1Complementation of Wild-Type and Drug-Resistant Hepatitis B Virus Genomes to Maintain Viral Replication and Rescue Virion Production under Nucleos(t)ide Analogs显示文摘As the open reading frames of hepatitis B virus(HBV)genomes are overlapping,resistance mutations(MTs)in HBV polymerase may result in stop codon MTs in hepatitis B surface proteins,which are usually detected as a mixed population with wild-type(WT)HBV.The question was raised how the coexistence of nucleos(t)ide analogs(NAs)resistance MTs and WT sequences affects HBV replication.In the present study,HBV genomes with frequently detected reverse transcriptase(RT)/surface truncation MTs,rtA181T/sW172^*,rtV191I/sW182^*and rtM204I/sW196^*,were phenotypically characterized alone or together with their WT counterparts in different ratios by transient transfection in the absence or presence of Nas.In the absence of Nas,RT/surface truncation MTs impaired the expression and secretion of HBV surface proteins,and had a dose-dependent negative effect on WT HBV virion secretion.However,in the presence of Nas,coexistence of MTs with WT maintained viral replication,and the presence of WT was able to rescue the production of MT HBV virions.Our findings reveal that complementation of WT and MT HBV genomes is highly effective under drug treatment.Chunchen Wu Baolin Li Xiaoyong Zhang Kaitao Zhao Yingshan Chen Yifei Yuan Yan Liu Rongjuan Chen Dongping Xu Xinwen Chen Mengji Lu 2019Virologica Sinica2019,34,4:4
2A practical method utilizing multi-spectral LiDAR to aid points cloud matching in SLAM显示文摘Light Detection and Ranging(LiDAR)sensors are popular in Simultaneous Localization and Mapping(SLAM)owing to their capability of obtaining ranging information actively.Researchers have attempted to use the intensity information that accompanies each range measurement to enhance LiDAR SLAM positioning accuracy.However,before employing LiDAR intensities in SLAM,a calibration operation is usually carried out so that the intensity is independent of the incident angle and range.The range is determined from the laser beam transmitting time.Therefore,the key to using LiDAR intensities in SLAM is to obtain the incident angle between the laser beam and target surface.In a complex environment,it is difficult to obtain the incident angle robustly.This procedure also complicates the data processing in SLAM and as a result,further application of the LiDAR intensity in SLAM is hampered.Motivated by this problem,in the present study,we propose a Hyperspectral LiDAR(HSL)-based-intensity calibration-free method to aid point cloud matching in SLAM.HSL employed in this study can obtain an eight-channel range accompanied by corresponding intensity measurements.Owing to the design of the laser,the eight-channel range and intensity were collected with the same incident angle and range.According to the laser beam radiation model,the ratio values between two randomly selected channels’intensities at an identical target are independent of the range information and incident angle.To test the proposed method,the HSL was employed to scan a wall with different coloured papers pasted on it(white,red,yellow,pink,and green)at four distinct positions along a corridor(with an interval of 60 cm in between two consecutive positions).Then,a ratio value vector was constructed for each scan.The ratio value vectors between consecutive laser scans were employed to match the point cloud.A classic Iterative Closest Point(ICP)algorithm was employed to estimate the HSL motion using the range information from the matched point clouds.According to the test results,we found that pink and green papers were distinctive at 650,690,and 720 nm.A ratio value vector was constructed using 650-nm spectral information against the reference channel.Furthermore,compared with the classic ICP using range information only,the proposed method that matched ratio value vectors presented an improved performance in heading angle estimation.For the best case in the field test,the proposed method enhanced the heading angle estimation by 72%,and showed an average 25.5%improvement in a featureless spatial testing environment.The results of the primary test indicated that the proposed method has the potential to aid point cloud matching in typical SLAM of real scenarios.Changhui Jiang Yuwei Chen Wenxin Tian Ziyi Feng Wei Li Chunchen Zhou Hui Shao Eetu Puttonen Juha Hyyppä 2020Satellite Navigation2020,1,1:2

5Microbial community coexisting with harmful alga Karenia mikimotoi and microbial control of algal bloom in laboratory显示文摘Algicidal bacteria have been frequently isolated from algal blooming areas.However,knowledge regarding the microbial communities coexisting with microalgae and their potential application in preventing harmful algal blooms(HABs)is limited.In this study,we investigated the composition of the microbial community coexisting with harmful alga Karenia mikimotoi and its responses to algal control via nutrient stimulation or by adding algicidal strain in microcosms.The microorganisms inhabiting the K.mikimotoi culture consisted of 24 identifi ed phyla,including dominant Proteobacteria(relative abundance 76.24%±7.28%)and Bacteroidetes(22.67%±8.32%).Rhodobacteraceae,Phaeodactylibacter,and Maritimibacter predominated during the