5Enhancing survival, engraftment, and osteogenic potential of mesenchymal stem cells显示文摘Mesenchymal stem cells(MSCs)are promising candidates for bone regeneration therapies due to their plasticity and easiness of sourcing.MSC-based treatments are generally considered a safe procedure,however,the long-term results obtained up to now are far from satisfactory.The main causes of these therapeutic limitations are inefficient homing,engraftment,and osteogenic differentiation.Many studies have proposed modifications to improve MSC engraftment and osteogenic differentiation of the transplanted cells.Several strategies are aimed to improve cell resistance to the hostile microenvironment found in the recipient tissue and increase cell survival after transplantation.These strategies could range from a simple modification of the culture conditions,known as cell-preconditioning,to the genetic modification of the cells to avoid cellular senescence.Many efforts have also been done in order to enhance the osteogenic potential of the transplanted cells and induce bone formation,mainly by the use of bioactive or biomimetic scaffolds,although alternative approaches will also be discussed.This review aims to summarize several of the most recent approaches,providing an up-to-date view of the main developments in MSCbased regenerative techniques.Daniel García-Sánchez Darío Fernández JoséC Rodríguez-Rey Flor M Pérez-Campo 2019World Journal of Stem Cells2019,11,10:4
6Cytotoxic effect of Spirulina platensis extracts on human acute leukemia Kasumi-1 and chronic myelogenous leukemia K-562 cell lines显示文摘Objective: To evaluate the cytotoxic effects of Spirulina platensis extracts on acute leukemia Kasumi-1 and chronic leukemia K-562 cancer cell lines.Methods: Various concentrations of Spirulina platensis extracts(0.25–50.00 mg/m L)obtained with different solvents were used to treat cell lines for 72 h. For cytotoxic effect studies, cell viability test with trypan blue solution, MTT assay and microscopic cytomorphological assessment were done.Results: Spirulina extract obtained with 70% ethanol showed significant cytotoxicity in K562 and Kasumi-1 cell lines. With trypan blue solution, IC_(50) values were found to be4.64 mg/m L for K-562 and 3.68 mg/m L for Kusumi-1 cell lines. Spirulina aqueous extract also showed cytotoxicity with trypan blue method, at a slightly higher dose; where IC_(50) values were 12.68 mg/m L for K-562 and 2.13 mg/m L for Kusumi-1 cell lines. The IC_(50) values were found 0.40 mg/m L for K-562 and 0.31 mg/m L for Kusumi-1 cell lines for the 70% ethanol extract according to the MTT assay. Spirulina extract obtained with water also showed cytotoxicity but the dose was a little higher where IC_(50) values were15.77 mg/m L for K-562 and 9.44 mg/m L for Kusumi-1 cell lines. The effect of cytotoxicity with ethanol extract is quite comparable with that observed for cyclophosphamide, which is a chemical used as anticancer agent.Conclusions: The cytotoxicity exhibited by Spirulina extract to cancer cell lines might be due to the presence of phytopigments(carotenoids, chlorophyll, phycocyanin) as well as polysaccharides that were reported previously as constituents of the extract. 10Simulation Tools for a Fiber-Optic Based Structural Health Monitoring System显示文摘Probability of detection(POD)graphics allow for a change from qualitative to quantitative assessment for every damage detection system,and as such it is a main request for conventional non-destructive testing(NDT)techniques.Its availability can greatly help towards the industrialization of the corresponding Structural health monitoring(SHM)system.But having in mind that for SHM systems the sensors are at fixed positions,and the location of a potential damage would change its detectability.Consequently robust simulation tools are required to obtain the model assisted probability of detection(MAPOD)which is needed to validate the SHM system.This tool may also help for the optimization of the sensor distribution,and finally will allow a probabilistic risk management.INDEUS,simulation of ultrasonic waves SHM system,was a main milestone in this direction.This article deals with the simulation tools for a strain based SHM system,using fiber optic sensors(FOS).FOS are essentially strain/temperature sensors,either with multi-point or with distributed sensing.The simulation tool includes the finite element model(FEM)for the original and damaged structure,and algorithms to compare the strain data at the pre-established sensors locations,and from this comparison to extract information about damage occurrence and location.The study has been applied to the structure of an all-composite unmanned aircraft vehicle(UAV)now under construction,designed at Universidad Politecnica de Madrid for the inspection of electrical utilities networks.Distributed sensing optical fibers were internally bonded at the fuselage and wing.Routine inspection is planned to be done with the aircraft at the test bench by imposing known loads.From