| 1 | Ustilaginoidea virens: A Fungus Infects Rice Flower and Threats World Rice Production显示文摘Rice false smut disease, which is caused by the fungus Ustilaginoidea virens, is currently one of the most devastating rice fungal diseases in the world. Rice false smut disease not only causes severe yield loss and grain quality reduction, but also threatens food safety due to its production of mycotoxins. In this review, the most recent progresses regarding the life cycle, infection processes, genome and genetic diversity, pathogenic gene and disease resistance in rice were summarized in order to provide theoretical basis for the control of U. virens. | 2 | Identification of genes contributing to quantitative disease resistance in rice显示文摘Despite the importance of quantitative disease resistance during a plant’s life, little is known about the molecular basis of this type of host-pathogen interaction, because most of the genes underlying resistance quantitative trait loci (QTLs) are unknown. To identify genes contributing to resistance QTLs in rice, we analyzed the colocalization of a set of characterized rice defense-responsive genes and resistance QTLs against different pathogens. We also examined the expression patterns of these genes in response to pathogen infection in the parents of the mapping populations, based on the strategy of validation and functional analysis of the QTLs. The results suggest that defense-responsive genes are important resources of resistance QTLs in rice. OsWRKY45-1 is the gene contributing to a major resistance QTL.NRR,OsGH3-1,and OsGLP members on chromosome 8 contribute alone or collectively to different minor resistance QTLs. | 3 | Warm temperature compromises JA-regulated basal resistance to enhance Magnaporthe oryzae infection in rice显示文摘Changes in global temperatures profoundly affect the occurrence of plant diseases.It is well known that rice blast can easily become epidemic in relatively warm weather.However,the molecular mechanism remains unclear.In this study,we show that enhanced blast development at a warm temperature(22C)compared with the normal growth temperature(28C)is rice plant-determined.Comparative transcriptome analysis revealed that jasmonic acid(JA)biosynthesis and signaling genes in rice could be effectively induced by Magnaporthe oryzae at 28C but not at 22C.Phenotypic analyses of the osaoc1 and osmyc2 mutants,OsCOI1 RNAi lines,and OsMYC2-OE plants further demonstrated that compromised M.oryzaeinduced JA biosynthesis and signaling lead to enhanced blast susceptibility at the warm temperature.Consistent with these results,we found that exogenous application of methyl jasmonate served as an effective strategy for improving blast resistance under the warm environmental conditions.Furthermore,decreased activation of JA signaling resulted in the downregulated expression of some key basal resistance genes at 22C when compared with 28C.Among these affected genes,OsCEBiP(chitin elicitorbinding protein precursor)was found to be directly regulated by OsMYB22 and its interacting protein OsMYC2,a key component of JA signaling,and this contributed to temperature-modulated blast resistance.Taken together,these results suggest that warm temperature compromises basal resistance in rice and enhances M.oryzae infection by reducing JA biosynthesis and signaling,providing potential new strategies for managing rice blast disease under warm climate conditions. | Jiehua Qiu Junhui Xie Ya Chen Zhenan Shen Huanbin Shi Naweed I.Naqvi Qian Qian Yan Liang Yanjun Kou | 2022 | Molecular Plant2022,15,4: | 5 |
