2Overexpression of Slug is associated with malignant progression of esophageal adenocarcinoma显示文摘AIM: To characterise expression of known E-cadherin repressors; Snail, Slug and Twist in the development of esophageal adenocarcinoma. METHODS: E-cadherin, Slug, Snail and Twist mRNA expression in Barrett's metaplasia and esophageal adenocarcinoma specimens was examined by real-time reverse transcription-polymerase chain reaction (RT-PCR). Semi-quantitative immunohistochemistry was used to examine cellular localisation and protein levels. The effect of Slug on epithelial mesenchymal transition (EMT) markers was examined by transfection of Slug into an adenocarcinoma line OE33.RESULTS: Cellular localisation of Slug in Barrett's metaplasia was largely cytoplasmic whilst in adenocarcinoma it was nuclear. Semi-quantitative analysis indicated that Slug was more abundant in adenocarcinoma compared to matched Barrett's metaplastic specimens. Snail and Twist were expressed in adenocarcinoma but were cytoplasmic in location and not induced compared to Barrett's mucosa. These observations were supported by mRNA studies where only Slug mRNA was shown to be over-expressed in adenocarcinoma and inversely correlated to E-cadherin expression. 3湿邪致病大鼠相关T淋巴细胞受体β链可变区基因谱系的研究显示文摘目的:采用实时荧光定量PCR方法检测湿邪致病大鼠(TCRVβ)亚家族基因表达谱,从免疫识别的角度探讨湿邪致病'蒙蔽性'特点。方法:将32只SD大鼠按随机数字表随机分为正常对照组、内湿大鼠模型组、外湿大鼠模型组和内外湿大鼠模型组4组,造模20d后,各组按随机数字表选3只大鼠,检测其TCRVβ亚家族表达水平。结果:与正常对照组比较,外湿大鼠模型组的TCRVβ1、TCRVβ7、TCRVβ9、TCRVβ13表达升高(P<0.05,P<0.01,P<0.001)。与正常对照组比较,内湿大鼠模型组的TCRVβ8、TCRVβ14、TCRVβ18表达降低(P<0.05,P<0.01),内外湿大鼠模型组的TCRVβ8、TCRVβ18表达也降低(P<0.05,P<0.01)。结论:在外湿大鼠模型组部分TCRVβ亚家族比正常对照组表达升高,而在内湿大鼠模型组及内外湿大鼠模型组中部分TCRVβ比正常对照组表达低下,提示内湿之邪可能具有'蒙蔽性'有关,在内湿的环境中,机体可能对外在相关抗原相对不敏感,进而导致湿邪致病大鼠免疫功能障碍而死亡率升高,其机制值得进一步研究。Carlini Fan Hardi 张诗军 陈泽雄 朱宝益 王伟 胡斌 2010中华中医药杂志2010,25,2:14
4Regulation of AMP-activated protein kinase by natural and synthetic activators显示文摘The AMP-activated protein kinase(AMPK)is a sensor of cellular energy status that is almost universally expressed in eukaryotic cells.While it appears to have evolved in single-celled eukaryotes to regulate energy balance in a cell-autonomous manner,during the evolution of multicellular animals its role has become adapted so that it also regulates energy balance at the whole body level,by responding to hormones that act primarily on the hypothalamus.AMPK monitors energy balance at the cellular level by sensing the ratios of AMP/ATP and ADP/ATP,and recent structural analyses of the AMPK heterotrimer that have provided insight into the complex mechanisms for these effects will be discussed.Given the central importance of energy balance in diseases that are major causes of morbidity or death in humans,such as type 2 diabetes,cancer and inflammatory disorders,there has been a major drive to develop pharmacological activators of AMPK.Many such activators have been described,and the various mechanisms by which these activate AMPK will be discussed.A particularly large class of AMPK activators are natural products of plants derived from traditional herbal medicines.While the mechanism by which most of these activate AMPK has not yet been addressed,I will argue that many of them may be defensive compounds produced by plants to deter infection by pathogens or grazing by insects or herbivores,and that many of them will turn out to be inhibitors of mitochondrial function.David Grahame Hardie 2016Acta Pharmaceutica Sinica B2016,6,1:11
