1The first offshore natural gas hydrate production test in South China Sea显示文摘Natural gas hydrates (NGH)is one of key future clean energy resources.Its industrialized development will help remit the huge demand of global natural gas,relieve the increasing pressure of the environment, and play a vital role in the green sustainable growth of human societies.Based on nearly two decades' studying on the reservoir characteristics in the South China Sea (SCS)and the knowledge of reservoir system,the China Geological Survey (CGS)conducted the first production test on an optimal target selected in Shenhu area SCS in 2017.Guided by the 'three-phase control'exploitation theory which focused on formation stabilization,technologies such as formation fluid extraction,well drilling and completing, reservoir stimulating,sand controlling,environmental monitoring,monitoring and preventing of secondary formation of hydrates were applied.The test lasted for 60 days from May 10^th when starting to pump, drop pressure and ignite to well killing on July 9^th,with gas production of 3.09×10^5 m^3 in total,which is a world record with the longest continuous duration of gas production and maximal gas yield.This successful test brings a significant breakthrough on safety control of NGH production.Jin-fa Li Jian-liang Ye Xu-wen Qin Hai-jun Qiu Neng-you Wu Hai-long Lu Wen-wei Xie Jing-an Lu Fei Peng Zhen-qiang Xu Cheng Lu Zeng-gui Kuang Jian-gong Wei Qian-yong Liang Hong-feng Lu Bei-bei Kou 2018China Geology2018,1,1:117
2Inhibition of SARS-CoV-2 (previously 2019-nCoV) infection by a highly potent pan-coronavirus fusion inhibitor targeting its spike protein that harbors a high capacity to mediate membrane fusion显示文摘The recent outbreak of coronavirus disease(COVID-19)caused by SARS-CoV-2 infection in Wuhan,China has posed a serious threat to global public health.To develop specific anti-coronavirus therapeutics and prophylactics,the molecular mechanism that underlies viral infection must first be defined.Therefore,we herein established a SARS-CoV-2 spike(S)protein-mediated cell-cell fusion assay and found that SARS-CoV-2 showed a superior plasma membrane fusion capacity compared to that of SARS-CoV.We solved the X-ray crystal structure of six-helical bundle(6-HB)core of the HR1 and HR2 domains in the SARS-CoV-2 S protein S2 subunit revealing that several mutated amino acid residues in the HR1 domain may be associated with enhanced interactions with the HR2 domain.We previously developed a pan-coronavirus fusion inhibitor,EK1,which targeted the HR!domain and could inhibit infection by divergent human coronaviruses tested,including SARS-CoV and MERS-CoV.Here we generated a series of lipopeptides derived from EK1 and found that EK1C4 was the most potent fusion inhibitor against SARS-CoV-2 S protein-mediated membrane fusion and pseudovirus infection with IC50s of 1.3 and 15.8 nM,about 241-and 149-fold more potent than the original EK1 peptide,respectively.EK1C4 was also highly effective against membrane fusion and infection of other human coronavirus pseudoviruses tested,including SARS-CoV and MERS-CoV,as well as SARSr-CoVs,and potently inhibited the replication of 5 live human coronaviruses examined,including SARS-CoV-2.Intranasal application of EK1C4 before or after challenge with HCoV-OC43 protected mice from infection,suggesting that EK1C4 could be used for prevention and treatment of infection by the currently circulating SARS-CoV-2 and other emerging SARSr-CoVs.Shuai Xia Meiqin Liu Chao Wang Wei Xu Qiaoshuai Lan Siliang Feng Feifei Qi Linlin Bao Lanying Du Shuwen Liu Chuan Qin Fei Sun Zhengli Shi Yun Zhu Shibo Jiang Lu Lu 2020Cell Research2020,30,4:81

