1Genetic susceptibility to autoimmune liver disease显示文摘Autoimmune hepatitis(AIH),primary sclerosing cholangitis(PSC) and primary biliary cirrhosis(PBC) are considered as putative autoimmune diseases of the liver.Whereas strong evidence that bacterial infection may trigger PBC exists,the etiologies for PSC and AIH remain unknown.Although there have been significant discoveries of genetic polymorphisms that may underlie the susceptibility to these liver diseases,their associations with environmental triggers and the subsequent implications have been difficult to elucidate.While single nucleotide polymorphisms within the negative costimulatory molecule cytotoxic T lymphocyte antigen 4(CTLA-4) have been suggested as genetic susceptibility factors for all three disorders,we discuss the implications of CTLA-4 susceptibility alleles mainly in the context of PBC,where Novosphingobium aromaticivorans,an ubiquitous alphaproteobacterium,has recently been specifically associated with the pathogenesis of this devastating liver disease.Ultimately,the discovery of infectious triggers of PBC may expand the concept of genetic susceptibility in immune-mediated liver diseases from the concept of aberrant immune responses against self-antigens to insufficient and/or inappropriate immunological defense mechanisms allowing microbes to cross natural barriers,establish infection and damage respective target organs.Jochen Mattner 2011World Journal of Hepatology2011,3,1:3

5The interplay between dendritic cells and CD8 T lymphocytes is a crucial component of SARS-CoV-2 immunity显示文摘SARS-CoV-2,a novel beta-coronavirus(CoV),causes the coronavirus disease 2019(COVID-19)pandemic,which manifests with a wide clinical spectrum ranging from asymptomatic carriage or mild disease to hospitalization and even death.The highly divergent and variable clinical outcomes,as well as the fluctuating involvement of different organ systems,link SARS-CoV-2 pathogenesis to the immune response of an individual host.Thus,detailed knowledge of the cellular and humoral mechanisms underlying antiviral immunity against SARS-CoV-2 is pivotal for understanding the concept of host susceptibility,evaluating therapeutic regimens and designing vaccination strategies.However,most studies so far have focused only on the analysis of immune responses in convalescent individuals.In contrast,Zhou and colleagues explored in detail different innate and adaptive immune cells in the peripheral blood of patients with acute disease in their latest report published in Immunity.Compared to convalescent individuals,they observed that acute SARS-CoV-2 infection quantitatively and qualitatively impairs dendritic cell(DC)and CD8 T-cell responses and interactions,despite rapid and abundant antibody production targeting dominant viral antigen epitopes.Jonas Buttenschön Jochen Mattner 2021Cellular & Molecular Immunology2021,18,2:0
6Intestinal eosinophils:characterization of elusive granulocytes as anti-bacterial and immunomodulatory effector cells in colitis显示文摘In a recent study published in Nature,Gurtner and colleagues identify active eosinophils(A-Eos)as eosinophil subset with anti-bacterial and immune regulatory functions during intestinal disease.1 Due to the combination of human singe cell transcip-tomics with pre-clinical experimental studies,the authors suggest eosinophils as promising targets for clinical intervention in inflammatory bowel disease(IBD).Eosinophils are known to exhibit versatile functions at mucosal surfaces.Thus,they can sustain the intestinal epithelial barrier as well as intestinal tissue architecture,communicate with other cell subsets and regulate local immune responses.2 However,despite having been associated with a panoply of different diseases including eosinophilic esophagitis or IBD,3 their role and plasticity in gastrointestinal disorders have not been well characterized.Tim Holland Jochen Mattner 2023Signal Transduction and Targeted Therapy2023,8,9:0
