3VDR基因敲除鼠一氧化氮合酶和内皮素-1基因的表达显示文摘目的探讨维生素D受体(VDR)基因敲除鼠血压变化以及与血压水平密切相关的内皮型一氧化氮合酶、内皮素-1基因在主动脉的表达情况,了解维生素D、维生素D受体与血压的联系及可能机制。方法小鼠血压采用动脉血管插管法直接测定,主动脉内皮型一氧化氮合酶、内皮素-1基因表达采用实时定量聚合酶链式反应检测。结果内皮型一氧化氮合酶、内皮素-1基因均能在野生型正常鼠及维生素D受体基因敲除鼠主动脉表达。与野生型正常鼠比较,纯合子维生素D受体基因敲除鼠收缩压、舒张压明显升高,主动脉内皮型一氧化氮合酶基因表达明显下调,内皮素-1基因表达明显上调(P均<0.05),杂合子维生素D受体基因敲除鼠收缩压、舒张压及主动脉内皮型一氧化氮合酶、内皮素-1基因表达则无明显改变(P均>0.05)。相关性研究发现主动脉内皮型一氧化氮合酶基因表达水平与收缩压、舒张压水平呈负相关,与收缩压、舒张压水平呈正相关(P均<0.05)。结论纯合子维生素D受体基因敲除鼠存在与血压密切相关的主动脉内皮型一氧化氮合酶、内皮素-1基因表达水平明显异常。向伟 蒋湘玲 符生苗 Wei Zheng Uskokovic M Jian Kong YC Li 2005中国热带医学2005,5,5:7

5Current knowledge and future directions of TLR and NOD signaling in sepsis显示文摘The incidence of sepsis is increasing over time, along with an increased risk of dying from the condition. Sepsis care costs billions annually in the United States. Death from sepsis is understood to be a complex process, driven by a lack of normal immune homeostatic functions and excessive production of proinflammatory cytokines, which leads to multi-organ failure. The Toll-like receptor(TLR) family, one of whose members was initially discovered in Drosophila, performs an important role in the recognition of microbial pathogens. These pattern recognition receptors(PRRs), upon sensing invading microorganisms, activate intracellular signal transduction pathways. NOD signaling is also involved in the recognition of bacteria and acts synergistically with the TLR family in initiating an efficient immune response for the eradication of invading microbial pathogens. TLRs and NOD1/NOD2 respond to different pathogenassociated molecular patterns(PAMPs). Modulation of both TLR and NOD signaling is an area of research that has prompted much excitement and debate as a therapeutic strategy in the management of sepsis. 6Age at diagnosis of type 2 diabetes and cardiovascular risk factor profile:A pooled analysis显示文摘BACKGROUND The diagnosis of type 2 diabetes(T2D)in younger adults,an increasingly common public health issue,is associated with a higher risk of cardiovascular complications and mortality,which may be due to a more adverse cardiovascular risk profile in individuals diagnosed at a younger age.AIM To investigate the association between age at diagnosis and the cardiovascular risk profile in adults with T2D.METHODS A pooled dataset was used,comprised of data from five previous studies of adults with T2D,including 1409 participants of whom 196 were diagnosed with T2D under the age of 40 years.Anthropometric and blood biomarker measurements included body weight,body mass index(BMI),waist circumference,body fat percentage,glycaemic control(HbA1c),lipid profile and blood pressure.Univariable and multivariable linear regression models,adjusted for diabetes duration,sex,ethnicity and smoking status,were used to investigate the association between age at diagnosis and each cardiovascular risk factor.RESULTS A higher proportion of participants diagnosed with T2D under the age of 40 were female,current smokers and treated with glucose-lowering medications,compared to participants diagnosed later in life.Participants diagnosed with T2D under the age of 40 also had higher body weight,BMI,waist circumference and body fat percentage,in addition to a more adverse lipid profile,compared to participants diagnosed at an older age.Modelling results showed that each one year reduction in age at diagnosis was significantly associated with 0.67 kg higher body weight[95%confidence interval(CI):0.52-0.82 kg],0.18 kg/m^(2) higher BMI(95%CI:0.10-0.25)and 0.32 cm higher waist circumference(95%CI:0.14-0.49),after adjustment for duration of diabetes and other confounders.Younger age at diagnosis was also significantly associated with higher HbA1c,total cholesterol,low-density lipoprotein cholesterol and triglycerides.CONCLUSION The diagnosis of T2D earlier in life is associated with a worse cardiovascular risk factor profile,compared to those diagnosed later in life.Mary M Barker Francesco Zaccardi Emer M Brady Gaurav S Gulsin Andrew P Hall Joseph Henson Zin ZinHtike Kamlesh Khunti Gerald P McCann Emma L Redman David R Webb Emma G Wilmot Tom Yates Jian Yeo Melanie J Davies Jack A Sargeant 2022World Journal of Diabetes2022,13,3:2