4Treatment outcomes for different subgroups of nasopharyngeal carcinoma patients treated with intensity-modulated radiation therapy显示文摘Although many studies have investigated intensity-modulated radiation therapy (IMRT) for nasopharyngeal carcinoma (NPC),sample sizes in the reported studies are usually small and different in outcomes in different T and N subgroups are seldom analyzed.Herein,we evaluated the outcomes of NPC patients treated with IMRT and further explored treatment strategy to improve such outcome.We collected clinical data of 865 NPC patients treated with IMRT alone or in combination with chemotherapy,and classified all cases into the following prognostic categories according to different TNM stages:early stage group (T1-2N0-1M0),advanced local disease group (T3-4N0-1M0),advanced nodal disease group (T1-2N2-3M0),and advanced locoregional disease group (T3-4N2-3M0).The 5-year overall survival (OS),local relapse-free survival (LRFS),and distant metastases-free survival (DMFS) were 83.0%,90.4%,and 84.0%,respectively.The early disease group had the lowest treatment failure rate,with a 5-year OS of 95.6%.The advanced local disease group and advanced nodal disease group had similar failure pattern and treatment outcomes as well as similar hazard ratios for death (4.230 and 4.625,respectively).The advanced locoregional disease group had the highest incidence of relapse and death,with a 5-year DMFS and OS of 62.3% and 62.2%,respectively,and a hazard ratio for death of 10.402.Comparing with IMRT alone,IMRT in combination with chemotherapy provided no significant benefit to locoregionally advanced NPC.Our results suggest that the decision of treatment strategy for NPC patients should consider combinations of T and N stages,and that IMRT alone for early stage NPC patients can produce satisfactory results.However,for advanced local,nodal,and locoregional disease groups,a combination of chemotherapy and radiotherapy is recommended.Sheng-Fa Su 1,2,3,Fei Han 1,2,Chong Zhao 1,2,Ying Huang 1,2,Chun-Yan Chen 1,2,Wei-Wei Xiao 1,2,Jia-Xin Li 1,2 and Tai-Xiang Lu 1,2 1 State Key Laboratory of Oncology in South China,Guangzhou,Guangdong 510060,P.R.China 2 Department of Radiation Oncology,Sun Yat-sen University Cancer Center,Guangzhou,Guangdong 510060,P.R.China 3 Department of Oncology,Guiyang Medical College Hospital,Tumor Hospital of Guizhou,Guiyang,Guizhou 550003,P.R.China. 2011Chinese Journal of Cancer2011,30,8:40
5Magnifying narrow-band imaging endoscopy is superior in diagnosis of early gastric cancer显示文摘AIM:To evaluate the diagnostic effectiveness of white light endoscopy,magnifying endoscopy(ME),and magnifying narrow-band imaging endoscopy(ME-NBI) in detecting early gastric cancer(EGC).METHODS:From March 2010 to June 2012,a total of 3616 patients received screening for gastric cancer by magnifying endoscopy. There were 3675 focal gastric lesions detected using conventional high definition white light endoscopy(HD-WLE) in four different referentialhospitals that were recruited for further investigation using ME and ME-NBI. The images obtained from HD-WLE,ME,and ME-NBI were reviewed by four experienced endoscopists to evaluate their diagnostic effectiveness for EGC. The diagnosis of cancerous and non-cancerous lesions was conducted by evaluating the microvascular and microsurface patterns using the VS classification system. The final endoscopic diagnosis of each lesion was determined by consultation when a disagreement occurred. We used histopathological results as the gold standard for the diagnosis of EGC.RESULTS:Among the 3675 lesions found,1508 were validated by pathological findings as chronic gastritis,1279 as chronic gastritis with intestinal metaplasia,631 as low-grade neoplasia,and 257 as EGC. The sensitivity,specificity,positive predictive value,negative predictive value,and accuracy of HD-WLE for the diagnosis of EGC were 71.2%,99.1%,85.5%,97.9% and 97.1%,respectively. The results of ME for diagnosing EGC were 81.3%,98.8%,83.3%,98.6% and 97.6%,respectively. The results of ME-NBI for the diagnosis of EGC were 87.2%,98.6%,82.1%,99.0% and 97.8%,respectively. The diagnostic sensitivity and accuracy of paired ME and ME-NBI were significantly better than those of HD-WLE(P < 0.05).CONCLUSION:HD-WLE has a relatively high accuracy for diagnosing EGC and is an effective screening tool. 7Size distribution of chemical elements and their source apportionment in ambient