1STAT3 mediates resistance of CD44^+CD24^(-/low) breast cancer stem cells to tamoxifen in vitro显示文摘We sought to determine whether STAT3 mediated tamoxifen resistance of breast cancer stem cells in vitro.The capacities for mammosphere formation and STAT3 expression of CD44+CD24-/low MCF-7 and MCF-7 were observed.The CD44+CD24-/low subpopulation ratio and its sensitivity to adriamycin were analyzed in MCF-7 and TAM resistant(TAM-R) cells.Cell cycle,apoptosis,STAT3 and phospho-STAT3 changes were observed af-ter treatment with tamoxifen.Small interference RNA-mediated knockdown of STAT3 in TAM-R cells was also performed.CD44+CD24-/low MCF-7 showed higher capacities for mammosphere formation and STAT3 expression than total MCF-7.The CD44+CD24-/low subpopulation was also upregulated in TAM-R cells with less sensitivity to adriamycin than MCF-7.Cell cycle changes,anti-apoptotic effects and STAT3 changes were also found.Mean-while,the knock-down of STAT3 in TAM-R resulted in an increase in sensitivity to tamoxifen.It is concluded that STAT3 plays an essential role in breast cancer stem cells,which correlated with tamoxifen resistance.Xiaoyan Wang Guozhu Wang Yi Zhao Xiaoan Liu Qiang Ding Jingping Shi Yin Ding Shui Wang 2012The Journal of Biomedical Research2012,26,5:6
2Effects of moxibustion on heat-shock protein 70 expression in the spinal cord and colonic mucosa in a rat model of ulcerative colitis显示文摘Pathological changes in the colon are closely associated with the spinal cord,and innervation of spinal cord can regulate cellular functions.Our previous studies verified that moxibustion protects and restores the colonic mucosa,but the mechanisms of action remain unknown.The present study observed the effects of moxibustion and salicylazosulfapyridine on expression of heat-shock protein 70(HSP70) and its mRNA in the spinal cord and colonic mucosa of ulcerative colitis rats.Results demonstrated that moxibustion and salicylazosulfapyridine increased HSP70 mRNA expression in the spinal cord and colonic mucosa of ulcerative colitis rats.The decreased transcriptional activity of HSP70 in the spinal cord and colonic mucosa might participate in damage to the colonic mucosa in ulcerative colitis rats.Moxibustion exerted protective effects on colonic mucosa by up-regulating HSP70 transcriptional activity in the spinal cord and colonic mucosa.Li Qi Yin Shi Luyi Wu Jingping Mu Linying Tan Xiaopeng Ma Huirong Liu Shifen Xu Huangan Wu 2010Neural Regeneration Research2010,5,22:3
3Non-Hermitian control of confined optical skyrmions in microcavities formed by photonic spin-orbit coupling显示文摘Optical skyrmions formed by photonic spin–orbit(SO)coupling are of significant interest in high-dimensional optical information processing.We report the formation mechanism and non-Hermitian properties of skyrmionlike states in a circular confinement potential with photonic SO coupling,which is preferably realized in a concave-planar microcavity system.We show that the effective photonic gauge field leads to two split manifolds of degenerate skyrmions whose spin textures can be controlled via the non-Hermitian properties by introducing circularly polarized gain and loss,exhibiting dramatically discrepant evolutions at the two sides of the exceptional point(EP).Furthermore,the lifetime degeneracy can be lifted by spatially inhomogeneous pumping according to the non-Hermitian mechanism,enabling the possibility for the skyrmion laser.By introducing shape asymmetry of the confinement potential,a double EP evolution can be achieved,which allows non-Hermitian control of the SO coupled states with higher degrees of freedom.These results open the way for the non-Hermitian control of photonic spin in confined systems,which would be of great significance for the fundamentals of advanced optical information processing.XIAOXUAN LUO YIN CAI XIN YUE WEI LIN JINGPING ZHU YANPENG ZHANG FENG LI 2023Photonics Research2023,11,4:1

