1Soybean AP1 homologs control flowering time and plant height显示文摘Flowering time and plant height are key agronomic traits that directly affect soybean(Glycine max)yield.APETALA1(AP1)functions as a class A gene in the ABCE model for floral organ development,helping to specify carpel,stamen,petal,and sepal identities.There are four AP1 homologs in soybean,all of which are mainly expressed in the shoot apex.Here,we used clustered regularly interspaced short palindromic repeats(CRISPR)–CRISPR-associated protein 9 technology to generate a homozygous quadruple mutant,gmap1,with loss-of-function mutations in all four GmAP1 genes.Under short-day(SD)conditions,the gmap1 quadruple mutant exhibited delayed flowering,changes in flower morphology,and increased node number and internode length,resulting in plants that were taller than the wild type.Conversely,overexpression of GmAP1a resulted in early flowering and reduced plant height compared to the wild type under SD conditions.The gmap1 mutant and the overexpression lines also exhibited altered expression of several genes related to flowering and gibberellic acid metabolism,thereby providing insight into the role of GmAP1 in the regulatory networks controlling flowering time and plant height in soybean.Increased node number is the trait with the most promise for enhancing soybean pod number and grain yield.Therefore,the mutant alleles of the four AP1 homologs described here will be invaluable for molecular breeding of improved soybean yield.Liyu Chen Haiyang Nan Lingping Kong Lin Yue Hui Yang Qingsong Zhao Chao Fang Haiyang Li Qun Cheng Sijia Lu Fanjiang Kong Baohui Liu Lidong Dong 2020Journal of Integrative Plant Biology2020,62,12:9
2Parallel selection of distinct Tof5 alleles drove the adaptation of cultivated and wild soybean to high latitudes显示文摘Photoperiod responsiveness is a key factor limiting the geographic distribution of cultivated soybean and its wild ancestor.In particular,the genetic basis of the adaptation in wild soybean remains poorly understood.In this study,by combining whole-genome resequencing and genome-wide association studies we identified a novel locus,Time of Flowering 5(Tof5),which promotes flowering and enhances adaptation to high latitudes in both wild and cultivated soybean.By genomic,genetic and transgenic analyses we showed that Tof5 en-codes a homolog of Arabidopsis thaliana FRUITFULL(FUL).Importantly,further analyses suggested that different alleles of Tof5 have undergone parallel selection.The Tof5H1 allele was strongly selected by humans after the early domestication of cultivated soybean,while Tof5H2 allele was naturally selected in wild soybean,and in each case facilitating adaptation to high latitudes.Moreover,we found that the key flowering repressor E1 suppresses the transcription of Tof5 by binding to its promoter.In turn,Tof5 physically associates with the promoters of two important FLOWERING LOCUS T(FT),FT2a and FT5a,to upregulate their transcription and promote flowering under long photoperiods.Collectively,ourfindings provide insights into how wild soybean adapted to high latitudes through natural selection and indicate that cultivated soybean underwent changes in the same gene but evolved a distinct allele that was artificially selected after domestication.Lidong Dong Qun Cheng Chao Fang Lingping Kong Hui Yang Zhihong Hou Yongli Li Haiyang Nan Yuhang Zhang Qingshan Chen Chunbao Zhang Kun Kou Tong Su Lingshuang Wang Shichen Li Haiyang Li Xiaoya Lin Yang Tang Xiaohui Zhao Sijia Lu Baohui Liu Fanjiang Kong 2022Molecular Plant2022,15,2:5
