| 1 | Oxidative stress, mitochondrial damage and neurodegenerative diseases显示文摘Oxidative stress and mitochondrial damage have been implicated in the pathogenesis of several neurodegenerative diseases, including Alzheimer's disease, Parkinson's disease and amyotrophic lateral sclerosis. Oxidative stress is characterized by the overproduction of reactive oxygen species, which can induce mitochondrial DNA mutations, damage the mitochondrial respiratory chain, alter membrane permeability, and influence Ca2+ homeostasis and mitochondrial defense systems. All these changes are implicated in the development of these neurodegenerative diseases, mediating or amplifying neuronal dysfunction and triggering neurodegeneration. | 2 | Oxidative stress in neurodegenerative diseases显示文摘Reactive oxygen species are constantly produced in aerobic organisms as by-products of normal oxygen metabolism and include free radicals such as superoxide anion (O2-) and hydroxyl radical (OH-), and non-radical hydrogen peroxide (H2O2). The mitochondrial respiratory chain and enzymatic reactions by various enzymes are endogenous sources of reactive oxygen species. Exogenous reactive oxygen species -inducing stressors include ionizing radiation, ultraviolet light, and divergent oxidizing chemicals. At low concentrations, reactive oxygen species serve as an important second messenger in cell signaling; however, at higher concentrations and long-term exposure, reactive oxygen species can damage cellular macromolecules such as DNA, proteins, and lipids, which leads to necrotic and apoptotic cell death. Oxidative stress is a condition of imbalance between reactive oxygen species formation and cellular antioxidant capacity due to enhanced ROS generation and/or dysfunction of the antioxidant system. Biochemical alterations in these macromolecular components can lead to various pathological conditions and human diseases, especially neurodegenerative diseases. Neurodegenerative diseases are morphologically featured by progressive cell loss in specific vulnerable neuronal cells, often associated with cytoskeletal protein aggregates forming inclusions in neurons and/or glial cells. Deposition of abnormal aggregated proteins and disruption of metal ions homeostasis are highly associated with oxidative stress. The main aim of this review is to present as much detailed information as possible that is available on various neurodegenerative disorders and their connection with oxidative stress. A variety of therapeutic strategies designed to address these pathological processes are also described. | 3 | Ground Surface Ruptures and Near-Fault,Large-Scale Displacements Caused by the Wenchuan Ms8.0 Earthquake Derived from Pixel Offset Tracking on Synthetic Aperture Radar Images显示文摘The 12 May 2008 Wenchuan Ms8.0 earthquake produced surface displacements along the causative fault,the Yingxiu-Beichuan Fault,which are up to several meters near the fault.Because of the large gradient,satellite synthetic aperture radar(SAR) interferometric data are strongly incoherent;the usual SAR interferometry method does not allow such displacements to be measured.In the present study,we employed another approach,the technique based on pixel offset tracking,to solve this problem.The used image data of six tracks are from the Advanced Land Observing Satellite,Phased Array type L-band Synthetic Aperture Radar(ALOS/PALSAR) dataset of Japan.The results show that the entire surface rupture belt is 238 km long,extending almost linearly in a direction of 42° north-east.It is offset left laterally by a north-west-striking fault at Xiaoyudong,and turns at Gaochuan,where the rupture belt shifts toward the south by 5 km,largely keeping the original trend.In terms of the features of the rupture traces,the rupture belt can be divided into five sections and three types.Among them,the Beichuan-Chaping and Hongkou-Yingxiu sections are relatively complex,with large widths and variable traces along the trend.The Pingtong-Nanba and Qingping-Jingtang sections appear uniform,characterized by straight traces and small widths.West of Yingxiu,the rupture traces are not clear.North of the rupture belt,surface displacements are 2.95 m on average,mostly 2-3.5 m,with 7-9 m the maximum near Beichuan.South of the rupture belt,the average displacement is 1.75 m,dominated by 1-2 m,with 3-4 m at a few sites.In the north,the displacements in the radar line of sight are of subsidence,and in the south,they are uplifted,in accordance with a right-slip motion that moves the northern wall of the fault to the east,and the southern wall to the west,respectively.Along the Guanxian-Jiangyou Fault,there is a uplift zone in the radar line of sight,which is 66 km long,1.5-6 km wide,and has vertical displacements of approximately 2 m,but no observable rupture traces. | QU Chunyan SHAN Xinjian LIU Yunhua ZHANG Guohong SONG Xiaogang ZHANG Guifang GUO Liming HAN Yufei | 2012 | Acta Geologica Sinica(English Edition)2012,86,2: | 10 |
