1The Genesis Mechanism of the Mantle Fluid Action and Evolution in the Ore-Forming Process: A Case Study of the Laowangzhai Gold Deposit in Yunnan, China显示文摘Based on petrological studies of the wall rocks, mineralizing rocks, ores and veins from the Laowangzhai gold deposit, it is discovered that along with the development of silication, carbonation and sulfidation, a kind of black opaque ultra-microlite material runs through the spaces between grains, fissures and cleavages. Under observations of the electron microprobe, scanning electronic microscopy and energy spectrum, this kind of ultra-microlite material is confirmed to consist of ultra microcrystalline quartz, silicate, sulfides and carbonates, as well as rutile, scheelite and specularite (magnetite), showing characters of liquation by the analyses of SEM and energy spectrum. The coexistence of immiscibility and precipitating co-crystallization strongly suggests that the mineralizing fluid changed from the melt to the hydrothermal fluid. Combined with the element geochemical researches, it is realized that the ultra-microlite aggregate is the direct relics of the mantle fluid behaving like a melt and supercritical fluid, which goes along with the mantle-derived magma and will escape from the magma body at a proper time. 2C^2G^2FSnake: automatic tongue image segmentation utilizing prior knowledge显示文摘Extraction of the tongue body from digital images is essential for automated tongue diagnoses in traditional Chinese medicine. This paper presents a fully automated active contour initial method that utilizes prior knowledge of the tongue shape and its location in tongue images. Then colorspace information is introduced to control curve evolution. Combining the geometrical Snake model with the parameterized GVFSnake model, a novel approach for automatic tongue segmentation: C2G2FSnake (color control-geometric & gradient flow Snake) is proposed. This method increases the curve velocity but decreases the complexity. C2G2FSnake greatly extends practical usage to tongue segmentation, at the same time increasing the precision. 3Clinical classification and gene mutation of Chinese probands with Charcot-Marie-Tooth disease Analysis of 57 cases显示文摘Charcot-Marie-Tooth(CMT) disease is the most common inherited peripheral neuropathic disorder.CMT is clinically and genetically heterogeneous.To date,27 genes associated with the disease have been cloned.The present study carried out clinical classification according to clinical,electrophysiological and pathological features,conducted inheritance classification according to inheritance patterns,and performed mutation analysis of 13 CMT disease genes(PMP22,CX32,HSPB1,MNF2,MPZ,HSPB8,GDAP1,NFL,EGR2,SIMPLE,RAB7,LMNA,MTMR2) in 57 Chinese probands with CMT.Five cases of AD-CMT1 and 13 cases of sporadic CMT1 were diagnosed as CMT1A;five cases of X-CMT1,one case of X-CMT2 and one case of sporadic CMT1 were diagnosed as CMTX1;four cases of AD-CMT2 were diagnosed as CMT2F;one case of AD-CMT2 and one case of sporadic CMT2 were diagnosed as CMT2A2;one case of AD-CMT2 was diagnosed as CMT2L;one case of AD-CMT2 was diagnosed as CMT2J;one case of AR-CMT1 was diagnosed as CMT4A.Among the 57 CMT probands,seven genotypes were determined among 34 patients,with a detection rate of 59.6%.The results indicated that the clinical classification and inheritance classification are indispensable for selecting potential disease genes for mutation detection,and for efficient molecular diagnosis.Ruxu Zhang Xiaobo Li Xiaohong Zi Shunxiang Huang Fufeng Zhang Kun Xia Qian Pan Beisha Tang 2011Neural Regeneration Research2011,6,9:4
4Detecting Non-Stationarity for Auscultation Signal of Traditional Chinese Medicine显示文摘The information about the nonstationarity of the aus-cultation signal is utilized in this paper to objectively and auto-matically identify healthy people and patients with qi-deficiency or yin-deficiency. In order to characterize the nonstationarity of the sound signal,the nonlinear cross-prediction method is used to extract features from the signal. A feature selection method based on conditional mutual information maximization criterion (CMIM) is implemented to find an optimal feature set. By means of the support vector machine (SVM) classifier,three common states (healthy,qi-deficiency and yin-deficiency) in traditional Chinese medicine are distinguished using the feature set,and a satisfactory classification accuracy of 80% is achieved in the experiment. 