3A universal pipeline for mobile mRNA detection and insights into heterografting advantages under chilling stress显示文摘Heterografting has long been used to enhance the chilling tolerance of temperature-sensitive crops,including watermelon,whose mechanism is known to involve bidirectional long-distance mRNA movements.Despite several studies reporting on mobile mRNA(mb-mRNA)profiles in plants,accurate identification of mb-mRNAs is challenging owing to an array of technical problems.Here,we developed a bioinformatical pipeline that took most of the known technical concerns into consideration and is considered to be a universal tool for mb-mRNA detection in heterografts.By applying this pipeline to a commercial watermelon–bottle gourd heterografting system,we detected 130 and 1144 mb-mRNAs upwardly and 167 and 1051 mb-mRNAs downwardly transmitted under normal and chilling-stress conditions,respectively.Quantitative real-time PCR indicated a high accuracy rate(88.2%)of mb-mRNA prediction with our pipeline.We further revealed that the mobility of mRNAs was not associated with their abundance.Functional annotation and classification implied that scions may convey the stress signal to the rootstock,subsequently triggering energy metabolism reprogramming and abscisic acid-mediated stress responses by upward movement of effective mRNAs,ultimately leading to enhanced chilling tolerance.This study provides a universal tool for mb-mRNA detection in plant heterografting systems and novel insights into heterografting advantages under chilling stress.Ying Wang Lingping Wang Nailin Xing Xiaohua Wu Xinyi Wu Baogen Wang Zhongfu Lu Pei Xu Ye Tao Guojing Li Yuhong Wang 2020Horticulture Research2020,7,1:3
4Agronomical selection on loss-of-function of GIGANTEA simultaneously facilitates soybean salt tolerance and early maturity显示文摘Salt stress and flowering time are major factors limiting geographic adaptation and yield productivity in soybean(Glycine max).Although improving crop salt tolerance and latitude adaptation are essential for efficient agricultural production,whether and how these two traits are integrated remains largely unknown.Here,we used a genome-wide association study to identify a major salt-tolerance locus controlled by E2,an ortholog of Arabidopsis thaliana GIGANTEA(GI).Loss of E2 function not only shortened flowering time and maturity,but also enhanced salt-tolerance in soybean.E2 delayed soybean flowering by enhancing the transcription of the core flowering suppressor gene E1,thereby repressing Flowering Locus T(FT)expression.An E2 knockout mutant e2^(CR) displayed reduced accumulation of reactive oxygen species(ROS)during the response to salt stress by releasing peroxidase,which functions in ROS scavenging to avoid cytotoxicity.Evolutionary and population genetic analyses also suggested that loss-of-function e2 alleles have been artificially selected during breeding for soybean adaptation to high-latitude regions with greater salt stress.Our findings provide insights into the coupled selection for adaptation to both latitude and salt stress in soybean;and offer an ideal target for molecular breeding of early-maturing and salt-tolerant cultivars.Lidong Dong Zhihong Hou Haiyang Li Zhaobo Li Chao Fang Lingping Kong Yongli Li Hao Du Tai Li Lingshuang Wang Milan He Xiaohui Zhao Qun Cheng Fanjiang Kong Baohui Liu 2022Journal of Integrative Plant Biology2022,64,10:3

9Development and validation of InDel markers for identification of QTL underlying flowering time in soybean显示文摘Soybean [Glycine max(L.) Merrill] is a major plant source of protein and oil. An accurate and well-saturated molecular linkage map is a prerequisite for forward genetic studies of gene function and for modern breeding for many useful agronomic traits. Next-generation sequence data available in public databases provides valuable information and offers new insights for rapid and efficient development of molecular markers. In this study, we attempted to show the feasibility and facility of using genomic resequencing data as raw material for identifying putative In Del markers. First, we identified 17,613 In Del sites among 56 soybean accessions and obtained 12,619 primer pairs. Second, we constructed a genetic map with a random subset of 2841 primer pairs and aligned 300 polymorphic markers with the 20 consensus linkage groups(LG). The total genetic distance was 2347.3 c M and the number of mapped markers per LG ranged from 10 to 23 with an average of 15 markers. The largest and smallest genetic distances between adjacent markers were 52.3 c M and 0.1 cM, respectively. Finally, we validated the genetic map constructed by newly developed In Del markers by QTL analysis of days to flowering(DTF) under different environments. One major QTL(qDTF4) and four minor QTL(qDTF20, qDTF13, qDTF12,and q DTF11) on 5 LGs were detected. 