coarse, fine, and ultrafine particles in Shanghai urban summer atmosphere显示文摘Ambient coarse particles (diameter 1.8-10 μm), fine particles (diameter 0.1-1.8 μm), and ultrafine particles (diameter < 0.1 μm) in the atmosphere of the city of Shanghai were sampled during the summer of 2008 (from Aug 27 to Sep 08). Microscopic characterization of the particles was investigated by scanning electron microscopy coupled with energy dispersive X-ray spectroscopy (SEM/EDX). Mass concentrations of Si, P, S, Cl, K, Ca, Ti, V, Cr, Mn, Fe, Ni, Cu, Zn, As, Se, Br, Rb, Sr, and Pb in the size-resolved particles were quantified by using synchrotron radiation X-ray fluorescence (SRXRF). Source apportionment of the chemical elements was analyzed by means of an enrichment factor method. Our results showed that the average mass concentrations of coarse particles, fine particles and ultrafine particles in the summer air were 9.38 ± 2.18, 8.82 ± 3.52, and 2.02 ± 0.41 μg/m3, respectively. The mass percentage of the fine particles accounted for 51.47% in the total mass of PM10, indicating that fine particles are the major component in the Shanghai ambient particles. SEM/EDX results showed that the coarse particles were dominated by minerals, fine particles by soot aggregates and fly ashes, and ultrafine particles by soot particles and unidentified particles. SRXRF results demonstrated that crustal elements were mainly distributed in the coarse particles, while heavy metals were in higher proportions in the fine particles. Source apportionment revealed that Si, K, Ca, Fe, Mn, Rb, and Sr were from crustal sources, and S, Cl, Cu, Zn, As, Se, Br, and Pb from anthropogenic sources. 8Antibacterial Effect of Cinnamon Oil Combined with Thyme or Clove Oil显示文摘Antibacterial effect of cinnamon oil combined with thyme or clove oil was studied in this paper.Agar dilution method was used to determine the minimum inhibitory concentrations(MICs) of the essential oils(EOs) of cinnamon,thyme,and clove oil against three Gram-positive bacteria(Bacillus subtilis,Bacillus cereus,and Staphylococcus aureus),and two Gram-negative bacteria(Escherichia coli and Salmonella typhimurium).The results showed that cinnamon was a promising antibacterial substance with MIC ranged from 0.1 to 0.4 μL mL-1 for the five bacterial species.Agar dilution checkerboard method was used to test the combined antibacterial effect of cinnamon oil with thyme or clove oil.Combination of cinnamon and thyme oil showed an additive effect against all selected bacteria,and combination of cinnamon and clove oil displayed an additive effect against B.subtilis,B.cereus,S.aureus,and an indifferent effect against E.coli and S.typhimurium.Furthermore,gas chromatography-mass spectrometry(GC-MS) measurement was used to analyze the components of the EOs,and the main components of cinnamon,thyme,and clove were cinnamaldehyde,thymol,carvacrol and p-cymene,and eugenol,respectively.LU Fei DING Yi-cheng YE Xing-qian DING Yu-ting 2011Agricultural Sciences in China2011,10,9:19
9Long-term outcomes of a phase Ⅱ randomized controlled trial comparing intensity-modulated radiotherapy with or without weekly cisplatin for the treatment of locally recurrent nasopharyngeal carcinoma显示文摘Background:Salvage treatment for locally recurrent nasopharyngeal carcinoma(NPC) is complicated and relatively limited.Radiotherapy,combined with effective concomitant chemotherapy,may improve clinical treatment outcomes.We conducted a phase Ⅱ randomized controlled trial to evaluate the efficacy of intensity-modulated radiotherapy with concomitant weekly cisplatin on locally recurrent NPC.Methods:Between April 2002 and January 2008,69 patients diagnosed with non-metastatic locally recurrent NPC were randomly assigned to either concomitant chemoradiotherapy group(n = 34) or radiotherapy alone group(n = 35).All patients received intensity-modulated radiotherapy.The radiotherapy dose for both groups was 60 Gy in 27 fractions for 37 days(range 23-53 days).The concomitant chemotherapy schedule was cisplatin 30 mg/m^2 by intravenous infusion