| 4 | ERP dissociation and connection between implicit and explicit memory显示文摘ERP dissociations between implicit and explicit memory have been confirmed by a large amount of evidence in theories of human memory. However, similarities between the two forms of memory have scarcely been studied. A possibility is that while implicit memory and explicit memory have independ- ent components, they might additionally have shared components. To explore this question, an ERP experiment was conducted with a study-to-test paradigm, in which participants performed a 'shallow' (color) study task or a 'deep' (pleasant) study task, followed by either a lexical decision (implicit) test (Section 1) or a recognition (explicit) test (Section 2). An interference task was performed concurrently with either the encoding or the retrieval phase of the memory task for encoding interference condition or retrieval interference condition. We compared ERP signatures of implicit and explicit memory as a function of depth of processing or interference. Under the action of the same variables, 300―500 ms old/new ERP effects of implicit and explicit memory showed the same trend. These effects maybe all link with a perceptual representational system. 500―700 ms old/new ERP effects of the two memories were dissociated. They may probably reflect voluntary and involuntary recollection respectively. | 5 | Orientation to learning context modulates retrieval processing for unrecognized words显示文摘Explicit memory errors may occur when individuals fail to retrieve information about items previously studied (item memory) or about the learning context (source memory). We examined electrophysiological measures during recognition failure in order to determine the influence of retrieval orientation for item versus source information. Recognition failure was associated with brain potentials distinct from those associated with success. Furthermore, source-memory failures were associated with earlier-onset brain potentials with a more anterior distribution compared to item-memory failures. | 6 | Effects of a two-meal daily feeding pattern with varied crude protein levels on growth performance and antioxidant indexes in pigs显示文摘The present study aimed to evaluate the effects of daily feeding pattern on growth performance, blood biochemistry, and antioxidant indexes in pigs. One hundred and eighty female Duroc x Landrace x Yorkshire(DLY) pigs with similar body weight(11.00 ± 0.12 kg) were randomly assigned to 3 groups: the control group(fed 17.01% CP diet, twice daily); high-low group(H-L group, fed18.33% CP diet in the morning, followed by 15.70% CP diet in the afternoon); and low-high group(L-H group, fed 15.70% CP diet in the morning, followed by 18.33% CP diet in the afternoon)(n = 6). Comparable amounts of their respective diets were given at 05:30 and 15:00 throughout the experimental periods to make all the treatments consumed the same type of food and the same amount of calories on a daily basis. On day 30, one pig was randomly selected per litter for blood samples. Compared with the control group, ADG in the H-L and L-H groups increased by 8.11% and 16.23%, but not significant(P > 0.05); and blood urea nitrogen(BUN) in the H-L and L-H groups decreased by 26.76% and 41.04%(P < 0.05), respectively. The H-L group feeding pattern could significantly improve levels of serum superoxide dismutase(SOD), when compared with the control group. These findings suggest that the twomeal daily feeding pattern with varied levels of CP affects serum levels of BUN and SOD. | 7 | Baicalin and deferoxamine alleviate iron accumulation in different brain regions of Parkinson's disease rats显示文摘Previous studies found that iron accumulates in the substantia nigra of Parkinson's disease patients. However, it is still unclear whether other brain regions have iron accumulation as well. In this experiment, rats with rotenone-induced Parkinson's disease were treated by gastric perfusion of baicalin or intraperitoneal injection of deferoxamine. Immunohistochemical staining demonstrated that iron accumulated not only in the substantia nigra pars compacta, but also significantly in the striatum globus pallidus, the dentate gyrus granular layer of the hippocampus, the dentate-interpositus and the facial nucleus of the cerebellum. Both baicalin and deferoxamine, which are iron chelating agents, significantly inhibited iron deposition in these brain areas, and substantially reduced the loss of tyrosine hydroxylase-positive cells. These chelators also reduced iron content in the substantia nigra. In addition to the substantia nigra, iron deposition was observed in other brain regions as well. | 8 | An event-related potential study of working memory in children显示文摘To examine the neural mechanisms o working memory in children, event-related potentials (ERPs) were recorded from the 12―13 year-old while they performed a delayed match-to-sample task. The ERP results revealed that new and studied objects both evoked a late positive ERP componen peaking around 350 ms during the working memory process. New objects evoke a more positive ERP waveform than the studied objects. The scalp distri bution showed that the frontal-central electrode sites were associated with object working memory proc