1TNF-α Induces Transient Resistance to Fas-Induced Apoptosis in Eosinophilic Acute Myeloid Leukemia Cells显示文摘Tumor necrosis factor α (TNF-α) has been recognized as an activator of nuclear factor κB (NF-κB), a factor implicated in the protection of many cell types from apoptosis. We and others have presented evidence to suggest that Fas-induced apoptosis may be an important aspect of the resolution of inflammation, and that delayed resolution of inflammation may be directly associated with NF-κB-dependent resistance to Fas. Because TNF-α activates NF-κB in many cell types including inflammatory cells such as eosinophils, we examined effects of TNF-α signaling on the Fas-mediated killing of an eosinophilic cell line AML14. While agonist anti-Fas (CH11) treatment induced apoptosis in AML14 cells, no significant cell death occurred in response to TNF-α alone. Electrophoretic mobility shift assay (EMSA) revealed that TNF-α induced NF-κB transactivation in AML14 cells in a time- and dose-dependent fashion, and subsequent supershift assays indicated that the translocated NF-κB was the heterodimer p65 (RelA)/p50. Pre-treatment of cells with TNF-α dramatically decreased the CH11-induced cell death in a transient fashion, accompanied by suppression of activation of caspase-8 and caspase-3 activation. Inhibition of NF-κB transactivation by inhibitors, BAY 11-7085 and parthenolide, reversed the suppression of Fas-mediated apoptosis by TNF-α. Furthermore, TNF-α up-regulated X-linked inhibitor of apoptosis protein (XIAP) transiently and XIAP levels were correlated with the temporal pattern of TNF-α protection against Fas-mediated apoptosis. 3荨麻疹性皮炎的定义:皮肤过敏反应模式的一种亚型显示文摘Background: Urticarial dermatitis may represent a useful term for a subset of a reaction pattern designated most commonly as dermal hypersensitivity by pathologists. The term is not commonly used, and requires definition to determine whether it is clinically relevant. Objectives: To define urticarial dermatitis and distinguish it from other urticarial reaction patterns and to review the frequency with which dermatologists can recognize clinical settings that match the biopsy findings of urticarial dermatitis. Design: Retrospective analysis of clinical and/or histological diagnosis of urticarial dermatitis, applying strict histological criteria in a center using urticarial dermatitis as a diagnostic term in 190 archived reports. Setting: Tertiary referral dermatopathology service reporting for dermatological practices in Sydney, Australia. Main Outcome Measures: The correlation between clinical and histological diagnoses of urticarial dermatitis and alternate diagnoses was analyzed. The frequency of positive immunofluorescence findings for bullous pemphigoid was determined in a subset of patients with urticarial dermatitis in whom this test was ordered to exclude prodromal bullous pemphigoid. Results: Urticarial dermatitis was the histological diagnosis in at least 1 biopsy result in 148 patients, and matched the provisional clinical diagnosis in 49 (33.1% ) patients. Urticarial dermatitis was the only diagnosis provided in 21 patients. The main alternate clinical diagnoses provided were early bullous pemphigoid or dermatitis herpetiformis (47 patients [31.8% ]), dermatitis (39 patients [26.4% ]),drug reaction (35 patients [23.6% ]), urticarial vasculitis (24 patients [16.2% ]), and urticaria (12 patients [8.1% ]). In 91 patients with a clinical diagnosis of urticarial dermatitis, the histological diagnosis in at least 1 biopsy result was matched in 49 patients (53.8% ); other histological diagnoses included dermatitis (21 patients [23.1% ]), papular urticaria (12 patients [13.2% ]), drug reaction (6 patients [6.6% ]), and urticaria (3 patients [3.3% ]). Review of 38 direct immunofluorescent results for prodromal bullous pemphigoid and a biopsy finding of urticarial dermatitis revealed only 3 positive results (7.9% ).Conclusions: Urticarial dermatitis seems to be a useful histological and clinical term for a subset of the dermal hypersensitivityreaction pattern. Although the clinical presentation is not restricted to a specific entity, eczema and drug reactions seem to be the most frequent clinical associations; and in a subset of patients, urticarial dermatitis remains as a recognizable reaction pattern. Urticarial dermatitis without eosinophilic spongiosis is not a reliable indicator for bullous pemphigoid, because the findings of immunofluorescence are often negative.Kossard S. Hamann I. 7Plant responses to simulated warming and drought:a comparative study of functional plasticity between congeneric mid and high elevation species显示文摘Aims Effects of climate change,especially changes in temperatures and precipitation patterns,are particularly pronounced in alpine regions.In response,plants may exhibit phenotypic plasticity in key functional traits allowing short-term adjustment to novel conditions.However,little is known about the degree of pheno-typic plasticity of high elevation species relative to mid elevation congeners.Methods We transplanted 14 herbaceous perennial species from high eleva-tion into two common gardens(1050 and 2000 m.a.s.l.)in the Swiss Alps,and we examined plastic responses in key functional traits to changes in temperature and soil water availability.This design was replicated with 14 congeneric species from mid elevation to assess if the degree of phenotypic plasticity differs between mid and high elevation species.Survival was assessed across two growing seasons,while aboveground biomass and specific leaf area(SLA)were measured after the first growing season,and biomass alloca-tion to belowground and reproductive structures after the second.Moreover,a phenotypic plasticity index was calculated for the func-tional traits to compare the degree of plasticity between mid and high elevation species.Important Findings Aboveground biomass was higher in mid elevation species relative to high elevation congeners in all treatments,yet decreased for both with elevation and drought.Similarly,SLA decreased with elevation and drought.Root mass fraction(RMF)was generally higher in high ele-vation species,and decreased with drought at the lower site.Drought increased the allocation to reproductive structures,especially when plants were grown at their elevation of origin.Interestingly,no differ-ence was found in the degree of phenotypic plasticity averaged across mid and high elevation species for any of the studied functional traits.These results indicate that phenotypic plasticity in the focal traits did not depend on the elevation of origin of the species.Plasticity was not related to environmental heterogeneity,nor constrained by selective pressures at high elevation.However,both species groups showed a remarkable capacity for short-term acclimation to a prospective cli-mate through rapid adjustments in key functional traits.Elena Hamann Halil Kesselring Jürg Stöcklin 2018Journal of Plant Ecology2018,11,3:2
