1Expression,deleton and mnutation of ρ16 gene in human gastric cancer显示文摘AIM To investigate the relationship between the expression of p16 gene and the gastric carcinogenesis,depth of invasion and lymph node metastases, and to evaluate the deletion and mutation of exon 2 in p16 gene in gastric carcinoma.METHODS The expression of P16 protein was examined by streptavidin-peroxidase conjugated method (S-P); the deletion and mutation of p16 gene were respectively examined by polymerase chain reaction (PCR) and polymerase chain reaction single-strand conformation polymorphism analysis (PCR-SSCP) in gastric carcinoma.RESULTS Expression of P16 protein was detected in 96.25% (77/80) of the normal gastric mucosa, in 92.00% (45/50) of the dysplastic gastric mucosa and in 47.54% (58/122) of the gastric carcinoma. The positive rate of P16 protein expression in gastric carcinoma was significantly lower than that in normal gastric mucosa and dysplastic gastric mucosa (P<0.05). The positive rate of P16 protein expression in mucoid carcinoma 10.00% (1/ 10) was significantly lower than that in poorly differentiated carcinoma 51.22% ( 21/ 41 ),undifferentiated carcinoma 57.69% (15/26) and signet ring cell carcinoma 62.50% (10/ 16) (P<0.05). The positive rate of p16 protein in 30 cases paired primary and lymph node metastatic gastric carcinoma: There was 46.67% (14/30) in primary gastric carcinoma, 16.67% (5/30) in lymph node metastatic gastric carcinoma. The positive rate of lymph node metastatic carcinoma was significantly lower than that of primary carcinoma (P<0.05). There was of p16 gene mutation in exon 2, but 5 cases displayed deletion of p16 gene in exon 2 in the 25 primary gastric carcinomas.CONCLUSIONS The expression loss of P16 protein related to the gastric carcinogenesis, gastric carcinoma histopathological subtypes and lymph metastasis. The mutation of p16 gene in exon 2 may not be involved in gastric carcinogenesis. 3LncRNA SNHG12 ameliorates brain microvascular endothelial cell injury by targeting miR-199a显示文摘Long non-coding RNAs regulate brain microvascular endothelial cell death, the inflammatory response and angiogenesis during and after ischemia/reperfusion and oxygen-glucose deprivation/reoxygenation(OGD/R) insults. The long non-coding RNA, SNHG12, is upregulated after ischemia/reperfusion and OGD/R in microvascular endothelial cells of the mouse brain. However, its role in ischemic stroke has not been studied. We hypothesized that SNHG12 positively regulates ischemic stroke, and therefore we investigated its mechanism of action. We established an OGD/R mouse cell model to mimic ischemic stroke by exposing brain microvascular endothelial cells to OGD for 0, 2, 4, 8, 16 or 24 hours and reoxygenation for 4 hours. Quantitative real-time polymerase chain reaction showed that SNHG12 levels in brain microvascular endothelial cells increased with respect to OGD exposure time. Brain microvascular endothelial cells were transfected with pc DNA-control, pc DNA-SNHG12, si-control, or si-SNHG12. After exposure to OGD for 16 hours, these cells were then analyzed by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide, trypan blue exclusion, western blot, and capillary-like tube formation assays. Overexpression of SNHG12 inhibited brain microvascular endothelial cell death and the inflammatory response but promoted angiogenesis after OGD/R, while SNHG12 knockdown had the opposite effects. miR-199a was identified as a target of SNHG12, and SNHG12 overexpression reversed the effect of miR-199a on brain microvascular endothelial cell death, the inflammatory response, and angiogenesis. 5Clinical role of (18)~F-fluorodeoxyglucose positron emission tomography/computed tomography in post-operative follow up of gastric cancer: Initial results显示文摘AIM: To evaluate the clinical role of 18F-fluorodeo-xyglucose positron emission and computed tomography (18F-FDG PET/CT) in detection of gastric cancer recurrence after initial surgical resection. METHODS: In the period from January 2007 to May 2008, 23 patients who had previous surgical resection of histopathologically diagnosed gastric cancer underwent a total of 25 18F-FDG PET/CT scans as follow-up visits in our center. The standard of reference for tumor recurrence consisted of histopathologic confirmation or clinical follow-up information for at least 5 mo after PET/CT examinations. RESULTS: PET/CT was positive in 14 patients (61%) and negative in 9 (39%). When correlated with final diagnosis, which was confirmed by