esses. When tracking new or studied targets among visual distracters, ERPs of targets and distracters revealed differential responses as early as 150 ms The visual targets evoked larger and more positive ERP responses than the distracters. The typica old-new effect was observed between ERPs of stud ied and new distracters. However, ERPs of new and studied targets differed at about 250 ms, in which new targets evoked more positive-going and slightly earlier ERP responses. | 9 | Baicalin suppresses iron accumulation after substantia nigra injury: relationship between iron concentration and transferrin expression显示文摘Previous studies have shown that baicalin prevented iron accumulation after substantia nigra injury, reduced divalent metal transporter 1 expression, and increased ferroportin 1 expression in the substantia nigra of rotenone-induced Parkinson's disease rats. In the current study, we investigated the relationship between iron accumulation and transferrin expression in C6 cells, to explore the mechanisms of the inhibitory effect of baicalin on iron accumulation observed in Parkinson's disease rats. Iron content was detected using inductively coupled plasma-atomic emission spectroscopy. Results showed that iron content decreased 41% after blocking divalent metal transporter 1 and ferroportin 1 proteins. After treatment with ferric ammonium citrate of differing concentrations(10, 50, 100, 400 μg/mL) in C6 glioma cells, cell survival rate and ferroportin 1 expression were negatively correlated with ferric ammonium citrate concentration, but divalent metal transporter 1 expression positively correlated with ferric ammonium citrate concentration. Baicalin or deferoxamine reduced divalent metal transporter 1 expression, but increased ferroportin 1 expression in the 100 μg/mL ferric ammonium citrate-loaded C6 cells. | 13 | Enhanced production of seed oil with improved fatty acid composition by overexpressing NAD^(+)-dependent glycerol-3-phosphate dehydrogenase in soybean显示文摘There is growing interest in expanding the production of soybean oils(mainly triacylglycerol, or TAG) to meet rising feed demand and address global energy concerns. We report that a plastidlocalized glycerol-3-phosphate dehydrogenase(GPDH), encoded by GmGPDHp1 gene, catalyzes the formation of glycerol-3-phosphate(G3 P), an obligate substrate required for TAG biosynthesis.Overexpression of GmGPDHp1 increases soybean seed oil content with high levels of unsaturated fatty acids(FAs), especially oleic acid(C18:1), without detectably affecting growth or seed protein content or seed weight. Based on the lipidomic analyses, we found that the increase in G3 P content led to an elevated diacylglycerol(DAG) pool, in which the Kennedy pathwayderived DAG was mostly increased, followed by PC-derived DAG, thereby promoting the synthesis of TAG containing relatively high proportion of C18:1. The increased G3 P levels induced several transcriptional alterations of genes involved in the glycerolipid pathways. In particular, genes encoding the enzymes responsible for de novo glycerolipid synthesis were largely upregulated in the transgenic lines, in-line with the identified biochemical phenotype. | 16 | Effect of Lamb Age and in vitro Culture System of Oocytes on JIVET Technology显示文摘[Objectives] This study was conducted to investigate the effects of lamb age and in vitro culture system of oocytes on the results of juvenile in vitro embryo transfer( JIVET). [Methods]Ten Dorper × small-tailed Han lambs aged 5 to 10 weeks were induced to superovulate via i. p. injection of pregnant mare's serum gonadotropin( PMSG). The oocytes were matured in basal maturation solution or modified maturation solution,which was prepared by adding 200 μmol/L cysteine to the basal maturation solution. Then,the oocytes were fertilized in fertilization medium I containing 2% estrus sheep serum( ESS) or fertilization medium II containing 3 mg/ml bull serum albumin( BSA). Finally,the number of oocytes,oocyte maturation rate and cleavage rate of the lambs of different ages were determined. [Results]The average number of oocytes recovered per lamb was( 111. 00 ± 16. 97),( 139. 50 ± 28. 99),( 108. 50 ± 17. 68) and( 42. 00 ± 11. 31) for5-,7-,8-and 10-week-old Dorper × small-tailed Han lambs,respectively. The number of oocytes obtained from 5-,7-and 8-week-old lambs was significantly higher than that from 10-week-old lambs( P < 0. 05),but there was no significant difference among 5-,7-and 8-week-old lambs( P > 0. 05). The maturation rate of oocytes cultured in modified maturation solution was 3. 64% higher than that in basal maturation solution. The cleavage rate of oocytes in fertilization medium I was very significantly higher than that in fertilization medium II( P < 0. 