1A novel STING agonist-adjuvanted pan-sarbecovirus vaccine elicits potent and durable neutralizing antibody and T cell responses in mice,rabbits and NHPs显示文摘The emergence of SARS-CoV-2 variants and potentially other highly pathogenic sarbecoviruses in the future highlights the need for pan-sarbecovirus vaccines.Here,we discovered a new STING agonist,CF501,and found that CF501-adjuvanted RBD-FC vaccine(CF501/RBD-FC)elicited significantly stronger neutralizing antibody(nAb)and T cell responses than Alum-and cGAMP-adjuvanted RBD-FC in mice.Vaccination of rabbits and rhesus macaques(nonhuman primates,NHPs)with CF501/RBD-FC elicited exceptionally potent nAb responses against SARS-CoV-2 and its nine variants and 41 S-mutants,SARS-CoV and bat SARSr-CoVs.CF501/RBD-FC-immunized hACE2-transgenic mice were almost completely protected against SARS-CoV-2 challenge,even 6 months after the initial immunization.NHPs immunized with a single dose of CF501/RBD-FC produced high titers of nAbs.The immunized macaques also exhibited durable humoral and cellular immune responses and showed remarkably reduced viral load in the upper and lower airways upon SARS-CoV-2 challenge even at 108 days post the final immunization.Thus,CF501/RBD-Fc can be further developed as a novel pan-sarbecovirus vaccine to combat current and future outbreaks of sarbecovirus diseases.Zezhong Liu Jie Zhou Wei Xu Wei Deng Yanqun Wang Meiyu Wang Qian Wang Ming Hsieh Jingming Dong Xinling Wang Weijin Huang Lixiao Xing Miaoling He Chunlin Tao Youhua Xie Yilong Zhang Youchun Wang Jincun Zhao Zhenghong Yuan Chuan Qin Shibo Jiang Lu Lu 2022Cell Research2022,32,3:5
2Antidepressant-like effect of active fraction of Polyrhachisvicina Roger in a rat depression model显示文摘OBJECTIVE: To investigate the antidepressant-likeeffect of active fraction of Polyrhachis vicina Roger(AFPR) in a rat depression model, and to elucidate the underlying mechanism.METHODS: AFPR was extracted with ethanol followed by petroleum ether. Its antidepressant-like effect was investigated in mice by tail suspension test(TST), forced swimming test(FST) and open field test(OPT). A repeated dose of reserpine(0.5 mg/kg, daily for 14 d) was used to establish a rat depression model. Fluoxetine was used as positive control agent. The effect of AFPR on reserpine-induced ptosis, hypothermia and akinesia, the levels of monoamines and their metabolites, and the activity of monoamine oxidase(MAO) in hippocampus and prefrontal cortex were determined.RESULTS: Administration of AFPR by gavage at 160 and 320 mg/kg significantly reduced the duration of immobility in the FST and TST, and did not affect locomotor activity in the OPT. In the reserpine-induced depression model, AFPR attenuated anhedonia, demonstrated by reversing hypothermia, akinesia and sucrose consumption. AFPR significantly increased the concentration of monoamines, including dopamine, serotonin, noradrenaline and acetylcholine.CONCLUSION: AFPR normalized the metabolism rates of noradrenaline, serotonin and dopamine,and the activity of MAO, which were altered by chronic reserpine exposure. 4A gain-of-function mutation in Msl10 triggers cell death and wound-induced hyperaccumulation of jasmonic acid in Arabidopsis显示文摘Jasmonates(JAs) are rapidly induced after wounding and act as key regulators for wound induced signaling pathway. However, what perceives the wound signal and how that triggers JA biosynthesis remains poorly understood. To identify components involved in Arabidopsis wound and JA signaling pathway, we screened for mutants with abnormal expression of a luciferase reporter, which is under the control of a wound-responsive promoter of an ethylene response factor(ERF) transcription factor gene, RAP2.6(Related to APetala 2.6).The rea1(RAP2.6 expresser in shoot apex) mutant constitutively expressed the RAP2.6-LUC reporter gene in young leaves.Along with the typical JA phenotypes including shorter petioles,loss of apical dominance, accumulation of anthocyanin pigments and constitutive expression of JA response gene, rea1 plants also displayed cell death and accumulated high levels of JA in response to wounding. The phenotype of rea1 mutant is caused by a gain-of-function mutation in the C-terminus of a mechanosensitive ion channel MscS-like 10(MSL10). MSL10 is localized in the plasma membrane and is expressed predominantly in root tip, shoot apex and vascular tissues. 