10Wide-field aurora imager onboard Fengyun satellite:Data products and validation显示文摘New observations of auroras based on the wide-field aurora imager(WAI)onboard Fengyun-3D(FY-3D)satellite are exhibited in this paper.Validity of the WAI data is analyzed by comparing auroral boundaries derived from WAI observations with results obtained from data collected by the Special Sensor Ultraviolet Spectrographic Imager(SSUSI)aboard the Defense Meteorological Satellite Program(DMSP F18).Dynamic variations of the aurora with the solar wind,interplanetary magnetic field(IMF)parameters,and the SYM-H index are also investigated.The comparison of auroral boundaries indicates that the WAI data are morphologically valid and suitable to the study of auroral dynamics.Effective responses to solar wind parameters indicate that the WAI data can be useful to monitor and predict the Earth’s space weather.Since the configuration of aurora is a good indicator of the solar wind–magnetosphere–ionosphere(SW-M-I)coupling system,and can reflect the disturbance of the space environment,the WAI will provide important data to help us to study the physical processes in space.GuangXing Ding JiaWei Li XiaoXin Zhang Fei He LingPing He KeFei Song Liang Sun Shuang Dai ShiJie Liu Bo Chen Chao Yu XiuQing Hu SongYan Gu ZhongDong Yang Peng Zhang 2021Earth and Planetary Physics2021,5,1:1

12Stent-assisted coiling of intracranial carotid ophthalmic segment aneurysm segment aneurysms:Long-term follow-up from a single center显示文摘Background:To evaluate the efficacy of stent-assisted coiling(SAC)for the treatment of carotid ophthalmic segment aneurysm segment aneurysms(OSAs)of the internal carotid artery(ICA)through detailed long-term follow-up of a large patient cohort.Methods:We retrospectively analyzed 88 consecutive patients with OSAs between January 2009 and January 2020 at our center.Angiographic results were evaluated using the modified Raymond grading system and clinical outcomes were evaluated using the mRS scale.The primary endpoints were major aneurysm recurrence and poor clinical outcomes for at least 18 months of follow-up.The patients were asked to attend clinical follow-up assessments and possibly undergo DSA or MR via telephone.Results:We enrolled 88 patients with 99 OSAs treated with coiling,of whom 76 were treated with SAC.The coiling procedures were successful in all 88 patients.Overall,complications occurred in 8 patients(9.1%).No procedure-related mortality was observed.67(76.1%)experienced immediate aneurysm occlusion at the end of the procedure.Long-term angiographic follow-up(18 months)was available in 45/88 aneurysms(51%)(average 18.7±5.2 months).Four patients continued their follow-up for 5 years after initial aneurysm treatment.After a clinical follow-up time of 28.7 months(range,12-51 months),85 patients(95.5%)achieved favorable clinical outcomes(mRS scores of 0-2).Conclusions:This study indicates that SAC treatment is a safe and effective therapeutic alternative for ruptured and unruptured OSAs.The procedural risks are low with relatively long-term effectiveness.Wenquan Gu Geng Zhou Aizada Aldiyarova Tengyue Liu Yi Zhang Weidong Liu Lingping Meng Binxian Gu MingHua Li Ming Su Chen Su Aihua Liu Wu Wang 2023Journal of Interventional Medicine2023,6,3:0