01). | 17 | Interactions between working memory and selective attention显示文摘Event-related potential (ERP) was used to examine the interactions between working memory and se- lective attention. We combined two unrelated tasks, one requiring working memory and the other se- lective attention, which were performed by some undergraduates. The ERP results revealed that both congruent and incongruent stimuli in the selective attention task evoked an N400 component, reaching the peak point at around 500 ms. The N400 evoked by incongruent stimuli was more negative than that of congruent, which indicated the difference of semantic N400. Furthermore, working memory load had a significant influence on the N400 evoked by selective attention task in parietal region. And working memory load showed difference in the ERPs of working memory retrieval in central and parietal regions. The ERPs of probe under high working memory load were more positive from 350 to 550 ms post-stimulus; however, stimulus type of selective attention had no influence on working memory re- trieval. The present study shows that working memory does not play a major role in the selective at- tention, especially in ignoring distracter, but it influences the performance of the selective attention as the background. | 18 | The Surface Rupture Zone and Coseismic Deformation Produced by the Yutian Ms7.3 Earthquake of 21 March 2008,Xinjiang显示文摘On 21 March 2008,a Ms7.3 earthquake occurred at Quickbird,Yutian County,Xinjiang.We attempt to reveal the features of the causative fault of this shock and its coseismic deformation field.Our work is based on analysis and interpretation to high-resolution satellite images as well as differential interferometric synthetic aperture radar(D-InSAR) data from the satellite Envisat SAR,coupled with seismicity,focal mechanism solutions and active tectonics in this region.The result shows that the 40km-long,nearly NS trending surface rupture zone by this event lies on a range-front alluvial platform in Qira County.It is characterized by distinct linear traces and simple structure with 1-3-m-wide individual seams and maximum 6.5m width of a collapse fracture.Along the rupture zone many secondary fractures and fault-bounded blocks are seen,exhibiting remarkable extension.The coseismic deformation affected a large area 100×100km2.D-InSAR analysis indicates that the interferometric deformation field is dominated by extensional faulting with a small strike-slip component.Along the causative fault,the western wall fell down and the eastern wall,that is the active unit,rose up,both with westerly vergence.Because of the big deformation gradients near the seismogenic fault,no interference fringes are seen on images,and what can be determined is a vertical displacement 70cm or more between the two fault walls.According to the epicenter and differential occurrence times from the National Earthquake Information Center,China Earthquake Network Center,Harvard and USGS,it is suggested that the seismic fault ruptured from north to south. | SHAN Xinjian QU Chunyan WANG Chisheng ZHANG Guifang ZHANG Guohong SONG Xiaogang GUO Liming LIU Yunhua | 2012 | Acta Geologica Sinica(English Edition)2012,86,1: | 2 |
| 19 | BUB1B and circBUB1B_544aa aggravate multiple myeloma malignancy through evoking chromosomal instability显示文摘Multiple myeloma(MM)is an incurable plasma cell malignancy in the bone marrow characterized by chromosome instability(CIN),which contributes to the acquisition of heterogeneity,along with MM progression,drug resistance,and relapse.In this study,we elucidated that the expression of BUB1B increased strikingly in MM patients and was closely correlated with poor outcomes.Overexpression of BUB1B facilitated cellular proliferation and induced drug resistance in vitro and in vivo,while genetic targeting BUB1B abrogated this effect.Mechanistic studies unveiled that enforced expression of BUB1B evoked CIN resulting in MM poor outcomes mainly through phosphorylating CEP170.Interestingly,we discovered the existence of circBUB1B_544aa containing the kinase catalytic center of BUB1B,which was translated by a circular RNA of BUB1B.The circBUB1B_544aa elevated in MM peripheral blood samples was closely associated with MM poor outcomes and played a synergistic effect with BUB1B on evoking CIN.In addition,MM cells could secrete circBUB1B_544aa and interfere the MM microenvironmental cells in the same manner as BUB1B full-length protein.Intriguingly,BUB1B siRNA,targeting the kinase catalytic center of both BUB1B and circBUB1B_544aa,significantly inhibited MM malignancy in vitro and in vivo.Collectively,BUB1B and circBUB1B_544aa are promising prognostic and therapeutic targets of MM. | Xiaozhu Tang Mengjie guo Pinggang Ding Zhendong Deng Mengying Ke Yuxia Yuan Yanyan Zhou Zigen Lin Muxi Li Chunyan Gu Xiaosong Gu Ye Yang | 2021 | Signal Transduction and Targeted Therapy2021,6,11: | 2 |
| 20 | Global well-posedness of the fractional Klein-Gordon-Schr¨odinger system with rough initial data显示文摘We investigate the low regularity local and global well-posedness of the Cauchy problem for the coupled Klein-Gordon-Schr¨odinger system with fractional Laplacian in the Schr¨odinger equation in R^(1+1). We use Bourgain space method to study this problem and prove that this system is locally well-posed for Schr¨odinger data in H^(s_1) and wave data in H^(s_2) × H^(s_2-1)for 3/4- α < s_1≤0 and-1/2 < s_2 < 3/2, where α is the fractional power of Laplacian which satisfies 3/4 < α≤1.
