| 1 | Effects of aminoguanidine on nitric oxide production induced by inflammatory cytokines and endotoxin in cultured rat hepatocytes显示文摘AIM To study the effects of aminoguanidine(AG) and two L-arginine analogues Nω-nitro-L-arginine methyl ester (L-NAME) and Nω-nitro-L-arginine (L-NNA) on nitric oxide (NO) productioninduced by cytokines (TNF-α, IL-11β, and IFN-γ)and bacterial lipopolysaccharide (LPS) mixture(CM) in the cultured rat hepatocytes, andexamine their mechanisms action.METHODS Rat hepatocytes were incubatedwith AG, L-NAME, L-NNA, Actinomycin D (ActD)and dexamethasene in a medium containing CM(LPS plus TNF-α, IL-1β, and IFN-γ) for 24 h. NOproduction in the cultured supernatant wasmeasured with the Griese reaction. IntracellularcGMP level was detected with radioimmunoasey.RESULTS NO production was markedlyblocked by AG and L-NAME in a dose-dependentmanner under inflammatory stimuli conditiontriggered by CM in vitro. The rate of themaximum inhibitory effects of L-NAME (38.9%)was less potent than that obtained with AG(53.7%, P<0.05). There was no significantdifference between the inhibitory effects of AGand two L-arginine analogues on intracellularcGMP accumulation in rat cultured hepatocytes.Non-specific NOS expression inhibitordexamethasone ( DEX ) and iNOS mRNAtranscriptional inhibitor ActD also significantlyinhibited CM-induced NO production. AG(0.1mmol.L-1) and ActD (0.2ng@Lt) wereequipotent in decreasing NO production inducedby inflammatory stimuli in vitro, and botheffects were more potent than that induced bynon-selectivity NOS activity inhibitor L-NAME(0. 1 mmol@ L- 1) under similar stimuli conditions(P | Guo Liang Zhang Ye Hong Wang Hui Ling Teng Zhi Bin Lin Department of Pharmacology,School of Basic Medical Sciences,Beijing University,Beijiog 100083,ChinaDr.Guo Liang Zhang graduated from Xinxiang Medical College in 1982,got Ph.D.at Nagoya City University Medical School,Japan in 1994,finished postdoctoral research at Beijing Medical Univcrsity in 1996,now an associate professor of pharmacology, specialized in hepatic pharmacology, having 15 papers published. | 2001 | World Journal of Gastroenterology2001,7,3: | 20 |
| 2 | Mycoplasma infections and different human carcinomas显示文摘AIM To explore relationships between human carcinomas and mycoplasma infection.METHODS Monoclonal antibody PD4, which specifically recognizes a distinct protein from mycoplasma hyorhinis, was used to detect mycoplasma infection in different paraffinembedded carcinoma tissues with immunohistochemistry. PCR was applied to amplify the mycoplasma DNA from the positive samples for confirming immunohistochemistry.RESULTS Fifty of 90 cases (56%) of gastric carcinoma were positive for mycoplasma hyorhinis. In other gastric diseases, the mycoplasma infection ratio was 28% (18/49) in chronic superficial gastritis, 30% (14/ 46) in gastric ulcer and 37% (18/ 49) in intestinal metaplasia. The difference is significant with gastric cancer (X2=12.06, P<0.05). In colon carcinoma, the mycoplasma infection ratio was 55.1% (32/58), but it was 20.9% (10/49) in adenomarous polyp (X2=13.46, P<0.005).Gastric and colon cancers with high differentiation had a higher mycoplasma infection ratio than those with low differentiation (P< 0.05). Mycoplasma infection in esophageal cancer, lung cancer, breast cancer and glioma was 50.9% (27/53), 52.6% (31/ 59), 39.7%(25/63) and 41% (38/91), respectively. The mycoplasma DNA was successfully amplified with the DNA extracted from the cancer tissues that were positive for mycoplasma infection (detected with antibody PD4).CONCLUSION There was high correlation between mycoplasma infection and different cancers, which suggests the possibility of an association between the two. The mechanism involved in oncogenesis by mycoplasma remains unknown. | Su Huang Ji You Li Jan Wu Lin Meng Cheng Chao Shou Beijing Institute for Cancer Research, Peking University School of Oncology, Beijing 100034. | 4 | microRNAs-powerful repression comes from small RNAs显示文摘microRNAs (miRNAs) encode a novel class of small, non-coding RNAs that regulate gene expression post-trancriptionally. miRNAs comprise one of the major non-coding RNA families, whose diverse bio- logical functions and unusual capacity for gene regulation