histopathologic evidence of tumor recurrence in 8 of the 23 patients (35%) and by clinical follow-up in 15 (65%), PET/CT was true positive in 12 patients, false positive in 2, true negative in 8 and false negative in 2. Overall, the accuracy of PET/CT was 82.6%, the negative predictive value (NPV) was 77.7%, and the positive predictive value (PPV) was 85.7%. The 2 false positive PET/CT findings were actually chronic inflammatory tissue lesions. For the two patients with false negativePET/CT, the f inal diagnosis was recurrence of mucinous adenocarcinoma in the anastomosis in one patient and abdominal wall metastasis in the other. Importantly, PET/CT revealed true-positive findings in 11 (47.8%) patients who had negative or no definite findings by CT. PET/CT revealed extra-abdominal metastases in 7 patients and additional esophageal carcinoma in one patient. Clinical treatment decisions were changed in 7 (30.4%) patients after introducing PET/CT into their conventional post-operative follow-up program. 7Casticin induces caspase-mediated apoptosis via activation of mitochondrial pathway and upregulation of DR5 in human lung cancer cells显示文摘Objective:To assess if casticin induces caspase-mediated apoptosis via activation of mitochondrial pathway and upregulation of DR5 in human lung cancer ceils.Methods:Human non-small-cell lung carcinoma cell lines H460,AS49 and H157 were cultured in vitro.The cytotoxic activities were determined using MTT assay.The apoptotic cells death was examined by flow cytometry using PI staining and DMA agarose gel electrophoresis.The activities of caspase-3, -8 and -9 were measured via ELISA.Cellular fractionation was determined by flow cytometry to assess release of cytochrome c and the mitochondrial transmembrane potential.Bcl-2/Bcl-XL/ XIAP/Bid/ DR5 and DR4 proteins were analyzed using western blot.Results:The concentrations required for a 50%decrease in cell growth(IC50) ranged from 1.8 to 3.2 Jt M.Casticin induced rapid apoptosis and triggered a series of effects associated with apoptosis by way of mitochondrial pathway,including the depolarization of the mitochondrial membrane,release of cytochrome c from mitochondria,activation of procaspase-9 and -3,and increase of DNA fragments.Moreover, the pan caspase inhibitor zVAD-FMK and the caspase-3 inhibitor zOEVD-FMK suppressed casticin-induced apoptosis.In addition,casticin induced XIAP and Bcl-XL down-regulation, Bax upregulation and Bid clearage.In H157 cell line,casticin increased expression of DRS at protein levels but not affect the expression of DR4.The prelreatmenl with DR5/Fc chimera protein effectively attenuated casticin-induced apoptosis in H157 cells.No correlation was found between cell sensitivity to casticin and that to p53 status,suggesting that casticin induce a p53- independent apoptosis.Conclusions:Our results demonstrate that casticin induces caspase-mediated apoptosis via activation of mitochondrial pathway and upregulation of DRS in human lung cancer cells.Yuan Zhou Yi Peng Qi-Qi Mao Xia Li Ming-Wu Chen Ting Su Li Tian Nai-Quan Mao Ling-Zhi Long Mei-Fang Quan Fei Liu Su-Fang Zhou Yong-Xiang Zhao 2013Asian Pacific Journal of Tropical Medicine2013,6,5:11

10Early initiation renal replacement therapy for fluid management to reduce central venous pressure is more conducive to renal function recovery in patients with acute kidney injury显示文摘Background:Acute kidney injury (AKI) is a serious complication in critically ill patients with septic shock treated in the intensive care unit. Renal replacement therapy (RRT) is a treatment for severe AKI;however, the time of initiation of RRT and factors that affect the recovery of kidney function remains unclear. This study was to explore whether early initiation of RRT treatment for fluid management to reduce central venous pressure (CVP) can help to improve patients5 kidney function recovery. Methods: A retrospective analysis of septic patients who had received RRT treatment was conducted. Patients received RRT either within 12 h after they met the diagnostic criteria of renal failure (early initiation) or after a delay of 48 h if renal recovery had not occurred (delayed initiation). Parameters such as patients5 renal function recovery at discharge, fluid balance, and levels of CVP were assessed. Results: A total of 141 patients were eligible for enrolment: 40.4% of the patients were in the early initiation group (57 of 141 patients), and 59.6% were in the delayed initiation group (84 of 141 patients). There were