algal cultivation.Both the added nutrient and fermentation broth of algicidal strain Pseudoalteromonas QF1 caused a massive death of K.mikimotoi and substantial changes in the coexisting microbial community,in which Rhodobacteraceae and Phaeodactylibacter signifi cantly decreased,while Halomonas and Alteromonas increased.Core operational taxonomic units(OTUs)analysis indicated that 13 OTUs belonging to Rhodobacteraceae,Maritimibacter,Marivita,Nisaea,Phaeodactylibacter,Citreicella,Halomonas,Alteromonas,Marinobacter,Muricauda,and Pseudoalteromonas dominated the changes of the microbial communities observed in the K.mikimotoi culture with or without treatments.Collectively,this study indicated that microbial community inhabiting K.mikimotoi culture includes potential algicidal bacteria,and improves our knowledge about microbial community succession during biocontrol of K.mikimotoi via nutrient stimulation or by adding isolated algicidal strains.Li SUN Peike GAO Yu LI Chao WANG Ning DING Junfeng CHEN Yuhao SONG Chunchen LIU Lun SONG Renjun WANG 2022Journal of Oceanology and Limnology2022,40,3:1
6Recent progress in side chain engineering of Y-series non-fullerene molecule and polymer acceptors显示文摘Organic solar cells(OSCs) have drawn considerable attention in the last decade due to the great potential of light weight,flexibility, and low-cost solution processing. Particularly, Y-series non-fullerene acceptors(NFAs) including small molecular acceptors(SMAs) and polymerized small molecular acceptors(PSMAs) have become research hot spots due to their excellent power conversion efficiency. Side chain engineering is crucial to adjust the solubility and crystallinity of NFAs, which will significantly affect the morphology of active layers and the efficiency of OSCs. However, the understanding of side chain engineering on NFAs is still limited and lacks a systematic review. This review aims to provide a brief summary of the recent developments in side chain engineering of NFAs, with a special focus on the design and application of Y-series SMAs and PSMAs for high-efficiency non-fullerene organic solar cells(NF-OSCs). 9Repurposing of Antazoline Hydrochloride as an Inhibitor of Hepatitis B Virus DNA Secretion显示文摘Hepatitis B virus(HBV) belongs to Hepadnaviridae family and mainly infects hepatocytes, which can cause acute or chronic hepatitis. Currently, two types of antiviral drugs are approved for chronic infection clinically: interferons and nucleos(t)ide analogues. However, the clinical cure for chronic infection is still rare, and it is a huge challenge for all researchers to develop high-efficiency, safe, non-tolerant, and low-toxicity anti-HBV drugs. Antazoline hydrochloride is a first-generation antihistamine with anticholinergic properties, and it is commonly used to relieve nasal congestion and in eye drops. Recently, an in vitro high-throughput evaluation system was constructed to screen nearly 800 compounds from the Food and Drug Administration(FDA)-approved Drug Library. We found that arbidol hydrochloride and antazoline hydrochloride can effectively reduce HBV DNA in the extracellular supernatant in a dose-dependent manner, with EC_(50) of4.321 lmol/L and 2.910 lmol/L in HepAD38 cells, respectively. Moreover, the antiviral effects and potential mechanism of action of antazoline hydrochloride were studied in different HBV replication systems. The results indicate that antazoline hydrochloride also has a significant inhibitory effect on HBV DNA in the extracellular supernatant of Huh7 cells,with an EC_(50) of 2.349 lmol/L. 10ARTICLE OPEN Viral dynamics and antibody responses in people with asymptomatic SARS-CoV-2 infection显示文摘Over 40% of the coronavirus disease 2019(COVID-19)COVID-19 patients were asymptomatically infected with severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)and the immune responses of these asymptomatic individuals is a critical factor for developing the strategy to contain the COVID-19 pandemic.Here,we determined the viral dynamics and antibody responses among 143 asymptomatic individuals identified in a massive screening of more than 5 million people in eight districts of Wuhan in May 2020.Asymptomatic individuals were admitted to the government-designated centralized sites in accordance with policy.The incidence rate of asymptomatic infection is〜2.92/100,000.These individuals had low viral copy numbers(peaked at 315 copies/mL)and short-lived antibody responses with the estimated diminish time of 69 days.The antibody responses in individuals with persistent SARS-CoV-2 infection is much longer with the estimated diminish time of 257 days.These results imply that the immune responses in the asymptomatic individuals are not potent enough for preventing SARS-CoV-2 re-infection,which has recently been reported in recovered COVID-19 patients.This casts doubt on the efficacy of forming'herd-immunity'through natural SARS-CoV-2 infection and urges for the development of safe and effective vaccines.Zhiwei Sui Xinhua Dai Qingbin Lu Yulan Zhang Min Huang Shufen Li Tao Peng Jie Xie Yongzhuo Zhang Chunchen Wu Jianbo Xia Lianhua Dong Jiayi Yang Wenfeng Huang Siyuan Liu Ziquan Wang Ke Li Qingfang Yang Xi Zhou Ying Wu Wei Liu Xiang Fang Ke Peng 2021Signal Transduction and Targeted Therapy2021,6,6:0
