1Tumor extracellular vesicles mediate anti-PD-L1 therapyresistance by decoying anti-PD-L1显示文摘PD-L1+tumor-derived extracellular vesicles(TEVs)cause systemic immunosuppression and possibly resistance to anti-PD-L1 antibody(αPD-L1)blockade.However,whether and how PD-L1+TEVs mediateαPD-L1 therapy resistance is unknown.Here,we show that PD-L1+TEVs substantially decoyαPD-L1 and that TEV-boundαPD-L1 is more rapidly cleared by macrophages,causing insufficient blockade of tumor PD-L1 and subsequentαPD-L1 therapy resistance.Inhibition of endogenous production of TEVs by Rab27a or Coro1a knockout reversesαPD-L1 therapy resistance.Either an increasedαPD-L1 dose or macrophage depletion mediated by the clinical drug pexidartinib abolishesαPD-L1 therapy resistance.Moreover,in the treatment cycle with the same total treatment dose ofαPD-L1,high-dose and low-frequency treatment had better antitumor effects than low-dose and highfrequency treatment,induced stronger antitumor immune memory,and eliminatedαPD-L1 therapy resistance.Notably,in humanized immune system mice with human xenograft tumors,both increasedαPD-L1 dose and high-dose and low-frequency treatment enhanced the antitumor effects ofαPD-L1.Furthermore,increased doses ofαPD-L1 andαPD-1 had comparable antitumor effects,butαPD-L1 amplified fewer PD-1+Treg cells,which are responsible for tumor hyperprogression.Altogether,our results reveal a TEV-mediated mechanism ofαPD-L1-specific therapy resistance,thus providing promising strategies to improveαPD-L1 efficacy.Jiming Chen Jie Yang Wenhui Wang Danfeng Guo Chengyan Zhang Shibo Wang Xinliang Lu Xiaofang Huang Pingli Wang Gensheng Zhang Jing Zhang Jianli Wang Zhijian Cai 2022Cellular & Molecular Immunology2022,19,11:1

7Detached eddy simulation on the structure of swirling jet flow field显示文摘The improved delayed detached eddy simulation method with shear stress transport model was used to analyze the evolution of vortex structure,velocity and pressure fields of swirling jet.The influence of nozzle pressure drop on vortex structure development and turbulence pulsation was investigated.The development of vortex structure could be divided into three stages:Kelvin-Helmholtz(K-H)instability,transition stage and swirling flow instability.Swirling flow could significantly enhance radial turbulence pulsation and increase diffusion angle.At the downstream of the jet flow,turbulence pulsation dissipation was the main reason for jet velocity attenuation.With the increase of pressure drop,the jet velocity,pulsation amplitude and the symmetry of velocity distribution increased correspondingly.Meanwhile the pressure pulsation along with the axis and vortex transport intensity also increased significantly.When the jet distance exceeded about 9 times the dimensionless jet distance,the impact distance of swirling jet could not be improved effectively by increasing the pressure drop.However,it could effectively increase the swirl intensity and jet diffusion angle.The swirling jet is more suitable for radial horizontal drilling with large hole size,coalbed methane horizontal well cavity completion and roadway drilling and pressure relief,etc.CHEN Jianxiang YANG Ruiyue HUANG Zhongwei LI Gensheng QIN Xiaozhou LI Jingbin WU Xiaoguang 2022Petroleum Exploration and Development2022,49,4:0
8Probabilistic model of disc-cutter wear in TBM construction:A case study of Chaoer to Xiliao water conveyance tunnel in China显示文摘Disc-cutters play a crucial role in the penetration process during construction using a tunnel boring machine(TBM).Their wear status has a significant impact on working efficiency and costs that accounts for a large part of the budget.Discovering the wear pattern and characteristics of disc-cutters could provide a valuable guide for field maintenance and cutter replacement work.It also provides insight into the wear mechanism of cutters at different positions for optimizing the disc-cutter arrangement design.In this study,the cutter wears data of 34 disc-cutters over 643 days in the Chaoer River to Xiliao River water diversion tunnel was collected.The dataset contains 21862 manual readings measured from 1079 disc-cutters replaced in this project.The raw data from the hard copy version was transformed into a digital twin database by eliminating abnormal data,filling in empty values,and performing linear interpolations.It has been found that the cutters can pass statistical testing for an exponential probability distribution function with respect to the wear rate(w).The regression ratios of R2are essentially greater than 0.8.These findings would help estimate the future service life of a currently working cutter,which means significant savings for the costly disccutters.The application of exponential distribution has the advantage of only one shape parameter,λ,whose reciprocal represents both the statistical mean and standard deviation of the wear rates.It is simple and practically amenable.A preliminary study was carried out to simulate the wear process between two neighboring cutters for drafting a replacement plan for disc-cutters by the Monte Carlo method.The prediction results agreed reasonably well with the measured information.YANG WenKun CHEN ZuYu WU GenSheng XING Hai 2023Science China(Technological Sciences)2023,66,12:0
9Progress on research and mitigation of wind-blown sand risk in Dunhuang Singing Sand Mountain and Crescent Spring Scenic area,China显示文摘The Singing Sand Mountain and Crescent Spring Scenic Spot in Dunhuang,Northwest China is a world-renowned desert attraction that is also an integral component of the Dunhuang UNESCO Global Geopark.This scenic area underwent a 30-year transformation,i.e.,from a severe sand risk with spring water threatened by sand burial due to dune deformation,to restoration of the original sand flow field and mitigation of the sand burial problem.The current paper summarizes the research on the intensive monitoring of the dynamic change of star dunes near the spring,observation of wind and sand flow movement,and then restoring the harmonic vibration of the sand particles(singing sand)that were previously silenced.The existing and prospective impacts of anthropogenic and natural forces on the deformation of the sand dunes are investigated by integrated methods,guiding the implementation of mitigating measures with significant ameliorative effects.Contrast to common sand control practices that aim to reduce wind speed and stop blown sands,our research highlights the importance of maintaining the natural wind flow field in stabilizing surrounding dunes.These mitigation measures consist of removing excessive vegetation and newly constructed buildings to recover the original wind flow field and sand transport activity.Such research and mitigation efforts ensure the scientific protection and restoration of the special desert landform,and contribute to the mutual enhancement of the conservation and exploitation of this desert scenic spot and similar sites.BenLi Liu KeCun Zhang JianJun Qu HaiJiang Li QingHe Niu ZhiShan An YingJun Pang LiHai Tan GenSheng Yang 2023Research in Cold and Arid Regions2023,15,3:0
