| 2 | New vacuum solar telescope and observations with high resolution显示文摘The New Vacuum Solar Telescope(NVST) is a one meter vacuum solar telescope that aims to observe fine structures on the Sun. The main goals of NVST are high resolution imaging and spectral observations, including measurements of the solar magnetic field. NVST is the primary ground-based facility used by the Chinese solar research community in this solar cycle. It is located by Fuxian Lake in southwest China, where the seeing is good enough to perform high resolution observations. We first introduce the general conditions at the Fuxian Solar Observatory and the primary science cases of NVST. Then, the basic structures of this telescope and instruments are described in detail. | 3 | A Genetic Pathway for Tapetum Development and Function in Arabidopsis显示文摘In anther development, tapetal cells take part in complex processes, including endomitosis and apoptosis (programmed cell death). The tapetum provides many of the proteins, lipids, polysaccharides and other molecules necessary for pollen development. Several transcription factors, including DYT1, TDF1, AMS, MS188 and MS1, have been reported to be essential for tapetum development and function in Arabidopsis thaliana. Here, we present a detailed cytological analysis of knockout mutants for these genes, along with an in situ RNA hybridization experiment and double mutant analysis showing that these transcription factors form a genetic pathway in tapetum development. DYT1, TDF1 and AMS function in early tapetum development, while MS188 and MS1 are important for late tapetum development. | 5 | Role of gut microbiota via the gut-liver-brain axis in digestive diseases显示文摘The gut-brain axis is a bidirectional information interaction system between the central nervous system(CNS) and the gastrointestinal tract, in which gut microbiota plays a key role. The gut microbiota forms a complex network with the enteric nervous system, the autonomic nervous system, and the neuroendocrine and neuroimmunity of the CNS, which is called the microbiota-gut-brain axis. Due to the close anatomical and functional interaction of the gut-liver axis, the microbiota-gut-liver-brain axis has attracted increased attention in recent years. The microbiota-gut-liver-brain axis mediates the occurrence and development of many diseases, and it offers a direction for the research of disease treatment. | 8 | Downregulation of lncRNA AFAP1-AS1 by oridonin inhibits the epithelial-to-mesenchymal transition and proliferation of pancreatic cancer cells显示文摘Recent studies have demonstrated that the expression of the long non-coding RNA(lncRNA)AFAP1-AS1 in pancreatic cancer is negatively correlated with survival and prognosis.However,the effects of oridonin and lncRNA AFAP1-AS1 on the epithelial-to-mesenchymal transition(EMT)and migration of pancreatic cancer cells have not been fully elucidated.Surgery is the only potentially curative method for pancreatic cancer,but postoperative recurrence and metastasis are common.The aim of the present study was to assess the effect of oridonin and lncRNA AFAP1-AS1 silencing on pancreatic cancer cells.The pancreatic cancer cell lines BxPC-3 and PANC-1 cells were transfected with siAFAP1-AS1 and its negative control(siNC).After that,oridonin was used to treat the siAFAP1-AS1-transfected cells.The expression of lncRNA AFAP1-AS1 was downregulated in the pancreatic cancer cell lines BxPC-3 and PANC-1.The apoptosis and cell cycle progression of pancreatic cancer cells were evaluated by flow cytometry and Hoechst 33258 staining.Metastasis and invasion of BxPC-3 and PANC-1 cells were detected by transwell migration assay,real-time cell analysis,and western blot analysis.Cells were transfected with the lentiviral siAFAP1-AS1 and siNC,and tumorigenesis was evaluated in BALB/C nude mice.Immunohistochemical examination was used to verify the effects of oridonin and siAFAP1-AS1 on pancreatic cancer.The results demonstrated that the combination of oridonin and siAFAP1-AS1 inhibited pancreatic cancer cell proliferation,induced apoptosis,arrested cell cycle progression,prevented the migration,regulated EMT-related protein expression in BxPC-3 and PANC-1 cells,and inhibited pancreatic cancer cell tumorigenicity and EMT in nude mice. | Songmei Lou Jian Xu Bili Wang Shuquan Li Jun Ren Zhengjun Hu Bin Xu Feng Luo | 2019 | Acta Biochimica et Biophysica Sinica2019,51,8: | 10 |

| 11 | Analysis of ESTs and gene expression patterns of the posterior silkgland in the fifth instar larvae of silkworm, Bombyx mori L.显示文摘The fibroin gene expression pattern and regulation of the posterior silkgland were studied by means of expressed sequence tags (ESTs) using the first and fifth day larvae of the fifth instar of silkworm, Bombyx mori L (strain: C 108). The results showed that there were 911 repetitive ESTs and 1950 single sequences (Singlets) among total 2861 consentient sequences, which were spliced. 1335 sequences were identified and the other 1526 were unknown. 5560 sequences (55.89%) in the posterior silkgland cell of the silkworm were new ESTs without homology with EST data published by Mita et al. The number of repetitive ESTs and single sequences from the first day larvae of the fifth instar was double more than that of the fifth day of the same instar in the silkworms. The unigenes which were more than 50 in repetitive EST size (contig size) came to only about 0.5% in total consentient sequences. There were significant differences between gene expression frequencies, and expressed genes were related to fibroin synthesis and its secretion and fibroin composition. Comparing the fifth day with the first day of the fifth instar, the genes-expressed quantity of fibroin heavy-chain gene was 18 fold higher, fibroin light-chain gene 9 fold and fibroin P52 gene 8 fold. 508 genes functioned for cellular component and 315 for enzyme after function tracing. These results implied that the gene expression of the first day was mainly for preparation for fibroin synthesis except for the growth of silkgland cells, and the gene expression of the fifth day of the fifth instar was mainly for synthesizing and excreting fibroin. Because the ratio of heavy chain, light chain and p25 of fibroin was not 6:6:1 as theoretically expected, or its special H-chain structure, the H-chain gene was not easy to detect through EST technique. Most of genes among total 2861 consentient sequences functioned for fibroin synthesis and secretion. | 13 | Do vehicular emissions dominate the source of C6–C8 aromatics in the megacity Shanghai of eastern China?