1Alcohol metabolites and lipopolysaccharide: Roles in the development and/or progression of alcoholic liver disease显示文摘The onset of alcoholic liver disease (ALD) is initiated by different cell types in the liver and a number of different factors including: products derived from ethanol-induced inflammation, ethanol metabolites, and the indirect reactions from those metabolites. Ethanol oxidation results in the production of metabolites that have been shown to bind and form protein adducts, and to increase inflammatory, fibrotic and cirrhotic responses. Lipopolysaccharide (LPS) has many deleterious effects and plays a significant role in a number of disease processes by increasing inflammatory cytokine release. In ALD, LPS is thought to be derived from a breakdown in the intestinal wall enabling LPS from resident gut bacterial cell walls to leak into the blood stream. The ability of adducts and LPS to independently stimulate the various cells of the liver provides for a two-hit mechanism by which various biological responses are induced and result in liver injury. 5颈动脉破损斑块的高分辨MR成像研究显示文摘目的评价破损颈动脉粥样硬化斑块的成分及其分布的特点。方法回顾性分析43例经超声诊断颈动脉狭窄50%~99%患者斑块高分辨MR图像,根据斑块表面纤维帽的完整性判定斑块破损情况,同时定量测量管腔、管壁、斑块成分的面积比例、纵向长度,并计算总管腔面积、血管负荷指数、偏心指数。采用SPSS12.0统计学软件对患者一般临床资料进行卡方检验,并且对定量测量所得到的计量资料采用Mann-Whitney秩和检验。结果MRI共发现17例破损斑块(破损斑块组)。破损斑块组和未破损斑块组在血管纵向分布上大面积脂质核心长度差异有统计学意义,中位脂质核心长度分别为6mm、0mm(U=126.5,P〈0.05)。血管负荷指数中位数分别为0.67、0.59,斑块内出血的发生率分别为82.4%和42.3%,差异均有统计学意义(U值分别为105、132,P值均〈0.05)。破损斑块组偏心分布更明显,狭窄范围更长。偏心指数分别为3.68、2.79,平均管腔面积为23.97、30.18mm^2,差异均有统计学意义(U值分别为126、138,P值均〈0.05)。结论破损斑块在血管横断面及纵向分布上具有明显的特点,高分辨MR多方向成像可提供更多易损斑块的信息。于薇 张兆琪 Hunter Underhill ThomasS.Hatsukami YuanChun 2008中华放射学杂志2008,42,8:11

9基于分布式大流域径流模型的中国西北黑河流域水文模拟(英文)显示文摘水资源短缺是中国西北干旱地区长期的问题,区域人口增加、城市化扩张,加之气候变化的影响进一步加剧了西北地区水资源短缺,也使生活用水、灌溉用水、工业用水和维持生态系统稳定的用水危险加剧.采用分布式大流域径流模型(DLBRM)模拟黑河流域水文(中国第二大内陆河,流域面积128 000 km2)来理解区域的冰川和积雪融化水、地下水、地表水、蒸散发等方面的分布,评估气候变化对水文的影响和冰川退缩对中游和下游来水量的影响.模拟结果表明,黑河流域的大部分产流那源于黑河上游地区的祁连山.模拟1990—2000年黑河河流日流量变化结果认为,黑河中游正义峡给下游的供水为10×108m3,其中地表径流占51%,层间流占49%.中游地区沙土具有较高的蒸腾发能力,近一半的地表水被蒸发掉.模拟实践证明,分布式大流域径流模型可以结合气候变化、水资源管理方面的成果,改进流域水文模拟的精度.贺缠生 Carlo DeMarchi Thomas E. 10Non-pharmacological therapies for inflammatory bowel disease: Recommendations for self-care and physician guidance显示文摘We performed a scoping review on sought-after complementary therapies for patients with inflammatory bowel disease(IBD), specifically diet, physical activity and exercise(PA/E), and psychotherapy. We aim to update patients with IBD on therapies for self-care and provide physicians with guidance on how to direct their patients for the management of IBD. A search of MEDLINE, EMBASE, and PUBMED was completed in Sept 2016. Studies on diet, PA/E, or psychotherapy in patients with IBD were included. Medical Subject Heading terms and Boolean operators were used. The search was limited to full-text English articles describing an adult population. This review included 67 studies: Diet(n = 19); PA/E(n = 19); and psychotherapy(n = 29). We have made the following recommendations:(1) Diet: Consumption of diets rich in vegetables, fruit and soluble fiber may be beneficial in IBD. A trial of a low FODMAP diet can be considered in those patients with functional gastrointestinal symptoms. Restrictive diets are lacking in evidence and should be avoided;(2) PA/E: Regular lowmoderate intensity activity, including cardiovascular and resistance exercise, has been shown to improve quality of life(QOL) and may improve inflammation; and(3) psychotherapy: Therapies such as cognitive-behavioural interventions, mindfulness, hypnosis, and stress management have been shown to improve QOL, but evidence is limited on their impact on anxiety, depression, and disease activity. 