10Investigation on the interaction length and access resistance of a nanopore with an atomic force microscopy显示文摘Nanopore devices have attracted a lot of attention for their potential application in DNA sequencing. Here, we study how an occluding object placed near a nanopore affects its access resistance by integrating an atomic force microscopy with a nanopore sensor. It is found that there exists a critical hemisphere around the nanopore, inside which the tip of an atomic force microscopy will affect the ionic current. The radius of this hemisphere, which is a bit smaller than the theoretical capture radius of ions, increases linearly with the applied bias voltage and quadratically with the nanopore diameter, but is independent of the operation modes and scanning speeds of the atomic force microscopy. 11Gp350-anchored extracellular vesicles: promising vehicles for delivering therapeutic drugs of B cell malignancies显示文摘Although chimeric antigen receptor-modified(CAR)T cell therapy has been successfully applied in the treatment of acute B lymphocytic leukemia,its effect on Burkitt lymphoma(BL)and chronic B lymphocytic leukemia(B-CLL)is unsatisfactory.Moreover,fatal side effects greatly impede CAR T cell application.Extracellular vesicles(EVs)are excellent carriers of therapeutic agents.Nevertheless,EVs mainly accumulate in the liver when administered without modification.As an envelope glycoprotein of Epstein–Barr viruses,gp350 can efficiently bind CD21 on B cells.Here,gp350 was directly anchored onto red blood cell EVs(RBC-EVs)via its transmembrane region combined with low-voltage electroporation.The results showed that gp350 could anchor to RBC-EVs with high efficiency and that the resulting gp350-anchored RBC-EVs(RBC-EVs/gp350^(Etp))exhibited increased targeting to CD21+BL and B-CLL relative to RBC-EVs.After the loading of doxorubicin or fludarabine,RBC-EVs/gp350^(Etp) had powerful cytotoxicity and therapeutic efficacy on CD21+BL or B-CLL,respectively.Moreover,RBC-EVs/gp350^(Etp) loaded with a drug did not exhibit any apparent systemic toxicity and specifically induced the apoptosis of tumor B cells but not normal Bcells.Therefore,our findings indicate that drug-loaded RBC-EVs/gp350^(Etp) may be adopted in the treatment of CD21+B cell malignancies.Huiqing Xiu Xi Nan Danfeng Guo JiaoliWang Jiahui Li Yanmei Peng Guirun Xiong Shibo Wang Changjun Wang Gensheng Zhang Yunshan Yang Zhijian Cai 2022Asian Journal of Pharmaceutical Sciences2022,17,3:0
12Preliminary Report on Species and Occurrence Regularity of Diseases,Pests and Weeds of Corn in Angola显示文摘[Objective]The paper was to understand species of diseases,pests and weeds and their occurrence periods and dynamic changes of quantity during corn growing period in rainy and dry seasons in Angola.[Method]Using systematic investigation method,the species and occurrence regularity of diseases,pests and weeds of corn in Pedras Negras farm(Malanje),Sanza Pombo farm(Uige)and Catete farm(Luanda)in the African continent,the Republic of Angola,were studied in two dry seasons and a rainy season from 2011 to 2012.[Result]The occurrence status of pests and diseases of corn during dry and rainy seasons were basically similar,and only individual diseases and pests had different incidence degrees.The corn varieties from the United States and Brazil had prominent advantages in resistance to diseases and barren.Due to different climate environments in dry and rainy seasons,the unearthed rate of weeds in fields was also different.Soil moisture had great impact on weed population and density.Soil layer depth greatly influenced unearthed speed and unearthed rate of weeds.[Conclusion]The study cleared species,occurrence period and harmful levels of pests,diseases and weeds of corn in three provinces of Angola.Xue Gensheng Liu Bin Wang Lusheng Liu Jianhua Liu Dalu Yang Hui 2013Plant Diseases and Pests2013,4,5:0
13Bubble dynamics characteristics and influencing factors on the cavitation collapse intensity for self-resonating cavitating jets显示文摘Based on bubble dynamics theory, a mathematic model describing the cavitation bubble size variation in the flow field of self-resonating cavitating jet was developed considering the pressure field and mass and heat exchange between cavitation bubble and ambient fluid. With this model, the influence factors on the cavitation intensity are investigated. The results show that the destructiveness of cavitating jet in breaking rocks depends on the bubble's first collapse, with decreasing intensity in the subsequent collapses. The self-resonating effect significantly enhances the cavitation intensity by promoting the collapse pressure and elongating its duration. Hydraulic parameters are proven to be the dominating factors influencing cavitation intensity: while collapse intensity monotonously increases with jet velocity, there exists an optimum ambient pressure where highest collapse intensity can be achieved. Conversely, the fluid properties show minor influences: cavitation intensity only slightly decreases with the increasing of fluid's density and barely changes with the variation of viscosity and surface tension. The results from this investigation help to uncover the mechanism of the enhanced erosion potential of self-resonating cavitating jet.
