| 1 | Antisense MMP-9 RNA inhibits malignant glioma cell growth in vitro and in vivo显示文摘The matrix-degrading metalloproteinases (MMPs), particularly MMP-9, play important roles in the pathogenesis and development of malignant gliomas. In the present study, the oncogenic role of MMP-9 in malignant glioma cells was investigated via antisense RNA blockade in vitro and in vivo. TJ905 malignant glioma cells were transfected with pcDNA3.0 vector expressing antisense MMP-9 RNA (pcDNA-AS-MMP9), which significantly decreased MMP-9 expression, and cell proliferation was assessed. For in vivo studies, U251 cells, a human malignant glioma cell line, were implanted subcutaneously into 4-to 6-week-old BALB/c nude mice. The mice bearing well-established U251 gliomas were treated with intratumoral pcDNA-AS-MMP9-Lipofectamine complex (AS-MMP-9-treated group), subcutaneous injection of endostatin (endostatin-treated group), or both (combined therapy group). Mice treated with pcDNA (empty vector)-Lipofectamine served as the control group. Four or eight weeks later, the volume and weight of tumor, MMP-9 expression, microvessel density and proliferative activity were assayed. We demonstrate that pcDNA-AS-MMP9 significantly decreased MMP-9 expression and inhibited glioma cell proliferation. Volume and weight of tumor, MMP-9 expression, microvessel density and proliferative activity in the antisense-MMP-9-treated and therapeutic alliance groups were significantly lower than those in the control group. The results suggest that MMP-9 not only promotes malignant glioma cell invasiveness, but also affects tumor cell proliferation. Blocking the expression of MMP-9 with antisense RNA substantially suppresses the malignant phenotype of glioma cells, and thus can be used as an effective therapeutic strategy for malignant gliomas. | Cuiyun Sun Qian Wang Hongxu Zhou Shizhu Yu Alain R. | 11 | Konjac glucomannan and xanthan gum as compression coat for colonic drug delivery:experimental and theoretical evaluations显示文摘Compression coated tablets for oral colon specific delivery systems were developed with a mixture polysaccharide of konjac glucomannan(KGM)and xanthan gum(XG)as the compression coat.Diffusion of cimetidine from compression coated tablets was investigated by release experiment in Vitro.0.22U/mLβ-mannanase was applied in the mimic colon solution.The structure of the mixture polysaccharide was studied by an atomic force microscope(AFM).The experimental results indicate that a KGM70 tablet with a 0.4 g coat is of good design,due to a less than 5%drug loss in the mimic upper gastrointestinal solution by the synergistic interaction between XG and KGM,and due to about 50%cumulative release in the mimic colon solution by degradation after 24 hours.The release mechanism and model are discussed based on different periods of drug release including the delay of the drug,the constant release without an enzyme and the delay of degradation.Under hydrolysis byβ-mannanase,drug release from the tablet with KGM coat shows an exponential increase,while that from the dosage with the mixture polysaccharide coat is an approximately zero-order process in which the constant release rate relates to the release velocity of a non-degraded system,the content of KGM within the coat and the average molecular weight ratio of KGM to XG.It was found that XG was the framework of the polysaccharide mixtures by AFM,which is similar to the analysis results from experiments on drug release. | Kang WANG Jiangyang FAN Yanjun LIU Zhimin HE | 2010 | Frontiers of Chemical Science and Engineering2010,4,1: | 0 |

| 13 | Enlarged fins of Tibetan catfish provide new evidence of adaptation to high plateau显示文摘The uplift of the Tibetan Plateau significantly altered the geomorphology and climate of the Euroasia by creating large mountains and rivers.Fishes are more likely to be affected relative to other organisms,as they are largely restricted to river systems.Faced with the rapidly flowing water in the Tibetan Plateau,a group of catfish has evolved greatly enlarged pectoral fins with more numbers of fin-rays to form an adhesive apparatus.However,the genetic basis of these adaptations in Tibetan catfishes remains elusive.In this study,we performed comparative genomic analyses based on the chromosome-level genome of Glyptosternum maculatum in family Sisoridae and detected some proteins with conspicuously high evolutionary rates in particular in genes involved in skeleton development,energy metabolism,and hypoxia response.We found that the hoxd12a gene evolved faster and a loss-of-function assay of hoxd12a supports a potential role for this gene in shaping the enlarged fins of these Tibetan catfishes.Other genes with amino acid replacements and signatures of positive selection included proteins involved in low temperature(TRMU)and hypoxia(VHL)responses.Functional assays reveal that the G.maculatum TRMU allele generates more mitochondrial ATP than the ancestral allele found in low-altitude fishes.Functional assays of VHL alleles suggest that the G.maculatum allele has lower transactivation activity than the low-altitude forms.These findings provide a window into the genomic underpinnings of physiological adaptations that permit G.maculatum to survive in the harsh environment of the Tibetan Himalayas that mirror those that are convergently found in other vertebrates such as humans. | Liandong Yang Ning Sun Honghui Zeng Ying Wang Wenjun Chen Zufa Ding Yang Liu Jing Wang Minghui Meng Yanjun Shen Jingliang Kang Xiuhui Ma Wenqi Lv Juan Chen Axel Meyer Baocheng Guo Shunping He | 2023 | Science China(Life Sciences)2023,66,7: | 0 |