weekly during radiotherapy.Results:The median follow-up period of all patients was 35 months(range 2-112 months).Between concomitant chemoradiotherapy and radiotherapy groups,there was only significant difference in the 3-year and 5-year overall survival(OS) rates(68.7%vs.42.2%,P = 0.016 and 41.8%vs.27.5%,P = 0.049,respectively).Subgroup analysis showed that concomitant chemoradiotherapy significantly improved the 5-year OS rate especially for patients in stage rT3-4(33.0%vs.13.2%,P = 0.009),stages Ⅲ-Ⅳ(34.3%vs.13.2%,P = 0.006),recurrence interval >30 months(49.0%vs.20.6%,P = 0.017),and tumor volume >26 cm^3(37.6%vs.0%,P = 0.006).Conclusion:Compared with radiotherapy alone,concomitant chemoradiotherapy can improve OS of the patients with locally recurrent NPC,especially those with advanced T category(rT3-4) and stage(lll-IV) diseases,recurrence intervals >30 months,and tumor volume >26 cm^3.Ying Guan Shuai Liu Han-Yu Wang Ying Guo Wei-Wei Xiao Chun-Yan Chen Chong Zhao Tai-Xiang Lu Fei Han 2016Chinese Journal of Cancer2016,35,4:19
10Effects of neutral sulfate berberine on LPS-induced cardiomyocyte TNF-(αsecretion,abnormal calcium cycling,and cardiac dysfunction in rats显示文摘Aim:To evaluate the effect of neutral sulfate berberine on cardiac function,tumornecrosis factor α(TNF-α)release,and intracellular calcium concentration([Ca^(2+)]_i)in cardiomyocytes exposed to lipopolysaccharide(LPS).Methods:Primary cul-tured rat cardiomyocytes were prepared from ventricles of 3-4-day old Sprague-Dawley rats.TNF-α concentrations in cell-conditioned media were measured byusing a Quantikine enzyme-linked immunosorbent assay kit,and cardiomyocyte[Ca^(2+)]_i was measured by using Fura-2/AM.The isolated rat hearts were perfusedin the Langendorff mode.Results:LPS at doses of 1,5,i0,and 20 μg/mL markedlystimulated TNF-α secretion from cardiomyocytes,and neutral sulfate berberineinhibited LPS-induced TNF-α production.Intracellular calcium concentration wassignificantly decreased after LPS stimulation for 1 h,and increased 2 h after LPStreatment.Pretreatment with neutral sulfate berberine reversed the LPS-induced[Ca^(2+)]_i alterations,although neutral sulfate berberine did not inhibit a rapid in-crease in cardiomyocyte[Ca^(2+)]_i induced by LPS.Perfusion of isolated hearts withLPS(100 μg/mL)for 20 min resulted in significantly impaired cardiac performanceat 120 min after LPS challenge:the maximal rate of left ventricular pressure rise andfall(±dp/dt_(max)decreased compared with the control.In contrast,±dp/dt_(max) at 120min in hearts perfused with neutral sulfate berberine(1 μmol/L)for 10 min followedby 20 rain LPS(100 μg/mL)was greater than the corresponding value in the LPSgroup.Conclusion:Neutral sulfate berberine inhibits LPS-stimulated myocardialTNF-α production,impairs calcium cycling,and improves LPS-induced contrac-tile dysfunction in intact heart.Jing YANG Hua-dong WANG Da-xiang LU Yan-ping WANG Ren-bin QI Jing LI Fei LI Chu-jie LI 2006Acta Pharmacologica Sinica2006,27,2:18
11Expression changes and bioinformatic analysis of Wallerian degeneration after sciatic nerve injury in rat显示文摘Wallerian degeneration (WD) remains an important research topic. Many genes are differentially expressed during the process of WD, but the precise mechanisms responsible for these differentiations are not completely understood. In this study, we used microarrays to analyze the expression changes of the distal nerve stump at 0, 1, 4, 7, 14, 21 and 28 days after sciatic nerve injury in rats. The data revealed 6 076 differentially-expressed genes, with 23 types of expression, specifically enriched in genes associated with nerve development and axonogenesis, cytokine biosynthesis, cell differentiation, cytokine/chemokine production, neuron differentiation, cytokinesis, phosphorylation and axon regeneration. Kyoto Encyclopedia of Genes and Genomes pathway analysis gave findings related mainly to the MAPK signaling pathway, the Jak-STAT signaling pathway, the cell cycle, cytokine-cytokine receptor interaction, the p53 signaling pathway and the Wnt signaling pathway. Some key factors were NGF, MAG, CNTF, CTNNA2, p53, JAK2, PLCB1, STAT3, BDNF, PRKC, collagen II, FGF, THBS4, TNC and c-Src, which were further validated by real-time quantitative PCR, Western blot, and immunohistochemistry. 