2Emotion processing in Parkinson's disease: a blood oxygenation level-dependent functional magnetic resonance imaging study显示文摘Parkinson's disease is a neurodegenerative disorder caused by loss of dopamine neurons in the substantia nigra pars compacta. Tremor, rigidity, and bradykinesia are the major symptoms of the disease. These motor impairments are often accompanied by affective and emotional dysfunctions which have been largely studied over the last decade. The aim of this study was to investigate emotional processing organization in the brain of patients with Parkinson's disease and to explore whether there are differences between recognition of different types of emotions in Parkinson's disease. We examined 18 patients with Parkinson's disease(8 men, 10 women) with no history of neurological or psychiatric comorbidities. All these patients underwent identical brain blood oxygenation level-dependent functional magnetic resonance imaging for emotion evaluation. Blood oxygenation level-dependent functional magnetic resonance imaging results revealed that the occipito-temporal cortices, insula, orbitofrontal cortex, basal ganglia, and parietal cortex which are involved in emotion processing, were activated during the functional control. Additionally, positive emotions activate larger volumes of the same anatomical entities than neutral and negative emotions. Results also revealed that Parkinson's disease associated with emotional disorders are increasingly recognized as disabling as classic motor symptoms. 3Sclareol and linalyl acetate are produced by glandular trichomes through the MEP pathway显示文摘Sclareol,an antifungal specialized metabolite produced by clary sage,Salvia sclarea,is the starting plant natural molecule used for the hemisynthesis of the perfume ingredient ambroxide.Sclareol is mainly produced in clary sage flower calyces;however,the cellular localization of the sclareol biosynthesis remains unknown.To elucidate the site of sclareol biosynthesis,we analyzed its spatial distribution in the clary sage calyx epidermis using laser desorption/ionization mass spectrometry imaging(LDI–FTICR-MSI)and investigated the expression profile of sclareol biosynthesis genes in isolated glandular trichomes(GTs).We showed that sclareol specifically accumulates in GTs’gland cells in which sclareol biosynthesis genes are strongly expressed.We next isolated a glabrous beardless mutant and demonstrate that more than 90%of the sclareol is produced by the large capitate GTs.Feeding experiments,using 1-13 C-glucose,and specific enzyme inhibitors further revealed that the methylerythritol-phosphate(MEP)biosynthetic pathway is the main source of isopentenyl diphosphate(IPP)precursor used for the biosynthesis of sclareol.Our findings demonstrate that sclareol is an MEP-derived diterpene produced by large capitate GTs in clary sage emphasing the role of GTs as biofactories dedicated to the production of specialized metabolites.Camille Chalvin Stephanie Drevensek Françoise Gilard Caroline Mauve Christel Chollet Halima Morin Edith Nicol Eva Heripre Lucie Kriegshauser Bertrand Gakiere Michel Dron Abdelhafid Bendahmane Adnane Boualem 2021Horticulture Research2021,8,1:2
4Dietary betaine activates hepatic VTGⅡexpression in laying hens associated with hypomethylation of GR gene promoter and enhanced GR expression显示文摘Background: Vitellogenin(VTG) is a precursor of egg yolk proteins synthesized within the liver of oviparous vertebrates. Betaine is an important methyl donor that is reported to improve egg production of laying hens with an unclear mechanism. In the present study, we fed betaine-supplemented diet(0.5%) to laying hens for 4 wk and investigated its effect on VTGII expression in the liver.Results: Betaine did not affect chicken weight, but significantly(P < 0.05) increased egg laying rate accompanied with a significant(P < 0.05) increase in hepatic concentration and plasma level of VTGI. Plasma estrogen level did not change,but the hepatic expression of estrogen receptor α(ERα) mR NA was significantly(P < 0.05) up-regulated. Betaine did not affect the protein content of ERα, but significantly(P < 0.05) increased hepatic expression of glucocorticoid receptor(GR)at both mR NA and protein levels. Also, ERα/GR interaction tended to be enhanced in the liver nuclear lysates of betainesupplemented hens as determined by co-immunoprecipitation. Furthermore, dietary betaine supplementation significantly increased(P < 0.05) the hepatic expression of methyl-transfer enzymes, such as BHMT, GNMT, and DNMT1,which was associated with higher SAM/SAH ratio and hypomethylation of GR promoter regions.Conclusions: Betaine activates hepatic VTGII expression in association with modified DNA methylation of GR gene promoter, GR expression and ERα/GR interaction. 5Preservation of Juniperus thurifera L.:a rare endangered species in Algeria through in vitro regeneration显示文摘Juniperus thurifera L.is an endemic Cupres saceae from the Aure`s Mountains of north eastern Algeria and endangered,in part,due to the scarcity of viable seeds It is threatened by other abiotic factors and the lack of an effective management strategy will increase its risk o extinction.The dearth of information on its in vitro regeneration impedes its application in forest managemen programs.We therefore developed a micropropagation protocol using microcuttings with auxiliary buds.Cuttings were grown on different combinations of media supplemented with plant growth regulators at different concentrations.The highest number of shoots and branches regenerated from original shoots was obtained on Woody Plant Medium(WPM)supplemented with 6-benzylaminopurine(BAP)(0.5 mg L^(-1))and 2,4-dichlorophe noxyacetic acid(2,4-D)(0.25 mg L^(-1)).The best elongation of shoots was achieved with WPM supplemented with0.5 mg L^(-1)of BAP and 0.25 or 1 mg L^(-1) of 2,4-D.On the second subculture,shoots had a higher number of branches than those of the first.The highest rooting rate,38.8%,was obtained with shoots cultured in 1/2 Murashige and Skoog(MS)medium supplemented with 5.0 mg L^(-1)each of indol-3-butyric(IBA)and naphthalene acetic acid(NAA).Similarly,the highest root numbers and lengths were produced on 1/2 MS medium supplemented with IBA and NAA(5.0 mg L^(-1)each).During transfer to acclimatization,rates of plant losses of 50% occurred.The second part of the experiment showed that the best shoot callusing was on WPM supplemented with BAP and 2,4-D,with either the combination 0.5+0.25 or 0.25+0.25 mg L^(-1).The results of this research provide a starting point for further studies on in vitro regeneration of J.thurifera for the sustainable management of its unique ecosystem in the Mediterranean basin.Nadia Khater Halima Benbouza 2019Journal of Forestry Research2019,30,1:2

13Functional properties of peptides obtained from whey proteins by ficin extract hydrolysis显示文摘Enzymatic whey proteins concentrates (WPC) hydrolysis under optimal conditions of ficin crude extract (FCE) in two ratios E/S (0.5 and 1%) was studied,assessing the impact of hydrolysis on WPC by determination degree of hydrolysis (DH),Fast proteins liquid chromatography (FPLC),Tricine SDS-PAGE,antioxidant activity at sampling time beside,and technological properties (solubility,emulsifying activity index (EAI) and foaming activity (FA)) at the best condition (maximum DH and antioxidant activity) other side.Results showed,in the first 30 min,DH reached 22.76% at E/S (1%) with a total degradation of α-lactalbumin (α-La) and almost of β-lactoglobulin (β-Lg).A significant increase in antioxidant activity and solubility was observed,while not for EAI,and FA.These results indicate for the first time that β-Lg is susceptible to hydrolyse by FCE that rarely found for other enzymes.Ficin hydrolysates showed very relevant performance,and suggest that could be appropriate production of functional ingredients.Afaf Kheroufi María E.Brassesco Débora A.Campos Halima Boughellout Manuela E.Pintado 2022Food Bioscience2022,47,3:1