| 4 | Putative Phosphatase UvPsr1 Is Required for Mycelial Growth, Conidiation, Stress Response and Pathogenicity in Ustilaginonidea virens显示文摘Ustilaginoidea virens is the causal agent of rice false smut,which can be a highly destructive disease of rice.The plasma membrane phosphatase Psr1 proteins,which act as a regulator of the salinity stress response in yeast,are widely distributed across fungi,but their functional characterization is sketchy.In this study,we characterized the functions of Psr1 protein,UvPsr1,in U.virens.Analyses of the AUvpsr1 and its complementation strain showed that UvPsr1 is required for normal mycelial growth,conidiation and tolerance to oxidative,osmotic and cell wall stresses.When rice panicles were inoculated with the AUvpsr1 strains,no symptoms of false smut disease developed,showing that UvPSR1 also contributes to the pathogenicity of the fungus.The deletion mutant of UvPSR1 also appeared to produce a smaller titer of toxic compounds able to inhibit elongation of the germinated seeds.In conclusion,our results indicated that UvPsr1 is a new pathogenic factor of U.virens. | Xiong Meng Meng Shuai Qiu Jiehua Shi Huanbin Shen Xiangling Kou Yanjun | 2020 | Rice science2020,27,6: | 2 |

| 6 | The E3 ubiquitin ligase OsRGLG5 targeted by the Magnaporthe oryzae effector AvrPi9 confers basal resistance against rice blast显示文摘Rice blast,caused by Magnaporthe oryzae,is one of the most devastating diseases of rice.During infection,M.oryzae secretes effectors to facilitate blast development.Among these effectors,the avirulence factor AvrPi9 is recognized by Pi9,a broad-spectrum blast resistance protein that triggers Pi9-mediated resistance in rice.However,little is known about the interaction between AvrPi9 and Pi9 and how AvrPi9 exerts virulence to promote infection.In this study,we found that ectopic expression of AvrPi9 in the Pi9-lacking cultivar TP309 suppressed basal resistance against M.oryzae.Furthermore,we identified an AvrPi9-interacting protein in rice,which we named OsRGLG5,encoding a functional RING-type E3 ubiquitin ligase.During infection,AvrPi9 was ubiquitinated and degraded by OsRGLG5.Meanwhile,AvrPi9 affected the stability of OsRGLG5.Infection assays revealed that OsRGLG5 is a positive regulator of basal resistance against M.oryzae,but it is not essential for Pi9-mediated blast resistance in rice.In conclusion,our results revealed that OsRGLG5 is targeted by the M.oryzae effector AvrPi9 and positively regulates basal resistance against rice blast. | Zhiquan Liu Jiehua Qiu Zhenan Shen Congcong Wang Nan Jiang Huanbin Shi Yanjun Kou | 2023 | Plant Communications2023,4,5: | 1 |

| 9 | Carbon Catabolite Repressor UvCreA is Required for Development and Pathogenicity in Ustilaginoidea virens显示文摘The rice false smut disease, caused by Ustilaginoidea virens, has emerged as a significantglobal threat to rice production. The mechanism of carbon catabolite repression plays a crucial role in theefficient utilization of carbon nutrients and enzyme regulation in the presence of complex nutritionalconditions. Although significant progress has been made in understanding carbon catabolite repression infungi such as Aspergillus nidulans and Magnaporthe oryzae, its role in U. virens remains unclear. Toaddress this knowledge gap, we identified UvCreA, a pivotal component of carbon catabolite repression,in U. virens. Our investigation revealed that UvCreA localized to the nucleus. Deletion of UvCreA resultedin decreased growth and pathogenicity in U. virens. Through RNA-seq analysis, it was found that theknockout of UvCreA led to the up-regulation of 514 genes and down-regulation of 640 genes. Moreover,UvCreA was found to be involved in the transcriptional regulation of pathogenic genes and genesassociated with carbon metabolism in U. virens. | 10 | Development of plant cytosine base editors with the Cas12a system显示文摘Base editors of the Cas9 system have been widely used for precise nucleotide substitution in crops. In this study, Cas12a was applied to construct plant cytosine base editors(CBEs). The main elements of Cas12aCBEs were engineered and their efficiency was evaluated in stably transformed rice cells. An optimized ttCas12a-hyA3Bctd editor, consisting of a LbCas12a variant carrying catalytic inactive D832A and temperature-tolerance D156R double mutations, a truncated human APOBEC3B deaminase, a human RAD51 single-stranded DNA-binding domain, and double copies of UGI, outperformed other Cas12aCBEs in base editing efficiency. In T0transgenic rice plants, ttCas12a-hyA3Bctd edited an average of42.01% and a maximum of 68.75% of lines at six genomic targets. A-to-G conversions were generated in rice by an adenine base editor with a similar architecture to the optimized CBE.