12The effects of nitrogen fertilizer application on methane and nitrous oxide emission/uptake in Chinese croplands显示文摘The application of nitrogen(N) fertilizer to increase crop yields has a significant influence on soil methane(CH_4) and nitrous oxide(N_2O) emission/uptake.A meta-analysis was carried out on the effect of N application on(i) CH_4 emissions in rice paddies,(ii) CH_4 uptake in upland fields and(iii) N_2O emissions.The responses of CH_4 emissions to N application in rice paddies were highly variable and overall no effects were found.CH_4 emissions were stimulated at low N application rates(<100 kg N ha^(-1)) but inhibited at high N rates(>200 kg N ha^(-1)) as compared to no N fertilizer(control).The response of CH_4 uptake to N application in upland fields was 15%lower than control,with a mean CH_4 uptake factor of-0.001 kg CH_4-C kg^(-1) N.The mean N_2O emission factors were 1.00 and 0.94%for maize(Zea mays) and wheat(Triticum aestivum),respectively,but significantly lower for the rice(Oryza sativa)(0.51%).Compared with controls,N addition overall increased global warming potential of CH_4 and N_2O emissions by 78%.Our result revealed that response of CH_4 emission to N input might depend on the CH_4concentration in rice paddy.The critical factors that affected CH_4 uptake and N_2O emission were N fertilizer application rate and the controls of CH_4 uptake and N_2O emission.The influences of application times,cropping systems and measurement frequency should all be considered when assessing CH_4 and N_2O emissions/uptake induced by N fertilizer.SUN Bin-feng ZHAO Hong LU Yi-zhong Lü Fei WANG Xiao-ke 2016Journal of Integrative Agriculture2016,15,2:18
13Emission characteristics of dioxins,furans and polycyclic aromatic hydrocarbons during fluidized-bed combustion of sewage sludge显示文摘Pre-dried sewage sludge with high sulfur content was combusted in an electrically heated lab-scale fluidized-bed incinerator. The emission characteristics of polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs) and polycyclic aromatic hydrocarbons (PAHs) were studied. Coal and calcium oxide (CaO) were added during the sewage sludge combustion tests to optimize combustion conditions and control SO2 emission. The results indicated that the flue gases emitted during mono-combustion of sewage sludge were characterized by relatively high concentrations of SO2, NOx and organic pollutants, due to the high sulfur, nitrogen, and volatile matter content of sewage sludge. The total 16 USEPA priority PAHs and 2,3,7,8-substituted PCDD/Fs produced from sewage sludge combustion were found to be 106.14 μg/m3 and 8955.93 pg/m3 in the flue gas, respectively. In the case of co- combustion with coal (msludge/mcoal = 1:1), the 16 PAHs and 2,3,7,8-substituted PCDD/Fs concentrations were markedly lower than those found during mono-combustion of sewage sludge. 15Timing, distribution, and microbiology of infectious complications after necrotizing pancreatitis显示文摘BACKGROUND Acute pancreatitis(AP)is a common acute abdominal disease worldwide,and its incidence rate has increased annually.Approximately 20%of AP patients develop into necrotizing pancreatitis(NP),and 40%to 70%of NP patients have infectious complications,which usually indicate a worse prognosis.Infection is an important sign of complications in NP patients.AIM To investigate the difference in infection time,infection site,and infectious strain in NP patients with infectious complications.METHODS The clinical data of AP patients visiting the Department of General Surgery of Xuanwu Hospital of Capital Medical University from January 1,2014 to December 31,2018 were collected retrospectively.Enhanced computerized tomography or magnetic resonance imaging findings in patients with NP were included in the study.Statistical analysis