1ZmWRKY104 positively regulates salt tolerance by modulating ZmSOD4 expression in maize显示文摘Salinity impairs plant growth, limiting agricultural development. It is desirable to identify genes responding to salt stress and their mechanism of action. We identified a function of the Zea mays WRKY transcription factor, Zm WRKY104, in salt stress response. Zm WRKY104 was localized in the nucleus and showed transcriptional activation activity. Phenotypic and physiological analysis showed that overexpression of Zm WRKY104 in maize increased the tolerance of maize to salt stress and alleviated salt-induced increases in O;accumulation, malondialdehyde(MDA) content, and percent of electrolyte leakage. Further investigation showed that Zm WRKY104 increased SOD activity by regulating Zm SOD4 expression. Yeast onehybrid, electrophoretic mobility shift test, and chromatin immunoprecipitation–quantitative PCR assay showed that Zm WRKY104 bound directly to the promoter of Zm SOD4 by recognizing the W-box motif in vivo and in vitro. Phenotypic, physiological, and biochemical analysis showed that Zm SOD4 increased salt tolerance by alleviating salt-induced increases in O;accumulation, MDA content, and percent of electrolyte leakage under salt stress. 2Lx2-32c,a novel semi-synthetic taxane,exerts antitumor activity against prostate cancer cells in vitro and in vivo显示文摘Tubulin has been shown to be an effective target for the development of cytotoxic agents against prostate cancer. Previously, we reported that Lx2-32 c is an anti-tubulin agent with high binding affinity to tubulin. In this study, we investigated the potential of Lx2-32 c to act as an effective cytotoxic agent in the treatment of prostate cancer. MTT assays showed that Lx2-32 c was cytotoxic to all tested prostate cancer cell lines. The Lx2-32c-treated cells typically exhibited a rounded morphology associated with the onset of apoptosis, as evidenced by immunocytochemical staining. Human prostate cancer cell lines treated with Lx2-32 c arrest in the G2/M phase of the cell cycle and the treatment is associated with an increased ratio of cells in the sub-G0/G1 phase as determined by flow cytometry. Furthermore, expression of the cleaved form of poly(ADP-ribose) polymerase in prostate cancer cell lines treated with Lx2-32 c was shown by Western blotting assay. Xenograft implants of LNCa P and PC3-derived tumors in nude mice showed that Lx2-32 c treatment significant inhibited tumor growth with effects equivalent to those of docetaxel. 3Essential sequence of the N-terminal cytoplasmic localization-related domain of huntingtin and its effect on huntingtin aggregates显示文摘Huntington's disease(HD) is caused by abnormal CAG repeat expansion in the 5′-end of the Huntingtin(HTT) gene.In addition to the canonical C-terminal full-length huntingtin(htt) nuclear export signal,a cytoplasmic localization-related domain(CLRD) in the N-terminus of htt has recently been reported.Here,we analyzed this domain by introducing deletion and substitution mutations in a truncated N-terminal htt protein and subsequently monitored htt expression,aggregation and subcellular localization by immunocytochemistry and Western blot analysis.We demonstrated that Htt4-17 was the essential sequence for htt cytoplasmic localization.We also found that the subcellular distribution of htt was altered when Htt1-17 was mutated to contain amino acids of different charges,suggesting a structural requirement of Htt1-17 for the cytoplasmic localization of htt. 4Genome-wide identification of RNA editing in seven porcine tissues by matched DNA and RNA high-throughput sequencing显示文摘Background: RNA editing is a co/posttranscriptional modification mechanism that increases the diversity of transcripts, with potential functional consequences. The advent of next-generation sequencing technologies has enabled the identification of RNA edits at unprecedented throughput and resolution. However, our knowledge of RNA editing in swine is still limited.Results: Here, we utilized RES-Scanner to identify RNA editing sites in the brain, subcutaneous fat, heart, liver,muscle, lung and ovary in three 180-day-old Large White gilts based on matched strand-specific RNA sequencing and whole-genome resequencing datasets. In total, we identified 74863 editing sites, and 92.1% of these sites caused adenosine-to-guanosine(A-to-G) conversion. Most A-to-G sites were located in noncoding regions and generally had low editing levels. In total, 151 A-to-G sites were detected in coding regions(CDS), including 94 sites that could lead to nonsynonymous amino acid changes. We provide further evidence supporting a previous observation that pig transcriptomes are highly editable at PRE-1 elements. The number of A-to-G editing sites ranged from 4155(muscle) to 25001(brain) across the seven tissues. The expression levels of the ADAR enzymes could explain some but not all of this variation across tissues. The functional analysis of the genes with tissuespecific editing sites in each tissue revealed that RNA editing might play important roles in tissue function.Specifically, more pathways showed significant enrichment in the fat and liver than in other tissues, while no pathway was enriched in the muscle.Conclusions: This study identified a total of 74863 nonredundant RNA editing sites in seven tissues and revealed the potential importance of RNA editing in tissue function. 