5刺梨自然发酵过程中非酿酒酵母多样性分析显示文摘【目的】分析刺梨果实自然发酵过程中非酿酒酵母菌群特征,为筛选优质刺梨非酿酒酵母提供参考。【方法】基于Illumina MiSeq高通量测序技术和WL营养琼脂鉴定培养基纯种分离技术,分析刺梨果实自然发酵1 d(F1)、3 d(F3)、5 d(F5)和15 d(F15)4个阶段及YPD培养基富集培养样本中非酿酒酵母种群组成和多样性。【结果】高通量测序分析结果共获得182个OTUs(operational taxonomic units,OTUs),归属于81个属107个种;物种多样性分析结果表明,刺梨果实自然发酵前期,优势非酿酒酵母为汉逊酵母(Hanseniasporasp.)和伯顿丝孢毕赤酵母(Hyphopichiaburtonii),二者在样本F1中分别占42.59%和26.85%;随着自然发酵的不断进行,二者的比例逐渐降低,在第15天(F15),Hanseniaspora sp.和H.burtonii比例降低至7.73%和0.52%。相反,Pichia sporocuriosa和未培养的酵母,随着自然发酵不断进行所占比例逐渐增大,分别由F1中的0.23%和0.33%增至F15中的37.26%和32.62%。此外,采用WL营养琼脂鉴定培养基纯种分离和鉴定技术,从刺梨上分离到Hanseniasporasp.、H.burtonii、克鲁维毕赤酵母(Pichia kluyveri)、P.sporocuriosa和异常威克汉姆酵母(Wickerhamomyces anomalus)5种类型的可培养非酿酒酵母。【结论】刺梨果实上存在着丰富的非酿酒酵母菌资源,研究刺梨自然发酵过程中非酿酒酵母多样性,为酵母资源开发和利用奠定基础。刘晓柱 李银凤 于志海 HARDIE William James 黄名正 2020微生物学报2020,60,8:10
6Micro-Channel Heat Sink:A Review显示文摘The method to cool a high heat flux device is an important research direction for the heat exchanger design.Micro-channels are an eflfective heat exchange structure both for single-phase and two-phase flow.In this paper,the heat transfer correlations of single-phase,two-phase and nanofluid in a micro-channel are discussed and analyzed.The correlations of pressure drop for single-phase and two-phase fluids are also presented.Excluding the different working fluids used in the micro-channel,the diameter and aspect ratio,shape and structure,surface roughness,internal and external factor and layout of micro-channel pipe are considered to analyze their influence on the heat transfer performance and pressure drop.Micro-channel technology applications include industry,air-conditioning,solar energy systems,heat pipe technology and computer data center cooling.Compared to the conventional heat exchangers used in these fields,a micro-channel heat sink showed a much better heat transfer coefficient and low volume,indicating that it is a good choice and has huge potential for cooling application.Finally,existing problems and future scopes are described,and drawing up design standard,experimental and simulated methods for evaluating its performance are the urgent actions which need to be carried out.This review paper serves as guidance for researchers to design and predict the performance of micro-channel heat sinks.ZHOU Jinzhi CAO Xiaoling ZHANG Nan YUAN Yanping ZHAO Xudong HARDY David 2020Journal of Thermal Science2020,29,6:9
7Indicators for prediction of Mycobacterium tuberculosis positivity detected with bronchoalveolar lavage fluid显示文摘Background:The diagnosis of active pulmonary tuberculosis(TB)remains a challenge in clinic,especially for sputum negative pulmonary TB.Bronchoalveolar lavage fluid(BALF)has higher sensitivity than sputum for detection of Mycobacterium tuberculosis(Mtb).However,bronchoscopy is invasive and costly,and not suitable for all patients.In order to make TB patients get more benefit from BALF for diagnosis,we explore which indicator might be used to optimize the choice of bronchoscopy.Methods:A total of 1539 sputum-smear-negative pulmonary TB suspects who underwent bronchoscopy were recruited for evaluation.The sensitivity,specificity and accuracy of Mtb detection in sputum and BALF were compared.Odds ratios