of infectious bacteria,infection site,and infection time in NP patients with infectious complications was performed,because knowledge about pathogens and their antibiotic susceptibility patterns is essential for selecting an appropriate antibiotic.In addition,the factors that might influence the prognosis of patients were analyzed.RESULTS In this study,539 strains of pathogenic bacteria were isolated from 162 patients with NP infection,including 212 strains from pancreatic infections and 327 strains from extrapancreatic infections.Gram-negative bacteria were the main infectious species,the most common of which were Escherichia coli and Pseudomonas aeruginosa.The extrapancreatic infection time(9.1±8.8 d)was earlier than the pancreatic infection time(13.9±12.3 d).Among NP patients with early extrapancreatic infection(<14 d),bacteremia(25.12%)and respiratory tract infection(21.26%)were predominant.Among NP patients with late extrapancreatic infection(>14 d),bacteremia(15.94%),respiratory tract infection(7.74%),and urinary tract infection(7.71%)were predominant.Drug sensitivity analysis showed that P.aeruginosa was sensitive to enzymatic penicillins,thirdand fourth-generation cephalosporins,and carbapenems.Acinetobacter baumannii and Klebsiella pneumoniae were sensitive only to tigecycline;Staphylococcus epidermidis and Enterococcus faecium were highly sensitive to linezolid,tigecycline,and vancomycin.CONCLUSION In this study,we identified the timing,the common species,and site of infection in patients with NP.Jiong-Di Lu Feng Cao Yi-Xuan Ding Yu-Duo Wu Yu-Lin Guo Fei Li 2019World Journal of Gastroenterology2019,25,34:17
16Combined treatment of hepatocellular carcinoma with partial splenic embolization and transcatheter hepatic arterial chemoembolization显示文摘AIM: To prospectively evaluate the effi cacy and safety of partial splenic embolization (PSE) combined with transcatheter hepatic arterial chemoembolization (TACE) in treatment of hepatocellular carcinoma (HCC). METHODS: Fifty patients suffering from primary HCC associated with hypersplenism caused by cirrhosis were randomly assigned to 2 groups: group A receiving PSE combined with TACE (n = 26) and group B receiving TACE alone (n = 24). Follow-up examinations included calculation of peripheral blood cells (leukcytes, platelets and red blood cells) and treatment-associated complications. RESULTS: Prior to treatment, there was no signifi cant difference in sex, age, Child-Pugh grade, tumor diameter, mass pathology type and peripheral blood cell counts between the 2 groups. After treatment, leukocyte and platelet counts were significantly higher in group A during the 3-mo follow-up period (P < 0.05), but lower in group B (P < 0.05). Severe complications occurred in 3 patients (11.5%) of group A and in 19 patients (79.2%) of group B (P < 0.05), and there was no significant difference in symptoms of post-embolization syndrome, including abdominal pain, fever, mild nausea and vomiting between the 2 groups (P > 0.05). 17Higher CO_2-insufflation pressure inhibits the expression of adhesion molecules and the invasion potential of colon cancer cells显示文摘AIM:To investigate the influence of CO2-insufflation pressure on adhesion,invasion and metastatic potential of colon cancer cells based on adhesion molecules expression.METHODS:With an in vitro artificial pneumoperitoneum model,SW1116 human colon carcinoma cells were exposed to CO2-insufflation in 5 different pressure groups:6 mmHg,9 mmHg,12 mmHg,15 mmHg and control group,respectively for 1 h.Expression of E-cadherin,ICAM-1,CD44 and E-selectin was measured at 0,12,24,48 and 72 h after CO2-insufflation using flow cytometry.The adhesion and invasion capacity of SW1116 cells before and after exposure to CO2-insufflation was detected by cell adhesion/invasion assay in vitro.Each group of cells was injected intraperitoneally into 16 BALB/C mice.The number of visible abdominal cavity tumor nodules,visceral metastases and survival of the mice were recorded in each group.RESULTS:The expression of E-cadherin,ICAM-1,CD44 