no significant differences in the characteristics at baseline between the two groups, and there were no differences in 28-day mortality between the two groups (χ^2 = 2.142, P = 0.143);however, there was a significant difference in the recovery rate of renal function between the two groups at discharge (χ^2 = 4.730, P < 0.001). More importantly, early initiation of RRT treatment and dehydration to reduce CVP are more conducive to the recovery of renal function in patients with AKI. 11Historic distribution and driving factors of human-caused fires in the Chinese boreal forest between 1972 and 2005显示文摘Aims The pattern and driving factors of forest fires are of interest for fire occurrence prediction and forest fire management.The aims of the study were:(i)to describe the history of human-caused fires by season and size of burned area over time;(ii)to identify the spatial patterns of human-caused fires and test for the existence of‘hotspots’to determine their exact locations in the Daxing’an mountains;(iii)to determine the driving factors that determine the spatial distribution and the possibility of human-caused fire occurrence.Methods In this study,K-function and Kernel density estimation were used to analyze the spatial pattern of human-caused fires.The analysis was conducted in s-plus and arcgIs environments,respectively.The analysis of driving factors was performed in SPSS 19.0 based on a logistic regression model.The variables used to identify factors that influence fire occurrence included vegetation types,meteorological conditions,socioeconomic factors,topography and infrastructure factors,which were extracted and collected through the spatial analysis mode of arcgIs and from official statistics,respectively.Important Findings The annual number of human-caused fires and the area burnt have declined since 1987 due to the implementation of a forest fire protection act.There were significant spatial heterogeneity and seasonal variations in the distribution of human-caused fires in the Daxing’an mountains.The heterogeneity was caused by elevation,distance to the nearest railway,forest type and temperature.a logistic regression model was developed to predict the likelihood of human-caused fire occurrence in the Daxing’an mountains;its global accuracy attained 64.8%.The model was thus comparable to other relevant studies.Futao Guo John L.Innes Guangyu Wang Xiangqing Ma Long Sun Haiqing Hu Zhangwen Su 2015Journal of Plant Ecology2015,8,5:10

14An overview of resistive random access memory devices显示文摘With recent progress in material science, resistive random access memory (RRAM) devices have attracted interest for nonvolatile, low-power, nondestructive readout, and high-density memories. Relevant performance parameters of RRAM devices include operating voltage, operation speed, resistance ratio, endurance, retention time, device yield, and multilevel storage. Numerous resistive-switching mechanisms, such as conductive filament, space-charge-limited conduction, trap charging and discharging, Schottky Emission, and Pool-Frenkel emission, have been proposed to explain the resistive switching of RRAM devices. In addition to a discussion of these mechanisms, the effects of electrode materials, doped oxide materials, and different configuration devices on the resistive-switching characteristics in nonvolatile memory applications, are reviewed. 15High power linearly polarized fiber laser:Generation, manipulation and application显示文摘Linearly-polarized(LP) fiber lasers, which could find wide potential applications such as coherent detection, coherent/spectral beam combining, nonlinear frequency conversion, have been a research focus in recent years. In this paper, we will present a general review on the achievements of various kinds of high power LP fiber laser and its applications for the first time. The recent progress in high power LP fiber oscillator, including fiber oscillator based on active fiber, Raman fiber laser and Random distributed feedback fiber laser are summarized. Power scaling of LP fiber laser by using active-fiber based power amplifier, passive-fiber based Raman amplifier, and active/passive fiber based hybrid fiber amplifier has been achieved. Polarization-maintained active fiber based power amplifier and non-polarization-maintained active fiber based power amplifier incorporating active polarization control are specially introduced in detail. High power LP fiber laser with diversified property, such as narrow-linewidth, wavelength-tunable and ultrashort pulse operation, are summarized. 