and 95%confidence intervals were used to assess variables that associated with positive acid-fast bacilli(AFB)smear,Mtb culture and nucleic acid amplification test(NAAT)of BALF in sputum-negative and non-sputum-producing pulmonary TB suspects.Results:BALF has significantly higher sensitivity(63.4%)than sputum(43.5%)for Mtb detection by culture and NAAT.19.7%(122/620)sputum-negative and 40.0%(163/408)non-sputum-producing suspects had positive bacteriological results in BALF.Among sputum-negative and non-sputum-producing pulmonary TB suspects,the positivity of Mtb detection in BALF is associated with a younger age,the presence of pulmonary cavities and a positive result of interferon-gamma release assay(IGRA).Sputum-negative patients under 35 years old with positive IGRA and pulmonary cavity had 84.8%positivity of Mtb in BALF.Conclusions:Our study indicated that combination of age,the presence of pulmonary cavity,and the result of IGRA is useful to predict the positivity of Mtb detection in BALF among sputum-negative and non-sputum producing pulmonary TB suspects.Those who are under 35 years old,positive for the presence of pulmonary cavity and IGRA,should undergo bronchoscopy to collect BAFL for Mtb tests,as they have the highest possibility to get bacteriologically confirmation of TB.Xi Liu Xing-Fang Hou Lei Gao Guo-Fang Deng Ming-Xia Zhang Qun-Yi Deng Tao-Sheng Ye Qian-Ting Yang Bo-Ping Zhou Zhi-Hua Wen Hai-Ying Liu Hardy Kornfeld Xin-Chun Chen 2018Infectious Diseases of Poverty2018,7,1:9
8An electron microscopic-cytochemical localization of plasma membrane Ca^(2+)-ATPase activity in poplar apical bud cells during the induction of dormancy by short-day photoperiods显示文摘Plasma membrane (PM) Ca2+-ATPase activity in poplar apical bud meristematic cells during short-day (SD)-induced dormancy development was examined by a cerium precipitation EM-cytochemical method. Ca2+-ATPase activity, indicated by the status of cerium phosphate precipitated grains, was localized mainly on the interior face (cytoplasmic side) of the PM when plants were grown under long days and reached a deep dormancy. A few reaction products were also observed on the nuclear envelope.When plant buds were developing dormancy after 28 to 42 d of SD exposure, almost no reaction products were present on the interior face of the PM. In contrast, a large number of cerium phosphate precipitated grains were distributed on the exterior face of the PM. After 70 d of SD exposure, when buds had developed a deep dormancy, the reaction products of Ca2+-ATPase activity again appeared on the interior face of the PM. The results seemed suggesting that two kinds of Ca2+-ATPases may be present on the PM during the SD-induced dormancy in poplar.One is the Ca2+-punlping ATPase, which is located on the interior face of the PM, for maintaining and restoring the Ca2+homeostasis. 