the acquired strain data,damage occurrence may be calculated as slight deviations from the baselines.This is a fast inspection procedure without requiring trained specialists,and it would allow for detection of hidden damages.Simulation indicates that stringer partial debondings are detected before they become critical,while small delaminations as those produced by barely visible impact damages would require a prohibited number of sensing lines.These simulation tools may easily be applied to any other complex structure,just by changing the FEM models.From these results it is shown how a fiber optic based SHM system may be used as a reliable damage detection procedure.Alfredo Güemes Antonio Fernandez-Lopez Jaime García-Ramírez Maria Eugenia Reyes-Perez Flor Criado Zurita 2018Transactions of Nanjing University of Aeronautics and Astronautics2018,35,2:2
11Central line-associated bloodstream infection among children with biliary atresia listed for liver transplantation显示文摘BACKGROUND Pre-transplant nutrition is a key driver of outcomes following liver transplantation in children.Patients with biliary atresia(BA) may have difficulty achieving satisfactory weight gain with enteral nutrition alone,and parenteral nutrition(PN) may be indicated.While PN has been shown to improve anthropometric parameters of children with BA listed for liver transplantation,less is known about the risks,particularly infectious,associated with this therapy among this specific group of patients.AIM To describe the incidence,microbiology,and risk factors of central line-associated bloodstream infection(CLABSI) among children with BA listed for liver transplantation.METHODS Retrospective review of children aged ≤ 2-years of age with BA who were listed for primary liver transplantation at Texas Children's Hospital from 2008 through2015(n = 96).Patients with a central line for administration of PN(n = 63) were identified and details of each CLABSI event were abstracted.We compared the group of patients who experienced CLABSI to the group who did not,to determine whether demographic,clinical,or laboratory factors correlated with development of CLABSI.RESULTS Nineteen of 63 patients(30%,95%CI:19,43) experienced 29 episodes of CLABSI during 4800 line days(6.04 CLABSI per 1000 line days).CLABSI was predominantly associated with Gram-negative organisms(14/29 episodes,48%)including Klebsiella spp.,Enterobacter spp.,and Escherichia coli.The sole polymicrobial infection grew Enterobacter cloacae and Klebsiella pneumoniae.Grampositive organisms(all Staphylococcus spp.) and fungus(all Candida spp.)comprised 9/29(31%) and 6/29(21%) episodes,respectively.No demographic,clinical,or laboratory factors were significantly associated with an increased risk for the first CLABSI event in Cox proportional hazards regression analysis CONCLUSION There is substantial risk for CLABSI among children with BA listed for liver transplantation.No clinical,demographic,or laboratory factor we tested emerged as an independent predictor of CLABSI.While our data did not show an impact of CLABSI on the short-term clinical outcome,it would seem prudent to implement CLABSI reduction strategies in this population to the extent that each CLABSI event represents potentially preventable hospitalization,unnecessary healthcare dollar expenditures,and may exact an opportunity cost,in terms of missed allograft offers.Nicole D Triggs Stacey Beer Sonam Mokha Kat Hosek Danielle Guffey Charles G Minard Flor M Munoz Ryan W Himes 2019World Journal of Hepatology2019,11,2:2
12Mesenchymal stem cells secretome:The cornerstone of cell-free regenerative medicine显示文摘Mesenchymal stem cells(MSCs)are the most frequently used stem cells in clinical trials due to their easy isolation from various adult tissues,their ability of homing to injury sites and their potential to differentiate into multiple cell types.However,the realization that the beneficial effect of MSCs relies mainly on their paracrine action,rather than on their engraftment in the recipient tissue and subsequent differentiation,has opened the way to cell-free therapeutic strategies in regenerative medicine.All the soluble factors and vesicles secreted by MSCs are commonly known as secretome.MSCs secretome has a key role in cell-to-cell communication and has been proven to be an active mediator of immunemodulation and regeneration both in vitro and in vivo.Moreover,the use of secretome has key advantages over cell-based therapies,such as a lower immunogenicity and easy production,handling and storage.Importantly,MSCs can be modulated to alter their secretome composition to better suit specific therapeutic goals,thus,opening a large number of possibilities.Altogether these advantages now place MSCs secretome at the center of an important number of investigations in different clinical contexts,enabling rapid scientific progress in this field.Alberto González-González Daniel García-Sánchez Monica Dotta JoséC Rodríguez-Rey Flor M Pérez-Campo 2020World Journal of Stem Cells2020,12,12:2
