| 3 | Regulation of actin cytoskeleton via photolithographic micropatterning显示文摘Actin cytoskeleton plays crucial roles in various cellular functions.Extracellular matrix(ECM)can modulate cell morphology by remodeling the internal cytoskeleton.To define how geometry of ECM regulates the organization of actin cytoskeleton,we plated individual NIH 3T3 cells on micropatterned substrates with distinct shapes and sizes.It was found that the stress fibers could form along the nonadhesive edges of T-shaped pattern,but were absent from the opening edge of V-shaped pattern,indicating that the organization of actin cytoskeleton was dependent on the mechanical environment.Furthermore,a secondary actin ring was observed on 50μm circular pattern while did not appear on 30μm and 40μm pattern,showing a size-dependent organization of actin cytoskeleton.Finally,osteoblasts,MDCK and A549 cells exhibited distinct organization of actin cytoskeleton on T-shaped pattern,suggesting a cell-type specificity in arrangement of actin cytoskeleton.Together,our findings brought novel insight into the organization of actin cytoskeleton on micropatterned environments. | Fulin Xing Haimei Zhang Mengyu Li Hao Dong Xuehe Ma Shiyu Deng Fen Hu Imshik Lee Leiting Pan Jingjun Xu | 2023 | Journal of Innovative Optical Health Sciences2023,16,2: | 1 |

| 7 | Ultraviolet light A irradiation induces immunosuppression associated with the generation of reactive oxygen species in human neutrophils显示文摘Ultraviolet blood irradiation has been used as a physical therapy to treat many nonspeci¯c diseases in clinics;however,the underlying mechanisms remain largely unclear.Neutrophils,the first line of host defense,play a crucial role in a variety of in°ammatory responses.In the present work,we investigated the effects of ultraviolet light A(UVA)on the immune functions of human neutrophils at the single-cell level by using an inverted°uorescence microscope.N-Formylmethionyl-leucyl-phenylalanine(FMLP),a classic physiological chemotactic peptide,was used to induce a series of immune responses in neutrophils in vitro.FMLP-induced calcium mobilization,migration,and phagocytosis in human neutrophils was significantly blocked after treatment with 365 nm UVA irradiation,demonstrating the immunosuppressive effects of UVA irradiation on neutrophils.Similar responses were also observed when the cells were pretreated with H2O2,a type of reactive oxygen species(ROS).Furthermore,UVA irradiation resulted in an increase in NAD(P)H,a member of host oxidative stress in cells.Taken together,our data indicate that UVA irradiation results in immunosuppression associated with the production of ROS in human neutrophils. | Cunbo Li Xuechen Shi Mincai Chen Guangxue Xu Xinglei Su Pengchong Jiang Leiting Pan | 2016 | Journal of Innovative Optical Health Sciences2016,9,1: | 0 |
| 8 | Regulation of intracellular Ca^2+/CaMKII signaling by TRPV4 membrane translocation during osteoblastic differentiation显示文摘Bone constantly remodels between resorption by osteoclasts and formation by osteoblasts;therefore the functions of osteoblasts are pivotal for maintaining homeostasis of bone mass.Transient receptor potential vanilloid 4(TRPV4),a type of mechanosensitive channel,has been reported to be a key regulator in bone remodeling.Howevei;the relationship between TRPV4 and osteoblast function remains largely elusive.Only little is known about the spatial distribution change of TRPV4 during osteoblastic differentiation and related signal events.Based on three-dimensional super-resolution microscopy,our results clearly showed a different distribution of TRPV4 in undifferentiated and differentiated osteoblasts,which reflected the plasma membrane translocation of TRPV4 along with prolonged differentiation.GSK1016790A(GSK101),the most potent agonist of TRPV4,triggered rapid calcium entry and calmodulin-dependent protein kinase II(CaMKII)phosphorylation via TRPV4 acti・vation in a differentiation-dependent manner,indicating that the abundance of TRPV4 at the cell surface resulting from differentiation may be related to the modulation of Ca2+response and CaMKII activity.These data provide compelling evidences for the plasma membrane translocation of TRPV4 during osteoblastic differentiation as well as demonstrate the regulation of downstream Ca2+/CaMKII signaling. | Fen Hu Yali Zhao Zhenhai Hui Fulin Xing Jianyu Yang Imshik Lee Xinzheng Zhang Leiting Pan Jingjun Xu | 2019 | Biophysics Reports2019,5,5: | 0 |