10Hierarchical activation of compartmentalized pools of AMPK depends on severity of nutrient or energy stress显示文摘AMPK, a master regulator of metabolic homeostasis, is activated by both AMP-dependent and AMP-independent mechanisms. The conditions under which these different mechanisms operate, and their biological implications are unclear. Here, we show that, depending on the degree of elevation of cellular AMP, distinct compartmentalized pools of AMPK are activated, phosphorylating different sets of targets. Low glucose activates AMPK exclusively through the AMP-independent, AXIN-based pathway in lysosomes to phosphorylate targets such as ACC1 and SREBP1c, exerting early anti-anabolic and pro-catabolic roles. Moderate increases in AMP expand this to activate cytosolic AMPK also in an AXIN-dependent manner. In contrast, high concentrations of AMP, arising from severe nutrient stress, activate all pools of AMPK independently of AXIN. Surprisingly, mitochondrion-localized AMPK is activated to phosphorylate ACC2 and mitochondrial fission factor (MFF) only during severe nutrient stress. 11Vitamin B12 modulates Parkinson’s disease LRRK2 kinase activity through allosteric regulation and confers neuroprotection显示文摘Missense mutations in Leucine-Rich Repeat Kinase 2(LRRK2)cause the majority of familial and some sporadic forms of Parkinson’s disease(PD).The hyperactivity of LRRK2 kinase induced by the pathogenic mutations underlies neurotoxicity,promoting the development of LRRK2 kinase inhibitors as therapeutics.Many potent and specific small-molecule LRRK2 inhibitors have been reported with promise.However,nearly all inhibitors are ATP competitive—some with unwanted side effects and unclear clinical outcome—alternative types of LRRK2 inhibitors are lacking.Herein we identify 5′-deoxyadenosylcobalamin(AdoCbl),a physiological form of the essential micronutrient vitamin B12 as a mixed-type allosteric inhibitor of LRRK2 kinase activity.Multiple assays show that AdoCbl directly binds LRRK2,leading to the alterations of protein conformation and ATP binding in LRRK2.STD-NMR analysis of a LRRK2 homologous kinase reveals the contact sites in AdoCbl that interface with the kinase domain.Furthermore,we provide evidence that AdoCbl modulates LRRK2 activity through disrupting LRRK2 dimerization.Treatment with AdoCbl inhibits LRRK2 kinase activity in cultured cells and brain tissue,and prevents neurotoxicity in cultured primary rodent neurons as well as in transgenic C.elegans and D.melanogaster expressing LRRK2 disease variants.Finally,AdoCbl alleviates deficits in dopamine release sustainability caused by LRRK2 disease variants in mouse models.Our study uncovers vitamin B12 as a novel class of LRRK2 kinase modulator with a distinct mechanism,which can be harnessed to develop new LRRK2-based PD therapeutics in the future.Adam Schaffner Xianting Li Yacob Gomez-Llorente Emmanouela Leandrou Anna Memou Nicolina Clemente Chen Yao Farinaz Afsari Lianteng Zhi Nina Pan Keita Morohashi Xiaoluan Hua Ming-Ming Zhou Chunyu Wang Hui Zhang Shu G.Chen Christopher J.Elliott Hardy Rideout Iban Ubarretxena-Belandia Zhenyu Yue 2019Cell Research2019,29,4:7

13和解法治疗乙型肝炎相关性肝源性糖尿病的临床观察显示文摘目的探讨和解法治疗乙型肝炎相关性肝源性糖尿病的临床疗效。方法将39例乙型肝炎相关性肝源性糖尿病患者随机分为治疗组和对照组2组,治疗组20例采用和解法治疗,对照组19例采用护肝、抗病毒、降糖综合治疗,观察治疗后外周血肝功能、血糖水平及症状的改善情况。结果两组治疗后丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)水平和血清总胆红素(TB)水平较治疗前明显降低(P<0.01),空腹血糖(FBG)、餐后2小时血糖(PBG)水平均较治疗前明显降低(P<0.01),治疗组还能明显升高白蛋白与球蛋白比率(P<0.05)。两组治疗后症状积分均较治疗前明显降低,但治疗组治疗前后3个月和6个月的积分差值要明显高于对照组(P<0.05,P<0.01)。结论和解法治疗乙型肝炎相关性肝源性糖尿病具有一定疗效,而且副作用较小,值得进一步推广应用。郝尧坤 Carlini Fan Hardi 程永华 张诗军 2008北京中医药2008,27,8:6