12SFRP4 expression correlates with epithelial mesenchymal transitionlinked genes and poor overall survival in colon cancer patients显示文摘BACKGROUND Colon cancer is among the most commonly diagnosed cancers in the United States with an estimated 97220 new cases expected by the end of 2018.It affects 1.2 million people around the world and is responsible for about 0.6 million deaths every year.Despite decline in overall incidence and mortality over the past 30 years,there continues to be an alarming rise in early-onset colon cancer cases(<50 years).Patients are often diagnosed at late stages of the disease and tend to have poor survival.We previously showed that the WNT“gatekeeper”gene,secreted frizzled-related protein 4(SFRP4),is over-expressed in early-onset colon cancer.SFRP4 is speculated to play an essential role in cancer by inhibiting the epithelial mesenchymal transition(EMT).AIM To investigate the correlation between SFRP4 expression and EMT-linked genes in colon cancer and how it affects patient survival.METHODS SFRP4 expression relative to that of EMT-linked genes and survival analysis were performed using the University of California Santa Cruz Cancer Browser interface.RESULTS SFRP4 was found to be co-expressed with the EMT-linked markers CDH2,FN1,VIM,TWIST1,TWIST2,SNAI1,SNAI2,ZEB1,ZEB2,POSTN,MMP2,MMP7,MMP9,and COL1A1.SFRP4 expression negatively correlated with the EMTlinked suppressors CLDN4,CLDN7,TJP3,MUC1,and CDH1.The expression of SFRP4 and the EMT-linked markers was higher in mesenchymal-like samples compared to epithelial-like samples which potentially implicates SFRP4-EMT mechanism in colon cancer.Additionally,patients overexpressing SFRP4 presented with poor overall survival(P=0.0293).CONCLUSION Considering the implication of SFRP4 in early-onset colon cancer,particularly in the context of EMT,tumor metastasis,and invasion,and the effect of increased expression on colon cancer patient survival,SFRP4 might be a potential biomarker for early-onset colon cancer that could be targeted for diagnosis and/or disease therapy.Landry E Nfonsam Jana Jandova Hunter C Jecius Pamela N Omesiete Valentine N Nfonsam 2019World Journal of Gastrointestinal Oncology2019,11,8:4

14Dysfunctional stem and progenitor cells impair fracture healing with age显示文摘Successful fracture healing requires the simultaneous regeneration of both the bone and vasculature;mesenchymal stem cells (MSCs) are directed to replace the bone tissue, while endothelial progenitor cells (EPCs) form the new vasculature that supplies blood to the fracture site. In the elderly, the healing process is slowed, partly due to decreased regenerative function of these stem and progenitor cells. MSCs from older individuals are impaired with regard to cell number, proliferative capacity, ability to migrate, and osteochondrogenic differentiation potential. The proliferation, migration and function of EPCs are also compromised with advanced age. Although the reasons for cellular dysfunction with age are complex and multidimensional, reduced expression of growth factors, accumulation of oxidative damage from reactive oxygen species, and altered signaling of the Sirtuin-1 pathway are contributing factors to aging at the cellular level of both MSCs and EPCs. Because of these geriatric-specific issues, effective treatment for fracture repair may require new therapeutic techniques to restore cellular function. Some suggested directions for potential treatments include cellular therapies, pharmacological agents, treatments targeting age-related molecular mechanisms, and physical therapeutics. 17Colour compound lenses for a portable fluorescence microscope显示文摘In this article,we demonstrated a handheld smartphone fluorescence microscope(HSFM)that integrates dualfunctional polymer lenses with a smartphone.The HSFM consists of a smartphone,a field-portable illumination source,and a dual-functional polymer lens that performs both optical imaging and filtering.Therefore,compared with the existing smartphone fluorescence microscope,the HSFM does not need any additional optical filters.Although fluorescence imaging has traditionally played an indispensable role in biomedical and clinical applications due to its high specificity and sensitivity for detecting cells,proteins,DNAs/RNAs,etc.,the bulky elements of conventional fluorescence microscopes make them inconvenient for use in point-of-care diagnosis.The HSFM demonstrated in this article solves this problem by providing a multifunctional,miniature,small-form-factor fluorescence module.This multifunctional fluorescence module can be seamlessly attached to any smartphone camera for both bright-field and fluorescence imaging at