and E-selectin in SW1116 cells were changed significantly following exposure to CO2 insufflation at different pressures(P<0.05).The expression of E-cadherin,CD44 and ICAM-1 decreased with increasing CO2-insufflation pressure.The adhesive/invasive cells also decreased gradually with increasing pressure as determined by the adhesion/invasion assay.In animal experiments,the number of abdominal cavity tumor nodules in the 15 mmHg group was also signif icantly lower than that in the 6 mmHg group(29.7±9.91 vs 41.7±14.90,P=0.046).However,the survival in each group was not statistically different.CONCLUSION:CO2-insufflation induced a temporary change in the adhesion and invasion capacity of cancer cells in vitro.Higher CO2-insufflation pressure inhibited adhesion,invasion and metastatic potential in vitro and in vivo,which was associated with reduced expression of adhesion molecules.Jun-Jun Ma Bo Feng Yi Zhang Jian-Wen Li Ai-Guo Lu Ming-Liang Wang Yuan-Fei Peng Wei-Guo Hu Fei Yue Min-Hua Zheng 2009World Journal of Gastroenterology2009,15,22:16

19Bone marrow mesenchymal stem cells repair spinal cord ischemia/reperfusion injury by promoting axonal growth and anti-autophagy显示文摘Bone marrow mesenchymal stem cells can differentiate into neurons and astrocytes after transplantation in the spinal cord of rats with ischemia/reperfusion injury. Although bone marrow mesenchymal stem cells are known to protect against spinal cord ischemia/reperfusion injury through anti-apoptotic effects, the precise mechanisms remain unclear. In the present study, bone marrow mesenchymal stem cells were cultured and proliferated, then transplanted into rats with ischemia/reperfusion injury via retro-orbital injection. Immunohistochemistry and immunofluorescence with subsequent quantification revealed that the expression of the axonal regeneration marker, growth associated protein-43, and the neuronal marker, microtubule-associated protein 2, significantly increased in rats with bone marrow mesenchymal stem cell transplantation compared with those in rats with spinal cord ischemia/reperfusion injury. Furthermore, the expression of the autophagy marker, microtubule-associated protein light chain 3B, and Beclin 1, was significantly reduced in rats with the bone marrow mesenchymal stem cell transplantation compared with those in rats with spinal cord ischemia/reperfusion injury. Western blot analysis showed that the expression of growth associated protein-43 and neurofilament-H increased but light chain 3B and Beclin 1 decreased in rats with the bone marrow mesenchymal stem cell transplantation. Our results therefore suggest that bone marrow mesenchymal stem cell transplantation promotes neurite growth and regeneration and prevents autophagy. 20Calcium channel blockers reduce severe fever with thrombocytopenia syndrome virus (SFTSV) related fatality显示文摘Severe fever with thrombocytopenia syndrome (SFTS),an emerging tick-borne infectious disease caused by a novel phlebovirus (SFTS virus, SFTSV), was listed among the top 10 priority infectious diseases by the World Health Organization due to its high fatality of 12%-50% and possibility of pandemic transmission. Currently, effective anti-SFTSV intervention remains unavailable. Here, by screening a library of FDA-approved drugs, we found that benidipine hydrochloride, a calcium channel blocker (CCB), inhibited SFTSV replication in vitro. Benidipine hydrochloride was revealed to inhibit virus infection through impairing virus internalization and genome replication. Further experiments showed that a broad panel of CCBs, including nifedipine, inhibited SFTSV infection. The anti-SFTSV effect of these two CCBs was further an a lyzed in a humanized mouse model in which CCB treatment resulted in reduced viral load and decreased fatality rate. Importantly, by performing a retrospective clinical investigation on a large cohort of 2087 SFTS patients, we revealed that nifedipine administration enhaneed virus clearance, improved clinical recovery, and remarkably reduced the case fatality rate by >5-fold.
