1Exogenous Application of Abscisic Acid (ABA)Enhances Chilling Tolerance in Seedlings ofNapier Grass (Pennisetum purpureum Schum.)显示文摘Napier grass, an important forage crop with potentials in multi-purpose applications, is widely grown throughout the tropics and subtropics. Low temperature severely limits its productivity and geographical distribution in temperate regions of the world. In this study, we investigated the effect of exogenous abscisic Acid(ABA) on chilling tolerance of napier grass(Pennisetum purpureum Schum.) seedlings. Seven-day-old napier grass seedlings were cultured in dd H_2O or ABA solution at different concentrations and exposed to 1 ℃ for different time durations. The chilling injury, membrane stability index(MSI) and proline content were estimated from leaf samples. The results showed that there was obvious morphological injury of leaf blighting and restrained growth for the seedlings under chilling stress, but this damage can be largely reduced(by 2/3) when the seedlings were treated by 100 μmol/L ABA in the culture solution, and that the application of exogenous ABA can help to maintain a good stability of leaf cell membrane as expressed by a high MSI value and a low level of proline in leaf cells. These results suggested that exogenous ABA can significantly alleviate chilling injury in napier grass seedlings by maintaining the stability of leaf cell membrane during chilling stress, and that the chilling tolerance was not ensured by a proline accumulation although a passive accumulation of proline was observed in the seedlings under chilling stress. 2菌草种质资源AFLP多样性分析显示文摘利用AFLP技术对48份菌草种质资源进行了遗传多样性分析。首先,在海南象草等4份菌草材料上检测了20个AFLP引物组合的扩增情况。然后,从中筛选出6个扩增效果较好的组合用来进一步分析48份菌草种质材料的DNA,共产生了208个多态性条带。用NTSYS-pc 2.1软件结合UPGMA聚类分析方法对数据进行分析,结果显示,48份种质材料间遗传相似系数(SM)在0.48~0.98,在遗传相似系数为0.57时可将48份材料分成4个类群。以上研究结果表明,AFLP分子标记技术的分析结果能直观地表现菌草种质材料之间的亲缘关系,为菌草种质资源的收集和评价提供了一项新的技术手段。梅嘉洺 葛红柳 刘翠翠 朱专为 ALAM Intikhab 阳宴清 卢运海 2017热带农业科学2017,37,11:2
3菌草种质资源iPBS多样性分析(英文)显示文摘[目的]对47份菌草种质资源进行遗传多样性分析.[方法]用iPBS(Inter Primer Binding Site Amplification,引物结合位点间扩增)分子标记技术对47份菌卓种质资源材料进行遗传多样性分析。利用从28个iPBS引物中共挑选出的11个扩增效果较好的iPBS引物,对47份菌草种质材料的DNA进行PCR扩增和电泳分析。[结果]11个iPBS引物在47份菌草种质材料上共给出了208个多态片段,平均每个引物给扩增出18,9个多态片段,聚类分析表明,供试材料之间的遗传相似性系数(SM)分布在0.58~0.99之间,在遗传相似系数0.67时可将47份菌草种质材料分成10大类群,所有47份材料被区分开来。[结论]iPBs分子标记技术可以有效用于菌草种质资源材料的品种鉴定和遗传多样性分析,本研究为科学管理和利用菌草种质资源提供理论指导和技术支撑。李婵 赵永红 梅嘉洺 黄小霞 王咏 朱美兰 Intikhab Alam 阳宴清 卢运海 2016Agricultural Science & Technology2016,17,9:2
4Soil and Rhizosphere Associated Fungi in Gray Mangroves(Avicennia marina) from the Red Sea—A Metagenomic Approach显示文摘Covering a quarter of the world's tropical coastlines and being one of the most threatened ecosystems, mangroves are among the major sources of terrestrial organic matter to oceans and harbor a wide microbial diversity. In order to protect, restore, and better understand these ecosystems, researchers have extensively studied their microbiology, yet few surveys have focused on their fungal communities. Our lack of knowledge is even more pronounced for specific fungal populations, such as the ones associated with the