| 3 | A LINEAR THEORY FOR DISTURBANCE OF COHERENT STRUCTURE AND MECHANISM OF SAND WAVES IN OPEN-CHANNEL FLOW显示文摘The formation of sand wave is such a process in which the roughness and discontinuity of the original bed surface cause the disturbance of the bottom laminar flow in an open channel, and the development of the disturbance in turn leads to instability of the flow and the appearance of the coherent structure. The evolution and development of the coherent structure further promote the undulations of bed surface and sand waves rise finally. The sand wave is explained as a result of action that the bed sediment particles are disturbed by the coherent structure. | 8 | EXPERIMENTS ON FLOW AROUND A CYLINDER IN A SCOURED CRANNEL BED显示文摘The flow pattern around a cylinder, installed in a scourd channel bed, was experimentally investigated. Detailed measurements of the instantaneous 3D velocities were performed by using an Acoustic Doppler Velocity Profiler (ADVP), from which the profiles of the time^averaged velocities and turbulence stresses were obtained. It is shown that the influence of thC cylinder and of the scour hole alters the approach flow; this is essentially confined to the viciulty of the cylinder and to the inside of the scour hole. | 10 | Tomato fruit as a model for tissue-specific gene silencing in crop plants显示文摘Use of CRISPR-Cas9(Clustered Regularly Interspaced Short Palindromic Repeats(CRISPR)-CRISPR-associated 9)-mediated genome editing has proliferated for use in numerous plant species to modify gene function and expression,usually in the context of either transient or stably inherited genetic alternations.While extremely useful in many applications,modification of some loci yields outcomes detrimental to further experimental evaluation or viability of the target organism.Expression of Cas9 under a promoter conferring gene knockouts in a tissue-specific subset of genomes has been demonstrated in insect and animal models,and recently in Arabidopsis.We developed an in planta GFP(green fluorescent protein)assay system to demonstrate fruit-specific gene editing in tomato using a phosphoenolpyruvate carboxylase 2 gene promoter.We then targeted a SET-domain containing polycomb protein,SlEZ2,previously shown to yield pleiotropic phenotypes when targeted via ^(35)S-driven RNA interference and we were able to characterize fruit phenotypes absent additional developmental perturbations.Tissue-specific gene editing will have applications in assessing function of essential genes otherwise difficult to study via germline modifications and will provide routes to edited genomes in tissues that could not otherwise be recovered when their germline modification perturbs their normal development. | Ari Feder Sarah Jensen Anquan Wang Lance Courtney Lesley Middleton Joyce Van Eck Yongsheng Liu James J.Giovannoni | 2020 | Horticulture Research2020,7,1: | 2 |
| 11 | Evaluation of mercury methylation in fresh and pre dried sediments using ^(203)Hg radioisotope显示文摘Mercury methylation rates in fresh and pre dried river sediments were determined in sediment water systems spiked with 203 HgCl 2 at a level of 0.75—0.95 ppm as Hg. At the end of the incubation period (21—38 days), methylmercury production in the fresh sediments was in the order of 3.0% to 13.75%, whereas in the pre dried sediments the methylation was much lower, at 0.6% to 3.9%. The highest methylation levels were generally recorded in the uppermost, 1 cm layer of the sediment column. Total mercury concentration in water in equilibrium with the 203 Hg spiked fresh sediments was in the range of 44.7 to 634 ng/L, whereas in equilibrium with the pre dried sediments the concentration was in the range of 19.5 to 34.5 ng/L. The proportion of methylmercury to total mercury in the water varied from 9.4% to 66.6% over the fresh sediments, and from 7.1% to 10.8% over the pre dried sediments. In the system consisting of water, sediment and sediment inverterbrates the concentrations of methylmercury were 8.61 to 15.69 ng/g in the sediment, 0.011 ng/ml in the water, and 9.22 to 40.69 ng/g in the inverterbrates. Methylmercury bioaccumualtion factors (BAFs) were in the order of 274 to 8087 relative to the water, and 0.25 to 7.31 relative to the sediment.