13A recent retrotransposon insertion of J caused E6 locus facilitating soybean adaptation into low latitude显示文摘Soybean(Glycine max) is an important legume crop that was domesticated in temperate regions.Soybean varieties from these regions generally mature early and exhibit extremely low yield when grown under inductive short-day(SD) conditions at low latitudes. The long-juvenile(LJ) trait, which is characterized by delayed flowering and maturity,and improved yield under SD conditions, allowed the cultivation of soybean to expand to lower latitudes. Two major loci control the LJ trait: J and E6. In the current study, positional cloning, sequence analysis, and transgenic complementation confirmed that E6 is a novel allele of J, the ortholog of Arabidopsis thaliana EARLY FLOWERING 3(ELF3). The mutant allele e6^(PG), which carries a Ty1/Copia-like retrotransposon insertion, does not suppress the legume-specific flowering repressor E1, allowing E1 to inhibit Flowering Locus T(FT) expression and thus delaying flowering and increasing yields under SD conditions. The e6^(PG)allele is a rare allele that has not been incorporated into modern breeding programs.The dysfunction of J might have greatly facilitated the adaptation of soybean to low latitudes. 14Dt1 inhibits SWEET-mediated sucrose transport to regulate photoperiod-dependent seed weight in soybean显示文摘Soybean is a photoperiod-sensitive short-day crop whose reproductive period and yield are markedly affected by day-length changes.Seed weight is one of the key traits determining the soybean yield;how-ever,the prominent genes that control the final seed weight of soybean and the mechanisms underlying the photoperiod's effect on this trait remain poorly understood.In this study,we identify SwW19 as a major locus controlling soybean seed weight by QTL mapping and determine Dt1,an orthologous gene of Arabidopsis TFL1 that is known to govern the soybean growth habit,as the causal gene of the SW19 locus.We showed that Dt1 is highly expressed in developing seeds and regulates photoperiod-dependent seed weight in soybean.Further analyses revealed that the Dt1 protein physically interacts with the sucrose transporter GmSWEET10a to negatively regulate the import of sucrose from seed coat to the embryo,thus modulating seed weight under long days.However,Dt1 does not function in seed development under short days due to its very low expression.Importantly,we discovered a novel natural allelic variant of Dt1(H4 haplotype)that decouples its pleiotropic effects on seed size and growth habit;i.e.,this variant remains functional in seed development but fails to regulate the stem growth habit of soybean.Collectively,our findings provide new insights into how soybean seed development responds to photoperiod at different latitudes,offering an ideal genetic component for improving soybean's yield by manipulating its seed weightandgrowth habit.Xiaoming Li Zhonghui Chen Haiyang Li Lin Yue Cuirong Tan Hongjie Liu Yilong Hu Yuhua Yang Xiani Yao Lingping Kong Xiang Huangi Bin Yu Chunyu Zhang Yuefeng Guan Baohui Liu Fanjiang Kong Xingliang Hou 2024Molecular Plant2024,17,3:0

16Apoptotic vesicles rejuvenate mesenchymal stem cells via Rab7-mediated autolysosome formation and alleviate bone loss in aging mice显示文摘Aging skeletons display decreased bone mass,increased marrow adiposity,and impaired bone marrow mesenchymal stem cells(MSCs).Apoptosis is a programmed cell death process that generates a large number of apoptotic vesicles(apoVs).Dysregulated apoptosis has been closely linked to senescence-associated diseases.However,whether apoVs mediate agingrelated bone loss is not clear.In this study,we showed that young MSC-derived apoVs effectively rejuvenated the nuclear abnormalities of aged bone marrow MSCs and restored their impaired self-renewal,osteo-/adipo-genic lineage differentiation capacities via activating autophagy.Mechanistically,apoptotic young MSCs generated and enriched a high level of Ras-related protein 7(Rab7)into apoVs.Subsequently,recipient aged MSCs reused apoV-derived Rab7 to restore autolysosomes formation,thereby contributing to autophagy flux activation and MSC rejuvenation.Moreover,systemic infusion of young MSC-derived apoVs enhanced bone mass,reduced marrow adiposity,and recused the impairment of recipient MSCs in aged mice.Our findings reveal the role of apoVs in rejuvenating aging-MSCs via restoring autolysosome formation and provide a potential approach for treating age-related bone loss.Fangcao Lei Zhiqing Huang Qianmin Ou Jiaqi Li Manqing Liu Lan Ma Lingping Tan Zhengmei Lin Xiaoxing Kou 2023Nano Research2023,16,1:0