5A novel transgenic mouse model of Chinese CharcotMarie-Tooth disease type 2L显示文摘We previously found that the K141N mutation in heat shock protein B8(HSPB8) was responsible for Charcot-Marie-Tooth disease type 2L in a large Chinese family.The objective of the present study was to generate a transgenic mouse model bearing the K141N mutation in the human HSPB8 gene,and to determine whether this K141NHSPB8 transgenic mouse model would manifest the clinical phenotype of Charcot-Marie-Tooth disease type 2L,and consequently be suitable for use in studies of disease pathogenesis.Transgenic mice overexpressing K141NHSPB8 were generated using K141N mutant HSPB8 cDNA cloned into a pCAGGS plasmid driven by a human cytomegalovirus expression system.PCR and western blot analysis confirmed integration of the K141NHSPB8 gene and widespread expression in tissues of the transgenic mice.The K141NHSPB8 transgenic mice exhibited decreased muscle strength in the hind limbs and impaired motor coordination,but no obvious sensory disturbance at 6 months of age by behavioral assessment.Electrophysiological analysis showed that the compound motor action potential amplitude in the sciatic nerve was significantly decreased,but motor nerve conduction velocity remained normal at 6 months of age.Pathological analysis of the sciatic nerve showed reduced myelinated fiber density,notable axonal edema and vacuolar degeneration in K141NHSPB8 transgenic mice,suggesting axonal involvement in the peripheral nerve damage in these animals.These findings indicate that the K141NHSPB8 transgenic mouse successfully models Charcot-Marie-Tooth disease type 2L and can be used to study the pathogenesis of the disease.Ruxu Zhang Fufeng Zhang Xiaobo Li Shunxiang Huang Xiaohong Zi Ting Liu Sanmei Liu Xuning Li Kun Xia Qian Pan Beisha Tang 2014Neural Regeneration Research2014,9,4:1

8Molecular basis of cross-interactions between Aβ and Tau protofibrils probed by molecular simulations显示文摘Amyloid β-protein(Aβ) and Tau, two common pathogenic proteins associated with Alzheimer’s disease(AD), cross-interact, and thus co-assemble into hybrid aggregates. However, molecular mechanism of the cross-interactions remains unclear. To explore the issue, docking and molecular dynamics(MD) simulations were coupled to study the cross-interactions between Aβ pentamer and Tau pentamer. Four stable hybrid decamer conformations including double layer, single layer, block, and part-in were obtained by protein-protein docking software HADDOCK 2.2. Then, MD simulations were used to explore the molecular mechanism of cross-interactions between Aβ pentamer and Tau pentamer. The results of MD simulations showed that the part-in structure was the most stable among all the above four representative ones. The binding energy between Aβ and Tau was about-759.77 kJ·mol-1in the part-in structure. Moreover, the part-in conformation would undergo conformational transition, which would improve its hydrophobicity and make the structure more compact. 9Paclitaxel-induced activation of murine peritoneal macrophage in vitro显示文摘Objective:To study the effects of paclitaxel on macrophage activation.Methods:Mouse macrophages were isolated by peritoneal lavage and cultured in RPMI 1640 medium according to the following groups:paclitaxel(5μmol/L) group,IFN-γ(5U/L) group,paclitaxel (5μmol/L) and IFN-γ (5U/L) combination group, and control group(without paclitaxel and IFN-γ).24 hours later,supernatants were collected for nitric oxide(NO) assessment using the Griess reagent, and tumor necrosis factor-α(TNF-α) assessment using the enzyme linked immunosorbent assay.Antibody-dependent cell-mediated cytotoxicity(ADCC) of the macrophages was assessed using the method of hemoglobin-enzyme release assay (Hb-ERA).Results:Paclitaxel induced the production of higher levels of NO(8.86±1.16μmol/L) and TNF-α(120.2±10.2pg/ml),and enhanced the ADCC of macrophages[(20.61±1.13)%].The differences were significant compared with the control group[no NO and TNF-α detected,ADCC (15.37±1.93)%](P<0.01).Paclitaxel and IFN-γ in combination induced the production of higher levels of NO(22.85±0.91μmol/L) and TNF-α(358.6±27.5pg/ml),and enhanced the ADCC of macrophages[(42.49±3.09)%].The differences were significant compared with paclitaxel or IFN-γ[NO 8.09±1.13μmol/L,TNF-α 124.8±9.6pg/ml,ADCC (23.32±2.63)%] alone(P<0.01).Conclusion:These findings indicate that paclitaxel can promote NO and TNF-α production,enhance ADCC of macrophages,and induce macrophage activation.The active effects are more significant with paclitaxel and IFN-γ combination.Li Zhongxiang Wang Fufeng Qiao Yuhuan 2004现代妇产科进展2004,13,2:0
