| 1 | Intergrin gene expression profiles of human hepatocellular carcinoma显示文摘AIM: To investigate gene expression profiles of intergringenes in hepatocellular carcinoma (HCC) through theusage of Atlas Human Cancer Array membranes, semi-quantitative reverse transcription polymerase chainreaction (RT-PCR) and Northern blot.METHODS: Hybridization of cDNA array membrane wasperformed with α 32P-labeled cDNA probes synthesizedfrom RNA isolated from hepatocellular carcinoma andadjacent non-cirrhotic liver. AtlasImage, which is asoftware specific to array, was used to analyze theresult. RT-PCR of 24 pairs specimen and Northern blotof 4 pairs specimen were used to confirm the expressionpattern of some intergrin genes identified by Atlasarrays hybridization.RESULTS: Among 588 genes spotted in membrane, 17genes were related to intergrin. Four genes were up-regulated, such as intergrin alpha8, beta1, beta7 andbeta8 in HCC. Whereas there were no genes down-regulated in HCC. RT-PCR and Northern blot analysisof intergrin beta1 gene gave results consistent withcDNA array findings.CONCLUSION: Investigation of these intergrin genesshould help to disclose the molecular mechanism of thecell adhesion, invasive and metastasis of HCC. A fewgenes are reported to have changed in HCC for the firsttime. The quick and high-throughout method of profilinggene expression by cDNA array provides us overviewof key factors that may involved in HCC, and may findthe clue of the study of HCC metastasis and moleculartargets of anti-metastasis therapy. | 3 | Relationship between petroleum hydrocarbon and plankton in a mesocosm experiment显示文摘A mesocosm experiment was carried out from May 26 to June 1, 1998 at the location of 30’52’N, 122’36’E, the Changjiang Estuary, in order to measure the influence of petroleum hydrocar- bon to the ecosystem, furthermore, to estimate the environmental capacity. In the experiment, it is found that the petroleum hydrocarbon can, in some degree, enhance the growth of diatom, but inhibit the growth of dinoflagellate. In general, the petroleum hydrocarbon can inhibit the growth of both Phytoplankton and zooplankton, and can the inhibit the growth of total plankton (including phytoplankton and zooplankton) as well. A kinetic model was presented to estimate the uptake/release rate constants of petroleum hydrocarbon by plankton, and thereafter, the uptake and release rate constants (k1, k2), bioconcentration factor (BCF) as well as the petroleum bydrocarbon influenced influenced uptake and release rate constants of nutrients by phytoplankton (kup, krel) were obtained. The results imply that the bioconcentration of petroleum hydrocarbon by plankton is fairly large and petroleum hydrocarbon caused no significant influence on the uptake of N- NO3, but significant influence on that of P -PO4. | 4 | Expression of p53, p16 and COX-2 in pancreatic cancer with tissue microarray显示文摘BACKGROUND: Pancreatic cancer development and progression is driven by the accumulation of genetic changes. In this study we constructed tissue microarray containing specimens from pancreatic cancer, adjacent non-cancer tissue and normal tissue to survey the expression of p53, p16 and cyclooxyganase-2 (COX-2). METHODS: Tissue microarray containing 337 specimens from different stages of pancreatic cancer, adjacent noncancer tissue and normal tissues was constructed, and the expression of p53, p16 and COX-2 was assayed by immunohistochemistry to consecutive formalin-fixed tissue microarray sections. RESULTS: The expression of p53, p16 and COX-2 was significantly higher in tumorous tissues than in non-tumorons ones. A significant relationship was observed between p53 and COX-2, or p16 and COX-2. But no obvious correlation was seen between p53 and p16 expressions. Logistic regression analysis showed p53 and COX-2 as dependent predictors in pancreatic carcinogenesis, and a reciprocal relationship to neoplastic progression between p53 and COX-2. | 5 | Semi-analytical analysis of the response of the air temperature over the land surface to the global vegetation distribution显示文摘Response of the air temperature over the land surface to the global vegetation distribution is investigated, using a three-dimensional governing equation to simulate the steady, large-scale, limited amplitude perturbation of the free, inviscid and adiabatic atmosphere. The adoption of the static equation leads to a temperature governing equation in the terrain following coordinate. With the prescribed temperature as the upper boundary condition and the radiation balance as the lower boundary condition, the semi-analytical solution of the global circulation temperature can be calculated. In this article, only the air temperature (at 2 m height) over the land surface is analyzed, and the result suggests that this model can simulate the air temperature