5Eupalinolide B inhibits hepatic carcinoma by inducing ferroptosis and ROS-ER-JNK pathway显示文摘Primary hepatic carcinoma is a common malignant tumor.The classic molecular targeted drug sorafenib is costly and is only effective for some patients.Therefore,it is of great clinical significance to search for new molecular targeted drugs.Eupalinolide B(EB)from Eupatorium lindleyanum DC.is used to treat chronic tracheitis in clinical practice.However,the role of EB in hepatic carcinoma is unknown.In this study,we first measure the effect of EB on tumor growth in a xenograft model and PDX model.The cell proliferation and migration are also detected in human hepatocarcinoma cell lines(SMMC-7721 and HCCLM3).Then,we investigate cell cycle,cell apoptosis,cell necrosis,cell autophagy,and ferroptosis by flow cytometry,western blot analysis and electron microscopy.The results demonstrate that EB exerts anti-proliferative activity in hepatic carcinoma by blocking cell cycle arrest at S phase and inducing ferroptosis mediated by endoplasmic reticulum(ER)stress,as well as HO-1 activation.When HO-1 is inhibited,EB-induced cell death and ER protein expression are rescued.The migration-related mechanism consists of activation of the ROS-ER-JNK signaling pathway and is not connected to ferroptosis.In summary,we first discover that EB inhibits cell proliferation and migration in hepatic carcinoma,and thus EB is a promising anti-tumor compound that can be used for hepatic carcinoma.Yonghui Zhang Haoyang Zhang Jinage Mu Meiyue Han Zhihao Cao Feng Dong Tingting Wang Lian Pan Wujing Luo Jiaxin Li Huan Liu Lishan Jin Wenxuan Ding Yong Wei Xuesong Deng Dan Liu Xiuzhen He Yi Pang Xiao Mu Zhongjun Wu Dilong Chen 2022Acta Biochimica et Biophysica Sinica2022,54,7:1
6Light-modulated vertical heterojunction phototransistors with distinct logical photocurrents显示文摘The intriguing carrier dynamics in graphene heterojunctions have stimulated great interest in modulating the optoelectronic features to realize high-performance photodetectors.However,for most phototransistors,the photoresponse characteristics are modulated with an electrical gate or a static field.In this paper,we demonstrate a graphene/C_(60)/pentacene vertical phototransistor to tune both the photoresponse time and photocurrent based on light modulation.By exploiting the power-dependent multiple states of the photocurrent,remarkable logical photocurrent switching under infrared light modulation occurs in a thick C_(60) layer(11 nm)device,which implies competition of the photogenerated carriers between graphene/C_(60) and C_(60)/pentacene.Meanwhile,we observe a complete positive-negative alternating process under continuous 405 nm irradiation.Furthermore,infrared light modulation of a thin C_(60)(5 nm)device results in a photoresponsivity improvement from 3425 A/W up to 7673 A/W,and we clearly probe the primary reason for the distinct modulation results between the 5 and 11 nm C_(60) devices.In addition,the tuneable bandwidth of the infrared response from 10 to 3×10^(3) Hz under visible light modulation is explored.Such distinct types of optical modulation phenomena and logical photocurrent inversion characteristics pave the way for future tuneable logical photocurrent switching devices and high-performance phototransistors with vertical graphene heterojunction structures.Jiayue Han Meiyu He Ming Yang Qi Han Fang Wang Fang Zhong Mengjian Xu Qing Li He Zhu Chongxin Shan Weida Hu Xiaoqing Chen Xinran Wang Jun Gou Zhiming Wu Jun Wang 2020Light(Science & Applications)2020,9,1:1
7Sequence analysis of peptides with biological activities using electrospray-Fourier transform ion cyclotron resonance mass spectrometry显示文摘The mass spectra of five peptides with biological activities are reported. All mass spectra were recorded using a 4.7-T Fourier transform ion cyclotron resonance mass spectrometer equipped with an external electrospray source. The accurate molecular weights for the five peptides prepared by solid phase synthesis were measured as 1765.9013, 1063.5420, 1092.5254, 820.3804 and 1078.5193, respectively. All the data were obtained with the external calibration. Differences between observed and theoretical monoisotopic molecular weights were in the (0.2-1.0)×10-6 range. The complete primary sequence for the five polypep-tides were determined using the method of in-source electrospray ionization/collision induced dissociation (ESI/CID). All the intact y series ions and b series ions were obtained from various peptides respectively, thus determining the sequences of the five polypeptides. 