16Clinical usefulness of ^(18) F-FDG PET/CT in the restaging of esophageal cancer after surgical resection and radiotherapy显示文摘AIM:To evaluate the clinical usefulness of 18F-fluorodeoxyglucose positron emission and computed tomography(18F-FDG PET/CT)in restaging of esophageal cancer after surgical resection and radiotherapy. METHODS:Between January 2007 and Aug 2008, twenty histopathologically diagnosed esophageal cancer patients underwent 25 PET/CT scans(three patients had two scans and one patient had three scans)for restaging after surgical resection and radiotherapy.The standard reference for tumor recurrence was histopathologic confirmation or clinical follow-up for at least ten months after 18F-FDG PET/CT examinations. RESULTS:Tumor recurrence was confirmed histopathologically in seven of the 20 patients(35%) and by clinical and radiological follow-up in 13(65%). 18F-FDG PET/CT was positive in 14 patients(68.4%) and negative in six(31.6%).18F-FDG PET/CT was true positive in 11 patients,false positive in three and true negative in six.Overall,the accuracy of 18F-FDG PET/CT was 85%,negative predictive value(NPV)was 100%,and positive predictive value(PPV)was 78.6%.The three false positive PET/CT findings comprised chronic inflammation of mediastinal lymph nodes(n =2)and anastomosis inflammation(n=1).PET/ CT demonstrated distant metastasis in 10 patients. 18F-FDG PET/CT imaging-guided salvage treatment in nine patients was performed.Treatment regimens were changed in 12(60%)patients after introducing 18F-FDG PET/CT into their conventional post-treatment follow-up program.CONCLUSION:Whole body 18F-FDG PET/CT is effective in detecting relapse of esophageal cancer after surgical resection and radiotherapy.It could also have important clinical impact on the management of esophageal cancer,influencing both clinical restaging and salvage treatment of patients.Long Sun Xin-Hui Su Yong-Song Guan Wei-Ming Pan Zuo-Ming Luo Ji-Hong Wei Long Zhao Hua Wu 2009World Journal of Gastroenterology2009,15,15:7
17Classification of the Gut Microbiota of Patients in Intensive Care Units During Development of Sepsis and Septic Shock显示文摘The gut microbiota of intensive care unit(ICU)patients displays extreme dysbiosis associated with increased susceptibility to organ failure,sepsis,and septic shock.However,such dysbiosis is difficult to characterize owing to the high dimensional complexity of the gut microbiota.We tested whether the concept of enterotype can be applied to the gut microbiota of ICU patients to describe the dysbiosis.We collected 131 fecal samples from 64 ICU patients diagnosed with sepsis or septic shock and performed 16S rRNA gene sequencing to dissect their gut microbiota compositions.During the development of sepsis or septic shock and during various medical treatments,the ICU patients always exhibited two dysbiotic microbiota patterns,or ICU-enterotypes,which could not be explained by host properties such as age,sex,and body mass index,or external stressors such as infection site and antibiotic use.ICU-enterotype I(ICU E1)comprised predominantly Bacteroides and an unclassified genus of Enterobacteriaceae,while ICU-enterotype II(ICU E2)comprised predominantly Enterococcus.Among more critically ill patients with Acute Physiology and Chronic Health Evaluation II(APACHE II)scores>18,septic shock was more likely to occur with ICU E1(P=0.041).Additionally,ICU E1 was correlated with high serum lactate levels(P=0.007).Therefore,different patterns of dysbiosis were correlated with different clinical outcomes,suggesting that ICU-enterotypes should be diagnosed as independent clinical indices.Thus,the microbial-based human index classifier we propose is precise and effective for timely monitoring of ICU-enterotypes of individual patients.This work is a first step toward precision medicine for septic patients based on their gut microbiota profiles.Wanglin Liu Mingyue Cheng Jinman Li Peng Zhang Hang Fan Qinghe Hu Maozhen Han Longxiang Su Huaiwu He Yigang Tong Kang Ning Yun Long 2020Genomics, Proteomics & Bioinformatics2020,18,6:7
