| 1 | Therapeutic targeting of epidermal growth factor receptor in human cancer: successes and limitations显示文摘Epidermal growth fac tor receptor (EGFR) is one of the most commonly altered genes in human cancer by way of over-expression, amplification, and mutation. Targeted inhibition of EGFR activity suppresses signal transduction pathways which control tumor cell growth, proliferation, and resistance to apoptosis. Small molecule tyrosine kinase inhibitors and monoclonal antibodies are among the most common EGFR-targeting agents and have been used clinically for treating various malignancies. This review discusses the successes and challenges of targeting EGFR in human cancer. The genetic alterations of EGFR tend to occur more often in some solid tumors than others, as do the mechanisms of resistance to targeted inhibition. | 2 | 鼻阻塞的主观评价与客观鼻测量的相关性研究显示文摘目的探讨鼻阻塞时检查者和患者的主观评价与客观鼻测量之间是否有相关性。方法检查者和患者均采用视觉模拟量表(visual analog scale,VAS)对鼻阻塞程度进行评价。客观鼻测量采用主动后鼻测压仪和鼻声反射测量仪进行。以配对t检验分析检查者的临床评价和患者VAS均数的差异,以Spearman等级相关分析评价客观鼻测量结果单侧鼻气流阻力与单侧鼻道容积、单侧鼻腔最小横截面积的关系,以及检查者的临床评价、患者VAS与上述客观鼻测量结果的相关性。结果本组病例经统计学分析,316例患者中,减充血前后单侧鼻气流阻力与单侧鼻道容积、单侧鼻腔最小横截面积间呈负相关(r值分别为-0.430、-0.554、-0.373、-0.600,P值均〈0.001)。减充血前后,检查者的临床评价与患者VAS均数呈正相关,差异均有统计学意义(r值分别为0.630、0.526,P值均〈0.001),二者均与鼻气流阻力有一定的正相关关系(减充血前:检查者的临床评价与鼻气流阻力r=0.530,P=0.000,患者VAS与鼻气流阻力r=0.351,P=0.000;减充血后:检查者的临床评价与鼻气流阻力r=0.452,P=0.000,患者VAS与鼻气流阻力r=0.216,P=0.000),与鼻道容积和鼻腔最小横截面积均有一定的负相关关系(减充血前:检查者的临床评价与鼻道容积r=-0.411,P=0.000,患者VAS与鼻道容积r=-0.325,P=0.000,检查者的临床评价与鼻腔最小横截面积r=-0.507,P=0.000,患者VAS与鼻腔最小横截面积r=-0.384,P=0.000;减充血后:检查者的临床评价与鼻道容积r=-0.391,P=0.000,患者VAS与鼻道容积r=-0.209,P=0.000,检查者的临床评价与鼻腔最小横截面积r=-0.471,P=0.000,患者VAS与鼻腔最小横截面积r=-0.286,P=0.000)。检查者的临床评价与客观鼻测量参数的相关系数大于患者VAS与客观鼻测量参数的相关系数。结论鼻阻力测压与鼻声反射测量的结果有一定的相关性。检查者的临床评价与患者VAS存在一定的正相关关系。两者均与客观鼻测量参数存在一定的直线相关关系。 | 张革化 Ronald S. | 3 | 鼻阻力测压与鼻声反射测量的相关性研究显示文摘目的探讨主动后鼻阻力测压仪的参数——鼻阻力(nasal resistance,NR)与鼻声反射测量仪的参数——鼻腔容积(nasal cavity volume,NV)、鼻腔最小横截面积(minimal cross sectional area,MCSA)间是否具有相关性。方法应用主动后鼻阻力测压法和鼻声反射测量法对316例有鼻阻塞的患者进行前瞻性研究。根据减充血前双侧鼻阻力(bilateral NR,BNR)将患者分为2组,组1:BNR减充血前≤0.25 Pa/cm^3·s,组2:BNR减充血前>0.25 Pa/cm^3·s。Spearman等级相关分析研究上述两种客观测量法的参数在全体研究对象及各组中的相关性。结果在使用减充血剂前、后,316例患者单侧鼻阻力(unilateral NR,UNR)与单侧鼻腔容积(unilateral NV,UNV)、单侧鼻腔最小横截面积(unilateral MCSA,UMCSA)间均存在显著性负相关(P值均=0.000);组1(104例)和组2(212例)在减充血前、后的UNR与UNV、UMCSA间均存在显著性负相关(P值均=0.000)。鼻阻力与鼻腔最小横截面积的相关系数均大于鼻阻力与鼻气道容积的相关系数。结论在有鼻阻塞的患者中,NR、NV及MCSA存在负相关,MCSA是影响NR的主要因素。 | 张革化 李源 Ronald S Fenton Philip Cole Richard Rival Philip Solomon | 2008 | 中国耳鼻咽喉头颈外科2008，15，10： | 8 |

| 12 | Increased presence of effector lymphocytes during Helicobacter hepaticus-induced colitis显示文摘AIM:To identify and characterize drosophila mothers against decapentaplegic(SMAD)3-dependent changes in immune cell populations following infection with Helicobacter hepaticus(H.hepaticus).METHODS:SMAD3 -/-(n=19)and colitis-resistant SMAD3+/-(n=24)mice(8-10 wk of age)were in-fected with H.hepaticus and changes in immune cell populations[T lymphocytes,natural killer(NK)cells,T regulatory cells]were measured in the spleen and mesenteric lymph nodes(MsLNs)at 0 d,3 d,7 d and 28 d post-infection using flow cytometry.Genotypedependent changes in T lymphocytes and granzyme B+ cells were also assessed after 28 d in proximal colon tissue using immunohistochemistry.RESULTS:As previously observed,SMAD3 -/-,but not SMAD3+/-mice,developed colitis,peaking at 4 wk post-infection.No significant changes in T cell subsets were observed in the spleen or in the MsLNs between genotypes at any time point.However,CD4+and CD8+/ CD62Llo cells,an effector T lymphocyte population,as well as NK cells(NKp46/DX5+)were significantly higher in the MsLNs of SMAD3-/-mice at 7 d and 28 d post-infection.In the colon,a higher number of CD3+cells were present in SMAD3-/-compared to SMAD3+/-mice at baseline,which did not significantly change during infection.However,the number of granzyme B+cells,a marker of cytolytic lymphocytes,significantly increased in SMAD3-/-mice 28 d post-infection compared to both SMAD3+/-mice and to baseline values.This was consistent with more severe colitis development in these animals.CONCLUSION:Data suggest that defects in SMAD3 signaling increase susceptibility to H.hepaticus-induced colitis through aberrant activation and/or dysregulation of effector lymphocytes. | Sarah J McCaskey Elizabeth A Rondini Jonathan F Clinthorne Ingeborg M Langohr Elizabeth M Gardner Jenifer I Fenton | 2012 | World Journal of Gastroenterology2012，18，13： | 2 |
