1The effects of biochar on soil nutrients status,microbial activity and carbon sequestration potential in two calcareous soils显示文摘Biochar as an organic amendment improves soil attributes,with a potentially significant effect on soil chemical fertility and quality.The main objective of this study was to quantify the effect of biochar addition on nutrients,carbon sequestra-tion and microbial activity and understand the mechanisms of controlling biochar effects in calcareous soils.Maize residue biochars produced at 200,400 and 600℃ were added at 5 and 10 g kg^(−1)rates to sandy loam and clayey texture calcareous soils.The soil properties measured were pH and electrical conductivity(EC),plant-available potassium(K)and available phosphorus(P),total nitrogen(TN),C sequestration;and the fluorescein diacetate(FDA)hydrolysis activity.Addition of raw material and biochars increased pH(0.15-0.46 units),EC(0.14-0.38 dS m^(−1)),TN(63-120%),K(12-41%)and FDA activity(27-280%),but tended to decrease plant-available P(23-86%).Increasing pyrolysis temperature increased soil C pool index(CPI),but decreased the FDA and the changes depended largely upon the application rate and soil texture.The positive effects of biochar addition and its pyrolysis temperature on soil C sequestration potential were more pronounced at high than low application rate and in sandy loam than clayey soils.Nevertheless,the effect of biochar addition and pyrolysis temperature on the FDA activity was higher at high than low application rates,but lower in sandy loam than clayey soils.Although biochar application may successfully improve soil processes and attributes and have a high potential for C seques-tration,its effects are controlled by soil texture,pyrolysis temperature and application rate.Allahyar Khadem Fayez Raiesi Hossein Besharati Mohammad Ali Khalaj 2021Biochar2021,3,1:4
2Molecular physiology of vesicular glutamate transporters in the digestive system显示文摘Glutamate is the major excitatory neurotransmitter in the mammalian central nervous system (CNS). Packaging and storage of glutamate into glutamatergic neuronal vesicles require ATP-dependent vesicular glutamate uptake systems, which utilize the electrochemical proton gradient as a driving force. Three vesicular glutamate transporters (VGLUT1-3) have been recently identified from neuronal tissue where they play a key role to maintain the vesicular glutamate level. Recently, it has been demonstrated that glutamate signaling is also functional in peripheral neuronal and non-neuronal tissues, and occurs in sites of pituitary, adrenal, pineal glands, bone, GI tract, pancreas,skin, and testis. The glutamate receptors and VGLUTs in digestivesystem have been found in both neuronal and endocrinal cells. The glutamate signaling in the digestive system may have significant relevance to diabetes and GI tract motility disorders. 3Split-dose menthol-enhanced PEG vs PEG-ascorbic acid for colonoscopy preparation显示文摘AIM:To compare the efficacy and palatability of 4L polyethylene glycol electrolyte(PEG)plus sugar-free menthol candy(PEG+M)vs reduced-volume 2 L ascorbic acid-supplemented PEG(Asc PEG).METHODS:In a randomized controlled trial setting,ambulatory patients scheduled for elective colonoscopy were prospectively enrolled.Patients were randomized to receive either PEG+M or Asc PEG,both splitdosed with minimal dietary restriction.Palatability was assessed on a linear scale of 1 to 5(1=disgusting;5=tasty).Quality of preparation was scored by assignment-blinded endoscopists using the modified Aronchick and Ottawa scales.The main outcomes were the palatability and efficacy of the preparation.Secondary outcomes included patient willingness to retake the same preparation again in the future and completion of the prescribed preparation.RESULTS:Overall,200 patients were enrolled(100patients per arm).PEG+M was more palatable than Asc PEG(76%vs 62%,P=0.03).Completing the preparation was not different between study groups(91%PEG+M vs 86%Asc PEG,P=0.38)but more patients were willing to retake PEG+M(54%vs 40%respectively,P=0.047).There was no significant difference between PEG+M vs Asc PEG in adequate cleansing on both the modified Aronchick(82%vs77%,P=0.31)and the Ottawa scale(85%vs 74%,P=0.054).However,PEG+M was superior in the left colon on the Ottawa subsegmental score(score0-2:94%for PEG+M vs 81%for Asc PEG,P=0.005)and received significantly more excellent ratings than Asc PEG on the modified Aronchick scale(61%vs 43%,P=0.009).Both preparations performed less well in afternoon vs morning examinations(inadequate:29%vs 15.2%,P=0.02).CONCLUSION:4 L PEG plus menthol has better palatability and acceptability than 2 L ascorbic acidPEG and is associated with a higher rate of excellentpreparations;Clinicaltrial.gov identifier:NCT01788709.Ala I Sharara Ali H Harb Fayez S Sarkis Jean M Chalhoub Rami Badreddine Fadi H Mourad Mahmoud Othman Omar Masri 2015World Journal of Gastroenterology2015,21,6:2

