1OTUD5 promotes innate antiviral and antitumor immunity through deubiquitinating and stabilizing STING显示文摘Stimulator of interferon genes(STING)is an adaptor protein that is critical for effective innate antiviral and antitumor immunity.The activity of STING is heavily regulated by protein ubiquitination,which is fine-tuned by both E3 ubiquitin ligases and deubiquitinases.Here,we report that the deubiquitinase OTUD5 interacts with STING,cleaves its K48-linked polyubiquitin chains,and promotes its stability.Consistently,knockout of OTUD5 resulted in faster turnover of STING and subsequently impaired type I IFN signaling following cytosolic DNA stimulation.More importantly,Lyz2-Cre Otud5^(fl/Y) mice and CD11-Cre Otud5^(fl/Y) mice showed more susceptibility to herpes simplex virus type 1(HSV-1)infection and faster development of melanomas than their corresponding control littermates,indicating that OTUD5 is indispensable for STING-mediated antiviral and antitumor immunity.Our data suggest that OTUD5 is a novel checkpoint in the cGAS-STING cytosolic DNA sensing pathway.Yunyun Guo Fei Jiang Lingli Kong Haifeng Wu Honghai Zhang Xiaorong Chen Jian Zhao Baoshan Cai Yanqi Li Chunhong Ma Fan Yi Lei Zhang Bingyu Liu Yi Zheng Lingqiang Zhang Chengjiang Gao 2021Cellular & Molecular Immunology2021,18,8:2

3Modeling the Warming Impact of Urban Land Expansion on Hot Weather Using the Weather Research and Forecasting Model: A Case Study of Beijing, China显示文摘The impacts of three periods of urban land expansion during 1990–2010 on near-surface air temperature in summer in Beijing were simulated in this study, and then the interrelation between heat waves and urban warming was assessed. We ran the sensitivity tests using the mesoscale Weather Research and Forecasting model coupled with a single urban canopy model,as well as high-resolution land cover data. The warming area expanded approximately at the same scale as the urban land expansion. The average regional warming induced by urban expansion increased but the warming speed declined slightly during 2000–2010. The smallest warming occurred at noon and then increased gradually in the afternoon before peaking at around 2000 LST—the time of sunset. In the daytime, urban warming was primarily caused by the decrease in latent heat flux at the urban surface. Urbanization led to more ground heat flux during the day and then more release at night, which resulted in nocturnal warming. Urban warming at night was higher than that in the day, although the nighttime increment in sensible heat flux was smaller. This was because the shallower planetary boundary layer at night reduced the release efficiency of near-surface heat. The simulated results also suggested that heat waves or high temperature weather enhanced urban warming intensity at night. Heat waves caused more heat to be stored in the surface during the day, greater heat released at night, and thus higher nighttime warming. 4ARF-mediated SUMOylation of Apak antagonizes ubiquitylation and promotes its nucleolar accumulation to inhibit 47S pre-rRNA synthesis显示文摘Ribosomes are among the most fundamental molecular machines in all cells,as they are required for protein synthesis.Most structural rRNA components are generated in the nucleolus and assembled into pre-ribosomal particles.Here we show Apak,a previously identified p53 inhibitor,as a novel ribosomal stress response protein.In unstressed cells,Apak is bound to the deSUMOylase SENP1 in the nucleoplasm and targeted for proteasomal degradation by MDM2 ubiquitin ligase.Upon ribosomal stress,SENP1 dissociates fromApak and the tumor suppressor protein ARF couplesUbc9 with Apak to promote Apak SUMOylation on zinc fingers.This results in Apak protein stabilization and translocation to the nucleolus,where Apak inhibits the pre-rRNA synthesis.These findings provide a molecular mechanism whereby ARF coordinates Apak to regulate ribosome biogenesis upon cellular stress.Shan Wang Siying Wang Lihua Yang Hua Guo Xue Kong Lin Yuan Guichun Xing Fuchu He Lingqiang Zhang 2015Journal of Molecular Cell Biology2015,7,2:0