rhizosphere. Likewise, the Red Sea gray mangroves(Avicennia marina) remain poorly characterized, and understanding of their fungal communities still relies on cultivation-dependent methods. In this study, we analyzed metagenomic datasets from gray mangrove rhizosphere and bulk soil samples collected in the Red Sea coast, to obtain a snapshot of their fungal communities. Our data indicated that Ascomycota was the dominant phylum(76%–85%), while Basidiomycota was less abundant(14%–24%), yet present in higher numbers than usually reported for such environments. Fungal communities were more stable within the rhizosphere than within the bulk soil, both at class and genus level. This finding is consistent with the intrinsic patchiness in soil sediments and with the selection of specific microbial communities by plant roots. Our study indicates the presence of several species on this mycobiome that were not previously reported as mangrove-associated. In particular, we detected representatives of several commercially-used fungi, e.g., producers of secreted cellulases and anaerobic producers of cellulosomes. 5基于甘蔗热带种(LA-purple)全基因组序列的SSR开发及特征分析显示文摘现代甘蔗栽培品种(2n=100~130)是由甘蔗热带种(2n=80)与割手密(2n=40~128)种间杂交而来,形成异源多倍体、非整倍体作物,使得甘蔗栽培品种中80%~90%的染色体来源于热带种。开发热带种基因组SSR分子标记,有助于甘蔗遗传多样性分析、分子标记辅助选择、遗传图谱的构建等。本研究基于热带种LA-purple的全基因组测序数据的255 398个预测基因序列(累计总长为1 029 222 285 bp),利用Perl程序与生物信息学软件结合,发掘SSR位点,获得了153 150个SSR位点,平均每1.67个基因有1个SSR位点,其中二、三核苷酸重复基序分别为39 556个和50 072个,占总SSR位点数的58.5%。在二核苷酸重复基序中,TA/AT所占比例最高,占41.4%,CG/GC所占比例最低,占4.6%;在三核苷酸碱基重复基序中,TGT/ACA所占比例最高,为15.6%。在TA/AT重复类型中选取100个基序重复次数在60~90之间的SSR位点,进行引物设计与合成,在12个甘蔗属材料中进行PCR扩增分析,从中筛选出52对具有多态性SSR引物,其中有27对引物在研究的2个甘蔗栽培品种间表现为多态。这些基因组SSR标记的开发,不仅可以用于甘蔗栽培品种DNA指纹图谱分析,而且为甘蔗属不同种的遗传图谱构建、遗传多样性分析和重要性状的遗传机制解析奠定基础,为甘蔗分子育种研究提供重要支撑。王恒波 肖乃衍 朱专为 刘翠翠 Intikhab ALAM 陈平华 卢运海 2018作物学报2018,44,9:2
6First Insights into the Viral Communities of the Deep-sea Anoxic Brines of the Red Sea显示文摘The deep-sea brines of the Red Sea include some of the most extreme and unique environments on Earth. They combine high salinities with increases in temperature, heavy metals,hydrostatic pressure, and anoxic conditions, creating unique settings for thriving populations of novel extremophiles. Despite a recent increase of studies focusing on these unusual biotopes, their viral communities remain unexplored. The current survey explores four metagenomic datasets obtained from different brine–seawater interface samples, focusing specifically on the diversity of their viral communities. Data analysis confirmed that the particle-attached viral communities present in the brine–seawater interfaces were diverse and generally dominated by Caudovirales,yet appearing distinct from sample to sample. With a level of caution, we report the unexpected finding of Phycodnaviridae, which infects algae and plants, and trace amounts of insect-infecting Iridoviridae. Results from Kebrit Deep revealed stratification in the viral communities present in the interface: the upper-interface was enriched with viruses associated with typical marine bacteria,while the lower-interface was enriched with haloviruses and halophages.