have attracted enormous interests in the RNA world. Over the past 16 years, genetic, biochemical and computational approaches have greatly shaped the growth of the field, leading to the identification of thousands of miRNA genes in nearly all metazoans. The key molecular machinery for miRNA biogenesis and silencing has been identified, yet the precise biochemical and regulatory mechanisms still remain elusive. However, recent findings have shed new light on how miRNAs are generated and how they function to repress gene expression. miRNAs provide a paradigm for endogenous small RNAs that mediate gene silencing at a genome-wide level. The gene silencing mediated by these small RNAs constitutes a major component of gene regu- lation during various developmental and physiological processes. The accumulating knowledge about their biogenesis and gene silencing mechanism will add a new dimension to our understanding about the complex gene regulatory networks. | MA Cong, LIU YuFei & HE Lin 535 LSA, Division of Cell and Developmental Biology, MCB Department, University of California at Berkeley, Berkeley, CA 94720-3200 | 2009 | Science China(Life Sciences)2009,52,4: | 7 |
| 5 | Sequencing, annotation and comparative analysis of nine BACs of the giant panda(Ailuropoda melanoleuca)显示文摘A 10-fold BAC library for the giant panda was constructed and nine BACs were selected to generate finish sequences. These BACs could be used as a validation resource for the de novo assembly accuracy of the whole genome shotgun sequencing reads of the giant panda newly generated by Illumina GA sequencing technology.Complete Sanger sequencing, assembly, annotation and comparative analysis were carried out on the selected BACs of a joint length 878 kb.Homologue search and de novo prediction methods were used to annotate genes and repeats.Twelve protein coding genes were predicted, seven of which could be functionally annotated. The seven genes have an average gene size of about 41 kb,an average coding size of about 1.2 kb and an average exon number of 6 per gene.Besides, seven tRNA genes were found.About 27 percent of the BAC sequence is composed of repeats.A phylogenetic tree was constructed using a neighbor-join algorithm across five species, including the giant panda, human, dog, cat and mouse, which reconfirms dog as the most closely related species to the giant panda. Our results provide detailed sequence and structure information for new genes and repeats of the giant panda, which will be helpful for further studies about the giant panda. | ZHENG Yang1,2,3,CAI Jing4,LI JianWen3,6,LI Bo3,LIN RunMao3,TIAN Feng3,WANG XiaoLing3 & WANG Jun3,5 1Beijing Institute of Genomics,Chinese Academy of Sciences(CAS),Beijing 100029,China 2College of Life Science,Graduate University of Chinese Academy of Sciences,Beijing 100049,China 3Beijing Genomics Institute at Shenzhen,Shenzhen 518083,China 4CAS-Max Planck Junior Research Group on Evolutionary Genomics,State Key Laboratory of Genetic Resources and Evolution,Kunming Institute of Zoology,Chinese Academy of Sciences(CAS),Kunming 650223,China 5Department of Biology,University of Copenhagen,Copenhagen DK-2200,Denmark 6School of Biological Science and Engineering,South China University of Technology,Guangzhou 510006,China | 2010 | Science China(Life Sciences)2010,53,1: | 5 |
| 6 | 2D/3D-QSAR comparative study on mutagenicity of nitroaromatics显示文摘Nitroaromatics are typical toxic organic pollutants and are ubiquitous in environment with diverse structures. They are widely used in many industries and formed during many natural and anthropogenic processes. Most of these pollutants are potentially carcinogenic and the as-sessment and prediction of the mutagenicity of nitroaromatics are of great interest. In this paper the structure-mutagenicity relationships of 219 nitroaromatics are investigated by molecular or-bital theory based classic structure-activity relationships and comparative molecular field analysis (CoMFA). A comparison is undertaken in respect of the interpretation of mechanism and predic-tive ability for these two categories of QSAR approaches and highly predictive QSAR models have been developed. | WANG Xiaodong1, LIN Zhifen2, YIN Daqiang1, LIU Shushen1 & WANG Liansheng1 1.