7Efficacy and safety of treatment of hepatitis C virus infection in renal transplant recipients显示文摘AIM: To assess the efficacy and safety of combinedpegylated interferon and ribavirin therapy in hepatitis C virus (HCV) infection in renal transplant recipients. METHODS: This is a retrospective chart review of post renal transplant patients who were positive for anti-HCV and HCV-RNA, and who have received treatment with combination of pegylated interferon and ribavirin between October 2003 and December 2008. Only patients with stable graft function and absence of evidence of cirrhosis and who received the therapy for continuous 48 wk were included. Nineteen patients (13 male and 6 female) were identified and included. The patient's complete blood count, liver and kidney prof ile, and cal- culated glomerular f iltration rate (GFR) were monitored every 6-8 wk while on treatment. HCV-RNA was tested at 12 wk for early virological response, at 48 wk for end of treatment response (ETR), and then retested at 24, and 48 wk after completion of therapy for sustained virological response (SVR). Liver biopsies were obtained before treatment from all patients and graft kidney biopsies were performed as required. RESULTS: Of the entire cohort, 9 patients (47.4%) showed an ETR and 8 had SVR (42.1%). Of the 8 patients with abnormal alanine aminotransferase (ALT) levels at baseline, 78.9% had their ALT normalized (including the virological non responders). ALT was normal in all responders at the end of therapy and at 24 wk post therapy (100%). Only one patient (5.3%) developed an increase in creatinine and decline in GFR from baseline towards the end of treatment. This patient's kidney biopsy revealed borderline rejection. There was no impact on response by HCV-genotype, initial HCV RNA load, age or sex of the patient or duration post transplant before commencement of therapy. All patients tolerated treatment in the same way as non-transplant with no unusual or increased occurrence of side effects. 8Short-term effects of maize residue biochar on kinetic and thermodynamic parameters of soil β-glucosidase显示文摘Soil β-glucosidase (BG), the rate-limiting enzyme in the final step of cellulose hydrolysis, plays a key role in microbial metabolism, carbon (C) cycling and sequestration in terrestrial ecosystems. Biochar application is known to affect soil BG activity;however, most of the biochar studies have focused on the potential activity of BG, and it is not clear how biochar influences the kinetic and thermodynamic behavior of BG in the soil. The objective of this study was to investigate the effect of maize residue biochar on soil BG kinetic and thermodynamic parameters. Soil BG kinetic (V_(max) and K_(m)) and ther-modynamic (E_(a), ΔH_(a) and Q_(10)) parameters were determined within soils (clayey and sandy loam soils) amended with either maize residue (as positive control) or its biochar (600℃) at 0.5 and 1.0% ratios (w/w), and the mixtures were incubated for 90 days. BG showed an increase in potential enzymatic activity (81%), enzyme concentration (higher V_(max) value) (25%) and substrate affinity (lower K_(m) value) (32%) in the biochar-amended sandy loam soil only at high addition rates compared with the control, and an increase by about 86% of the catalytic efficiency (V_(max)/K_(m)). In the clayey soil, biochar addition decreased potential BG activity (by 10-29%), increased the V_(max) value (by 20-25%) and had no impact on enzyme-substrate binding affinity, but still increased the catalytic efficiency by 47-72%. Adsorption of soil BG by biochar particles did not affect the catalytic efficiency in the soil. Generally, application of maize residue biochar to the soil decreased the E_(a), ΔH_(a) and Q_(10) values of BG compared with the negative controls at both biochar rates in the light-textured soil and only at low biochar rate in heavy-textured soil. The direction and magnitude of BG responses (activity, kinetics, and thermodynamics) to biochar were more related to the soil characteristics.
