| 1 | Thymic epithelial cell development and differentiation: cellular and molecular regulation显示文摘Thymic epithelial cells (TECs) are one of the most important components in thymic microenvironment supporting thymocyte development and maturation. TECs, composed of cortical and medullary TECs, are derived from a common bipotent progenitor, mediating thymocyte positive and negative selections. Multiple levels of signals including intracellular signaling networks and cell-cell interaction are required for TEC development and differentiation. Transcription factors Foxn1 and autoimmune regulator (Aire) are powerful regulators promoting TEC development and differentiation. Crosstalks with thymocytes and other stromal cells for extrinsic signals like RANKL, CD40L, lymphotoxin, fibroblast growth factor (FGF) and Wnt are also definitely required to establish a functional thymic microenvironment. | 3 | Biocompatible Multifunctional E-Skins with Excellent Self-Healing Ability Enabled by Clean and Scalable Fabrication显示文摘Electronic skins(e-skins) with an excellent sensing performance have been widely developed over the last few decades.However,wearability,biocompatibility,environmental friendliness and scalability have become new limitations. Self-healing ability can improve the long-term robustness and reliability of e-skins. However,self-healing ability and integration are hardly balanced in classical structures of self-healable devices. Here,cellulose nanofiber/poly(vinyl alcohol)(CNF/PVA),a biocompatible moisture-inspired self-healable composite,was applied both as the binder in functional layers and the substrate. Various functional layers comprising particular carbon materials and CNF/PVA were patterned on the substrate. A planar structure was beneficial for integration,and the active self-healing ability of the functional layers endowed self-healed e-skins with a higher toughness. Water served as both the only solvent throughout the fabrication process and the trigger of the self-healing process,which avoids the pollution and bioincompatibility caused by the application of noxious additives. Our e-skins could achieve real-time monitoring of whole-body physiological signals and environmental temperature and humidity. Cross-interference between di erent external stimuli was suppressed through reasonable material selection and structural design. Combined with conventional electronics,data could be transmitted to a nearby smartphone for post-processing. | 4 | Preparation, Characterization and in vitro Release of Chitosan-stavudine Conjugate Nano-prodrug显示文摘Chitosan-stavudine (d4T) conjugate with a succinic spacer was synthesized via carbodiimide coupling reaction and structurally characterized. In order to nanosize it for improving its therapeutic properties, the chitosan-5'-O-succinyl-d4T conjugate was crosslinked with sodium tripolyphosphate (TPP) to obtain the chitosan-d4T conjugate nano-prodrug. The morphologies of chitosan-d4T conjugate nanoparticles were observed by transmission electron microscopy (TEM), and their zeta potential, particle size, and polydispersity (size distribution) were measured by the dynamic light scattering (DLS) techniques. In vitro drug release studies at pH 1.1 and pH 7.4 indicate that the crosslinked chitosan-d4T conjugate nano-prodrug can prevent the coupled d4T from leaking out before entering the target viral reservoirs and provide a mild sustained release without the burst release. | 5 | 3D RNA nanocage for encapsulation and shielding of hydrophobic biomolecules to improve the in vivo biodistribution显示文摘Ribonucleic acid(RNA)nanotechnology platforms have the potential of harboring therapeutics for in vivo delivery in disease treatment.However,the nonspecific interaction between the harbored hydrophobic drugs and cells or other components before reaching the diseased site has been an obstacle in drug delivery.Here we report an encapsulation strategy to prevent such nonspecific hydrophobic interactions in vitro and in vivo based on a self-assembled three-dimensional(3D)RNA nanocage.By placing an RNA three-way junction(3WJ)in the cavity of the nanocage,the conjugated hydrophobic molecules were specifically positioned within the nanocage,preventing their exposure to the biological environment.The assembly of the nanocages was characterized by native polyacrylamide gel electrophoresis(PAGE),atomic force microscopy(AFM),and cryogenic electron microscopy(cryo-EM)imaging.The stealth effect of the nanocage for hydrophobic molecules in vitro was evaluated by gel electrophoresis,flow cytometry,and confocal microscopy.The in vivo sheathing effect of the nanocage for hydrophobic molecules was assessed by biodistribution profiling in mice.The RNA nanocages with hydrophobic biomolecules underwent faster clearance in liver and spleen in comparison to their counterparts.Therefore,this encapsulation strategy holds promise for in vivo delivery of hydrophobic drugs for disease treatment. | Congcong Xu Kaiming Zhang Hongran Yin Zhefeng Li Alexey Krasnoslobodtsev Zhen Zheng Zhouxiang Ji Sijin Guo Shanshan Li Wah Chiu Peixuan Guo | 2020 | Nano Research2020,13,12: | 0 |

| 7 | Heterogeneous distribution of Cardinium in whitefly populations is associated with host nuclear genetic background显示文摘Inherited bacterial symbionts are very common in arthropods, but infection frequency can vary widely among populations. Experiments and interpopulation comparisons suggest that host genetic background might be important in explaining this variation. Our extensive field investigation showed that the infection pattern of the facultative symbiont Cardinium was heterogeneous across geographical populations of the invasive whitefly Bemisia tabaci Mediterranean (MED) in China, with genetic nuclear differences evident in 2 of the populations: 1 with a low infection rate (SD line) and 1 with a high infection rate (HaN line). However, whether the heterogeneous frequency of Cardinium is associated with the host genetic background remains poorly understood. Here, we compared the fitness of the Cardinium-infected and uninfected sublines with similar nuclear genetic backgrounds from SD and HaN lines, respectively, and further determine whether host extranuclear or nuclear genotype influenced the Cardinium–host phenotype by performing 2 new introgression series of 6 generations between SD and HaN lines (i.e., Cardinium-infected females of SD were backcrossed with uninfected males of HaN, and vice versa). The results showed that Cardinium provides marginal fitness benefits in the SD line, whereas Cardinium provides strong fitness benefits in the HaN line. Further, both Cardinium and the Cardinium–host nuclear interaction influence the fecundity and pre-adult survival rate of B. tabaci, whereas the extranuclear genotype does not.