18MmNet:Identifying Mikania micrantha Kunth in the wild via a deep Convolutional Neural Network显示文摘Mikania micrantha Kunth is an invasive alien weed and known as a plant killer around the world.Accurately and rapidly identifying M.micrantha in the wild is important for monitoring its growth status,as this helps management officials to take the necessary steps to devise a comprehensive strategy to control the invasive weed in the identified area.However,this approach still mainly depends on satellite remote sensing and manual inspection.The cost is high and the accuracy rate and efficiency are low.We acquired color images of the monitoring area in the wild environment using an Unmanned Aerial Vehicle(UAV)and proposed a novel network-MmNet-based on a deep Convolutional Neural Network(CNN)to identify M.micrantha in the images.The network consists of AlexNet Local Response Normalization(LRN),along with the GoogLeNet and continuous convolution of VGG inception models.After training and testing,the identification of 400 testing samples by MmNet is very good,with accuracy of 94.50%and time cost of 10.369 s.Moreover,in quantitative comparative analysis,the proposed MmNet not only has high accuracy and efficiency but also simple construction and outstanding repeatability.Compared with recently popular CNNs,MmNet is more suitable for the identification of M.micrantha in the wild.However,to meet the challenge of wild environments,more M.micrantha images need to be acquired for MmNet training.In addition,the classification labels need to be sorted in more detail.Altogether,this research provides some theoretical and scientific basis for the development of intelligent monitoring and early warning systems for M.micrantha and other invasive species.QIAO Xi LI Yan-zhou SU Guang-yuan TIAN Hong-kun ZHANG Shuo SUN Zhong-yu YANG Long WAN Fang-hao QIAN Wan-qiang 2020Journal of Integrative Agriculture2020,19,5:7
19Rules of anti-infection therapy for sepsis and septic shock显示文摘Objective: Sepsis is a deadly infection that causes injury to tissues and organs. Infection and anti-infective treatment are the eternal themes of sepsis. The successful control of infection is a key factor of resuscitation for sepsis and septic shock. This review examines evidence for the treatment of sepsis. This evidence is combined with clinical experiments to reveal the rules and a standard flowchart of anti-infection therapy for sepsis. Data Sources: We retrieved information from the PubMed database up to October 2018 using various search terms and their combinations, including sepsis, septic shock, infection, antibiotics, and anti-infection. Study Selection: We included data from peer-reviewed journals printed in English on the relationships between infections and antibiotics. Results: By combining the literature review and clinical experience, we propose a 6Rs rule for sepsis and septic shock management: right patients, right time, right target, right antibiotics, right dose, and right source control. This rule encompasses rational decisions regarding the timing of treatment, the identification of the correct pathogen, the selection of appropriate antibiotics, the formulation of a scientifically based antibiotic dosage regimen, and the adequate control of infectious foci. 20Optimization of cantilevered piezoelectric energy harvester with a fixed resonance frequency显示文摘Piezoelectric energy harvesting is widely used to scavenge vibration energy in the environment.For some vibration sources with fixed frequency,cantilevered harvester can generate the energy effectively,so the optimization theory for cantilevered harvester in such an application is needed.In this article,we present the theoretical and experimental studies of the cantilevered piezoelectric energy harvester with a fixed resonance frequency.An analytical model based on energy method is used to estimate the open-circuit voltage and generated energy.Considering that the harvester may be subjected to the static force or steady-state sinusoidal vibration excitation,static and dynamic analysis is performed for device structure to achieve efficient energy.In the analysis,the effects of geometrical dimension on the energy harvesting performance are discussed comprehensively.Eventually,a prototype is designed and fabricated using(1-x)Pb(Mg1/3Nb2/3)O3-xPbTiO3(PMN-PT)single crystal with ultrahigh piezoelectric properties and coupling factor.Performances of the cantilever with different clamped length are evaluated under sinusoidal vibration excitation,proving the good consistency between experimental results and theoretical prediction.The established analysis can provide useful guidelines for the structure design of cantilevered piezoelectric energy harvester with a fixed resonance frequency.LIANG Zhu XU ChunDong REN Bo DI WenNing LI Long LUO HaoSu CHEN Zhen SU Jun 2014Science China(Technological Sciences)2014,57,6:6