6Relationship between the expression of TLR4 and TIRAP and sepsis in peripheral blood显示文摘Objective:To explore the relationship between the expression of TLR4 and TIRAP and sepsis severity.Methods:The study selected 30 healthy examinees as the control group and 53 patients with sepsis as the observation group.The patients in the observation group were assessed by Acute Physiology and Chronic Health Evaluation Ⅱ(APACHE Ⅱ).40 patients with APACHE Ⅱ score≤20,were classified into the low-score sepsis group;13 patients with APACHE Ⅱ score>20,were classified into the high-score sepsis group.The levels of TLR4 and TIRAP in venous serum were detected in all subjects by enzyme-linked immunosorbent assay(ELISA).Results:The levels of TLR4 and TIRAP in serum were 0.886±0.058 ng/ml and 5.216±0.410 ng/ml in the control group;2.253±0.379 ng/ml and 9.540±2.294 ng/ml in the low-score sepsis group;4.494±0.709 ng/ml and 19.206±1.755 ng/ml in the high-score sepsis group.The observation group(low-score and high-score sepsis groups)was significantly different from the control group(p=.000),and the low-score sepsis group was significantly different from the high-score sepsis group(p=.000).With APACHE II score of 20 as the cut-off point,the low-score sepsis group was consisted of low-risk patients and the high-score group was consisted of the high-risk,which can indicate the sepsis severity.According to Pearson’s correlation analysis,the level of TLR4 was positively correlated with sepsis severity(r=0.931,p<.05),the level of TIRAP was also positively correlated with the sepsis severity(r=0.972,p<.05);the level of TLR4 was positively correlated with the level of TIRAP(r=0.936,p<.05).Conclusions:The levels of TLR4 and TIRAP in septic patients can be used to predict and determine the severity of sepsis.Minglu Qi Jingping Yang Xue Yin Si Chen Xiyuan Xu 2016Discussion of Clinical Cases2016,3,4:0
7Transcriptome analysis reveals tumor antigen and immune subtypes of melanoma显示文摘Purpose:To screen potential tumor antigens for melanoma vaccine development and identify different immune subtypes.Methods:Transcriptional data(HTSEQ-FPKM)and clinical information of a 472 Melanoma cohort GDC TCGA Melanoma(SKCM)were downloaded from the UCSC XENA website(http://gffzz797b50e9334d4ed4hcuk596uk09uq69wo.ffgz.tsg.suse.edu.cn/).Subsequently,transcriptome data and clinical information of 210 melanoma cohort GSE65904 were downloaded from Gene Expression Omnibus(GEO),a large global public database.All the transcriptome expression data matrices were log2 transformed for subsequent analysis.GEPIA,TIMER,and IMMPORT databases are also used for analysis.Cell function experiments were performed to validate the role of the IDO1 gene in melanoma cell line A375.Results:Our study provides potential tumor antigens for vaccine development in melanoma patients:GZMB,GBP4,CD79A,APOBEC3F,IDO1,JCHAIN,LAG3,PLA2G2D,XCL2.In addition,we divide melanoma patients into two immune subtypes that have significant differences in tumor immunity and may have different responses to vaccination.In view of the unclear role of IDO1 in melanoma,we selected IDO1 for cell assay validation.Cell function assay showed that IDO1 was significantly overexpressed in the melanoma A375 cell line.After IDO1 knockdown,the activity,invasion,migration and healing ability of A375 cell lines were significantly decreased.Conclusion:Our study could provide a reference for the development of vaccines for melanoma patients.JIAHENG XIE MENGMENG OU PAN YU DAN WU QIKAI TANG YUAN CAO JING HANG LU YIN TINGHONG XIANG MING WANG JINGPING SHI 2023Oncology Research2023,31,3:0
8The Research of Carbon Emission and Carbon Sequestration Potential of Forest Vegetation in China显示文摘The study of China s carbon emissions and carbon sequestration potential is of great practical significance to the formulation of carbon neutrality strategies and methods of China.Carbon emission is an inevitable outcome of the initial stage of economic development,especially the right of developing countries existence and development.The carbon emission of China has been maintained at a low level for a long time and reached the top of the world in 2005.However,per capita carbon emission was still only 46.28%of the United States in 2016.China s total CO_(2) emissions are expected to reach 17-19 Gt/a by 2030.To achieve the goal of carbon neutrality in 2060,the main problems are the low technical capacity of emission reduction and the unreasonable structure of energy consumption.Therefore,replacing coal with gas is one of the most effective ways of emission reduction.By 2060,the carbon sequestration capacity of forest vegetation will reach or exceed 759.14 Mt/a and the CO_(2) sequestration capacity will reach 2783.5 Mt/a.According to that,China s carbon intensity must reduce by 95.39%on the basis of the carbon intensity in 2017,reaching 640 t/100 million yuan.The carbon sequestration capacity of terrestrial vegetation(forest,grassland)will reach or exceed 1380.3 Mt/a and the CO_(2) sequestration capacity will reach 5069.3 Mt/a.According to that,China s carbon intensity must reduce by 91.07%on the basis of the carbon intensity in 2017,reach 1152 t C per 100 million yuan.Yuxing ZHANG Jingping YIN Yao FU Xuejun WANG Guosheng WANG 2021Meteorological and Environmental Research2021,12,6:0