pattern over the land surface reasonably. A better simulation occurs when a simple feedback of the albedo on the temperature is included. Two sensitivity experiments are analyzed through this model. One suggests that the air temperature over the land surface descends obviously when the land surface is covered with ice all over, while another suggests that the air temperature rises a little when the land surface is covered with forest except the ice-covered area. This model appears to be a good tool to study the response of the air temperature to the vegetation distribution. Because the governing equations are nonlinear, some limited multiple equi-libria of cross-equatorial inertial jets may exist. | 9 | Safe and Objective Assay of Enterovirus 71 Neutralizing Antibodies via Pseudovirus显示文摘Current serum neutralization assays based on the inhibition of the cytopathic effect(Nt-CPE) need to ma nipulate live viruses, which are time-consuming, labor-intensive, and have the potential exposure to infectious agents, so a safe and objective assay via pseudovirus for the fast and efficient detection of enterovirus 71(EV71) neutralizing antibodies was developed. First, we generated EV71 pseudovirus containing firefly luciferase gene in place of the capsid gene P1 in EV71 genome. Vero cells infected with 200 CCID50(50% cell culture infective dose) of EV71 pseudovirus for 24 h were found to have the best performance. Seval sera were measured by EV71 pseudoparticle neutralization assay(Nt-PPN) and the conventional serological method Nt-CPE. Neutralizing antibody titers measured by Nt-PPN and those obtained by Nt-CPE demonstrate a high correlation between the two methods. Overall, the PPN assay represents a valid alternative to conventional serological methods for the evaluation of EV71 neutralizing anti bodies. Move analysis is one aspect that has conducted to describe RA abstract. This article analyzed the particular genre with the purpose of helping them work out good abstracts for academic writing to improve their ability of doing research. One hundred abstracts were selected randomly from all the 450 graduation papers written by Eng lish-major pre-service teachers in 2010 in Hunan First Normal University in China. These informative abstracts were analyzed with the framework of Hyland's 2000 Model. The compound blockage to Na + channel by injection of 2 5μl of 2% lidocaine and 2 5μl of 2% furosemide into the lateral ventricle reduced the selective death in CA3 caused by stress (P<0 01). | 13 | Formation of the thermal-driven boundary jet in an f-plane mesoscale basin显示文摘The paper adopts an f-plane quasi-geostrophic inertial model without linearization to investigate the perturbation temperature, boundary jet and upwelling (downwelling) in an idealized rectangular basin, under the consideration of west side friction layer and heat conservation. There is net heat input on the upper surface and equal quality heat dissipation on the west boundary, and without heat exchange on other boundaries, then the heat is conservation in the whole basin. Results show that there is thermal front due to denseness of the perturbation temperature in the west side boundary, the perturbation pressure and flow field are reversal on the upper layer and bottom layer. On the bottom layer, the west coastal current is northward, and the maximum perturbation pressure center is on the west, however, on the upper layer, the east coastal current is southward, and the maximum perturbation pressure center is on the east. There is strong vertical flow in narrow western boundary layer, and also in the central zone. | 14 | STUDY ON THE STANDARDIZED APPLICATION OF ACUPUNCTURE ANESTHESIA TO CRANIOCEREBRAL OPERATIONS ON TEMPORO-PARIETO-OCCIPITAL REGION AND POSTERIOR CRANIAL FOSSA显示文摘The effect of acupuncture anesthesia(AA) performed with unified protocol andmethod was evaluated by the National Unified Scaling Criteria. Among the 536 cases studied,there are 488 cases Grade I, the rate of Grade I (success rate) being 91.05%. Of the 357 casesof temporo-parieto-occipital operations, the success rate is 91.88%, with the success rates of bodyneedling and ear needling groups being 88.16% and 98.45% respectively. Of the 179 cases of po-sterior cranial fossa operations, the success rate is 89.38%, with the success rates of body needlingand ear needling groups being 85.58% and 95.59% respectively. Randomized controlled trials wereconducted among the simple 0.1% lidocaine group, AA plus 0.1% lidocaine group and AA plusnormal saline group, the results of which prove that acupuncture analgesia plays predominant rolein AA and adjuvants can remarkably enhance the AA effect. The neuroanatomical basis of thesuperiority of ear needling to body needling is discussed here, so is the mechanism of the poten-tiating effect of the adjuvants on AA.