8Measures of bioavailable serum sex hormone levels in aging Chinese by protein chip显示文摘The purpose of this study was to develop a protein chip technique based on receptor binding assays to measure bioavailable serum sex hormone levels (BSSHL). 224 aging healthy Chinese were investigated to get the referenced values of BSSHL for the first time. In the assays recombined sex hormone receptor proteins were jointed to polysaccharide coated slides to make protein chip, and the dose-dependence curves of sex hormone on chip were prepared. The data showed that this method had good precision (CV<16%) and accuracy (Bias<10%), and the sensitivity could reach 1 pmol/L. From the results, BSSHL of men and women declined with aging, but no significant differences were observed. The BSSHL of aging men were higher than those of women. The bioavailable serum androgen level of men was 52―112 pmol/L, women's was 3―70 pmol/L and the whole group was 41.9―81.4 pmol/L. The bioavailable serum estrogen level of men was 0.8―3.0 pmol/L, women's was 1.2―2.5 pmol/L and the whole group was 0.6―2.64 pmol/L. 9Sonic hedgehog signaling:Alternative splicing and pathogenic role in medulloblastoma显示文摘Alternative splicing(AS)produces the different mRNA splicing bodies,which are then translated into multiple protein isoforms and participate in various biological functions.With a deeper understanding of alternative splicing through the study of transcriptomes using high-throughput sequencing-based methods,the correlation between aberrant AS and diseases triggered a great concern,especially abnormal AS and cancer.Medulloblastoma(MB)is an intracranial tumor in children.Sonic hedgehog MB(SHH-MB)accounted for approximately 30%of MB,which is associated with the activation of SHH signaling.Growing evidence shows that aberrant AS is closely related to the tumorigenesis of MB.Here,we briefly introduced the AS and its mechanism.Next,we described canonical/noncanonical hedgehog signaling and its correlation with MB.The main description focused on AS of various regulators in canonical hedgehog signaling in MB.In addition,we also described AS of various regulators in noncanonical hedgehog signaling.Meanwhile,activated hedgehog signaling also induces AS in MB.Then,we pointed out that aberrant AS of hedgehog signaling is associated with different MB subgroups.Finally,we summarized the therapeutic applications of targeted AS in cancer treatment.In summary,further understanding of AS in SHH-MB could develop therapeutic targets for splicing factors which may be a novel therapeutic strategy.Meiyu Qu Qiangqiang He Jiahao Luo Tingyu Shen Ruibo Gao Yana Xu Chengyun Xu Muhammad Qasim Barkat Ling-Hui Zeng Ximei Wu 2023Genes & Diseases2023,10,5:0
10Redox-responsive phenyl-functionalized polylactide micelles for enhancing Ru complexes delivery and phototherapy显示文摘Poly(ethylene glycol)-poly(lactic acid)block copolymer(PEG-PLA)is one of the most widely used biomedical polymers in clinical drug delivery owing to its biocompatibility and biodegradability.However,endowing PEG-PLA micelles with high drug loading,self-assembly stability and fast intracellular drug release is still challenging.Redox-responsive diblock copolymers(MPEG-SS-PMLA)of poly(ethylene glycol)and phenyl-functionalized poly(lactic acid)with disulfide bond as the linker are synthesized to prepare PLA-based micelles that demonstrate excellent colloidal stability and high Ru loading.Notably,MPEGSS-PMLA achieved a remarkably high Ru loading efficiency of 84.3%due to the existence of strongπ-πstacking between phenyl and Ru complex.MPEG-SS-PMLA exhibited good colloidal stability in physiological condition but quickly destabilized by reductive tumor microenvironment.Interestingly,about 74%of Ru complex was released under 10 mmol/L GSH concentration.Ru-loaded MEPG-SS-PMLA showed efficient delivery and release of Ru complex into MCF-7 cancer cells,achieving enhanced in vitro and in vivo antitumor activity of photodynamic therapy.This feasible functionalization method of MPEG-PLA has appeared to be a clinically viable platform for controlled delivery therapeutic agents and enhanced phototherapy.Maomao He Zongwei Zhang Ziyue Jiao Meiyu Yan Pengcheng Miao Zhiyong Wei Xuefei Leng Yang Li Jiangli Fan Wen Sun Xiaojun Peng 2023Chinese Chemical Letters2023,34,3:0
