2Flavonoid content and radical scavenging activity in fruits of Chinese dwarf cherry(Cerasus humilis) genotypes显示文摘The Chinese dwarf cherry(Cerasus humilis(Bge.) Sok.) is a small shrub with edible fruits. It is native to northern and western China. This species was included as a medicinal plant in the ‘‘Chinese Pharmacopeia'' and has emerged as an economically important crop for fresh fruit consumption, processing into juice and wine and nutraceutical products as well. To gain a better understanding of flavonoid biosynthesis and help develop value added products and better cultivars with greater health benefits, we analyzed total flavonoid content(TFC), composition, and radical scavenging activities in fruit extracts of 16 Chinese dwarf cherry genotypes. Fruit peel TFC ranged from 33.5 to72.8 mg/g REáFW(RE: rutin equivalent, FW: fresh weight)while fruit flesh TFC ranged from 4.3 to 16.9 mg/g REáFW.An HPLC analysis revealed that fruit extracts contained 14 flavonoids with considerable variation in their profiles across genotypes. The most abundant flavonoids in most genotypes were proanthocyanidin B1(PA-B1), proanthocyanidin B2(PA-B2), phloretin 20-O-glucoside(PG), and phloretin 20,40-O-diglucoside(PDG). Principal component analysis showed that PG, PA-B1, and PA-B2 had large,positive factor loading values in the first principal component for each genotype. Increased scavenging activity of2,2-diphenyl-1-picrylhydrazyl(DPPH) radicals was apparent in genotypes ‘Nongda 4', ‘Nongda 3', ‘Nongda 6',‘Wenfenli', and '10-32', suggesting promising applications in the production of nutraceutical products. 3The intragastrointestinal fate of paclitaxel-loaded micelles:Implications on oral drug delivery显示文摘The goal of the present study is to elucidate the intragastrointestinal fate of micellar delivery systems by monitoring fluorescently labeled different micelles and the model drug paclitaxel(PTX).Both in vitro and ex vivo leakage studies showed fast PTX release in fluids while micelles remained intact,except in fedstate simulated intestinal fluid and fasted-state pig intestinal fluid,thus referring to the intact absorption of micelles and PTX leakage in the gastrointestinal tract with D-a-tocopherol polyethylene glycol 1000 succinate(TPGS)micelles showing higher stability than other micelles.All groups of micelles were absorbed intact in Caco-2 and Caco-2/HT29-MTX cell models and the absorption of TPGS micelles was found to be higher than other micelles.The transport of the micelles across Caco-2/Raji(1.6%-3.5%),Caco-2(0.8%-1%),and Caco-2/HT29-MTX(0.58%-1%)cell monolayers further verified the absorption of micelles and their subsequent transport;however,more TPGS micelles transported across cell monolayers than other groups.Moreover,the histological examination also confirmed that micelles ente red the enterocytes and were transported to basolateral tissues and TPGS showed the stronger ability of penetration than other groups.Thus,these results are succinctly presenting the absorption of intact micelles in GIT confirmed by imaging evidence with prior leakage of the drug,upta ke by enterocytes and the transport of micelles that survive the digestion by enterocytes and mainly by microfold cells in material nature dependent way with TPGS showing better results than other groups.In conclusion,these results identify the mechanism by which the gastrointestinal tract processes micelles and point to the likely use of this approach in the design of micelles-based therapies.Iqbal Zoya Haisheng He Luting Wang Jianping Qi Yi Lu Wei Wu 2021Chinese Chemical Letters2021,32,4:3
4Fishbone-like platinum-nickel nanowires as an efficient electrocatalyst for methanol oxidation显示文摘Platinum(Pt)-based electrocatalyst with low Pt content and high electrocatalytic performance is highly desired in fuel cell applications.Herein,we demonstrated that platinum-nickel(Pt-Ni)nanowires with an average composition of PtNi3 and a fishbone structure can be readily synthesized and used as an efficient electrocatalyst toward methanol oxidation reaction(MOR).The PtNi3 fishbone-like nanowires(PtNi3-FBNWs)present features such as richer Pt on the surface than in the bulk,high-index facets on the rough surface,and polyhedral facets at the ends of side chains.Such compositional and structural features could be determinative to the enhanced performance in the electrocatalysis of MOR.Compared with commercial 20%Pt/carbon black(Pt/C),the specific activity and mass activity of the PtNi3-FBNWs are enhanced by approximately 4.76 and 3.02 times,respectively.The stability of electrocatalysis is significantly improved as well.Such comprehensive enhancement indicates that the PtNi3-FBNWs would be a promising candidate toward MOR in fuel cells.Jinquan Chang Luting Song Yuanqing Xu Yanhong Ma Cheng Liang Wenyu Jiang Yong Zhang 2020Nano Research2020,13,1:3
5A new cell death program regulated by toll-like receptor 9 through p38 mitogen-activated protein kinase signaling pathway in a neonatal rat model with sepsis associated encephalopathy显示文摘Background:Sepsis,a serious condition with high mortality,usually causes sepsis associated encephalopathy(SAE)that involves neuronal cell death.However,the cell death programs involved and their underlying mechanisms are not clear.This study aimed to explore the regulatory mechanisms of different cell death programs in SAE.Methods:A neonatal rat model of SAE was established by cecal ligation and perforation.Survival rate and vital signs(mean arterial pressure and heart rate)were monitored,nerve reflexes were evaluated,and cortical pathological changes were observed by hematoxylin and eosin staining.The expression of pyroptosis,apoptosis,and necroptosis(PANoptosis)-related proteins,mitogen-activated protein kinase(MAPK),and its upstream regulator toll-like receptor 9(TLR9)were detected.The expression of TLR9 in neurons was observed by immunofluorescence staining.The ultrastructure of neurons was observed by transmission electron microscope.Results:First,PANoptosis was found in cortical nerve cells of the SAE rats.Meanwhile,the subunits of MAPKs,p38 MAPK,Jun N-terminal kinase,and extracellular signal-regulated kinase(ERK)were activated.After pharmacologically inhibiting each of the subunits,only p38 MAPK was found to be associated with PANoptosis.Furthermore,blocking the p38 MAPK signaling pathway activated necroptosis but inhibited apoptosis and pyroptosis.When necroptosis was pharmacologically inhibited,apoptosis and pyroptosis were reactivated.Finally,we found that the expression of TLR9,a regulator of MAPKs,was significantly increased in this model.After down-regulation of TLR9,p38 MAPK,and ERK signaling pathways were inhibited,which led to the inhibition of PANoptosis.Further analysis found that down-regulation of TLR9 improved the survival rate and reduced the pathological changes in SAE rats.Conclusions:Our study showed that the programs comprising PANoptosis are activated simultaneously in SAE rats.TLR9 activated PANoptosis through the p38 MAPK signaling pathway.TLR9 may work as a potential target for SAE treatment.Ruixi Zhou Junjie Ying Xia Qiu Luting Yu Yan Yue Qian Liu Jing Shi Xihong Li Yi Qu Dezhi Mu 2022Chinese Medical Journal2022,,12:3