5CRA-40/Atmosphere—The First-Generation Chinese Atmospheric Reanalysis(1979–2018):System Description and Performance Evaluation显示文摘Atmospheric reanalysis reproduces the past atmospheric conditions through assimilation of historical meteorological observations with fixed version of a numerical weather prediction(NWP)model and data assimilation(DA)system.It is widely used in weather,climate,and even business-related research and applications.This paper reports the development of CMA’s first-generation global atmospheric reanalysis(RA)covering 1979–2018(CRA-40;CRA refers to CMA-RA).CRA-40 is produced by using the Global Spectral Model(GSM)/Gridpoint Statistical Interpolation(GSI)at a 6-h time interval and a TL574 spectral(34-km)resolution with the model top at 0.27 hPa.A large number of reprocessed satellite data and widely collected conventional observations were assimilated during the reanalyzing process,including the reprocessed atmospheric motion vector(AMV)products from FY-2C/D/E/G satellites,dense conventional observations(at about 120 radiosonde and 2400 synoptic stations)over China,as well as MWHS-2 and GNSS-RO observations from FY-3C.The reanalysis fitting to observations is improved over time,especially for surface pressure with root-mean-square error reduced from 1.05 hPa in 1979 to 0.8 hPa,and for upper air temperature from 1.65 K in 1979 to 1.35 K,in 2018.The patterns of global analysis increments for temperature,specific humidity,and zonal wind are consistent with the changes in the observing system.Near surface temperature from the model’s 6-h forecast reflects the global warming trend reasonably.The CRA-40 precipitation pattern matches well with those of GPCP and other reanalyses.CRA-40 also successfully captures the QBO and its vertical and temporal development,hemispherical atmospheric circulation change,and moisture transport by the East Asian summer monsoon.CRA is now operationally running in near real time as a climate data assimilation system in CMA.Zhiquan LIU Lipeng JIANG Chunxiang SHI Tao ZHANG Zijiang ZHOU Jie LIAO Shuang YAO Jingwei LIU Minyan WANG Huiying WANG Xiao LIANG Zhisen ZHANG Yan YAO Ting ZHU Zhe CHEN Wenhui XU Lijuan CAO Hui JIANG Kaixi HU 2023Journal of Meteorological Research2023,37,1:4
6Cell wallβ-1,4-galactan regulated by the BPC1/BPC2-GALS1 module aggravates salt sensitivity in Arabidopsis thaliana显示文摘Salinity severely reduces plant growth and limits agricultural productivity.Dynamic changes and rearrangement of the plant cell wall is an important response to salt stress,but relatively little is known about the biological importance of specific cell wall components in the response.Here,we demonstrate a specific function ofβ-1,4-galactan in salt hypersensitivity.We found that salt stress induces the accumulation ofβ-1,4-galactan in root cell walls by up regulating the expression of GALACTAN SYNTHASE 1(GALS1),which encodes aβ-1,4-galactan synthase.The accumulation ofβ-1,4-galactan negatively affects salt tolerance.Exogenous application of D-galactose(D-Gal)causes an increase inβ-1,4-galactan levels in the wild type and GALS1 mutants,especially in GALS1 overexpressors,which correlated with the aggravated salt hypersensitivity.Furthermore,we discovered that the BARLEY B RECOMBINANT/BASIC PENTACYSTEINE transcription factors BPC1/BPC2 positively regulate plant salt tolerance by repressing GALS1 expression andβ-1,4-galactan accumulation.Genetic analysis suggested that GALS1 is genetically epistatic to BPC1/BPC2 with respect to the control of salt sensitivity as well as accumulation ofβ-1,4-galactan.Taken together,our results reveal a new regulatory mechanism by whichβ-1,4-galactan regulated by the BPC1/BPC2-GALS1 module aggravates salt sensitivity in Arabidopsis thaliana.Jingwei Yan Ya Liu Lan Yang Huan He Yun Huang Lin Fang Henrik Vibe Scheller Mingyi Jiang Aying Zhang 2021Molecular Plant2021,14,3:3

