| 2 | Dasatinib inhibits proliferation of liver cancer cells,but activation of Akt/mTOR compromises dasatinib as a cancer drug显示文摘Dasatinib is a multi-target protein tyrosine kinase inhibitor.Due to its potent inhibition of Src,Abl,the platelet-derived growth factor receptor(PDGFR)family kinases,and other oncogenic kinases,it has been investigated as a targeted therapy for a broad spectrum of cancer types.However,its efficacy has not been significantly extended beyond leukemia.The mechanism of resistance to dasatinib in a wide array of cancers is not clear.In the present study,we investigated the effect of dasatinib on hepatocellular carcinoma cell growth and explored the underlying mechanisms.Our results showed that dasatinib potently inhibited the proliferation of SNU-449 cells,but not that of other cell lines,such as SK-Hep-1,even though it inhibited the phosphorylation of Src on both negative and positive regulation sites in all these cells.Dasatinib activated the phosphoinositide-dependent protein kinase1(PDK1)/protein kinase B(Akt)/mammalian target of rapamycin(mTOR)signaling pathway in SK-Hep-1 cells,but not in SNU-449 cells.Blocking the Akt/mTOR signaling pathway strongly promoted the efficacy of dasatinib in SK-Hep-1 cells.In SNU-449 cells,dasatinib promoted apoptosis and the cleavage of caspase-3 and caspase-7,induced cell cycle arrest in the G1 phase,and inhibited the expression of Cyclin-dependent kinase(CDK4)/6/CyclinD1 complex.These findings demonstrate that dasatinib exerts its anti-proliferative effect on hepatocellular cell proliferation by blocking the Src family kinases;however,it causes Akt activation,which compromises dasatinib as an anti-cancer drug. | Chang Liu Xiaoxia Zhu Yuqi Jia Fenqing Chi Keru Qin Jinhong Pei Chan Zhang Xiuli Mu Hongwei Zhang Xiushan Dong Jun Xu Baofeng Yu | 2021 | Acta Biochimica et Biophysica Sinica2021,53,7: | 2 |
| 3 | A dynamic ejection coal burst model for coalmine roadway collapse显示文摘In this study,we established a dynamic ejection coal burst model for a coalmine roadway subject to stress,and held that the stress concentration zone at the roadway side is the direct energy source of this ejection.The formation and development of such burst undergoes three stages:(1)instability and propagation of the cracks in the stress concentration zone,(2)emerging of a layered energy storage structure in the zone,and(3)ejection of coal mass or coal burst due to instability.Moreover,we figured out the initial strength of periodic cracks is parallel to the maximal dominant stress direction in the stress concentration zone and derived from the damage strain energy within the finite area of the zone based on the Griffith energy theory.In addition,we analyzed the formation process of the layered energy storage structure in the zone,simplified it as a simply supported restraint sheet,and calculated the minimum critical load and the internally accumulated elastic energy at the instable state.Furthermore,we established a criterion for occurrence of the coal burst based on the variational principle,and analyzed the coal mass ejection due to instability and coal burst induced by different intensity disturbances.At last,with the stratum conditions of Junde Coalmine as the model prototype,we numerically simulated the load displacement distribution of the stress concentration zone ahead of the working face disturbed by the main roof-fracture-induced dynamic load during the mining process as well as their varying characteristics,and qualitatively verified the above model. | Dazhao Song Xueqiu He Enyuan Wang Zhenlei Li Menghan Wei Hongwei Mu | 2019 | International Journal of Mining Science and Technology2019,29,4: | 1 |