14Characterization of volatiles Tribolium castaneum(H.)in flour using solid phase microextraction–gas chromatography mass spectrometry(SPME–GCMS)显示文摘The objective of this study was to identify volatile organic compounds(VOCs)from flour,Tribolium castaneum(Herbst)and flour infested by T.castaneum separately,to confirm the difference of healthy flour and flour infested by T.castaneum and to explore the new technique to diagnose stored flour’s quality by its VOCs change.Headspace-solid phase microextraction(HS-SPME)coupled with gas chromatography(GC)and gas chromatography–mass spectrometry(GC–MS)were used to detect the VOCs of three different samples.Totally,71 different compounds were identified in flour,T.castaneum and T.castaneum infested flour.Therefore,27 VOCs were identified from flour alone,32 VOCs from T.castaneum and 39 VOCs from T.castaneum infested flour.The compound 2-ethyl-2,5-cyclohexadiene-1,4-dione is only found in T.castaneum infested flour.This suggests that 2-ethyl-2,5-cyclohexadiene-1,4-dione can be a useful VOC for detecting T.castaneum in flour.Yonghao Niu Giles Hardy Manjree Agarwal Lei Hua Yonglin Ren 2016Food Science and Human Wellness2016,5,1:6
15Increased apoptosis and decreased density of medial smooth muscle cells in human abdominal aortic aneurysms显示文摘Objective To determine the increase of apoptosis and the decrease of smooth muscle cells (SMCs) density in human abdominal aortic aneurysms (AAA). Methods In situ terminal transferase-mediated dUTP nick end labeling (TUNEL) was employed to detect apoptosis of SMCs in patients with AAA (n=25) and normal abdominal aortae (n=10). Positive cells were identified by specific cell marker in combination with immunohistochemistry. Meanwhile SMC counting was performed by anti-α-actin immunohistostaining to compare the SMC density. Results TUNEL staining revealed that there was significantly increased apoptosis in AAAs (average 8.6%) compared with normal abdominal aortae (average 0.95%, P <0.01). Double staining showed that most of these cells were SMCs. Counting of α-actin positive SMCs revealed that medial SMC density of AAAs (37.5±7.6 SMCs /HPF) was reduced by 79.1% in comparison with that of normal abdominal aortae (179.2±16.1 SMCs /HPF,P <0.01). Conclusions Significantly increased SMCs of AAA bear apoptotic markers initiating cell death. 16加氢反应器壁材料的氢损伤特性显示文摘研究了在模拟加氢反应器工况条件下 ,国产加氢反应器壁材料 [2 (1/ 4 )Cr - 1Mo及其上的 30 9L和 347L不锈钢堆焊层 ]的氢损伤特性 .结果表明 :热渗氢后 ,明显降低了它们的断裂应力 (σf)和塑性 .对于 30 9L和 347L堆焊层 ,未经热渗氢的原始试样 ,无论是光滑试样还是缺口试样 (除缺口位置在 30 9L区域试样外 )均断裂在 347L区域内 .而经热渗氢后 ,无论是起裂于 2 (1/ 4 )Cr - 1Mo和 30 9L的熔合线 ,还是 30 9L和 347L的熔合线处 ,最后都断在30 9L区域内 (除缺口位置在 347L区域试样外 ) .这是因为热渗氢使 30 9L和 347L的σf 分别从原始状态的 885MPa降至 4 78MPa和从原始状态的 799.9MPa降至 5 6 4MPa ;在对短裂纹的低周疲劳特性研究中 ,用da/dN =C(ΔJ) n计算了裂纹扩展速度 (da/dN)和门槛值 (ΔJth) .经热渗氢后 ,ΔJth有明显的下降 ,da/dN~ΔJ曲线也向左移 ,提高了da/dN .SEM断口分析结果与上述结论一致 .裂纹扩展以穿晶为主 ,热渗氢后有二次裂纹存在 .说明热渗氢降低了其力学性能 。徐坚 钟曼英 郭世行 D Hardie 2003中国腐蚀与防护学报2003,23,3:6

18基于IL-2、IL-8水平变化的湿邪致病机理研究显示文摘目的:通过湿邪致病大鼠IL-2、IL-8水平变化探讨湿邪致病与细胞免疫功能的关系。方法:将40只SD大鼠按随机数字表随机分为正常组、内湿组、外湿组、内外湿组及疲劳组5组,分别造模20天后,采用放射免疫(RLA)方法检测其IL-2、IL-8水平。结果:全组比较,IL-2和IL-8水平在各组的差异有非常显著意义(F=3.102,P=0.028;F=2.657,P=0.049)。两两比较,外湿组和内外湿组的IL-2水平都比正常组明显升高(P=0.005,P=0.028),外湿组的IL-2水平比内湿组明显升高(P=0.015);外湿组的IL-8水平明显升高,与正常组、内湿组、内外湿组相比较差异有显著性(P=0.036,P=0.004,P=0.034)。结论:外湿组的IL-2和IL-8水平比内湿组和正常组明显升高,提示湿邪致病初期外湿与机体的免疫激活和炎症反应密切有关,此阶段机体可能对外在湿邪较内在湿邪的刺激更为敏感,其机理有待于进一步研究。Carlini Fan Hardi 张诗军 陈泽雄 林佑武 董霄 2010中国中医药咨讯2010,,1:6