8The landscape of chromatin accessibility in skeletal muscle during embryonic development in pigs显示文摘Background:The development of skeletal muscle in pigs during the embryonic stage is precisely regulated by transcriptional mechanisms,which depend on chromatin accessibility.However,how chromatin accessibility plays a regulatory role during embryonic skeletal muscle development in pigs has not been reported.To gain insight into the landscape of chromatin accessibility and the associated genome-wide transcriptome during embryonic muscle development,we performed ATAC-seq and RNA-seq analyses of skeletal muscle from pig embryos at 45,70 and 100 days post coitus(dpc).Results:In total,21,638,35,447 and 60,181 unique regions(or peaks)were found across the embryos at 45 dpc(LW45),70 dpc(LW70)and 100 dpc(LW100),respectively.More than 91%of the peaks were annotated within−1 kb to 100 bp of transcription start sites(TSSs).First,widespread increases in specific accessible chromatin regions(ACRs)from embryos at 45 to 100 dpc suggested that the regulatory mechanisms became increasingly complicated during embryonic development.Second,the findings from integrated ATAC-seq and RNA-seq analyses showed that not only the numbers but also the intensities of ACRs could control the expression of associated genes.Moreover,the motif screening of stage-specific ACRs revealed some transcription factors that regulate muscle developmentrelated genes,such as MyoG,Mef2c,and Mef2d.Several potential transcriptional repressors,including E2F6,OTX2 and CTCF,were identified among the genes that exhibited different regulation trends between the ATAC-seq and RNA-seq data.Conclusions:This work indicates that chromatin accessibility plays an important regulatory role in the embryonic muscle development of pigs and regulates the temporal and spatial expression patterns of key genes in muscle development by influencing the binding of transcription factors.Our results contribute to a better understanding of the regulatory dynamics of genes involved in pig embryonic skeletal muscle development.Jingwei Yue Xinhua Hou Xin Liu Ligang Wang Hongmei Gao Fuping Zhao Lijun Shi Liangyu Shi Hua Yan Tianyu Deng Jianfei Gong Lixian Wang Longchao Zhang 2021Journal of Animal Science and Biotechnology2021,12,4:2
9Coarsening Behavior of Al_3(Sc, Zr) Precipitates and Its Influence on Recrystallization Temperature of Al-Mg-Sc-Zr Alloy显示文摘Effects of intermediate annealing temperature and time after hot working on recrystallization temperature (Tr) of Al-Mg-Sc-Zr alloy subjected to further cold working were investigated. It was found that Tr of cold worked alloy dropped with the rise of intermediate annealing temperature or extension of annealing time, due to coarsening of Al3(Sc, Zr) particles during intermediate annealing. After annealed at 550°C for 1 h or 475°C for 50 h, Tr is nearly same as that in a Sc, Zr free 5A06 alloy. Coarsening mechanism was also discussed. After hot rolling, residual dislocations promoted diffusion of Sc and Zr atoms at the elevated temperature of the intermediate annealing. 11The salt-activated CBF1/CBF2/CBF3-GALS1 module fine-tunes galactan-induced salt hypersensitivity in Arabidopsis显示文摘Plant growth and development are significantly hampered in saline environments,limiting agricultural productivity.Thus,it is crucial to unravel the mechanism underlying plant responses to salt stress.β-1,4-Galactan(galactan),which forms the side chains of pectic rhamnogalacturonan I,enhances plant sensitivity to high-salt stress.Galactan is synthesized by GALACTAN SYNTHASE1(GALS1).We previously showed that Na Cl relieves the direct suppression of GALS1 transcription by the transcription factors BPC1 and BPC2 to induce the excess accumulation of galactan in Arabidopsis(Arabidopsis thaliana).However,how plants adapt to this unfavorable environment remains unclear.Here,we determined that the transcription factors CBF1,CBF2,and CBF3 directly interact with the GALS1 promoter and repress its expression,leading to reduced galactan accumulation and enhanced salt tolerance.Salt stress enhances the binding of CBF1/CBF2/CBF3 to the GALS1 promoter by inducing CBF1/CBF2/CBF3 transcription and accumulation.Genetic analysis suggested that CBF1/CBF2/CBF3 function upstream of GALS1 to modulate salt-induced galactan biosynthesis and the salt response.CBF1/CBF2/CBF3 and BPC1/BPC2 function in parallel to regulate GALS1 expression,thereby modulating the salt response.Our results reveal a mechanism in which salt-activated CBF1/CBF2/CBF3 inhibit BPC1/BPC2-regulated GALS1 expression to alleviate galactan-induced salt hypersensitivity,providing an activation/deactivation fine-tune mechanism for dynamic regulation of GALS1 expression under salt stress in Arabidopsis.Jingwei Yan Ya Liu Jiawen Yan Zhihui Liu Heqiang Lou Jiasheng Wu 2023Journal of Integrative Plant Biology2023,65,8:2