cellular-scale resolutions without the use of additional bulky lenses/filters;in fact,the HSFM achieves magnification and light filtration using a single lens.Cell and tissue observation,cell counting,plasmid transfection evaluation,and superoxide production analysis were performed using this device.Notably,this lens system has the unique capability of functioning with numerous smartphones,irrespective of the smartphone model and the camera technology housed within each device.As such,this HSFM has the potential to pave the way for realtime point-of-care diagnosis and opens up countless possibilities for personalized medicine.Bo Dai Ziao Jiao Lulu Zheng Hunter Bachman Yongfeng Fu Xinjun Wan Yule Zhang Yu Huang Xiaodian Han Chenglong Zhao Tony Jun Huang Songlin Zhuang Dawei Zhang 2019Light(Science & Applications)2019,8,1:3
18A Web-based system enabling the integration,analysis,and 3D sub-surface visualization of groundwater monitoring data and geological models显示文摘The 3D Water Chemistry Atlas is an intuitive,open source,Web-based system that enables the three-dimensional(3D)sub-surface visualization of ground water monitoring data,overlaid on the local geological model(formation and aquifer strata).This paper firstly describes the results of evaluating existing virtual globe technologies,which led to the decision to use the Cesium open source WebGL Virtual Globe and Map Engine as the underlying platform.Next it describes the backend database and search,filtering,browse and analysis tools that were developed to enable users to interactively explore the groundwater monitoring data and interpret it spatially and temporally relative to the local geological formations and aquifers via the Cesium interface.The result is an integrated 3D visualization system that enables environmental managers and regulators to assess groundwater conditions,identify inconsistencies in the data,manage impacts and risks and make more informed decisions about coal seam gas extraction,waste water extraction,and water reuse.Jane Hunter Charles Brooking Lucy Reading Sue Vink 2016International Journal of Digital Earth2016,9,2:3
19Comparative WGBS identifies genes that influence non-ripe phenotype in tomato epimutant Colourless non-ripening显示文摘Whole-genome bisulfite sequencing(WGBS)allows single-base resolution and genome-wide profiling of DNA methylation in plants and animals.This technology provides a powerful tool to identify genes that are potentially controlled by dynamic changes of DNA methylation and demethylation.However,naturally occurring epimutants are rare and genes under epigenetic regulation as well as their biological relevances are often difficult to define.In tomato,fruit development and ripening are a complex process that involves epigenetic control.We have taken the advantage of the tomato epimutant Colourless non-ripening(Cnr)and performed comparative mining of the WGBS datasets for the Cnr and Sl CMT3-silenced Cnr fruits.We compared DNA methylation profiles for the promoter sequences of approximately 5,000 bp immediately upstream of the coding region of a list of20 genes.Differentially methylated regions were found for some of these genes.Virus-induced gene silencing(VIGS)of differentially methylated gene Sl DET1 or Sl PDS resulted in unusual brown pigmentation in Cnr fruits.These results suggest that comparative WGBS coupled with VIGS can be used to identify genes that may contribute to the colourless unripe phenotype of fruit in the Cnr epimutant.Weiwei Chen Zhiming Yu Junhua Kong Hui Wang Yichen Li Mei Zhao Xiaohong Wang Qianqian Zheng Nongnong Shi Pengcheng Zhang Silin Zhong Paul Hunter Mahmut Tor Yiguo Hong 2018Science China(Life Sciences)2018,61,2:3
20β-葡聚糖/柠檬酸喷剂与Optimoist^(TM)治疗口干的临床疗效比较显示文摘目的 对比 β-1 ,3葡聚糖 /柠檬酸喷剂与OptimoistTM喷剂对口腔粘膜干燥的临床治疗效果。方法选择 53例由于舍格伦综合征或头颈部放射治疗而导致中度或重度口腔粘膜干燥的患者 ,指导其使用 β -1 ,3葡聚糖 /柠檬酸和β -1 ,3葡聚糖以及OptimoistTM喷剂 ,分别记录首次用药和持续用药两周后不同时间点的口腔主观感觉指标和唾液流率 ,对三种不同制剂的效果进行对比分析。结果 主观感觉指标和测定的唾液流率均显示 ,三种喷剂都能较好地缓解口干症状。患者的总体满意度和首次用药后 1 0分钟的唾液流率 ,OptimoistTM 喷剂优于 β -1 ,3葡聚糖 /柠檬酸喷剂及 β -1 ,3葡聚糖喷剂。结论 β-1 ,3葡聚糖 /柠檬酸喷剂可以缓解口干 ,但其对唾液流率的提高和患者使用后的总体满意度均不及OptimoistTM喷剂 ,β -1 ,3葡聚糖在口干喷剂中的应用尚需进一步研究。张雷 王祖华 李雨琴 俞光岩 Cathy Hunter 徐韬 2001现代口腔医学杂志2001,15,4:3