显示文摘The characteristic ratios of volatile organic compounds(VOCs) to i-pentane, the indicator of vehicular emissions, were employed to apportion the vehicular and non-vehicular contributions to reactive species in urban Shanghai. Two kinds of tunnel experiments, one tunnel with more than 90% light duty gasoline vehicles and the other with more than 60% light duty diesel vehicles, were carried out to study the characteristic ratios of vehicle-related emissions from December 2009 to January 2010. Based on the experiments, the characteristic ratios of C6–C8aromatics to i-pentane of vehicular emissions were 0.53 ± 0.08(benzene), 0.70 ± 0.12(toluene),0.41 ± 0.09(m,p-xylenes), 0.16 ± 0.04(o-xylene), 0.023 ± 0.011(styrene), and 0.15 ± 0.02(ethylbenzene), respectively. The source apportionment results showed that around 23.3% of C6–C8 aromatics in urban Shanghai were from vehicular emissions, which meant that the non-vehicular emissions had more importance. These findings suggested that emission control of non-vehicular sources, i.e. industrial emissions, should also receive attention in addition to the control of vehicle-related emissions in Shanghai. The chemical removal of VOCs during the transport from emissions to the receptor site had a large impact on the apportionment results. | 17 | Preparation of a novel PAN/cellulose acetate-Ag based activated carbon nanofiber and its adsorption performance for low-concentration SO_2显示文摘Polyacrylonitrile(PAN), PAN/cellulose acetate(CA), and PAN/CA-Ag based activated carbon nanofiber(ACNF) were prepared using electrostatic spinning and further heat treatment. Thermogravimetry–differential scanning calorimetry(TG–DSC) analysis indicated that the addition of CA or Ag did not have a significant impact on the thermal decomposition of PAN materials but the yields of fibers could be improved. Scanning electron microscopy(SEM) analysis showed that the micromorphologies of produced fibers were greatly influenced by the viscosity and conductivity of precursor solutions. Fourier transform infrared spectroscopy(FT-IR) analysis proved that a cyclized or trapezoidal structure could form and the carbon scaffold composed of C=C bonds appeared in the PAN-based ACNFs. The characteristic diffraction peaks in X-ray diffraction(XRD) spectra were the evidence of a turbostratic structure and silver existed in the PAN/CA-Ag based ACNF. Brunner–Emmett–Teller(BET) analysis showed that the doping of CA and Ag increased surface area and micropore volume of fibers; particularly, PAN/CA-Ag based ACNF exhibited the best porosity feature. | 19 | De novo assembly of a Tibetan genome and identification of novel structural variants associated with high-altitude adaptation显示文摘Structural variants(SVs) may play important roles in human adaptation to extreme environments such as high altitude but have been under-investigated. Here, combining long-read sequencing with multiple scaffolding techniques, we assembled a high-quality Tibetan genome(ZF1), with a contig N50 length of 24.57 mega-base pairs(Mb) and a scaffold N50 length of 58.80 Mb. The ZF1 assembly filled 80 remaining N-gaps(0.25 Mb in total length) in the reference human genome(GRCh38). Markedly, we detected17 900 SVs, among which the ZF1-specific SVs are enriched in GTPase activity that is required for activation of the hypoxic pathway. Further population analysis uncovered a 163-bp intronic deletion in the MKL1 gene showing large divergence between highland Tibetans and lowland Han Chinese. This deletion is significantly associated with lower systolic pulmonary arterial pressure, one of the key adaptive physiological traits in Tibetans. Moreover, with the use of the high-quality de novo assembly, we observed a much higher rate of genome-wide archaic hominid(Altai Neanderthal and Denisovan) shared non-reference sequences in ZF1(1.32%–1.53%) compared to other East Asian genomes(0.70%–0.98%),reflecting a unique genomic composition of Tibetans. One such archaic hominid shared sequence-a662-bp intronic insertion in the SCUBE2 gene-is enriched and associated with better lung function(the FEV1/FVC ratio) in Tibetans. | 20 | Lijiang 2.4-meter Telescope and its instruments显示文摘The Lijiang 2.4-meter Telescope(LJT), the largest common-purpose optical telescope in China,has been available to the worldwide astronomical community since 2008. It is located at the Gaomeigu site,Lijiang Observatory(LJO), in the southwest of China. The site has very good observational conditions.During its 10-year operation, several instruments have been equipped on the LJT. Astronomers can perform both photometric and spectral observations. The main scientific goals of LJT include recording photometric and spectral evolution of supernovae, reverberation mapping of active galactic nuclei, investigating the physical properties of binary stars and near-earth objects(comets and asteroids), and identification of exoplanets and all kinds of transients. Until now, the masses of 41 high accretion rate black holes have been measured, and more than 168 supernovae have been identified by the LJT. More than 190 papers related to the LJT have been published. In this paper, the general observation conditions of the Gaomeigu site is introduced at first. Then, the structure of the LJT is described in detail, including the optical, mechanical, motion and control system. The specification of all the instruments and some detailed parameters of the YFOSC is also presented.