13Calcineurin B-like protein 5(SiCBL5)in Setaria italica enhances salt tolerance by regulating Na^(+)homeostasis显示文摘Salinity,a major abiotic stress,reduces plant growth and severely limits agricultural productivity.Plants regulate salt uptake via calcineurin B-like proteins(CBLs).Although extensive studies of the functions of CBLs in response to salt stress have been conducted in Arabidopsis,their functions in Setaria italica are still poorly understood.The foxtail millet genome encodes seven CBLs,of which only SiCBL4 was shown to be involved in salt response.Overexpression of SiCBL5 in Arabidopsis thaliana sos3-1 mutant rescued its salt hypersensitivity phenotype,but that of other SiCBLs(SiCBL1,SiCBL2,SiCBL3,SiCBL6,and SiCBL7)did not rescue the salt hypersensitivity of the Atsos3-1 mutant.SiCBL5 harbors an N-myristoylation motif and is located in the plasma membrane.Overexpression of SiCBL5 in foxtail millet increased its salt tolerance,but its knockdown increased salt hypersensitivity.Yeast two-hybrid and firefly luciferase complementation imaging assays showed that SiCBL5 physically interacted with SiCIPK24 in vitro and in vivo.Cooverexpression of SiCBL5,SiCIPK24,and SiSOS1 in yeast conferred a high-salt-tolerance phenotype.Compared to wild-type plants under salt stress conditions,SiCBL5 overexpressors showed lower accumulations of Na^(+) and stronger Na^(+) efflux,whereas RNAi-SiCBL5 plants showed higher accumulations of Na^(+) and weaker Na^(+) efflux.These results indicate that SiCBL5 confers salt tolerance in foxtail millet by modulating Na^(+) homeostasis.Jingwei Yan Lan Yang Ya Liu Yingdi Zhao Tong Han Xingfen Miao Aying Zhang 2022The Crop Journal2022,10,1:1

17Genome-wide search for candidate genes determining vertebrae number in pigs显示文摘Longer porcine carcasses may be expected to have more vertebrae. Therefore, vertebrae number in pigs is an economically important trait. To examine the genetic basis of this trait, we genotyped 578 F_2 Large White ?Minzhu pigs using the Porcine SNP60K Bead Chip. A genome-wide association study(GWAS) identified 36 significant single nucleotide polymorphisms(SNPs) on the chromosomes SSC1(294.28–300.32 Mb) and SSC7(102.22–109.39 Mb). A 6.04-Mb region that contained all 13 significant SNPs on SSC1 also contained the gene NR6A1, previously reported to influence the number of vertebrae in pigs. However, the reported putative casual mutation of NR6A1 c.748 C > T showed no genome-wide significant association with the trait, suggesting it was not a causal mutation in our population. The remaining 23 significant SNPs on SSC7 were concentrated in a 7.17-Mb region, which was within a quantitative trait locus interval for number of vertebrae. TMED10 was the closest gene to the most significant SNP and might be a candidate.Haplotype sharing and block analysis refined the QTL to an interval of about 3 Mb containing 29 candidate genes.Of these 29 genes, the previously reported possible casual mutation of VRTN g.19034 A > C was not found to be a causal mutation in our population. Exploration of these genes via additional genetic and functional studies in mammals revealed that TGFβ3 could be a good candidate on SSC7. 20Spatially distributed damage in sandstone under stress-freeze-thaw coupling conditions显示文摘In the present study,we tried to understand the spatially distributed damage in sandstone samples under the coupled stress-freeze-thaw(SFT)conditions.Firstly,uniaxial compressive stresses(i.e.0 MPa,10 MPa,20 MPa,and 25 MPa)were applied to the samples,and then freeze-thaw(FT)cycles(0,8,16,and 24)were performed on the uniaxially stressed samples to realize the SFT coupling.Next,real-time CT scanning was conducted to observe the induced damage.The total porosity was introduced to quantitatively evaluate the damage degree.The local porosity variation,with the distance from the center of the sandstone sample,was analyzed to understand the spatial distribution of damage.Finally,the coupling effects of SFT on the damage gradient were discussed.The results indicate that the porosity rises with FT cycles,and the applied stresses can accelerate the increase in porosity.The damage increases exponentially with the distance from the center of the sample.The damage presents a spatial gradient distribution,not the commonly used uniform distribution in various studies.The damage gradient increases with FT cycles,and the increasing rate in damage gradient decreases at uniaxial stress of 0 MPa and 10 MPa first,but the increasing rate in damage gradient increases with FT cycles then at stress increasing to 20 MPa.Lifeng Fan Yiding Fan Yan Xi Jingwei Gao 2022Journal of Rock Mechanics and Geotechnical Engineering2022,14,6:1