| 4 | Geological characteristics and metallogenic prospect of marine volcanic rock-type copper deposits in eastern Kunlun Mountains area, Xinjiang显示文摘The Katelixi Cu-Zn deposit is a marine volcanic rock-type copper deposit discovered for the first time in the Tokuzidaban Group in eastern Kunlun Mountains area. It is hosted in the Lower Carboniferous Tokuzidaban Group volcanic strata. The orebodies are obviously controlled by the strata and their ore-bearing rocks are a suite of greyish-green mafic tuffs, generally parallel-stratiform, stratoid and lenticular in form, occurring in limestone as well as in the contact between limestone and carbon-bearing siltstone. This ore deposit possesses distinct characteristics of marine volcanic rock sedimentaion. The geological, petrochemical and REE characteristics of its occurrence pro-vide strong evidence suggesting that this deposit is of marine volcanic rock sedimention origin, basically identical to those of some typical marine volcanic rock-type copper deposits in Xinjiang and other parts of China. Marine vol-canic rocks are well developed in the Lower Carboniferous Tokuzidaban strata in eastern Kunlun Mountains area. In addition to this deposit, we have also found a number of copper polymetallic ore deposits or occurrences in associa-tion with marine volcanc activities in many places where there is a good metallogenic prospect. | 5 | Channel Sandstone Architecture Characterization by Seismic Simulation显示文摘To describe the distribution of sandstone reservoirs between wells finely,this paper takes the sandstone reservoir of Layer Nm II-4 in Qinhuangdao 32-6 Oilfield for example,and uses the seismic forward simulation and well loggings to study the channel sandstone reservoir. Under the guidance of the modern rivers and outcrops sandstone distribution pattern,the predicting limitations of the seismic forward simulation about channel sandstone boundaries such as mudstone interlayer between channel sandstone and elevation distance between adjacent channel sandstone are clear. The research shows that the mudstone interlayer can be described by seismic forward simulation seismic when the mudstone interlayer is thicker than 2 m and the channel sandstone is thicker than 10 m because of the appearance of the seismic peak when the main frequency of seismic data is nearly 60 Hz. And the elevation distance between adjacent channel sandstone can be described by the seismic forward simulation when elevation distance is longer than 6 m and the channel sandstone is thicker than 10 m because of the appearance of the seismic peak. And the seismic waveform features of single channel sandstone boundaries such as elevation distance between channels,overbank sandstone and abandoned channels. Under the study mentioned above,the single channel boundaries are described in Layer Nm II-4 of Qinhuangdao 32-6 Oilfield,and the predicting boundaries are confirmed by the chemical tracers. | 8 | Physiological and Molecular Analyses of Low-Salinity Stress Response in the Cuttlefish(Sepia pharaonis)Juveniles显示文摘As a stenohaline species,the survival of Sepia pharaonis can be affected by salinity significantly.This study aimed to explore the function of decreasing salinity on the survival of Sepia pharaonis,which can provide an advanced production guide on the culture of S.pharaonis in the rainy season.Salinity was gradually decreased from 29 to 22 within 48 h to acclimate S.pharaonis to a low-salinity environment.After ten days of breeding under low-salinity of 22,the death rate was high.In this process,changes in tissue and cell structures in the larval liver,biochemical indicators,and osmoregulation-related gene expression were examined.In-terestingly,hepatocytes in the low-salinity group were irregular,had dissolved tissue inclusions,and contained vacuolized cells.There-fore,low salinity caused severe damages at a cellular level that can elevate the mortality rate.A gradual decline in salinity limited the full adaptation of S.pharaonis.Biochemical indicators and osmoregulation-related gene expression changed similarly.For instance,the trend of malondialdehyde(MAD)as a product of lipid peroxidation reflected the degree of damage to the body by free radicals.The antioxidant system of S.pharaonis could cope with oxidative stress caused by the change in salinity to a certain extent.Osmo-regulation-related genes’expression also showed an optimistic result,that is,S.pharaonis responded positively to the change in sali-nity by adjusting the expression of osmoregulation-related genes.Conversely,the increase in mortality at day 10 also proved the weak adaptation capability of S.pharaonis.This study indicated that S.pharaonis can adapt to a low-salinity environment with a li-mited extent. | XIN Hongwei WU Kunlan YUAN Yimeng ZHAO Yun SONG Weiwei WANG Chunlin MU Changkao LI Ronghua | 2022 | Journal of Ocean University of China2022,21,4: | 0 |
