| 3 | Arachidyl amido cholanoic acid improves liver glucose and lipid homeostasis in nonalcoholic steatohepatitis via AMPK and mTOR regulation显示文摘BACKGROUND Arachidyl amido cholanoic acid(Aramchol)is a potent downregulator of hepatic stearoyl-CoA desaturase 1(SCD1)protein expression that reduces liver triglycerides and fibrosis in animal models of steatohepatitis.In a phase IIb clinical trial in patients with nonalcoholic steatohepatitis(NASH),52 wk of treatment with Aramchol reduced blood levels of glycated hemoglobin A1c,an indicator of glycemic control.AIM To assess lipid and glucose metabolism in mouse hepatocytes and in a NASH mouse model[induced with a 0.1%methionine and choline deficient diet(0.1MCD)]after treatment with Aramchol.METHODS Isolated primary mouse hepatocytes were incubated with 20μmol/L Aramchol or vehicle for 48 h.Subsequently,analyses were performed including Western blot,proteomics by mass spectrometry,and fluxomic analysis with 13C-uniformly labeled glucose.For the in vivo part of the study,male C57BL/6J mice were randomly fed a control or 0.1MCD for 4 wk and received 1 or 5 mg/kg/d Aramchol or vehicle by intragastric gavage for the last 2 wk.Liver metabolomics were assessed using ultra-high-performance liquid chromatography-time of flight-MS for the determination of glucose metabolism-related metabolites.RESULTS Combination of proteomics and Western blot analyses showed increased AMPK activity while the activity of nutrient sensor mTORC1 was decreased by Aramchol in hepatocytes.This translated into changes in the content of their downstream targets including proteins involved in fatty acid(FA)synthesis and oxidation[PACCα/β(S79),SCD1,CPT1A/B,HADHA,and HADHB],oxidative phosphorylation(NDUFA9,NDUFB11,NDUFS1,NDUFV1,ETFDH,and UQCRC2),tricarboxylic acid(TCA)cycle(MDH2,SUCLA2,and SUCLG2),and ribosome(P-p70S6K[T389]and P-S6[S235/S236]).Flux experiments with 13Cuniformely labeled glucose showed that TCA cycle cataplerosis was reduced by Aramchol in hepatocytes,as indicated by the increase in the number of rounds that malate remained in the TCA cycle.Finally,liver metabolomic analysis showed that glucose homeostasis was improved by Aramchol in 0.1MCD fed mice in a dose-dependent manner,showing normalization of glucose,G6P,F6P,UDP-glucose,and Rbl5P/Xyl5P.CONCLUSION Aramchol exerts its effect on glucose and lipid metabolism in NASH through activation of AMPK and inhibition of mTORC1,which in turn activate FAβ-oxidation and oxidative phosphorylation. | David Fernández-Ramos Fernando Lopitz-Otsoa Laura Delacruz-Villar Jon Bilbao Martina Pagano Laura Mosca Maider Bizkarguenaga Marina Serrano-Macia Mikel Azkargorta Marta Iruarrizaga-Lejarreta Jesús Sot Darya Tsvirkun Sebastiaan Martijn van Liempd Felix M Goni Cristina Alonso María Luz Martínez-Chantar Felix Elortza Liat Hayardeny Shelly C Lu JoséM Mato | 2020 | World Journal of Gastroenterology2020,26,34: | 5 |
| 4 | The effects of exercise on cognition in Parkinson’s disease: a systematic review显示文摘Cognitive impairments are highly prevalent in Parkinson’s disease(PD)and can substantially affect a patient’s quality of life.These impairments remain difficult to manage with current clinical therapies,but exercise has been identified as a possible treatment.The objective of this systematic review was to accumulate and analyze evidence for the effects of exercise on cognition in both animal models of PD and human disease.This systematic review was conducted according to the Preferred Reporting Items for Systematic reviews and Meta-Analyses(PRISMA)statement.Fourteen original reports were identified,including six pre-clinical animal studies and eight human clinical studies.These studies used various exercise interventions and evaluated many different outcome measures;therefore,only a qualitative synthesis was performed.The evidence from animal studies supports the role of exercise to improve cognition in humans through the promotion of neuronal proliferation,neuroprotection and neurogenesis.These findings warrant more research to determine what roles these neural mechanisms play in clinical populations.The reports on cognitive changes in clinical studies demonstrate that a range of exercise programs can improve cognition in humans.While each clinical study demonstrated improvements in a marker of cognition,there were limitations in each study,including non-randomized designs and risk of bias.The Grading of Recommendations Assessment,Development and Evaluation(GRADE)system was used and the quality of the evidence for human studies were rated from“low”to“moderate”and the strength of the recommendations were rated from“weak”to“strong”.Studies that assessed executive function,compared to general cognitive abilities,received a higher GRADE rating.Overall,this systematic review found that in animal models exercise results in behavioral and corresponding neurobiological changes in the basal ganglia related to cognition.The clinical studies showed that various types of exercise,including aerobic,resistance and dance can improve cognitive function,although the optimal type,amount,mechanisms,and duration of exercise are unclear.With growing support for exercise to improve not only motor symptoms,but also cognitive impairments in PD,health care providers and policy makers should recommend exercise as part of routine management and neurorehabilitation for this disorder. | Danielle K Murray Matthew A Sacheli Janice J Eng A Jon Stoessl | 2014 | Translational Neurodegeneration2014,3,1: | 3 |

| 6 | Extent of lymphadenectomy has no impact on postoperative complications after gastric cancer surgery in Sweden显示文摘Objective: Curative gastric cancer surgery entails removal of the primary tumor with adequate margins including regional lymph nodes. European randomized controlled trials with recruitment in the 1990's reported increased morbidity and mortality for D2 compared to D1. Here, we examined the extent of lymphadenectomy during gastric cancer surgery and the associated risk for postoperative complications and mortality using the strengths of a population-based study.Methods: A prospective nationwide study conducted within the National Register of Esophageal and Gastric Cancer. All patients in Sweden from 2006 to 2013 who underwent gastric cancer resections with curative intent were included. Patients were categorized into D0, D1, or D1+/D2, and analyzed regarding postoperative morbidity and mortality using multivariable logistic regression.Results: In total, 349(31.7%) patients had a D0, 494(44.9%) D1, and 258(23.4%) D1+/D2 lymphadenectomy.The 30-d postoperative complication rates were 25.5%, 25.1% and 32.2%(D0, D1 and D1+/D2, respectively), and 90-d mortality rates were 8.3%, 4.3% and 5.8%. After adjustment for confounders, in multivariable analysis, there were no significant differences in risk for postoperative complications between the lymphadenectomy groups. For90-d mortality, there was a lower risk for D1 vs. D0.Conclusions: The majority of gastric cancer resections in Sweden have included only a limited lymphadenectomy(D0 and D1).
