness of p38 MAPK to ET-1,increased ETBR and Gai subunit but decreased Gaq and Ga11 expression.Inhibition of ERK1/2 or Gq signaling abrogated significantly the ET-1 induction of TXA_(2) in sham KCs but only slightly in BDL KCs.In contrast,inhibition of p38 MAPK and Gi signaling markedly attenuated the ET-1 induction of TXA_(2) in BDL KCs but had no effect in sham KCs.Lastly,inhibition of PLC or PKC abrogated ET-1 induction of TXA_(2) in KCs from both sham and BDL groups.The hepatic stress(such as BDL)induces significant modifications in the receptor and intermediates of ET-1 signaling in KC and subsequently alters ET-1 signaling mechanisms,particularly a shift from Gq induced signaling to Gi induced signaling,in the activation of cPLA_(2) and production of TXA_(2) in response to ET-1.Andrew M.Miller Jian X.Zhang 2009Cellular & Molecular Immunology2009,6,6:4
2Locomotor analysis identifies early compensatory changes during disease progression and subgroup classification in a mouse model of amyotrophic lateral sclerosis显示文摘Amyotrophic lateral sclerosis is a motoneuron degenerative disease that is challenging to diagnose and presents with considerable variability in survival.Early identification and enhanced understanding of symptomatic patterns could aid in diagnosis and provide an avenue for monitoring disease progression.Use of the m SOD1 G93 A mouse model provides control of the confounding environmental factors and genetic heterogeneity seen in amyotrophic lateral sclerosis patients,while investigating underlying disease-induced changes.In the present study,we performed a longitudinal behavioral assessment paradigm and identified an early hindlimb symptom,resembling the common gait abnormality foot drop,along with an accompanying forelimb compensatory mechanism in the m SOD1 G93 A mouse.Following these initial changes,m SOD1 mice displayed a temporary hindlimb compensatory mechanism resembling an exaggerated steppage gait.As the disease progressed,these compensatory mechanisms were not sufficient to sustain fundamental locomotor parameters and more severe deficits appeared.We next applied these initial findings to investigate the inherent variability in B6 SJL m SOD1 G93 A survival.We identified four behavioral variables that,when combined in a cluster analysis,identified two subpopulations with different disease progression rates:a fast progression group and a slow progression group.This behavioral assessment paradigm,with its analytical approaches,provides a method for monitoring disease progression and detecting m SOD1 subgroups with different disease severities.This affords researchers an opportunity to search for genetic modifiers or other factors that likely enhance or slow disease progression.Such factors are possible therapeutic targets with the potential to slow disease progression and provide insight into the underlying pathology and disease mechanisms.Melissa M.Haulcomb Rena M.Meadows Whitney M.Miller Kathryn P.Mc Millan Me Kenzie J.Hilsmeyer Xuefu Wang Wesley T.Beaulieu Stephanie L.Dickinson Todd J.Brown Virginia M.Sanders Kathryn J.Jones 2017Neural Regeneration Research2017,12,10:3

17Probing star formation and feedback using CCOSMA and archival data in the CFG028.68–0.28 quasi-sinusoidal filament显示文摘We have performed a multi-wavelength study toward a quasi-sinusoidal filament(CFG028.68–0.28). A new large-scale ^12CO J = 3-2 map was obtained from the China-Cologne Observation for Sub Millimeter Astronomy(CCOSMA) 3m radio telescope. Based on the ATLASGAL catalog, we have identified 27 dust clumps in the filament. Through the relationship between the mass and radius of these clumps, 67% of these clumps are dense and massive enough to potentially form massive stars. The obtained CFE is ~11% in the filament. The filament has a linear mass density of ~305 M⊙pc^-1, which is smaller than its critical mass to length ratio. This suggests that the external pressure from the neighboring H Ⅱ regions may help prevent the filament from dispersing under the effects of turbulence. 18Nucleation of recrystallization in magnesium alloy grains of varied orientation and the impacts on texture evolution显示文摘Selection of appropriate thermomechanical processing parameters is crucial for control of the crystallographic texture and grain size distribution in wrought magnesium alloys, which in turn dictate their mechanical properties. In this work, the recrystallization behavior of binary Mg-Zn alloys are examined after warm rolling as a function of alloy content and heat treatment temperature.
