| 3 | Insights into the Tectonic Fractures in the Yanchang Formation Interbedded Sandstone-Mudstone of the Ordos Basin Based on Core Data and Geomechanical Models显示文摘The distribution and intensity of tectonic fractures within geologic units are important to hydrocarbon exploration and development. Taken the Upper Triassic Yanchang Formation interbedded sandstone-mudstone in the Ordos Basin as an example, this study used the finite element method(FEM) based on geomechanical models to study the development of tectonic fractures. The results show that the sandstones tend to generate tectonic fractures more easily than mudstones with the same layer thickness, and the highest degree of tectonic fractures will be developed when the sandstone-mudstone thickness ratio is about 5.0. A possible explanation is proposed for the tectonic fracture development based on two important factors of rock brittleness and mechanical layer thickness. Generally, larger rock brittleness and thinner layer thickness will generate more tectonic fractures. | 6 | Discriminated sgRNAs-Based SurroGate System Greatly Enhances the Screening Efficiency of Plant Base-Edited Cells显示文摘The development of CRISPR/Cas9-mediated base editing has made genomic modification more efficient. However, selection of genetically modified cells from millions of treated cells, especially plant cells, is still challenging. In this study, an efficient surrogate reporter system based on a defective hygromycin resistance gene was established in rice to enrich base-edited cells. After step-by-step optimization, the Discriminated sgRNAs-based SurroGate system (DisSUGs) was established by artificially differentiating the editing abilities of a wild-type single guide RNA (sgRNA) targeting the surrogate reporter gene and an enhanced sgRNA targeting endogenous sites. The DisSUGs enhanced the efficiency of screening base-edited cells by 3- to 5-fold for a PmCDA1-based cytosine-to-tyrosine base editor (PCBE), and 2.5- to 6.5-fold for an adenine base editor (ABE) at endogenous targets. These targets showed editing efficiencies of <25% in the conventional systems. The DisSUGs greatly enhanced the frequency of homozygous substitutions and expanded the activity window slightly for both a PCBE and an ABE. Analyses of the total number of single-nucleotide variants from whole-genome sequencing revealed that, compared with the no-enrichment PCBE strategy, the DisSUGs did not alter the frequency of genome-wide sgRNA-independent off-target mutations, but slightly increased the frequency of target-dependent off-target mutations. Based on the characteristics of natural folds with a fractal pattern, the fold patterns are simulated to describe various types of folds quantitatively by means of fractal interpolation. The major factors affecting the fold pattern are elucidated in fractal simulation of folds, i.e. positions of interpolation points (x, y) and the disturbance coefficient d of folds (-1<d<1). The bigger the value d for a fold simulation is, the more complex or disturbed the folds are and the better developed the relative secondary folds are. If d>0, folds are up-convex. The new remote sensor is composed of three parts: a main instrument with a portable computer, an antenna with an amplifier and an external power.The new remote sensing technique is characterized by good stability and reproducibility at the same spot but at different times and high sensitivity and high signal-to-noise ratio, and can reveal geological and lithologic boundaries as well as strata and related mineral sources. | 13 | Wind vector retrieval using dual polarization imagery of ASAR显示文摘A new algorithm for retrieving wind vector using dual polarization imagery of advanced synthetic aperture radar (ASAR) is developed and tested. Based on the combination of co-polarization and cross-polarization, this new algorithm effectively eliminates the 180° ambiguity which occurs when using single imagery of the ASAR to retrieve wind vector. This algorithm also solves the problem that the retrieval results will break down on very small spatial scales. The results retrieved from dual polarization imagery of ASAR show that the wind speed and direction are in agreement with the data from Quikscat and buoy measurements. The root mean square errors of wind direction and speed between the retrieved results and data from Quikscat are 2.21° and 0.53 m/s, respectively.
